We previously observed that the transcription of some flagellar genes decreased in Salmonella Typhimurium tdcA mutant, which is a gene encoding the transcriptional activator of the tdc operon. Since flagella-mediated bacterial motility accelerates the invasion of Salmonella, we have examined the effect of tdcA mutation on the invasive ability as well as the flagellar biosynthesis in S. Typhimurium. A tdcA mutation caused defects in motility and formation of flagellin protein, FliC in S. Typhimurium. Invasion assays in the presence of a centrifugal force confirmed that the defect of flagellum synthesis decreases the ability of Salmonella to invade into cultured epithelial cells. In addition, we also found that the expression of Salmonella pathogenicity island 1 (SPI1) genes required for Salmonella invasion was down-regulated in the tdcA mutant because of the decreased expression of fliZ, a positive regulator of SPI1 transcriptional activator, hilA. Finally, the virulence of a S. Typhimurium tdcA mutant was attenuated compared to a wild type when administered orally. This study implies the role of tdcA in the invasion process of S. Typhimurium.
This study evaluated the effects of NaCl on heat resistance and Caco-2 cell invasion of Listeria monocytogenes in broth media and sausage. A 10-strain mixture of L. monocytogenes was inoculated in tryptic soy broth containing 0.6% yeast extract (TSBYE), and sausage formulated with 0, 2, 4, and 6% NaCl. The medium was stored at 7, 15, 20, and $25^{\circ}C$ for 3-16 d, and medium samples were withdrawn at the appropriate time and challenged to 55, 60, and $63^{\circ}C$ to evaluate the thermal resistance of the pathogen. Sausage samples were stored at 7 and $25^{\circ}C$, and they were exposed to $63^{\circ}C$ to evaluate thermal resistance. NaCl-habituated L. monocytogenes strains NCCP10811 and NCCP10943 were examined for 12 antibiotics and Caco-2 cell invasion assay (only L. monocytogenes NCCP10943). Bacterial populations of L. monocytogenes generally increased (p<0.05) during the heat challenge as NaCl concentrations increased in both TSBYE and sausage samples. The antibiotic resistance of L. monocytogenes was not observed ($p{\geq}0.05$) when it was exposed to a single concentration of NaCl in TSBYE, but the pathogen obtained resistance to some antibiotics when exposed to a sequential increase of NaCl concentration. Invasion efficiency of L. monocytogenes NCCP10943 was not increased ($p{\geq}0.05$) with NaCl concentration increase. These results indicate that NaCl may increase the resistance of L. monocytogenes to heat and to some antibiotics, but may not increase Caco-2 cell invasion of L. monocytogenes.
An important recent advance in the field of plant-microbe interactions has been the cloning of genes that confer resistance to specific viruses, bacteria, fungi or insects. Disease resistance (R) genes encode proteins with predicted structural motifs consistent with them having roles in signal recognition and transduction. Plant disease resistance is the result of an innate host defense mechanism, which relies on the ability of plant to recognize pathogen invasion and efficiently mount defense responses. In tomato, resistance to the pathogen Pseudomonas syringae pv. tomato is mediated by the specific recognition between the tomato serine/threonine kinase Pto and bacterial protein AvrPto or AvrPtoB. This recognition event initiates signaling events that lead to defense responses including an oxidative burst, the hypersensitive response (HR), and expression of pathogenesis- related genes.
When the tooth avulsion occur in accidents the drying damage to the periodontal ligament has extremely detrimental effects on healing. Pulp necrosis always occurs after an avulsion injury, but revascularization can only take place in teeth with immature apexes. Therefore complications after avulsion injuries are common, and treatment must be carried out in a timely and correct fashion to prevent or limit these complications. Every effort should be made to replant the tooth within the first 15 to 20 minutes. If doubt exists that the tooth can be replanted adequately, the tooth should quickly be stored in an appropriate medium until the patient can get to the dental office for replantation. A complication of inflammatory root resorption is occurred by bacterial infection of periodontal ligament and dental pulp. Therefore aseptic endodontic treatment must be carried out in a timely and systemic antibiotics given at the time of replantation and before endodontic treatment are effective in preventing bacterial invasion. Further studies are needed to establish the clinical importance of preparation of the socket and root.

Methicillin-resistant Staphylococcus aureus (MRSA) has been emerging worldwide as one of the most important hospital and community pathogens. Therefore, new agents are needed to treat the MRSA. In the present study, we investigated antimicrobial activity of ethyl acetate, methanol, and water extracts of Curcuma longa L. (C. longa) aganist clinical isolates of MRSA. The ethyl acetate extract of C. long a demonstrated a higher antibacterial activity than the methanol extract or water extract. Since the ethyl acetate extract was more active than other extracts, we examined whether ethyl acetate extract may restore the antibacterial activity of β-lactams and alter the adhesion and invasion of MRSA to human mucosal fibroblasts (HMFs). In the checkerboard test, ethyl acetate extract of C. longa markedly lowered the MICs of ampicillin and oxacillin against MRSA. In the bacterial adhesion and invasion assay, MRSA intracellular invasion were notably decreased in the presence of 0.125 - 2 mg/ml of C. longa extract compared to the control group. These results suggest that ethyl acetate extract of C. longa may have antibacterial activity and the potential to restore the effectiveness of β-lactams against MRSA, and inhibit the MRSA adhesion and invasion to HMFs.
Background: Liver-expressed antimicrobial peptide-2 (LEAP-2) is an important component of innate immune system in teleosts. In order to understand isoform-specific involvement and regulation of LEAP-2 genes in mud loach (Misgurnus mizolepis, Cypriniformes), a commercially important food fish, this study was aimed to characterize gene structure and expression characteristics of two paralog LEAP-2 isoforms. Results: Mud loach LEAP-2 isoforms (LEAP-2A and LEAP-2B) showed conserved features in the core structure of mature peptides characterized by four Cys residues to form two disulfide bonds. The two paralog isoforms represented a tripartite genomic organization, known as a common structure of vertebrate LEAP-2 genes. Bioinformatic analysis predicted various transcription factor binding motifs in the 5'-flanking regions of mud loach LEAP-2 genes with regard to development and immune response. Mud loach LEAP-2A and LEAP-2B isoforms exhibited different tissue expression patterns and were developmentally regulated. Both isoforms are rapidly modulated toward upregulation during bacterial challenge in an isoform and/or tissue-dependent fashion. Conclusion: Both LEAP-2 isoforms play protective roles not only in embryonic and larval development but also in early immune response to bacterial invasion in mud loach. The regulation pattern of the two isoform genes under basal and stimulated conditions would be isoform-specific, suggestive of a certain degree of functional divergence between isoforms in innate immune system in this species.
Cellulitis in broiler chickens is one of the economically important problems that facing the broiler industry due to the presence of the lesion leads to condemnation of part of /or the entire carcasses. Broiler with cellulitis lesions showed lower body weight. Cellulitis was recorded on different body regions including the head, dorsum, thighs, breast, legs, and abdomen. Cellulitis results from the invasion of subcutaneous (s.c.) tissues by bacteria through disruption of skin integrity. Lesions revealed the existence of the characteristic s.c colored exudate varies from yellowish to green, which were either serosanguineous, fibrinous s.c exudate yellowish, greenish or suppurative. Many bacterial isolates including E. coli, Staphylococci, Clostridia, Aeromonas spp., Enterobacter spp., Proteus mirabilis, P. aeruginosa, and Streptococci were isolated from the lesion. Chickens exposed to immunosuppression proved to have a greater probability of developing cellulitis. The condition was experimentally induced by s.c inoculation of 25-day-old broiler chickens with E. coli, S. aureus and clostridia. Usually, bacterial isolates were multidrug-resistant. The usage of Bifidobacterium bifidum or antibiotic with avoiding immunosuppression can reduce lesion and condemnation rate resulted from cellulitis. The objective of this review is to collect different literature written about cellulitis to be available to students, researchers, and veterinarians in poultry practical.
We found that the main bacterial metabolite M1 is an active component of orally administered protopanxadiol-type ginsenosides, and that the anti-metastatic effect by oral administration of ginsenosides may be primarily mediated through the inhibition of tumor invasion, migration and growth of tumor cells by their metabolite M1. Pharmacokinetic study after oral administration of ginsenoside Rb1 revealed that M1 was detected in serum for 24 h by HPLC analysis but Rb1 was not detected. M1, with anti-metastatic property, inhibited the proliferation of murine and human tumor cells in a time- and concentration-dependent manner in vitro, and also induced apoptotic cell death (the ladder fragmentation of the extracted DNA). The induction of apoptosis by M1 involved the up-regulation of the cyclin-dependent kinase(CDK) inhibitor $p27^{Kip1}$ as well as the down-regulation of a proto-oncogene product c-Myc and cyclin D1 in a time-dependent manner. Thus, M1 might cause the cell-cycle arrest (G1 phase arrest) in honor cells through the up/down-regulation of these cell-growth related molecules, and consequently induce apoptosis. The nucleosomal distribution of fluorescence-labeled M1 suggests that the modification of these molecules is induced by transcriptional regulation. Tumor-induced angiogenesis (neovascularization) is one of the most important events concerning tumor growth and metastasis. Neovascularization toward and into tumor is a crucial step for the delivery of nutrition and oxygen to tumors, and also functions as the metastatic pathway to distant organs. M1 inhibited the tube-like formation of hepatic sinusoidal endothelial (HSE) cells induced by the conditioned medium of colon 26-L5 cells in a concentration-dependent manner. However, M1 at the concentrations used in this study did not affect the growth of HSE cells in vitro.

In this study, we investigated the effects of linoleic acid (LA) treatment on Brucella abortus infection in professional phagocyte RAW264.7 cells, particularly during the pathogen's invasion and intracellular growth in these cells, as well as in murine model BALB/c mice focusing on bacterial splenic proliferation and immunoregulatory activities. LA inhibited the growth of Brucella in a dose- and time-dependent manner. The ability of the pathogen to enter the phagocytes was inhibited as was its survival within these cells. This was accompanied by increased nitrite accumulation in these cells at 24 h post-infection. The concentration of LA used in the present study did not affect the total body weight or liver function of the mice. During Brucella infection, the total splenic weight of these animals was not changed; rather, resistance to bacterial proliferation was enhanced in the spleen. Furthermore, mice treated with LA displayed elevated levels of IL-12 and IFN-γ but reduced levels of IL-10 during infection. The findings in this study showed the regulatory role of LA against B. abortus infection suggesting its potential use in designing intervention strategy for brucellosis.
Despite evidence that bacteria-sensing Toll-like receptors (TLRs) are activated in salivary gland tissues of Sjogren syndrome (SS) patients, the role of oral bacteria in SS etiopathogenesis is unclear. We previously reported that two SS-associated oral bacteria, Prevotella melaninogenica (Pm) and Rothia mucilagenosa (Rm), oppositely regulate the expression of major histocompatibility complex class I (MHC I) in human salivary gland (HSG) cells. Here, we elucidated the mechanisms underlying the differential regulation of MHC I expression by these bacteria. The ability of Pm and Rm to activate TLR2, TLR4, and TLR9 was examined using TLR reporter cells. HSG cells were stimulated by the TLR ligands, Pm, and Rm. The levels of MHC I expression, bacterial invasion, and viability of HSG cells were examined by flow cytometry. The hypoxic status of HSG cells was examined using Hypoxia Green. HSG cells upregulated MHC I expression in response to TLR2, TLR4, and TLR9 activation. Both Pm and Rm activated TLR2 and TLR9 but not TLR4. Rm-induced downregulation of MHC I strongly correlated with bacterial invasion and cell death. Rm-induced cell death was not rescued by inhibitors of the diverse cell death pathways but was associated with hypoxia. In conclusion, Pm upregulated MHC I likely through TLR2 and TLR9 activation, while Rm-induced hypoxia-associated cell death and the downregulation of MHC I, despite its ability to activate TLR2 and TLR9. These findings may provide new insight into how oral dysbiosis can contribute to salivary gland tissue damage in SS.
