Biological control of Flavobacterium psychrophilum infection in ayu (Plecoglossus altivelis altivelis) using a bacteriophage PFpW-3

  • The efficacy of using a bacteriophage (phage) to control Flavobacterium psychrophilum (F. psychrophilum) infection of ayu (Plecoglossus altivelis altivelis) was evaluated in this study. Intramuscular challenge failed to induce sufficient infection levels; therefore, a newly designed net-scratch challenge method was also used to induce bacterial infection. Administration of phage PFpW-3 in F. psychrophilum-infected ayu showed notable protective effects, increased survival rates and mean times to death. Additionally, the fate of inoculated bacteria and phage in ayu were investigated. Our results suggest that the phage PFpW-3 could be considered an alternative biocontrol agent against F. psychrophilum infections in ayu culture.

  • This study was conducted to determine the protein requirement of ayu (Plecoglossus altivelis). Two replicate groups of fish initially averaging 6.6 g were fed the five isocaloric diets containing different protein level from 29% to 57% in a flow-through freshwater system for 25 days. White fish meal was used as a sole protein source. Weight gain and feed efficiency of fish increased significantly with dietary protein level up to 43% (P<0.05) with no additional response above this level. Protein and lipid retention, moisture, protein and lipid contents of body were not affected by dietary protein levels (P>0.05). Daily protein intake increased significantly with dietary protein level, whereas protein efficiency ratio of fish fed the 57% dietary protein decreased (P<0.05). The data obtained in this study indicate that a 43% dietary protein level could be recommended for the optimum growth of ayu.

  • Directional (sinistral) asymmetry (DA) occurs when the traits of one side of a supposedly bilaterally symmetrical organism differ in a random way from those of the other side. We examined asymmetries in gonadal growth traits within both sexes of hatchery reared ayu, Plecoglossus altivelis. The pattern of the gonadal growth from hatching to 320 days after hatching was a type of DA except for the growth in gonad weight from 140 to 180 days after hatching, although the ovary of the right side tended to exhibit more pronounced DA phenomenon.

  • We investigated the genetic structure of Korean and Japanese ayu Plecoglossus altivelis populations by examining 669 individuals from 14 populations using three microsatellite loci. Genetic variation did not differ significantly among the populations examined in terms of allelic number and heterozygosity. Korean populations were genetically close to each other, implying that persistent gene flow has occurred in these populations. This suggests that eastern populations in Korea form a single large population and all of the Korean populations are distinct from the Japanese populations. Pairwise population $F_{ST}$ estimates, principal component analyses, and a neighbor-joining tree showed that genetic separation between the southern and pooled eastern coast populations was probably influenced by restricted gene flow. Hierarchical analysis of molecular variance (AMOVA) revealed a weak but significant genetic structure among three ayu groups (eastern and southern coasts of Korea and the Japan coast), and no genetic variation within groups. The estimated genetic population structure and potential applications of microsatellite markers may aid in the proper management of ayu populations.

  • 2008년 5월 제주지역 하천에 소하하는 은어 중 일부개체가 주둥이 부식 또는 체표궤양증상을 보이면서 폐사하였다. 어류 질병원인균 중 부식증 및 체표궤양과 연관성이 높은 Flavobac-terium 속 세균의 분리를 위해 Shu-Shott medium 및 R2A medium에 주둥이 부식 및 체표궤양 부위를 도말하여 배양 하였고, Flavobacterium 속 세균의 전형적인 집락인 pale yellow colony를 분리하였다. 16S rDNA 염기서열 분석결과 Flavobacterium succinicans DSM 4002(Accession NO. AM230492)와 98.27%의 상동성을 보여 F. succinicans JMFL55로 동정하였다.

Studies of Opsin Genes in a Smelt Fish, Ayu (Plecoglossus altivelis)

  • To investigate the visual and extra-ocular photoreception, we cloned the opsin genes in ayu (Plecoglossus allivelis). Amplified fragments encoding exon-4 (-5) of opsin cDNAs were cloned from the retina and brains of ayu, and sequenced. One clone was identified as rod (AYU-Rh), two as green cone (AYU-GI, -G2), one as red cone (A YU-R), two as ultraviolet cone (AYU-UVl, UV2), one as VA (AYU-VA), and one as extra-ocular rod (AYU-ExoRh) opsins. 335 amino acids sequence deduced from the full-length cDNA of AYU-Rh showed high identity with that of other fish. Southern blotting analysis indicated that ayu possess two 'rhodopsin' genes, one is visual rhodopsin and the other is non-visual extra-ocular rhodopsin. In situ hybridization showed that the mRNA of AYU-Rh was localized only in rod cells in the retina. On the other hands, AYU-ExoRh was expressed only in the pineal. We cloned two isoforms (AYU-VAM and -VAL) of VA opsin from ayu. The deduced amino acid sequences of these variants were identical to each other within the first 342 residues, but they showed divergence in the C-terminal sequence. AYU- VAL corresponded to the long isoform found in other fish, and AYU-VAM was identified as a new type of VA opsin variant. Pal-VAM is a new probably functional non-visual photoreceptive molecule in fish.

Effect of Dietary Sargassum Meal on Growth and Body Composition of Ayu (Plecoglossus altivelis) Reared in Seawater

  • This study was conducted to investigate the utilization of Sargassum meal in the diet on juvenile ayu (Plecoglossus altivelis) reared in seawater. White fish meal and wheat flour were used as the dietary protein and carbohydrate sources in the control diet. Wheat flour in the control diet was replaced with $5\%$ and $10\%$ Sargassum meal. Three replicate groups of fish average weighing 4.0 g were fed one of three isonitrogenous$(45\%)$ and isocaloric (14.5 MJ /kg diet) diets for 7 weeks. Survival of all groups were above $80\%$. Weight gain, feed efficiency and protein efficiency ratio were not significantly affected by dietary Sargassum meal levels (P>0.05). There were no significant differences (P>0.05) in moisture, crude protein, crude lipid, crude ash and fatty acid compositions of whole-body fish among groups. It is concluded that Sargassum meal could be used as a dietary additive or alternative low-cost dietary ingredient up to $10\%$ for juvenile ayu reared in seawater.

Genetic Variations of Natural and Hatchery Populations of Korean Ayu (Plecoglossus altivelis) by Isozyme Markers

  • Genetic variability and population structure of 11 natural ayu, Plecoglossus altivelis populations and one hatchery stock were assessed by starch gel electrophoretic analysis with 10 enzyme coding loci. Three loci were polymorphic (lower than 0.95 in major allele frequency) in natural populations,2 loci in hatchery stock. The average number of alleles per locus was 1.38. Observed heterozygosities ranged from 0.0235 to 0.088 (0.055 on the average) in natural population while 0.0925 in hatchery stock. The genetic distance among natural populations measured 0.000047-0.005407 and no significant differentiation was observed among them. On the other hand, a signifcant genetic distance was found between natural populations and the hatchery stock with measuring 0.002032-0.O08605. The results in this study suggest that the hatchery stock has diverged from natural populations, and also that careful to maintain sustainable and effective population size (parents number) should be made.

Notochord opacity in fry ayu, Plecoglossus altivelis

  • An ayu (Plecoglossus altivelis) farm in Korea recently experienced an epidemic of vertebral column deformities where about 90% of fry displayed a vertebral column that was grossly opaque along either the cranial part of the column or its entire length. Abnormal fish were lordotic, scoliotic and/or kyphotic about midway down the spine. Examination of serial sections of whole fish showed only histological lesions in the vertebral column and suggested some disturbance in the early development of the vertebral centrum. Such abnormalities included a frayed spinal or notochord sheaths with irregular thickening and compression, mal-absorbed notochord cells, thickening of around cell layer and hypercellularity on both facets of the notochord sheath. No parasites, fungi, or bacteria were detected. While this lesion has only been reported once in the past, this is the first report of histopathological findings.
