Avidin-biotin 복합체를 이용한 오제스키병과 돼지콜레라의 면역조직화학적 감별진단 (Avidin-biotin complex for immunohistochemical diagnosis of Aujeszky's disease and hog cholera)

  • 오제스키병바이러스 또는 돼지콜레라바이러스에 인공 또는 자연감염된 돼지 10두를 실험동물로 공시하였으며, 감염돈의 편도선, 비장, 대뇌 및 연층(buffy coat)의 냉동 및 파라핀절편에서, avidin-biotin-peroxidase complex(ABC)를 이용하여 이들 바이러스를 면역조직화학적으로 검출하였다. 오제스키병바이러스 항원은 임파구와 대식세포의 핵내 또는 세포질에서 검출되었으며, 돼지콜레라바이러스 항원은 이들 세포의 세포질에서 검출되었고, ABC법은 양성반응 부위에서 갈색의 색소 침착을 일으켰다. 오제스키병바이러스 양성세포는 편도선과 대뇌에서 가장 빈번히 검출되었음에 반해 돼지콜레라바이러스는 비장에서 가장 빈번하였다. 그리고 연충에서도 이들 두가지 바이러스항원이 모두 검출되었다. ABC법은 이을 바이러스의 면역조직화학적 검출에 있어 특이성이 높고 배경의 비특이염색성이 낮아, 바이러스 분리동정을 거치지않고 이들 질병을 확진할 수 있는 진단수단으로 활용될 수 있을 것으로 생각된다.

  • 소 theileriosis의 태반감염 사실을 입증하기 위하여 Theileria sergenti에 자연감염된 암소의 비장, 태반 그리고 유산된 태자의 비장과 태반조직으로부터 T sergenti를 면역화학적으로 검색 입증하고자 T sergenti 표피의 34KD 항원의 단크론 항체를 활용하여 avidin biotin complex 방법으로 면역화학적으로 염색하였던 바, 이들 formalin 고정 조직표본에서 T sergenti의 특이 항원성 물질을 관찰함으로써 태반감염을 증명할 수 있었다.

  • For a immunohistochemical diagnosis of the frozen and paraffin-embedded tissues against rabies virus, mice were intracerebrally inoculated with challege virus standard(CVS) rabies virus and then were used to detect the rabies viral antigen by the immunoperoxidase(IP) and the avidin-biotin complex(ABC) method. In this study, the results confirmed that ABC and IP methods, although the former showed more specific and sensitive than the latter, were reliable and effective for the demonstration of rabies virus in both frozen and paraffin-embedded brain tissues prepared from rabies-infected mice. Additionally, IP technique using the monoclonal antibody against rabies virus could be recommended as a standard diagnostic tool instead of the present immunofluorescent method for the local veterinary services in Korea.

  • The classical ABC (avidin-biotin peroxidase complex) method for immunohistochemistry in the paraffin-embedded tissues bring into being disadvantage such as low sensitivity of antigen detection and highly background. The biotinyl-tyramide conjugation recently introduced for sensitive immunohistochemistry was applied to light microscopy in paraffin-embedded pancreatic and liver tissues. The protocol consists of an indirect method in which 4-5㎛ tissue sections are reacted successively within a specific primary antibody, followed by a biotinylated secondary antibody, streptavidin-horseradich peroxidase (HRP), and then finally with biotinyl-tyramide. The labeling obtained for insulin and collagen antigen tested in pancreatic and liver tissues, respectively, was found to be highly specific with the labeling for each antigen confined to its particular cellular compartment. In this study, fish (flounder) serum was specially applied to remove nonspecific binding. Background levels and nospecific deposition of the staining were negligible. This results suggest that super-signal induction method by biotinyl-tyramide conjugate can readily applied to antigen detection of the paraffin-embedded tissues.

  • Glial fibrillary acidic proteins (GFAP) are a group of intermediate filaments that are distributed in the cytoplasm of glial cells. GFAP immunoreactivity (GFAP-IR) increase after central and peripheral nerve injuries. The purpose of this study was to determine change of GFAP-IR in rat trigeminal ganglion satellite cells following the axotomy of inferior alveolar nerve(IAN). The immunohistochemistry was carried out using the avidin-biotin-peroxidase complex(ABC) method. 1. Control group : Astrocytes in central root of trigeminal ganglion had strong GFAP-IR, but satellite cells of trigeminal ganglion occasionally had GFAP-IR. The patterns of reactivity in satellite cells of trigeminal ganglion were not concenturated in any specific region of trigeminal ganglion. 2. Three day group after IAN axotomy : There were highly GFAP-IR in satellite cells of trigeminal ganglion in mandibular region. GFAP-IR in maxillary and ophthalmic regions were less intense compared to mandibular region. 3. Seven day group after IAN axotomy : GFAP-IR that were increased compared to control group were seen in the mandibular region. But GFAP-IR were less intense compared to three day group. These results suggest that GFAP-IR increase in specific region of trigeminal ganglion following peripheral axotomy. therefore we suppose that GFAP study offer research tool in trigeminal neuralgia.

폐암 환자의 혈청과 조직 표본상에서 Alpha 1-Proteinase Inhibitor의 조사 연구 (Investigation of Alpha 1-Proteinase Inhibitor in Serum and Specimen of Lung Cancer Patients)

  • Alpha 1-Proteinase inhibitor[PI] was known as a major protective enzyme against to excessive hydrolytic and proteolytic reaction. So, it was suggested that Alpha 1-PI may implicated in growth of bronchogenic cancer. This study was undertaken to investigate the role of Alpha 1-PI in local invasion of bronchogenic cancer. Three groups of patients were studied; Preliminary research group of 15 bronchogenic cancer patients, Main research group of 13 bronchogenic cancer patients and Normal control group of 10 nephrectomy donor. Serum Alpha 1-PI level was observed in each group of patients during pre-and postoperative days. Pre-operative serum Alpha 1-PI level in preliminary research group [329.2$\pm$14.21mg/dl]and main research group[406.2$\pm$39.30mg/dl] were higher than in normal control group[236.2$\pm$19.55mg/dl] significantly[p<0.005]. Serial Alpha 1-PI level in each group during pre-and postoperative days shows peaked at 3rd. postoperative day in preliminary and main research group, thereafter decreased gradually. Immunohistochemical study for Alpha 1-antitrypsin[A1AT] was carried out by ABC[avidin-biotin peroxidase complex] method using Alpha-1 antitrypsin DAKOR to tumor tissues of 13 lung cancer patients in main research group. 6 cases[46.2%, squamous cell ca.;5, adenocarcinoma;1] of above 13 cases show positive immunoreactivity for A1AT. In conclusion, alpha 1-PI and elastase are disclosed that have defined actions for lung cancer growing or spreading.

랫드에서 스트레스에 의한 위궤양이 위발암화과정에 미치는 영향에 관한 연구 (Studies on the Effect of Stress-Induced Gastric Ulcer on Gastric Carcinogenesis in Rats treated with N-methyl-N'-nitro-N-nitrosoguanidine)

  • 랫드에서 속박 및 침수 스트레스에 의한 위궤양이 MNNG(N-methyl-N'-nitro-N-nitrosoguanidine)의 위발암과정에 어떠한 영향이 있는지 알아보기 위하여 본 실험을 실시 하였다. 1 군은 시험시작 8시간 전에 스트레스를 가한 후 MNNG를 투여하였으며, 2군은 MNNG를 음수 투여한 후 MNNG에 의한 위발암 기간 중 촉진기간이라고 알려진 3~12주 사이에 매주 발암 물질과 함께 스트레스를 1회 가하였다. 3군은 양성 대조군으로서 MNNG만을 투여하였고, 4군은 음성 대조군으로 발암제를 처치하지 않았다. 전군 공히 20주에 부검하여 acidine-biotin peroxidase complex(ABC) 방법으로 면역조직 화학적 염색을 한 수 PAPG({Pepsinogen isozyme 1 Altered Pyloric Gland)에 대한 통계 분석을 하였으며, 각 군간의 체중 변화와 병리 변화를 비교하여 다음과 같은 결과를 얻었다. 1. 발암 물질을 투여한 1·2·3군이 비투여군(4군)에 비해 증체율이 유의성있게 감소하였다. (p<0.05), 그러나 발암 물질 투여군간의 체중변화에는 유의성이 없었다. 2. MNNG 투여와 함께 스트레스를 가한 군이 스트레스를 가하지 않고 MNNG만을 투여한 군에 비하여 Pg 1 변이 유문선(PAPG)의 수가 유의성 있게 증가되었다. (p<0.01). 3. 위발암 과정 중 촉진기간에 스트레스를 가한 군이 스트레스를 가하지 않고 MNNG만을 투여한 군에 비해 점막 과형성(mucosal hyperplasia) 발생이 유의성있게 증가되었다. (P<0.05)

말초신경초 종양의 특징을 지닌 개 신경종의 조직병리학적 및 면역조직화학적 진단 (Canine nervous-tissue tumors with features of peripheral nerve sheath tumor: histopathological and immunohistochemical findings)

  • Canine peripheral nerve sheath tumors (PNSTs) are spindle cell tumors that arise from Schwann cells, perineural cells, fibroblasts or all of them. Based on the morphology and biologic behavior, PNSTs are divided into benign PNST (BPNST) and malignant PNST (MPNST) forms. The aim of this study is to diagnose the two cases of neoplastic tissue samples with features of PNSTs by the histopathology and immunohistochemistry. The study was performed using two specimens from small animal clinic. The first case, A was a mass, 3~4 cm in diameter, extruded from vaginal mucosa of 10-year-old spayed female mixed-breed dog. And the second case, B was a subcutaneous mass, 1.5 cm in diameter, which is originated from right hind leg of 9-year-old castrated male mixed-breed dog. Two cases were stained with hematoxylin and eosin (H&E) for histopathological examination. And also immunohistochemistry (IHC) was performed by the avidin-biotin peroxidase complex (ABC) method with antibodies specific for the following proteins: S-100 protein, smooth muscle actin (SMA) and epidermal growth factor receptor (EGFR). In results, Antoni B schwannoma pattern characterized by pleomorphic, round and fusiform polygonal cells was seen in A. In B, Antoni A pattern, densely packed spindle cells arranged in interlacing bundles was seen in addition to Antoni B pattern. In IHC, cytoplasms of neoplastic cells were diffusely labeled for S-100 expression in A and B. For SMA, both A and B show negative expression. And for EGFR, A shows negative expression but B shows partially positive expression in areas of Antoni B schwannoma pattern. The histopathologic features of two cases coupled with the S-100 immunoreactivity led to a diagnosis of PNST. For SMA, both A and B show negative expression. The diagnosis of A will be a BPNST with the negative result and B will be a MPNST with the positive result for EGFR.

Aujeszky's disease virus 국내분리주 접종자돈의 병리발생에 관한 연구 II. 면역조직화학 및 in situ hybridization 기법을 이용한 항원과 핵산 검출 (Studies on the pathogenesis of Korean isolate of Aujeszky's disease virus in experimentally infected piglets II. Immunohistochemistry and detection of viral nucleic acids by in situ hybridization)

  • This study was conducted to elucidate the distribution of Aujeszky's disease viral nucleic acids and antigens in the central nervous system (CNS) of piglets. The first Korean isolate of Aujeszky's disease virus(ADV) that isolated from naturally infected piglets in Yang San, was inoculated into 32 day old piglets with $10^{5.9}TCID_{50}/ml$ through intranasal or intramuscular route. These piglets were sacrificed at every 24hrs for 8 days. The immunohistochemistry (IHC) was conducted to detect the viral antigens in paraffin-embedded tissue sections using avidin-biotin-peroxidase complex (ABC) method. The viral nucleic acids were detected by in situ hybridization (ISH) using ADV specific DNA probe labeled with digoxigenin. The ADV antigens were detected in reticuloendothelial cells of spleen, lymph nodes and tonsil, alveolar walls, leptomeningeal vascular walls, inflammatory foci of each organ, and nerve cells. The viral nucleic acids were detected in the spinal trigeminal nucleus and its tracts of the pons and medulla oblongata by the ISH technique. The pathways of AD viruses in CNS were determined by IHC and ISH. In the intranasally inoculated group, the viruses in nasal mucosa moved to medulla oblongata and pons through the trigeminal nerve. In case of intramuscullarly inoculated group, viruses moved to brain via lymphoid organs or spinal nerves from sciatic nerves.

비소세포 폐암에서 HIF-$1{\alpha}$의 발현: 예후 및 종양표지자와의 관련성 (Expression of Hypoxia-inducible Factor-$1{\alpha}$ in Non-small Cell Lung Cancer: Relationship to Prognosis and Tumor Biomarkers)

  • 배경: 악성종양에서 신생혈관 생성 및 당분해의 증가는 저산소 상태의 미세환경을 나타내며, 이는 종양의 침습성, 전이 및 환자의 예후와 관련이 있는 것으로 알려져 있다. Hypoxia-inducible factor 1(HIF-1)는 당원 수송체, 당분해 효소, 혈관내피세포 성장인자 등의 유전자의 전사를 활성화한다고 알려져 있다. 그리고 HIF-1의 전사 활성도는 HIF-$1{\alpha}$ 아단위의 표현이 조절되는 정도에 의존한다. 비소세포 폐암에서 HIF-$1{\alpha}$의 발현이 혈관 생성능, 종양세포 증식능 및 이상형 p53의 축적 등 종양의 생물학적 특성에 미치는 영향과 환자의 수술 후 예후와의 관계를 규명하고자 한다. 대상 및 방법: 1997년부터 1999년까지 비소세포 폐암으로 진단받고 전폐절제술 혹은 폐엽절제술을 시행 받은 59명의 폐암 환자들에서 얻어진 파라핀 조직 블록을 대상으로 하였다. ABC(avidin-biotin complex) 방법에 기초한 면역조직화학검사를 이용하여 암조직과 정상조직에서 HIF-$1{\alpha}$, VEGF(vascular endothelial growth factor), p53 단백의 발현을 조사하고, Ki-67의 발현을 이용한 증식지수를 측정하였다. HIF-$1{\alpha}$ 발현과 환자의 생존기간을 포함한 임상적-병리학적 변수들과의 상관관계, VEGF, p53의 발현과 증식지수와의 상관관계를 분석하였다. 결과: HIF-$1{\alpha}$의 과발현은 40.7%(24예/59예)였다. HIF-$1{\alpha}$의 과발현은 병리학적 TNM병기(p=0.004), T병기(p=0.020), N병기(p=0.004), 림프관/혈관 침범(p=0.019) 등과 관련이 있었다. 또 혈관내피세포 성장인자의 발현(p<0.001) 및 이상형의 p53의 발현(p=0.040)과 관련성이 있었다. Kaplan-Meier 생존분석에서 HIF-$1{\alpha}$의 과발현이 있는 환자의 5년 생존울은 22%로 HIF-$1{\alpha}$의 저발현 환자의 5년 생존율 61%에 비해 불량한 생존율을 보였고, 단변량분석과 다변량분석에서 HIF-$1{\alpha}$의 발현은 불량한 예후를 나타내는 인자로 관찰되었다. 결론: 이상의 결과로 비소세포 폐암 환자에서 HIF-$1{\alpha}$의 과발현은 종양내 신생혈관의 생성과 림프절 전이와 관련이 있는 표지자로 여겨지며, 수술 후 불량한 예후를 나타내었다.
