Genetic Variation in the ABCB1 Gene May Lead to mRNA Level Chabge: Application to Gastric Cancer Cases

  • Background: One of the major mechanisms for drug resistance is associated with altered anticancer drug transport, mediated by the human-adenosine triphosphate binding cassette (ABC) transporter superfamily proteins. The overexpression of adenosine triphosphate binding cassette, sub-family B, member 1 (ABCB1) by multidrug-resistant cancer cells is a serious impediment to chemotherapy. In our study we have studied the possibility that structural single-nucleotide polymorphisms (SNP) are the mechanism of ABCB1 overexpression. Materials and Methods: A total of 101 gastric cancer multidrug resistant cases and 100 controls were genotyped with sequence-specific primed PCR (SSP-PCR). Gene expression was evaluated for 70 multidrug resistant cases and 54 controls by real time PCR. The correlation between the two groups was based on secondary structures of RNA predicted by bioinformatics tool. Results: The results of genotyping showed that among 3 studied SNPs, rs28381943 and rs2032586 had significant differences between patient and control groups but there were no differences in the two groups for C3435T. The results of real time PCR showed over-expression of ABCB1 when we compared our data with each of the genotypes in average mode. Prediction of secondary structures in the existence of 2 related SNPs (rs28381943 and rs2032586) showed that the amount of ${\Delta}G$ for original mRNA is higher than the amount of ${\Delta}G$ for the two mentioned SNPs. Conclusions: We have observed that 2 of our studied SNPs (rs283821943 and rs2032586) may elevate the expression of ABCB1 gene, through increase in mRNA stability, while this was not the case for C3435T.

미생물 성장을 억제하기 위하여 수용성 절삭유에 과다하게 첨가한 붕소와 아민 사례 연구 (Ethanolamine and boron abuse to limit microbial growth in water-synthetic metalworking fluids)

  • This study was conducted to examine whether a specific synthetic metalworking fluid (MWF), "A", in use for 10 months without replacement, displayed microbial resistance and to identify the additives associated with the control of microbial growth. Three synthetic MWF products ("A", "B", and "C") were studied every week for two months. Microbial deterioration of the fluids was assessed through evaluation by endotoxin, bacteria and fungi levels in the MWFs. In addition, formaldehyde, boron, ethanolamine, and copper levels were also studied to determine whether they influence microbial growth in water-based MWFs. Throughout the entire study in the sump where MWF "A" was used, bacteria counts were lower than 103 CFU/mL, and endotoxins never exceeded 103 EU/mL. These levels were significantly lower than levels observed in sumps badly deteriorated with microbes. Boron levels in MWF "A" ranged from 91.7 to 129.6 ppm, which was significantly higher than boron levels found in other MWF products. The total level of ethanolamine (EA) in MWF "A" ranged from 35,595 to 57,857 ppm (average 40,903 ppm), which was over ten times higher than that found in other MWFs. Monoethanolamine (MEA), diethanolamine (DEA) and triethanolamine (TEA) concentrations in MWF "A" were also significantly higher than seen in other MWFs. However, although EA and boron might improve anti-microbial performance, their abuse can pose a serious risk to workers who handle MWFs. From an industrial hygiene perspective, our study results stress that the positive synergistic effect of boron and EA in reducing microbial activity in MWF must be balanced with the potentially negative health effects of such additives. Our study also addresses the disadvantage of failing to comprehensively report MWF additives on Material Safety Data Sheets (MSDS). Future research in MWF formulation is needed to find the best level of EA and boron for achieving optimal synergistic anti-microbial effects while minimizing employee health hazards.

남산 주요 식생의 토양 미생물의 분포 및 생리적 특성 (Distribution and Properties of Microorganisms in Soil of Representative Vegetation of Mt. Nam)

  • Physicochemical factors, microbial population size and the properties of the bacterial isolates were estimated to find out the nature of soil ecosystem of Mt. Nam. Samples were obtained from the surface layer of soils on which specific plant community is developed. Average content of moisture and organic matter of the soils were 21.6% and 17.3%, respectively. These values were similar to those of developing forest soils, but were slightly lower than those of climax ecosystem such as Piagol in Mt. Chiri. Chiri. Content of phosphate was higher than those of other forest soils. The population size of soil bacteria ranged from 27.4 to 195.8 ${\times}\;10^5$ CFU/g. duy soil, and the size is somewhat dependent on the moisture and oranic matter content of soils. A large number of bacteria were able to decompose macromolecules such as starch, elastin and gelatin. Bacterial species composition of each soil was comparatively simple. Pseudomonas, Agrobacterium, Flavobacterium and Xanthomonas which are Gram-negative short rods were widely distributed in the forest soils. The endospore forming Bacillus species were also the main constituents of the soil microflroa. Actinomycetes were widely distributed in the forest soils, but the distribution pattern varied in each site. Most of the actinomycetes were also able to decompose organic macromolecules. The rate of resistant actinomycete strains to antibiotics and heavy metals were lower than those from cultivated soils, but higher than those from well-preserved forest soils. Antibiosis pattern of the actinomycete isolates was similiar to the resistance pattern. This means the forest soils of Mt. nam was somewhat interferred by artificial behabiour.

Comparative Genomic Analysis of Lactobacillus rhamnosus BFE5264, a Probiotic Strain Isolated from Traditional Maasai Fermented Milk

  • Lactobacillus rhamnosus BFE5264, isolated from a Maasai fermented milk product ("kule naoto"), was previously shown to exhibit bile acid resistance, cholesterol assimilation, and adhesion to HT29-MTX cells in vitro. In this study, we re-annotated and analyzed the previously reported complete genome sequence of strain BFE5264. The genome consists of a circular chromosome of 3,086,152 bp and a putative plasmid, which is the largest one identified among L. rhamnosus strains. Among the 2,883 predicted protein-coding genes, those with carbohydrate-related functions were the most abundant. Genome analysis of strain BFE5264 revealed two consecutive CRISPR regions and no known virulence factors or antimicrobial resistance genes. In addition, previously known highly variable regions in the genomes of L. rhamnosus strains were also evident in strain BFE5264. Pairwise comparison with the most studied probiotic strain L. rhamnosus GG revealed strain BFE5264-specific deletions, probably due to insertion sequence-mediated recombination. The latter was associated with loss of the spaCBA pilin gene cluster and exopolysaccharide biosynthetic genes. Comparative genomic analysis of the sequences from all available L. rhamnosus strains revealed that they were clustered into two groups, being within the same species boundary based on the average nucleotide identities. Strain BFE5264 had a sister group relationship with the group that contained strain GG, but neither ANI-based hierarchical clustering nor core-gene-based phylogenetic tree construction showed a clear distinctive pattern associated with the isolation source, implying that the genotype alone cannot account for their ecological niches. These results provide insights into the probiotic mechanisms of strain BFE5264 at the genomic level.

  • This study explores the effects of facemasks on respiratory, thermoregulatory, cardiovascular responses during exercise on a treadmill and at rest. Five male subjects (25.8 ± 0.8 y, 171.8 ± 9.2 cm in height, 79.8 ± 28.1 kg in weight) participated in the following five experimental conditions: no mask, KF80, KF94, KF99, and N95. Inhalation resistance was ranked as KF80 < KF94 < N95 < KF99 and dead space inside a mask was ranked as KF80 = KF94 < N95 < KF99. The surface area covered by a mask was on average 1.1% of the total body surface area. The results showed no significant differences in body core temperature, oxygen consumption (VO2), carbon dioxide production (VCO2), heart rate or subjective perception among the five experimental conditions; however, cheek temperature, respiratory ventilation and blood pressure were greater for KF80 or KF94 conditions when compared to KF99 or N95 conditions (p<0.05). The differences among mask conditions are attributed to the dead space or specific designs (cup type vs pleats type) rather than the filtration level. In addition, the results suggest that improving mask design can help mitigate respiratory resistance from increased filtration.

도시하수 슬러지의 탈수특성 연구 (Dewatering Characteristics of Sewage Sludge Produced by the Biological Treatment Process)

  • The dewatering characteristics of the sewage sludge was investigated through the experimental observations and model simulations. The activated sludge and the anaerobically digested sludge were examined for the dewaterability evaluation within the pressure range of $0{\sim}10^6N/m^2$. Modified Buchner funnel test and compression test by the consolidometer were conducted to evaluate average specific resistance, porosity, and moisture percentage of filter cake. Shirato's technique of compression-permeability test was followed for the pressure range lower than about $10^2N/m^2$. The flocculation effects on sludge dewatering was also examined for ferric chloride and polymeric flocculant. The application of hydrated lime which can be used for flue-gas desulfurization showed improved moisture percentage, and was thought to have positive feasibility in combined system of sludge dewatering and incineration. Determined characteristic constants were applied to Tiller's cake filtration model to simulate liquid pressure distribution and porosity distribution in cake. Model simulations showed a sharp drop of the porosity close to the cake-medium interface for the highly compressible material such as the activated sludge and the anaerobically digested sludge.

  • In this study, rheological properties of ternary system cement containing ground fly ash(F3, Blaine specific surface area $8,100\;cm^2/g$) were investigated using mini slump, coaxial cylinder viscometer and conduction calorimeter. In the results, the segregation resistance was observed at high W/B and PC area while the replacement ratio of F3 was increasing. The 2:5:3 system was shown in higher fluidity and lower hydration heat than 3:4:3 system. The segregation range of cement pastes occurred over 175 mm in average diameter by mini slump and below $10\;dynesec/cm^2$ of the plastic viscosity or below 50 cP of the yield stress by coaxial cylinder viscometer. It was observed that even if BFS and FA blended together admixture properties would remaine as they were separately. The properties of admixture would not be changed. On the above results, the decreased replacement ratio of OPC and increased replacement ratio of admixtures would be possible.

Constant Output Power Control Methods for Variable-Load Wireless Power Transfer Systems

  • This study proposes a comprehensive mathematical model that includes coil-system circuit and loss models for power converters in wireless power transfer (WPT) systems. The proposed model helps in understanding the performance of WPT systems in terms of coil-to-coil efficiency, overall efficiency, and output power capacity and facilitates system performance optimization. Three methods to achieve constant output power for variable-load systems are presented based on system performance analysis. An optimal method can be selected for a specific WPT system by comparing the efficiencies of the three methods calculated with the proposed model. A two-coil 1 kW WPT system is built to verify the proposed mathematical model and constant output power control methods. Experimental results show that when the load resistance varies between 5 and $25{\Omega}$, the system output power can be maintained at 1 kW with a maximum error of 6.75% and an average error of 4%. Coil-to-coil and overall efficiencies can be maintained at above 90% and 85%, respectively, with the selected optimal control method.

Cap truss and steel strut to resist progressive collapse in RC frame structures

  • In order to improve the efficiency of the Reinforced Concrete, RC, structures against progressive collapse, this paper proposes a procedure using alternate path and specific local resistance method to resist progressive collapse in intermediate RC frame structures. Cap truss consists of multiple trusses above a suddenly removed structural element to restrain excessive collapse and provide an alternate path. Steel strut is used as a brace to resist compressive axial forces. It is similar to knee braces in the geometry, responsible for enhancing ductility and preventing shear force localization around the column. In this paper, column removals in the critical position at the first story of two 5 and 10-story regular buildings strengthened using steel strut or cap truss are studied. Based on nonlinear dynamic analysis results, steel strut can only decrease vertical displacement due to sudden removal of the column at the first story about 23%. Cap truss can reduce the average vertical displacement and column axial force transferred to adjacent columns for the studied buildings about 56% and 61%, respectively due to sudden removal of the column. In other words, using cap truss, the axial force in the removed column transfers through an alternate path to adjacent columns to prevent local or general failure or to delay the progressive collapse occurrence.

  • 유기용액계 전해질을 갖는 전기이중층캐패서터의 성능향상을 위해 마이크로파 처리를 통하여 활성탄소를 개질하고 이에 따른 전기화학적 특성의 변화를 조사하였다. 대상 시료로 petroleum cokes와 pitch cokes를 사용하여 NaOH 활성화에 의해 제조한 활성탄과 시판용 활성탄 BP-25를 사용하였다. 세 종류의 활성탄 모두 마이크로파 처리를 통해 산소를 포함하는 친수성관능기들이 표면으로부터 제거되었으며, 처리 시간이 증가함에 따라 비표면적과 세공부피는 감소하고, 평균세공직경은 증가하였다. 이러한 영향으로 표면 개질된 활성탄소로 제조한 전기이중층캐패시터는 계면저항이 개질하지 않은 활성탄소를 사용한 전기이중층 캐패서터에 비해 크게 감소하였으며, 비표면적의 감소에도 불구하고 방전용량은 개질하지 않은 경우보다 크게 증가하였다.
