  • Two subspecies, japonica and indica, have been reported in rice, which differ in several ecotypic traits. However, reproductive barriers in hybrid progenies between subspecies have been major obstacles in breeding programs using inter-subspecific hybridization. As the first step to elucidate the reproductive barriers, we developed subspecies-specific(SS) STS markers in this study. A total of 765 STS primers were designed through comparing DNA sequences at every $2{\sim}3$cM interval between japonica and indica rices, which are available at Web DBs such as IRGSP, NCBI, TIGR, and GRAMENE, and tested for subspecies-specificity using 15 indica and 15 japonica varieties of diverse origin. Of them, 67 STS markers were identified as SS STS markers and their subspecies-specificity scores were estimated. The SS markers were dispersed throughout the genome along chromosomes. Of them, 64 SS markers were mapped on an RIL population derived from a Dasanbyeo(indica)/TR22183(japonica) cross. Genomic inclination of RILs was evaluated based on the genotyping with different types of markers. Association test between markers and segregation distortion revealed that segregation distortion might not be the cause of generating SS markers. The SS markers will be applicable to estimate the genomic inclination of varieties or lines and to study the differentiation of indica and japonica, and ultimately to breed true hybrid rice varieties in which desirable characters from both subspecies are recombined.

  • PURPOSES : In this study, field performance evaluation of crack treatment of pavement and the feasibility of surface treatment of pavement are presented. The performance and cost of preventive maintenance methods have been previously verified, and the methods are being used in many developed countries and cities. However, the performance and cost of the system have not been verified in domestic, field applications. Therefore, in order to improve performance, the field performance is evaluated, and a reasonable cost is proposed. METHODS : Visual Inspection was conducted to evaluate the field application and performance of the preventive maintenance method. In addition, the PCI index was calculated from the results of visual inspection of the application area of the surface treatment method, and the performance life of each method was predicted. For the economic evaluation, life cycle cost analysis was performed using the life cycle cost analysis program. RESULTS :In order to evaluate and quantify the field performance of crack repair material, the residue condition of the pavement surface after crack treatment, rather than the performance of the material, is evaluated. In addition, the crack resistance and performance life of surface treatment methods are evaluated. The cost of currently available treatment methods are compared to the common pavement cut and overlay method, and it is determined that the preventive method is not economical based on life cycle cost analysis. CONCLUSIONS :Because of the characteristics of cracking, it is necessary to conduct the evaluation of currently applied methods and the analysis of the cause of damage, by visual inspection. Moreover, in order to evaluate the performance and economic suitability of the currently applied surface treatment methods, it is necessary to acquire information on application sections by monitoring their long-term conditions and performance.

  • Cryo-electron microscopy (cryo-EM) is now the first choice to determine the high-resolution structures of huge protein complexes. Grids with two-dimensional arrays of holes covered with a carbon film are typically used in cryo-EM. Although semi-automatic plungers are available, notable trial-and-error is still required to obtain a suitable grid specimen. Herein, we introduce a new method to obtain thin ice specimens using real-time measurement of the liquid amounts in cryo-EM grids. The grids for cryo-EM strongly diffracted laser light, and the diffraction intensity of each spot was measurable in real-time. The measured diffraction patterns represented the states of the liquid in the holes due to the curvature of the liquid around them. Using the diffraction patterns, the optimal time point for freezing the grids for cryo-EM was obtained in real-time. This development will help researchers rapidly determine high-resolution protein structures using the limited resource of cryo-EM instrument access.

  • (S)-Carbamic acid 2-[4-(4-fluoro-benzoyl)-piperidin-1-yl]-1-phenyl-ethyl ester hydrochloride (YKP1447) is a novel "atypical" antipsychotic drug which selectively binds to serotonin (5-$HT_{2A}$, Ki=0.61 nM, 5-$HT_{2C}$, Ki=20.7 nM) and dopamine ($D_2$, Ki=45.9 nM, $D_3$, Ki=42.1 nM) receptors with over $10\sim100$-fold selectivity over the various receptors which exist in the brain. In the behavioral studies using mice, YKP1447 antagonized the apomorphine-induced cage climbing ($ED_{50}$=0.93 mg/kg) and DOI-induced head twitch ($ED_{50}$=0.18 mg/kg) behavior. In the dextroamphetamine-induced hyperactivity and conditioned avoidance response (CAR) paradigm in rats, YKP1447 inhibited the hyperactivity induced by amphetamine ($ED_{50}$=0.54 mg/kg) and the avoidance response ($ED_{50}$=0.48 mg/kg); however, unlike other antipsychotic drugs, catalepsy was observed only at much higher dose ($ED_{50}$=68.6 mg/kg). Based on the CAR and catalepsy results, the therapeutic index (TI) value for YKP1447 is over 100 (i.p.). These results indicate that YKP1447 has an atypical profile and less undesirable side effects than currently available drugs.

  • Competition among anion species in solution for same sorption sites and soil surface can be of major significance in determining the effective mobility of any potentially adsorbing species. Major soil anion species include $OH^-$, $F^-$, $Cl^-$, $HCO_3{^-}$, $CO_3{^-}$, $NO_3{^-}$, $SO_4{^{2-}}$, and $PO_4{^{3-}}$. And some micro nutrients such as boron and molybdenum exist as $H_2BO_3{^-}$ and $MoO_4{^{2-}}$, as do some heavy metals such as chrome and arsenic as $CrO_4{^{2-}}$ and $HAsO_4{^{2-}}$. Pesticides such as 2,4,5-T and 2,4-D also exist as anions. Many anion species are retained by more complex mechanisms than the simple electrostatic attractions involved in most cation adsorption reactions. In binary system composed of two anions, the adsorption of one anion is influenced by the other anion due to the competition for the available and limited binding sites in soil constituents. The specifically adsorbed anions may compete more effectively for sorption sites than that of nonspecifically adsorbed anion. In this study, we aim to evaluate the mathematical models to determine the magnitude of concentration variations in adsorption due to competitive interactions between anions introduced to a system in binary mixtures.

  • Two genera of the Rhodymeniales, Halopeltis and Leptofauchea, are here reported for the first time from the Hawaiian Islands and represent the deepest records for both genera. Molecular phylogenetic analyses of cytochrome oxidase subunit I (COI), rbcL, and large subunit ribosomal DNA (LSU) sequences for Hawaiian specimens of Leptofauchea revealed one well-supported clade of Hawaiian specimens and three additional lineages. One of these clades is described here as Leptofauchea huawelau sp. nov., and is thus far known only from mesophotic depths at Penguin Bank in the Main Hawaiian Islands. L. huawelau sp. nov. is up to 21 cm, and is the largest known species. An additional lineage identified in the LSU and rbcL analyses corresponds to the recently described L. lucida from Western Australia, and is a new record for Hawai'i. Hawaiian Halopeltis formed a well-supported clade along with H. adnata from Korea, the recently described H. tanakae from mesophotic depths in Japan, and H. willisii from North Carolina, and is here described as Halopeltis nuahilihilia sp. nov. H. nuahilihilia sp. nov. has a distinctive morphology of narrow vegetative axes that harbor constrictions along their length. The current distribution of H. nuahilihilia includes mesophotic depths around W. Maui, W. Moloka'i, and the island of Hawai'i in the Main Hawaiian Islands. Few reproductive characters were observed because of the small number of specimens available; however, both species are distinct based on phylogeny and morphology. These descriptions further emphasize the Hawaiian mesophotic zone as a location harboring many undescribed species of marine macroalgae.

  • The formation of uniform vitreous ice is a crucial step in the preparation of samples for cryogenic electron microscopy (cryo-EM). Despite the rapid technological progress in EM, controlling the thickness of vitreous ice on sample grids with reproducibility remains a major obstacle to obtaining high-quality data in cryo-EM imaging. The commonly employed classical blotting process faces the problem of excess water that cannot be absorbed by the filter paper, resulting in the formation of thick and heterogeneous ice. In this study, we propose a novel approach that combines the recently developed nanowire self-wicking technique with the classical blotting method to effectively control the thickness and homogeneity of vitrified ice. With simple procedures, we generated a copper oxide spike (COS) grid by inducing COSs on commercially available copper grids, which can effectively remove excess water during the blotting procedure without damaging the holey carbon membrane. The ice thickness could be controlled with good reproducibility compared to non-oxidized grids. Incorporated into other EM techniques, our new modification method is an effective option for obtaining high-quality data during cryo-EM imaging.

  • As a silicate source to rice, a coal ash was selected and mixed with phosphor-gypsum (50:50, wt $wt^{-1}$) to reduce the potential of boron toxicity and to supply calcium element. We expected that high con tent of calcium in this mixture might convert water-soluble phosphorus to less soluble forms and then reduce the release of soil phosphorus to surface runoff. The mixture was applied with the rate of 0, 20, 40, and 60 Mg $ha^{-1}$ in paddy soil (Nagdong series, a somewhat excessively drained loamy fine sand) in Daegok, Jinju, Korea The mixture reduced significantly water-soluble phosphorus (W-P) in the surface soils by shifting from W-P and Fe-P to Ca-P and Al-P during whole rice cultivation. In contrast with W-P, plant available phosphorus increased significantly with the mixture application due to high content of phosphorus and silicate in the mixture. The mixture of coal ash and phosphor-gypsum (50:50, wt $wt^{-l}$) would be a good alternative to reduce a phosphorus export in rice paddy soil together with increasing rice yields.

  • Convergence in technology and media makes it possible to augment nurturing by making blended infrastructures with new digital resources available in learning environments at schools. A game-based provision of educationally-valuable content within a collective, virtual environment combined with an analysis of events in the game after it has been played (blended nurture environment) may help to better communicate human values especially where old-fashioned discussion methods fail or do not fulfill the promise of effective, educational institution-based methods of bringing up. In this paper we discuss the use of behavioural metaphors in game-based blended-nurture scenarios for computer games and pedagogical-psychological tests. Upbringing demands an active intervention in the process of children growing up and the courage to place higher expectations on ourselves and our own lifestyles. Decisions and the behaviour of the younger generation are often irrational and the resulting effects can have destructive consequences. Nowadays, games take on the role of modern storytellers. With the help of complex analogies in games, a variety of situations can be depicted in an eye-catching way and later discussed.
