Antioxidant effect of Vitamin-C / alginate gel-entrapped liposomes for resistance of DHA autoxidation

  • The resistance of docosahexaenoic acid (DHA) incorporated $L-{\alpha}-phosphatidyl$ -choline (PC) liposomes against autoxidation was studied for application to food and cosmetic industry. For the preparation of vitamin-C/calcium alginate gel entrapped DHA-PC-liposomes (AVDLs), DHA incorporated PC bilayer was hydrated with vitamin-C containing calcium alginate solution, and the fraction containing liposome was suspended in $CaCl_2$ solution. DHA loading efficiency was calculated by TLC scanning method The morphological examination of AVDLs was performed with transmission electron microscopy (TEM) and lipid peroxidation was measured with an assay for thiobarbituric acid reactive substance (TBARS). DHA loading efficiency was about 17 % of initial loading volume, and when AVDLr containing 0.2 % vitamin C, lipid oxidation was minimized.

  • In order to study for Onion ethanolic extract on the Heavy Metal Elimination and antioxidation, the peroxide values of oil and eliminated metal were analyzed. The results are follows: It was very effective as a retardant for autoxidation processing of the soybean and olive oil by the Onion ethanolic extract. Quercetin in the Onion ethanolic extract was affected as a ligand for chelating with some metals. Inhibition of Human Hemoglobin Autoxidaiton by Sodium n-Dodecyl Sulphate

  • The effect of sodium n-dodecyl sulphate (SDS) on hemoglobin autoxidation was studied in the presence of a 100mM phosphate buffer (pH 7.0) by different methods. These included spectorphotometry, fluorescence technique, cyclic voltametry, differential scanning calorimetry, and densitometry. Spectroscopic studies showed that SDS concentrations up to 1 mM increased deoxy-, decreases oxy-, and had no significant effect on the met- conformation of hemoglobin. Therefore, a SDS concentration up to 1 mM increased the deoxy form of hemoglobin as the folded, compact state and decreases the oxy conformation. The turbidity measurements and differential scanning calorimetry techniques indicated a more stable conformation for hemoglobin in the presence of SDS up to 1mM. Electrochemical studies also confirmed a more difficult oxidation under these conditions. The induction of the deoxy form in the presence of SDS was confirmed by densitometry techniques. The compact structure of deoxyhemoglobin blocks the formation of met-conformation in low SDS concentrations.

Storage Stability of Refined Sardine Oil (정제정어리유의 저장안정성에 관한 연구)

  • For the utilization of polyunsaturated fatty acids in sardine (Sardinops melanosticta) oil, mainly as a source for dietary supplement of eicosapentaenoic acid(EPA) and docosahe-xaenoic acid(DHA), the effects of refining and storage, conditions, and addition of stabili-zing agents on the storage stability of EPA condensed and refined sardine oil were tested. Refined, sardine oil deacidified, decolored, and deodorized, was more stable to autoxidation than crude or partially purified oils such as deacidified or/and decolored. The refined sardine oil must be kept at $5^{\circ}C$ or lower temperture for longer than two month storage. The addition of $0.018\%\;\alpha-tocopherol$ or BHT could enhance the stability to autoxidation, and EDTA or citric acid was useful as synergist. Especially the addition of $0.02\%$ citric acid to the oil still hot after deodorizing process ($125^{\circ}C$) was of benefit to stabilize the refined oil.

Formation of Oxidative Cleavage Products from Lycopene (Lycopene으로부터 산화개열산물의 생성)

  • Lycopene was subjected to ozonolysis in ice-cold dichloromethane. The ozonolysis products were fractionated with a silica column and the carbonyl fraction was analyzed by ODS-HPLC with a photodiode array detector and by LC-MS. UV-vis spectra and $[M+H]^+$ of the carbonyl compound peaks showed clearly that acycloretinal, apo-14'-lycopenal, apo-12'-lycopenal, apo-10'-lycopenal, apo-8'-lycopenal and apo-6'-lycopenal were formed by ozonolysis of lycopene. Lycopene was solubilized in toluene and aqueous Tween 40, and then oxidized by incubating at $37^{\circ}C$ under atmospheric oxygen. Carbonyl compounds were produced. In comparison with autoxidation and ozonolysis, each compound showed the same retention time and UV-vis spectra are identical to the reference cleavage products prepared by ozonolysis of lycopene. Thus, eccentric cleavage of lycopene was confirmed to occur in vitro under oxidation condition.

Effect of Tocopherols and Carotene on the Oxidation of Purified Pinenut Oil in the Model System (토코페롤 및 카로틴이 정제 잣 지방질의 산화에 미치는 효과)

  • The oxidation of purified pinenut oil containing various concentration of tocopherols and β-carotene were studied. α-tocopherol revealed an antioxidant activity at the concentration of lower than 0.05%, however, it showed a prooxidant activity when the concentration was higher than 0.05%. The antioxidant activity of γ-tocopherol was not affected by the concentraitons in the range of 0.01∼0.10% in pinenut oil. γ -Tocopherol resulted in higher antioxidant activity than that of α-tocopherol. β -carotene seemed to be a prooxidant when 0.01% of β-carotene was added. The fatty acids composition of purified pinenut oil have been changed during autoxidation. The concentration of linoleic acid decreased readly while oleic acid seemed to increase. And the concentration of saturated fatty acid has'nt been changed much during autoxidation.

Assessment of Antioxidant Activity and Residue Level of Phenolic Antioxidants in Autoxidation of Linoleic Acid (리놀레인산 자동산화에 미치는 페놀계 산화방지제의 활성 및 잔존량 평가)

  • This study assessed the antioxidant activity and residue level of phenolic antioxidants in autoxidation of linoleic acid. The antioxidant activity of phenolic antioxidants was measured based on peroxide value of linoleic acid at $50^{\circ}C$ for 8 days. We further evaluated the residue level of phenolic antioxidants in the autoxidation period by HPLC-UV. The residue level of antioxidants changed with time starting on day 0 (100%) and was determined by 100-remaining of antioxidants (%). Our results showed that peroxide values ranged from 0.33 to 10.18 meq/kg in propyl gallate, from 0.67 to 11.01 meq/kg in dodecyl gallate, from 0.01 to 10.34 meq/kg in octyl gallate, from 0.01 to 4.17 meq/kg in butylated hydroxytoluene (BHT), from 1.00 to 5.85 meq/kg in butylated hydroxyanisole (BHA), from 0.33 to 4.18 meq/kg in 2,4,5-trihydroxybutyrophenone, and from 1.00 to 11.01 meq/kg in tert-butylhydroquinone (TBHQ). Among the residue levels of antioxidants, on day 8, BHT showed the highest level while TBHQ showed the lowest. BHT showed the highest correlation coefficient, whereas BHA showed the lowest. This study proves that the residual level of phenolic antioxidants has a good correlation with the degree of autoxidation in linoleic acid.
