A method to estimate the autotrophic maximum specific growth rate is presented in this paper. First of all, the concentration of nitrifier is simulated based on the amount of N nitrified, the sludge age and the default value for the decay coefficient. Secondly the OUR of the sludge with access of ammonia is measured. The maximum specific growth rate can be calculated as ${\mu}_{max,A}\;=\;OUR_{max,A}/Y_A$. It was demonstrated that the maximum specific growth rate of autotrophic biomass is not a constants but a time variable parameter. It is concluded that using $OUR_{max,A}$ for dynamic estimating maximum specific growth rate is a good approach and that using a constant value for the maximum specific growth rate over a longer period of time could not predict the performance of activated sludge plants.
Caustic (NaOH) solution is used to remove $H_2S$ from hydrocarbon streams in petroleum refining industry, gradually being, so called, spent sulfidic caustic (SSC) which has high levels of $H_2S$ and alkalinity. Thus, SSC can be used as an electron donor and a buffering agent for autotrophic denitrification. As SSC, however, contains some non-biodegradable organics, air stripping was conducted to remove the non-biodegradable organics. As a result, over 93 % of the non-biodegradable organics was removed within 30 min of aeration. Then, $Na_2S_2O_3{\cdot}5H_2O$, methanol and organic matters, which are produced from a biodiesel production plant, were added to reform the air-stripped SSC and their products being referred to new sulfidic caustics (NSCs) I, II and III, respectively. Thereafter, to investigate the effect of these products on the removal of COD and TN, these products were injected to a biological nitrogen removal (BNR) process, resulting in additional 44 % TN removal without noticeable increase in the effluent COD level. Therefore, it can be said that the BNR process is a promising option to treat NSC as demonstrated in this study whose results can be useful for developing resource recovery technologies.
This study was conducted to investigate the effect of supporting material and ionic strength of the MS medium on growth of strawberry plantlets cultured in vitro for the rapid mass production. Explants of $Fragaria$$ananassa$ 'Houkouwase' in vitro were planted in the agar or Tosilee (commercial plug medium) medium as the supporting material and supplied with 1/4 MS, 1/2 MS or basal (as the control) MS medium in an autotrophic micropropagation. Plant height and root length were significantly greater when they were cultured in 1/2 MS medium as compared to those grown in the agar medium. Also, shoot fresh and dry weights, and leaf area in the 1/2 MS medium were greater than in 1/4 MS or basal MS medium. When plantlets were cultured in Tosilee medium and fed with the basal MS medium, plant height, root length, shoot fresh and dry weights, root fresh and dry weights, and leaf area were promoted and greater than those in plantlets cultured in the agar medium.
In this study, a process combined biofiltration with sulfur-utilizing autotrophic denitrification and membrane separation was proposed to examine the efficiency of nitrogen removal. As an experimental device, hollow-fiber module was installed in the center of reactor to generate the flux forward sulfur layer in the cylinder packed with granular sulfur. In addition, a simple module was installed in activated sludge aeration tank which inside and outside of sulfur-using denitrification module was covered with microfilter and the module was considered as an alternative of clarifier. The experiment for developing new MBR process was carried out for three years totally. As the results of first two-year experiment, successful nitrogen removal performance was revealed with lab-scale test and pliot scale plant using artificial wastewater and actual plating wastewater. In this year, pilot scale test using actual domestic wastewater was performed to prove field applicability. As the results, high-rate nitrogen removal performance was confirmed with about 0.19 kg ${NO_3}^--N/m^3$ day of rate. Also significant fouling and pressure increase were not found during the experiment. And, the production ratio of sulfate and the consumption ratio of alkalinity showed a slightly higher value about 311 mg ${SO_4}^{2-}/L$ and 369 mg $CaCO_3$/L, respectively. In conclusion, the developed MBR process can be utilized as an alternative for retrofiting existing wastewater plants as well as new construction of advanced sewage wastewater treatment plants, with cost-effective merit.
Recently microalgae have been proposed as a promising biodiesel feedstock, owing to their higher lipid productivity and non-arable land based cultivation system. Biomass and lipid productivities of microalgae are largely affected by various environmental and nutritional factors. In this study, the effects of nitrogen (nitrate and ammonium) and organic carbon (glucose and glycerol) sources on the cell growth and lipid production of Chlorella sp. KR-1 were examined in flask cultures. Under autotrophic culture conditions for 15 days, overall cell growth and lipid (fatty acid methyl ester, FAME) production with nitrate were better than those of ammonium, resulting in 1.06 g cell/L and 333 mg FAME/L, respectively. Maximal intracellular lipid contents (348 - 352 mg FAME/g cell) were observed at low concentrations of 1 mM for both nitrate and ammonium. In the supply of light, addition of glucose in the range of 1 - 20 g/L showed higher cell densities than the autotrophic cell growth condition. Higher lipid accumulation of 375 mg FAME/g cell could achieved at 5 g glucose/L albeit of relatively short incubation of 7 days. With glycerol, intracellular lipid contents were ~1.9 times lower than glucose cases although similar cell growths were observed for both carbon sources.
The growth of the algae strain Chlorella vulgaris under mixotrophic conditions in the presence of saccharified acorn-starch (acorn-glucose) was evaluated with the objective of increasing biomass growth and triacylglycerols (TAGs) content. The results indicated that 81.3% of starch was converted to glucose in acorns. C.vulgaris algal strains grown with acorn-glucose produced higher biomass and TAGs content than with autotrophic growth. The highest biomass production and TAGs content with 3 g/L acorn-glucose were 12.44 g/L and 32.9%, respectively. Biomass production with 3 g/L acorn-glucose was 16.4 fold higher than under autotrophic growth condition. These findings suggested that 3 g/L acorn-glucose is economic and efficient for biomass production/productivity and TAGs content of microalgae. This study provides a feasible way to reduce the cost of bioenergy production from microalgae.
Changes in the phosphorylation of Chlorella cells during the life cycle the aulotrophic and micotrophic synchronous culture were followed under the light and dark. 1. In the autotrophic culture of Chlorella the amounts of esterified phosphate compounds of the algal cell under the light increased during the growing period and decreased strikingly in the ripening period showing a peak at the $L_1$ i/-cell stage. 2. TRhe amount of total esterified phosphate compounds of the cell under the dark, however, decreased during the growing period and then kept fairly constnat during the ripening nad division periods showing the greates activity of the oxidative phosphorylation in the early growing stage. 3. It is presumed that the energy requirement of the dividing algal cell in the autotrophic culture is fulfilled prior to the nuclear division mostly by the photosynthetic phosphorylation. 4. In the mixotrophic culture, the amount of esterified phosphate compounds of the algal cells under the light increased during the growing period and decreased during the late ripening and early division periods showing a peak in the $L_2$-cell stage as in the case of the phosphorylation under the dark. 5. The phosphorylation of the fell grown in the glucose medium is more active under the dark than under the light in the stages of the growing and early ripening periods. 6. It is considered that the excess glucose in the algal cell not only promotes the oxidative phosphorylation but also inhibits the photosynthetic phosphorylation of the cell. 7. It is presumed that the energy requirement of the dividing algal cell in the glucose medium is fulfilled prior to the nuclear division by the combined action of oxidative and photosynthetic phosphorylation, mostly by the oxidative phosphorylation.

In this study, a new submerged membrane bioreactor process packed with granular sulfur (MBR-GS) was operated to identify the biological nitrogen removal behaviors with plating wastewater containing high-strength $NO_3{^-}$ concentration. The continuous denitrification was carried out at $20^{\circ}C$ with various nitrogen loading rates using synthetic wastewater, which composed of $NO_3{^-}$ and $HCO_3{^-}$, but also actual plating wastewater, which was collected from the effluent of the H metal plating company. As a result, high-rate denitrification in the range of $0.8kg\;NO_3{^-}-N/m^3\;day$ was accomplished at nitrogen loading rate of $0.9kg\;NO_3{^-}-N/m^3\;day$ using synthetic wastewater. Also, higher-rate denitrification with actual plating wastewater was achieved up to $0.91kg\;NO_3{^-}-N/m^3\;day$ at the loading rate of $1.11kg\;NO_3{^-}-N/m^3\;day$. Additionally, continuous filtration was possible during up to 30 days without chemical cleaning in the range of 20 cmHg of transmembrane pressure. On the basis of the proposed stoichiometry, ${SO_4}^{2-}$ production could be estimated efficiently, while observed alkalinity consumption was somewhat lower than theoretical value. Consequently, a new process, MBR-GS is capable of high-rate autotrophic denitrification by compulsive flux and expected to be utilized as an alternative of renovation techniques for nitrogen removal from not only plating wastewater but also municipal wastewater with low C/N ratio.
The culture conditions and cell composition of a hydrogen bacterica, Alcaligenes eutrophus ATCC 17697. were investigated. Optimum pH and temperature for cell growth under autotrophic condition ($H_2$ as energy source, $CO_2$, as crabon source) were around 7.0 and $30^{\circ}C$, respectively. Effect of oxygen partial pressure in the range of 0.059 atm and 0.27 atm on cell growth was also studied. Maximum specific growth rate $({\mu}max=0.31hr^{-1})$ was observed at 0.11 atm of oxygen partial pressure $(H_2:O_2:CO_2=7:1:1)$. The contents of crude protein, nucleic acid and ash in cells were 69.2%, 17.6%, and 3.62%, respectively.

Hypoxia can indeed impact the survival of protists, which play a crucial role in marine ecosystems. To better understand the protistan community structure and species that can thrive in hypoxic waters, we collected samples from both the surface and bottom waters during the hypoxic period in Jinhae and Masan Bays and the non-hypoxic period in Jinhae Bay. Subsequently, we utilized metabarcoding techniques to identify the protistan species. During hypoxia, with dissolved oxygen concentrations of 0.8 mg L-1 in Jinhae Bay and 1.8 mg L-1 in Masan Bay within the bottom waters, the phylum Dinoflagellata exhibited the highest amplicon sequence variants richness among the identified protist phyla. Following the Dinoflagellata, Ochrophyta and Ciliophora also displayed notable presence. In hypoxic waters of Jinhae and Masan Bays, we identified a total of 36 dinoflagellate species that exhibited various trophic modes. These included one autotrophic species, 14 mixotrophic species, 9 phototrophic species with undetermined trophic modes (either autotrophic or mixotrophic), 2 kleptoplastidic species, and 10 heterotrophic species. Furthermore, the hypoxic bottom water exhibited a greater number of heterotrophic dinoflagellate species compared to the non-hypoxic surface water within the same water column or the non-hypoxic bottom water. Therefore, feeding by mixotrophic and heterotrophic dinoflagellates may be partially responsible for their dominance in terms of the number of species surviving in hypoxic waters. This study not only introduces the initial documentation of 26 dinoflagellate species surviving in hypoxic conditions but also establishes a foundation for a more comprehensive understanding of the ecophysiology of dinoflagellates in hypoxic marine environments.
