The bioleaching experiment under $42^{\circ}C$ was effectively carried out to leach the more valuable element ions from the pyrite in the Gangyang mine waste. Bacteria can survive at this temperature, as indigenous acidophilic bacteria were collected in the Hatchobaru acidic hot spring, in Japan. To enhance the bacterial activity, yeast extract was added to the pyrite-leaching medium. The indigenous acidophilic bacteria appeared to be rod-shaped in the growth-medium which contained elemental sulfur and yeast extract. The rod-shaped bacteria ($0.7\times2.6\;{\mu}m$, $0.6\times7\;{\mu}m$, $0.8\times5\;{\mu}m$ and $0.7\times8.4\;{\mu}m$) were attached to the pyrite surface. The colonies of the rod-shaped bacteria were selectively attached to the surroundings of a hexagonal cavity and the inner wall of the hexagonal cavity, which developed on a pyrite surface. Filament-shaped bacteria ranging from $4.92\;{\mu}m$ to $10.0\;{\mu}m$ in length were subsequently attached to the surrounding cracks and inner wall of the cracks on the pyrite surface. In the XRD analysis, the intensity of (111), (311), (222) and (320) plane on the bacteria pyrite sample relatively decreased in plane on the control pyrite sample, whereas the intensity of (200), (210) and (211) increased in these samples. The microbiological leaching content of Fe ions was found to be 3.4 times higher than that of the chemical leaching content. As for the Zn, microbiological leaching content, it was 2 times higher than the chemical leaching content. The results of XRD analysis for the bioleaching of pyrite indicated that the indigenous acidophilic bacteria are selectively attacked on the pyrite specific plane. It is expected that the more valuable element ions can be leached out from the mine waste, if the temperature is increased in future bioleaching experiments.

Purpose: The aim of this study was to evaluate the antimicrobial effect of a newly devised toothbrush with light-emitting diodes (LEDs) on Porphyromonas gingivalis attached to sandblasted and acid-etched titanium surfaces. Methods: The study included a control group, a commercial photodynamic therapy (PDT) group, and 3 test groups (B, BL, and BLE). The disks in the PDT group were placed in methylene blue and then irradiated with a diode laser. The B disks were only brushed, the BL disks were brushed with an LED toothbrush, and the BLE disks were placed into erythrosine and then brushed with an LED toothbrush. After the different treatments, bacteria were detached from the disks and spread on selective agar. The number of viable bacteria and percentage of bacterial reduction were determined from colony counts. Scanning electron microscopy was performed to visualize bacterial alterations. Results: The number of viable bacteria in the BLE group was significantly lower than that in the other groups (P<0.05). Scanning electron microscopy showed that bacterial cell walls were intact in the control and B groups, but changed after commercial PDT and LED exposure. Conclusions: The findings suggest that an LED toothbrush with erythrosine treatment was more effective than a commercial PDT kit in reducing the number of P. gingivalis cells attached to surface-modified titanium in vitro.

Statement of problem: The microbial adhesion on the surface of materials used in prosthodontics and restorative dentistry significantly influences microbial infection. Purpose: The purpose of this study was to evaluate the effect of how the degree of surface roughness of acrlyic resin affect the adhesion of bacteria. Material and methods: Resins were finished with $50{\mu}m$ and $250{\mu}m$ aluminium oxide particles by using sandblaster, by using stone point, and high polished with $Opa^{(R)}$ and Lace $motor^{(R)}$. The surface of acrylic resin attached by bacteria was directly touched on the surface of BHI agar, which was incubated. Bacteria colonies formed on BHI agar were counted in accordance with the degree of the surface roughness. Results: 1. The viable cell number of Streptococcus mutans increased on the acrylic resins incubated in BHI broth than in PBS. 2. The viable cell number of Streptococcus mutans increased on the acrylic resins incubated without agitation than with agitation, washed three times than six times, and incubated in broth added with 5% sucrose than without sucrose. 3. When Streptococcus mutans incubated in BHI broth, the number of Streptococcus mutans colonies formed on BHI agar was the largest on the acrylic resins finished with $250{\mu}m$ aluminium oxide particle using sandblaster. But when incubated in BHI broth containing sucrose, the number of colonies formed on that was the largest on the acrylic resins high polished using $Opal^{(R)}$ and Lace $motor^{(R)}$. 4. When Streptococcus sanguis was incubated in BHI broth with or without sucrose, the number of Streptococcus mutans colonies formed on BHI agar was the largest on the acrylic resins finished with $250{\mu}m$ aluminium oxide particle using sandblaster. 5. When Actinomyces viscosus was incubated in BHI broth with or without sucrose, the number of Streptococcus mutans colonies formed on BHI agar was the largest on the acrylic resins high polished using $Opal^{(R)}$ and Lace $motor^{(R)}$. Conclusion: These results indicated that when acrylic resins attached by bacteria were touched on the surface of BHI agar, the number of bacterial colonies formed on the agar was dependent on the bacterial species. Also, the result of this study was showed that increase in the surface roughness and the addition of sucrose increased retention of microbial cells.

Bio-filtration processes using honeycomb tubes (process 1) and aeration and manganese-sand filtration (process 2) were evaluated for the biological manganese removal efficiency. The concentration of manganese at effluent was stabilized after 20days operation in process 1. It was estimated the required time for attaching and growing microorganisms to honeycomb tubes. In long term of operation periods, manganese removal efficiency was dropped for the excessively attached biofilm and manganese dioxide to honeycomb tubes. It took several days for normal operation in process 2, after that manganese removal efficiency was increased to 98% and stabilized for 1.5 years. Microorganisms in process 1 and 2 were isolated and cultured to characterize manganese-oxidizing bacteria. Among the four types of colony, light brown colony was turned blue color by leuco crystal violet spot test. Stenotropomonas genus, known as manganese-oxidizing bacteria, was identified by 16S rDNA partial sequencing analysis which was isolated in process 1 and 2. For the biological treatment to remove manganese, these two considerations are important. One is to choose the proper media attaching manganese oxidant, another one is to define the cultural condition of isolated manganese-oxidizing bacteria.

This study performed in order to evaluate the effects of pipe materials on corrosion and bacteria regrowth using a laboratory scale batch test. Two varieties of feed water with different microbial conditions were selected: tap water, surface river water (Han River water), and five pipe materials; carbon steel, copper, galvanized iron, stainless steel, and PVC, Carbon steel and galvanized iron pipes showed higher corrosion rates than other materials. In terms of attached bacterial growth, pipes with PVC and stainless steel showed higher bacteria concentration compared to other materials. Pseudomonas vesicularis was the predominant bacteria found on biofilm. The behavior of bacterial growth in the pipes was observed using a scanning electron microscope.
Slime sampless from 10 ratating biological contactor (RBC) plants in New Jersey were microscopically examined. Filamentous bacteria such as Type 1701, Type 0041, Type 021N, Nocardia, Beggiatoa, and Sphaerotilus, which are commonly present as suspended forms also were found in RBC slimes growing as attached forms. However, the abundance wwas much different from that of activated sludge. In RBC slimes, Beggiatoa was most frequently observed filamentous bacteria and Sphaerotilus, Type 0041, Type 1701, Type 021N and Nocardia were present in decreasing order of abundance. There were morphologically two different types of Behhiatia in RBC slimes. The statistical anaysis shows that filamentous bacterial poulations between the 100 cm inside and the outside the RBC were different in most cases with significant interactions between the location and stage.
The mammalian glycosylphosphatidylinositol (GPI) anchor consists of three mannoses attached to acylated GlcN-(acyl)PI to form $Man_3$-GlcN-(acyl)PI. The first of the three mannose groups is attached to an intermediate to generate Man-GlcN-(acyl)PI by the first mannosyltransferase (GPI-MT-I). Mammalian and protozoan GPI-MT-I have different substrate specificities. PIG-M encodes the mammalial GPI-MT-I which has 423 amino acids and multiple transmembrane domains. In this work we cloned PIG-M homologues from humans, Plasmodium falciparum (PfPIG-M), and Saccharomyces cerevisiae (GPI14), to test whether they could complement GPI-MT-I-deficient mammalian cells, since this biosynthetic step is likely to be a good target for selective screening of inhibitors against many pathogenic organisms. PfPIG-M partially restored cell surface expression of the GPI-anchored protein CD59 in PIG-M deficient mammalian cells, and first mannose transfer activity in vitro; however, this was not the case for GPI14.
This study was carried out to leach valuable metals from galena using indigenous bacteria with no optimum pH conditions at room temperature. Even in these conditions, the rod-shaped indigenous bacteria, ranging from $0.4{\times}0.2{\mu}m$ to $0.5{\times}1.7{\mu}m$, were attached to the surface of the galena. For the 19 days of the bioleaching experiment, the content of Ph, Fe, Zn ions was found to be 347, 222 and 1.7 times higher than that of the control leaching agent, respectively. Numerous hexagonal column crystals were observed on the surface of galena. Those crystals may be formed from the biooxidation of galena by the indigenous bacteria. XRD analysis showed the peaks of anglesite observed in the bioleached galena. It is expected that more valuable elements can be leached out of the galena, if the bacteria is used under optimum pH and temperature conditions in future bioleaching experiments.
Four dead wild racoons found in Chonnam province were examined parasitologically, pathologically and bacteriologically. Using standard flotation technique with feces many number of ancylostoma eggs which were oval in shape and $60-70{\times}35-45 {\mu}m$ in size were detected. Grossly, severe anemia, multiple congestion and consolidation of lung and catarrhal exudate in small intestine were observed. One of racoons had multifocal white to yellow nodules in the liver. Histopathologically, ancylostoma larvas were found in intestinal lumen. Some of them penetrated and attached to intestinal mucosa. Lungs revealed multiple severe fibrinopurulent pneumonia in which bacterial colonies were scattered. Liver had multiple microabscesses containing peripheral bacterial colony. By Gram stain using paraffin sections of liver and lung, bacterial colony was Gram negative. Isolated bacteria from lung and liver was Gram negative rod. This bacteria was identical with Escherichia coli by biochemical tests. From these results, ancylostomiasis caused severe problems in Korean wild racoons. This is the first report about ancylostomiasis in wild racoons in Korea.
A study was made of diurnal changes in the ruminal bacteria associated with feed particles, i.e., non-associated (NAB), loosely associated (LAB), and tightly associated with particles (TAB), and the TAB concentration in different particle sizes from sheep fed orchardgrass (OG) or alfalfa (ALF) hay. Diaminopimelic acid (DAPA) was used to determine the TAB mass. Results showed that the bacterial masses in NAB and LAB were small, but comprised over 90% in TAB. The TAB mass in the ALF group sharply increased within 2 h after feeding and decreased afterward. The TAB mass showed the same trend in the OG group, increasing from 0 h to 2 h, but remained at the same level up to 14 h after feeding. The peak bacterial mass was, however, lower in the OG than the ALF group. The TAB concentration reflected the changes in total particulate tightly associated bacterial masses in both groups of hay fed sheep. Number of bacterial colonies per particle increased as the particulate size decreased in both groups. This difference, however, tended to decline as the postprandial period was prolonged. DAPA, however, tended to overestimate the TAB mass in the reticulo-rumen digesta of the hay fed sheep.
