A study on the optimal variable transformation method to identify the correlation between ATP and APC (ATP와 APC 간의 관련성 규명을 위한 최적의 변수변환법에 관한 연구)

  • In order to secure safe meals, the hazards of microorganisms associated with food poisoning accident should be monitored and controlled in real situations. It is necessary to determined the correlation between existing common bacteria number (aerobic plate count; APC) and RLU (relative light unit) in cookware. In this paper, we investigate the correlation between ATP (RUL) and APC (CFU) by using three types of transform (inverse, square root, log transforms) of raw data in two steps. Among these transforms, the log transform at the first step has been found to be optimal for the data of cutting board, knife, soup bowl (stainless), and tray (carbon). The square root-inverse and the square root-square root transform at the second step have been shown to be optimal respectively for the cup and for the soup bowl (carbon) data.

Studies on the Processing of Rapid- and low Salt-Fermented Liquefaction of Anchovy(Engrulis japonica)(III) - Changes in ATP-related compounds, TMAO, TMA, Creatine, and Creatinine during Fermentation - (저식염 속성 멸치 발효액화물 가공에 관한 연구(III) - 숙성 중 ATP관련화합물, TMAO, TMA, creatine 및 creatinine 함량변화 -)

  • Changes in ATP and related compounds, TMAO, TMA, creatine and creatinine were analyzed to establish the processing conditions for rapid- and low salt-fermented liquefaction of anchovy(Engrulis japonica) extracts during fermentation. Experimental sample A: chopped whole anchovy, adding 20% water, heating at $50^{\circ}C$ for 9 hrs and then adding 10% NaCl. Sample B: chopped whole anchovy, adding 20% water, heating at $50^{\circ}C$ for 9 hrs and then adding 13% NaCl. Sample C: chopped whole anchovy adding 13% NaCl. Sample D: whole anchovy adding 17% NaCl. ATP, ADP, AMP and IMP were broken down during fermentation period, while inosine and hypoxanthine or hypoxanthine were detected in each fermented liquefaction of anchovy. However the amounts of them were varied from collection to collection according to the pretreatment methods. Possibly ATP and their related compounds will not make a great contribution to the umami taste in fermented liquefaction of anchovy. The contents of TMAO were decreased during fermentation period, ranging from 3 to 15 mg/100g in the fermented liquefaction of anchovy after 180 days. The TMA contents were increased slowly during fermentation period, ranging from 60 to 114 mg/100g in the 180 days specimens, however their contents were varied from sample to sample. The contents of creatine and creatinine were increased during early fermentation period, and then they were decreased in the last period. As for distribution of nitrogen in the anchovy extracts, the contribution of creatine and creatinine to the extractive nitrogen was occupying 6.8, 5.7, 4.6 and 5.7% in the experimental sample A, B, C and D, respectively. The contribution of ATP and related compounds to the extractive nitrogen was occupying 2.1, 2.4, 2.2 and 2.7% in the experimental sample A, B, C and D, respectively. The contribution of TMAO and TMA to the extractive nitrogen was very low as they are occupying $0.7{\sim}1.2%$ in the four experimental samples.

The Relationship of Pulmonary Artery Copper Concentrations and Genes Involved in Copper Homeostasis in Cattle, Swine, and Goats

  • Liver and pulmonary artery tissue from 5 Angus cross bred steers, 6 goats, and 6 pigs were collected at a commercial abattoir to examine the relationship of pulmonary artery copper (Cu) concentrations and genes involved in copper homeostasis. Liver and pulmonary artery samples were collected at the time of harvest and snap frozen. Liver and pulmonary artery Cu concentrations were determined via flame atomic absorption spectrophotometry and gene expression was determined via real time PCR. Liver Cu concentrations (mg Cu/kg DM${\pm}$SE) were higher (p<0.01) in cows ($396.4{\pm}109.1$) and goats ($181.4{\pm}37.0$) than in pigs ($19.2{\pm}3.5$). All liver Cu concentrations were within normal ranges and considered adequate for each species. Liver Cu concentration was more variable in cows and goats compared to pig liver Cu concentrations. Pulmonary artery ${\beta}$-hydroxylproline was higher (p<0.01) in cow and pig than goat. Real Time PCR revealed that goat liver atp7a was positively correlated ($r^2$ = 0.92; p<0.01) to liver Cu concentrations while cow and pig atp7a was not correlated to liver Cu concentration. In the pig, liver atp7a concentration was positively correlated to atp7b ($r^2$ = 0.66; p<0.05). Pulmonary artery Cu concentration was highest in cows ($14.9{\pm}4.7$), intermediate in pigs ($8.9{\pm}3.3$), and lowest in goats ($3.9{\pm}1.1$). Goat pulmonary artery Cu concentration was not correlated to ctr1 concentration, however, atp7a concentration was positively correlated with ctr1 ($r^2$ = 0.90; p<0.01). In cow pulmonary artery, loxl1 concentration was positively correlated to eln mRNA concentration ($r^2$ = 0.91; p<0.02). Pulmonary artery CTR1 protein concentration was positively correlated to pulmonary artery Cu ($r^2$ = 0.85; p = 0.03) concentration while negatively correlated to liver Cu ($r^2$ = -0.79; p<0.04). Pulmonary artery Cu concentration was not correlated to concentration of Cu homeostatic genes in the pig. These data indicate that genes involved in Cu homeostasis (ctr1, atp7A, atp7B, loxl1 and eln) are differently regulated in different species.

Extractive Nitrogenous Constituents of Dried Layer, Porphyra dentata (잇바디돌김 건제품의 함질소 엑스성분 조성)

  • In order to investigate the composition of dried layer, Porphyra dentata cultured at the south coast of Korea, the dried layer was analyzed for extractive nitrogen, free amino acids, combined amino acids, ATP and its related compounds and quaternary ammonium basis, The extractive nitrogen contents of dried laver extracts were 670-1,304 mg/100 g (on dry basis). From twenty-eight to twenty-nine kinds of free amino acids were found in the dried laver extracts and their total amounts were $2,796\~6,277\;mg/100\;g$ (on dry basis). The extracts were rich in free amino acids such as alanine, taurine, glutamic acid, glutamine and phosphoserine, From eighteen to twenty-one kinds of combined amino acids were found in the extracts and their total amounts were $1,406\~2,142\;mg/100\;g$ (on dry basis). The amounts of ATP and its related compounds were $65.7\~124,7\;mg/100\;g(2.13\sim3.68{\mu}mol/g$ on dry basis), Homarine was detected in all samples but $\beta$-alaninebetaine, $\gamma$-bufobetaine and trigonelline disappeared during processing. TMAO and TMA were detected in all samples. During processing of dried layer, free amino acids, TMAO and TMA were increased but the other constituents such as combined amino acids, ATP and its related compounds and betaines were decreased in all specimens.

Microbial Activity Analysis for the Selectively Sterilizing of Government-controlled Bulk Public Archives (대량 공공 기록물의 선별적 소독을 위한 미생물 활성도 분석 연구)

  • An archive is a collection of documents or records. Currently, most archived documents are made of paper. Paper is susceptible to biological damage and deterioration due to its material properties. To control the biological damage, treatment with chemical disinfectants and control of the storage environment are often used. In government-controlled bulk public archives, all documents are chemically sterilized before storage. However, an extremely large quantity of public records have been produced, and storage space and conservation management are gradually reaching their limits. In this study, 60 species of microbes were identified using a genetic method. We successfully applied the adenosine triphosphate (ATP) bioluminescence method to detect microbial contamination on paper documents. A calibration curve of the ATP bioluminescence as a function of the microbe quantity was obtained, and the microbial activity on non-sterilized paper archives from 1951 was analyzed using an ATP luminometer. It was found that the microbial activity was suppressed or reduced in climate-controlled storage environments at $22^{\circ}C$ and 55% relative humidity. We anticipate that these results will be used to establish selective sterilization systems for government-controlled bulk public archives.

Changes of Components in Salt-Fermented Anchovy, Engraulis Japonicus Sauce during Fermentation (숙성기간에 따른 멸치액젓의 성분변화)

  • To investigate changes of components in salt-fermented anchovy, Engraulis japonims sauce during fermentation, various chemical properties were examined at $1.5{\~}3$ months interval during 18 months fermentation. Moisture content and pH were decreased slightly, but the contents of VBN and crude protein, total and amino nitrogen, hydrolytic degree and absorbance at 453 nm were increased gradually during fermentation. On the other hand, ash content and salinity showed almost no change. Hk and uric acid were the most abundant in ATP related compounds, ranging from $80.1{\%}\;to\;92.7{\%}$ in salt-fermented anchovy sauce during the fermentation, After 18 months of fermentation the sauce was rich in free amino acids, such as glutamic acid, leucine, alanine, lysine, isoleucine, valine in that order.

  • To investigate changes of components in salt-fermented blenny, Enedrias nebulosus sauce during fermentation, various chemical properties were examined at 2$\~$3 months intervals during 18 months of fermentation. The degree of hydrolysis increased sharply until 6 months of fermentation and showed the gentle increasement after that. On the other hand, the contents of total and amino nitrogen, total ATP related compounds increased gradually during 18 months of fermentation, The hypoxanthine and uric acid were the most abundant in ATP related compounds, ranging from $80.1\% to $90.5\%. After 18 months of fermentation, sauce was rich in free amino acids such as glutamic acid, alanine, lysine, valine, leucine in that order.

  • Asparagine biosynthesis by soybean sprouts grown under the dark conditions has been demonstrated. The amount of free asparagine synthesized in ten day-old soybean sprouts increases to 22.7% on the dry weight base. The effects of nitrogen compounds such as $NH_4Cl,\;(NH_4)_2SO_4$ and urea on asparagine synthesis during the sprouting were examined and the results showed that urea was more effective than other two compounds. Glutamine-dependent asparagine synthetase was partially purified (8.6 folds) through ammonium sulfate fractionation, followed by Sephadex G-150 gel filtration. The enzyme was very labile and required protection by thiol groups or high level of glycerol. The mixture of ATP and $Mg^{++}$ ion also stabilized the enzyme activity. The enzyme utilized glutamine more effectively than ${NH_4}^+$ as an amide donor for the formation of asparagine. The enzyme required L-aspartate (Km=3.1 mM), L-glutamine, ATP and $Mg^{++}$. It showed pH optimum of 7.5 and catalyzed the formation of ${\beta}-aspartyl$ hydroxamate in the presence of L-aspartate, ATP, $Mg^{++}$ and $NH_2OH$ in the reaction mixture.

The Effects of Electroacupuncture on Mechanical Allodynia and Its Involvement with the Sympathetic Nervous System (Tail model의 기계적 이질통에 대한 전침 자극의 효과 및 교감신경계의 관여기전)

  • Objective : This study was intended to investigate the analgesic effects of electroacupuncture(EA) on mechanical allodynia according to the frequency and intensity of EA. Also to know if mechanical allodynia and the analgesic effects of EA is related to the sympathetci nervous system and/or the purinergic system. Methods : mechanical allodynia-induced rats were produced by resecting S1-S2 nerve. The zusanli(ST36) was used for acupoint and the rats were divided into 4 groups. Each group was given different stimuli[low frequency low intensity-EA(LFLI-EA), low frequency high intensity-EA(LFHI-EA), high frequency low intensity-EA(LFHI-EA), high frequency high intensity-EA(HFHI-EA)]. Futhermore, to make sympathectomy6-OHDA and phentolamine were administered intraperitonially and the concentration of norepinephrine(NE) were measured. As a ATP blocker, suramin was applied for this study. Results : Comparing to control group, each of the 4 groups(LFLI-EA, LFHI-EA, HFLI-EA, HFHI-EA) showed a significant reduction of response frequency of mechanical allodynia. LFHI-EA was more effective than that of LFLI-EA. The LFHI-EA group also had longer lasting effects from the stimulation than the other groups. Sympathectomy didn't show any reduction of response frequency of mechanical allodynia.(Each n=6, n=4). Nor did both sympathectomy and ATP block. The response frequency wasn't reduced by sympathectomy or by sympathectomy and ATP block, but was significantly reduced with LFHI-EA Conclusions : These results suggest that EA has a significant analgesic effect on mechanical allodynia which has no connection with NE and/or ATP.

Effect of Biphenyl Dimethyl Dicarboxylate on Chemical-Induced Hepatotoxicity

  • To know the mechanism of biphenyl dimethyl dicarboxylate (DDB) in the protection of chemically induced hepatotoxicity, the activity of glutamic pyruvic tran.saminase (GPT) and the level of lipid peroxidation metabolite (malondialdehyde, MDA) and ATP content in hepatocytes were determined in serum and primarily cultured hepatocytes. For in vibo study, rats were pretreated with DDB (300 mg/ kg, p.o.)for 7 days. DDB pretreatment efficiently reduced the elevation of serum GPT activity induced by carbon tetrachloride (1.6 ml/kg, s.c.) and acetaminophen administration (1500 mg/kg, i.p.). In ex vivo study, hepatocytes were isolated from the rats pretreated with DDB (300 mg/kg, p.o.)for 7 days and cultured for 12 hrs before inducing cytotoxicity with chemicals. The MDA formation and the GPT release induced by adriamycin $(1\times10^{-4} mg/ml)$ and cisplatin $(2\times10^{-4} mg/ml)$ were markedly decreased in the hepatocytes from the rats pretreated with DDB as compared to vehicle only. However, DDB pretreatment did not prevent the decrease of ATP contents of hepatocytes induced by cisplatin and adriamycin. In in vitro experiment, DDB was pretreated in primary cultured hepatocytes for 3 days. DDB enhanced the decreases of ATP contents induced by cisplatin and adriamycln. These results suggest that DDB may protect the hepatocytes from injury induced by hepatotoxlcants through inhibiting the lipid peroxidation.
