Aspergillus Endocarditis after Open Herat Surgery (VSD Closure) (심실중격결손증 수술후 발생한 Aspergillus 심내막염)

  • A 15-year-old boy having a small VSD was readmitted with clinical manifestations of acute endocarditis and aortic regurgitation one month after open heart surgery. In spite of vigorous treatment with broad-spectrum antibiotics, high fever persisted. Pseudomonas aeruginosa was isolated just one time among several blood cultures. Progressive pulmonary infarction due to embolization. Progressive congestive heart failure and D.I.C. caused patient`s death. Aspergillus was found in autopsy specimen.

Extraction of β-glucosidase from Bagasse Fermented by Mixed Culture under Solid State Fermentation

  • Various parameters such as solvent selection, concentration, solid/liquid ratio, soaking time, temperature, stationary, shaking conditions, and repeated extractions were investigated in order to determine the optimum extraction conditions of ${\beta}$-glucosidase from bagasse fermented by mixed culture of Aspergillus niger NRC 7A and Aspergillus oryzae NRRL 447. Among various solvents tested, non ionic detergents gave the best results than the inorganic or organic salt solutions and distilled water. The optimum conditions for extraction of ${\beta}$-glucosidase were 30 min soaking time at $40^{\circ}C$ under shaking condition at 150 rpm, with solid/liquid ratio 1:15 (w/v), which yielded $2882.74{\pm}95.52U/g$ fermented culture (g fc) of enzyme activity. With repeated washes under the above optimum conditions, the results showed that enzyme extracted in the $1^{st}$ and $2^{nd}$ washes represents about 90% of the total activity.

  • A. niger H-18 and A. fumigatus E-29 wrer selected for their strong abilities to produce cellulase. The λd numerical values of the cotton fabrics inoculated with A. niger H-18 and A. fumigatus E-29 were 580 nm for the both strains of molds. By the growth of molds, lightness, original color scale, and grey scale of the fabrics gradually decreased while chroma increased. The minimum inhibitory concentrations of mold-proofing agents, such as Leperon WL, 8-hydroxyquinoline copper acetate, trimethylol melamine and dimethyl ethylene urea was 50 ppm. Glycoxale was not effective at the above mentioned concentration. Since Leperon WL, trimethylol melamine and dimethyl ethylene urea effectively inhibited the growth of A. niger H-18 and A. fumigatus E-29, tensile strength and elongation of the fabrics were not changed. however, cotton fabrics treated with glycoxale of the fabrics were not changed. However, cotton fabrics treated with glycoxale and inoculated with A. niger H-18 and A. fumigatus E-29 showed decreased in tensile strength by 31.1% and 33.9%, respectively.

  • In order to establish the enzyme linked immunosorbent assay(ELISA) of sterigmatocystin produced by Aspergillus versicolor, we experimented and obtained following results. Two of three rabbits which had been immunized with sterigmatocystin-hemiacetal-BSA produced antibodies against sterigmatocystin at 15 weeks. The produced antibodies were specific for sterigmatocystin and sterigmatocystin-hemiacetal but didn't cross react with other sterigmatocystin analogues in a significant degree. DMF : 4% KC1 (18 : 2) mixed solution was most effective to dissolve sterigmatocystin. For the preparation of sample solution to determine sterigmatocystin by ELISA, sample was extracted with CHC13 and dried, than the dried sample was redissolved with 100 ${mu}ell$ DMF + 4% KC1 mixture. 10~1,000 ng/$m\ell$ level of standard sterigmatocystin could be applied to the established ELISA. When artifically contaminated rice were assayed by the ELISA, the average recovery of sterigmatocystin spiked to 25~500 ng/g was 109% (97~116%), and mean interwell coefficient of variation was 21% (11~28%).

Semicontinuous Decolorization of Azo Dyes by Rotating Disc Contactor Immobilized with Aspergillus sojae B-10

  • Aspergillus sojae B-l0 was immobilized and used to treat model dye compounds. The model wastewater, containing 10 ppm of azo dyes such as Amaranth, Sudan III, and Congo Red, was treated with cells attached to a rotating disc contactor (RDC). Amaranth was decolorized more easily than were Sudan III and Congo Red. Decolorization of Amaranth began within a day, and the dye was completely decolorized within 5 days of incubation. Both Sudan III and Congo Red were almost completely decolorized after 5 days of incubation. Semicontinuous decolorization of azo by reusing attached mycelia resulted in almost complete decolorization in 20 days. This experiment indicated that decolorization was successfully conducted by removing azo dyes with Aspergillus sojae B-10.

  • A Nuluk, a Korean traditional Koji for brewing, was made with wheat flour and Aspergillus oryzae L2 which had a good aroma and strong abilities In producing saccharogenic and dextrogenic enzymes. The cultural conditions for the production of saccharogenic and proteolytic enzymes were tested. The productivity of dextrogenic enzyme was improved when Nuluk was made with unsteamed wheat flour as compared with steamed one, but that of proteolytic enzyme was reduced. The addition of water containing 0.5% hydrochloric acid was unfavorable for the production of those two enzymes. The optimum ratio of water added to wheat flour for the production of those two enzymes was 28$^{\circ}C$ on the basis of wheat flour, The productivity of saccharogenic enzyme was enhanced when the Nuluk was molded after 20 hours of precultivation, but that of proteolytic enzyme was reduced as compared with no molding. The optimum temperatures for the production of saccharogenic enzyme and proteolytic enzyme were 36$^{\circ}C$ and 28$^{\circ}C$, respectively.

  • Aspergillus fumigatus (A. fumigatus) is one of the most important mycotic infection in wild animals stressed by captivity and concurrent disease. A 14-year-old Carribean flamingo controlled bumble foot with antibiotics died after having dyspnea, depression, and anorexia. Necropsy findings revealed pyogranulomatous inflammations of lung and greenish gray-colored mold on air sacs. A carribean flamingo was diagnosed as pyogranulomatous pneumoniae due to A.fumigatus infection by results of lung tissue culture and microscopic examination.

  • We have determined the sequence of $tRNA^{Arg}$ of A. nidulans partially by enzymatic rapid RNA sequencing technique. The sequence was 5'GGCCGGCUGGCCCAAXUGGCAAGGXUCUGAXUACGAAXCAGGAGAUUGCACXXXXXGAGCXXUXXGUCGGUCACCA3' The cloverleaf structure was made from above data. As a result, the anticodon sequence was identified as ACG. This result was confirmed with charging test. The complete sequence was proposed by supplementing the DNA sequence to and by assigning the position of minor bases to this RNA sequence.

Studies on acid protease produced from Aspergillus tubingensis I (Aspergillus tubingensis의 acid protease에 관한 연구 I)

  • The strain of Aspergillus, 6368A, producing acid protease showing high activity was isolated from soil, as a result of wide research about mold group. This strain was identified as a species of Aspergillus tubingensis by the investigation of morphological characteristics. The change of the enzyme production under the various media and culture condition was also studied. The optimum pH and stability of crude acid protease are 2.5, 2.0~4.5 and the optimum temeprature and thermal inactivation waas shown $50^{\circ}C,\;55^{\circ}C$, respectively. From the result of the study on the effects of metal ions, it was found that $MnCl_2,\;CoCl_2,\;CuCl_2,\;SrCl_2,\;and\;NiCl_2$ slightly increased the enzyme activity, on the other hand $ZnCl_2,\;CaCl_2,\;MgCl_2,\;SLS,\;and\;KMnO_4$ decreased it.

  • In this study, th effective extraction of 5 vegetables, which have been shown to inhibit the growth of the aflatoxin production of Aspergillus parasiticus R-716, was investigated and their effects were examined. Radish, cabbage, garlic and zinger were effectively with water-chloroform, but crown daisy with n-hexane. Among them, water-chloroform extract of radish was remarkably effective, and garlic extract only inhibited the growth strongly. The growth and the aflatoxin production of the strain were showed 0.758g/25ml, 763ug/25ml with the addition of water-chloroform extract equivalent 15g of raw radish, and an increase in the level of radish extract resulted in a decrease both the growth and the aflatoxin production per mycerial weight.
