Effect of Priming on Germination of Aged Soybean Seeds

  • The object of this study is to determine the effect of priming on the germination ability and seedling emergence of aged soybean seeds in lab and field conditions. Artificial or natural procedure for seed aging was applied in this study. One seed lot was artificially aged for 3 to 5 days at 42$^{\circ}C$ with high relative humidity (nearly RH 100%), and the other one was stored at room temperature for 17 months. Aged seeds were osmoconditioned in -1.1 MPa polyethylene glycol 8000 (PEG) solution for 3 days at $25^{\circ}C$ and air-dried. When Danyeobkong was aged for 4 days average germination was 61.5%, however, this improved to 98.5% after the priming treatment. Improvement of seed germination by priming the aged seed was consistent with large seed sized Jangyeobkong cultivar, indicating that the priming was effective in enhancing seed germinability regardless of seed size. Priming aged seeds also resulted in good stand establishment in the field trials. Germination of aged seeds of Danyeobkong without priming was 17.0%, whereas that of primed ones was 66.4%.

  • This study was conducted to research how UCD(Ultra Constant Discharge) lamp lighting affects plant cultivation. To get the results within a short time, lettuce seedling was selected as a plant and nutrient hydroponic beds were used in a dark room for 100% artificial lighting. Two beds were installed for same distance lighting condition with UCD lamp and Hi-pressure Sodium lamp each and another two beds were installed for same light intensity condition with each type of lamp. After 15 days of cultivation, for both conditions, the yields under UCD lamp were weighed more than that under Hi-pressure Sodium lamp. The result can be analyzed that the spectrum of UCD lamp having near sunlight characteristic is much superior to that of Hi-pressure Sodium lamp having a line spectrum characteristic on yellow and red wavelengths for plant cultivation.

  • Jicon scallop (Chlamys farreri), as one of new culture species, has been taken a growing interest in Korea recently. Artificial seeds are needed since seeds in natural waters are absolutely short. To develop the better methods for the artificial spawning and the rearing larvae on a large scale, experiments were carried out from July 1993 to May 1994. Mother shells were collected from the area near Tong-yeong and Geoje-do, the southern part of the Korean Penninsula. It was estimated that a period of 2-years was needed to be mother shells after the fertilization. Generally, spawning occourred intermittently from March to August and the maximum occurred in July. A best way for the artificial spawning was a chemical incentive, a 'serotonin injection' in parallel with the exposure and the temperature stimulation. Densities of the fertilized eggs for the normal development was from 30 to 40 per mililiter. It took 27-days from the fertilization to the settling larva averaging $155{\mu}m$ in size with water temperature 11.5\~13.0^{\circ}C$. Isochrysis galvana and Chaetoceros calcitrans with a density of $1,500\times10^4$ cells per ililiter were supplied for rearing the larvae. A proper density of larvae was 10 individuals per mililiter, and the survival rate of larvae from the flowing water system was much better than that of larvae from the still water.

Protein Synthesis during Somatic Embryo Development and Artificial Seed Germination of Apium graveolens L. after Abscisic Acid or Cold Treatment (쎌러리(Apium graveolens L.)의 체세포배 발생 및 인공종자 발아에 있어서 앱시스산 및 저온처리에 의한 단백질 합성)

  • To understand the molecular mechanism of hardening process in somatic embryo development and artificial seed germination in celery (Apium graveolens L.), the changes of protein synthesis by ABA or cold teatment at early globular stage were examined. Protein content and nitrate reductase activity in ABA- or cold-treated somatic embryo and seedlings were higher than that in unheated ones. The protein content and nitrate reductase activity were more prominent in somatic embryos than in seedlings. From two-dimensional electrophoresis, several protein spots specific to ABA or cold treatment were identified: 30 KD, 32 KD, 171 KD and 205 KD at heart-shaped stage; and 29 KD, 33 KD, 37 KD, 38 KD, 41 KD, 55 KD, 66 KD, and 110 KD at cotyledonary stage were the most specifically synthesized However the synthesis of certain polypeptides were repressed at heart-shaped or cotyledonary stage: 42 KD, 44 KD, 59 KD, 64 KD, 101 KD, 104 KD, and 190 KD at heart-shaped stage; and 29 KD and 116 KD at cotyledonary stage. The protein pattern changes by ABA or cold treatment occurred simultaneously and mainly in acid-soluble proteins during somatic embryo development and artificial seed germination. Therefore it is suggested that the metabolic changes for adaptation to environmental change occur during somatic embryo development and the germination and growth of seedling from embryo.

  • Sedirea japonica is one of the critically endangered species in South Korea mostly due to artificial harms such as illegal collection and habitat destruction. Therefore, artificial propagation through improving germination rate, increasing growth, and controlling flowering is meaningful for the conservation and reintroduction of S. japonica. It is suggested that cytokinins are one of the multi-factors that contribute to plant growth and floral responses. Especially, exogenous cytokinins have been known to induce or promote shoot growth or earlier flowering in orchids. Therefore, it was investigated how the application of 6-benzylaminopurine (BA) influenced the growth and inflorescence of S. japonica. A foliar spray containing BA at 100, 200, 300, and 400 ppm was applied from 1st July to 30th December 2021. Leaf length, leaf length growth rate, leaf width, and width and length ratio were measured as growth-related factors. Visible inflorescence rate, inflorescence length, the number of flowers per inflorescence, and the distance between the stalks were measured as flowering-related factors. Growth-related factors except for leaf growth rate were not affected by BA treatments, while leaf growth rate was significantly increased by 200 ppm of BA treatment. The visible inflorescence rate increased by 200 ppm of BA treatment, and there seems an optimal concentration and threshold of BA treatment. An iterative experiment with more seedlings and measurement factors would be helpful to figure out the effects of exogenous BA treatment on S. japonica, and it can be applied for mass propagation.

Kernel Characteristics and Germination Rate during . the Grain Filling in Super Sweet Corn (초당옥수수 등숙시기에 따른 종실특성 변화와 발아율)

  • The higher sugar and lower starch in super sweet corn may be due to modified endosperm genes sh, bt series, but its seeds have major limiting factors causing low germination and low seedling vigor, This study was conducted to determine what measurable kernel characteristics during the grain tiling period might be more useful as a guide to optimize harvest date for good seed quality in hybrid super sweet corn production. Artificial crossing in super sweet corn hybrid (Chodangok 1) was made on the same day, and ears were harvested from 18 days to 53 days after pollination at weekly intervals. Kernel weight, moisture content, hardness, endosperm rate, seedling height and storage nutrients such as sugars, protein, starch, and germination rate were measured for the kernels at each harvest. Super sweet corn hybrid, Chodangok 1 presented satisfactory germination rate above 70% when harvested on 39 to 46 days after pollination. Its storage nutrients at that time were 23.7-24.2% in starch content 5.5-5.9% in total sugars, 38.9-46.6% in kernel moisture, and 62.7-64.2% in endosperm rate. Germination rate was extremely high when harvested on 39 days after pollination. The black layer of Chodangok 1 could not be used as an indicator for seed harvest. These results suggested that optimum harvest date seems to be 39 days, and kernel moisture and starch content could be used as indications of kernel maturity in deciding when to harvest fer good seed quality in super sweet corn.

Ecological Studies on Lettuce Drop Disease Occurring under Controlled Cultivation Conditions in Drained Paddy Fields (답리작 상치 시설재배지에서의 균핵병 발생생태에 관한 연구)

  • Incidence of lettuce drop was observed throughout the growing season in the vinylhouse at the southern part of Korea, Kimhai. Occurrence of this disease was especially severe at the seedling stage. Number of sclerotia in surface soil $(30\times30\times5cm)$ was 22.0 at the seedling stage, and 5.3 at harvest in the infected area. Temperature for mycelial growth ranged from 5 to $30^{\circ}C$ with optimum temperature at $25^{\circ}C$. Sclerotia were formed fewer at low temperature, but their size was larger resulting in heavier dry weight than that at high temperature. The apothecia were formed from the sclerotia that were buried in March, April and September upto 3cm soil depth, but formed from those buried only 1 em soil depth in October. Sclerotia buried in June and December did not form apothecia regardless of soil depth by 90 days. The sclerotia buried in the 5cm of soil depth did not form apothecia. Sclerotia that were embedded in wet or flooded soil at $25^{\circ}C$ and $30^{\circ}C$ for 5 weeks lost their viability. Infection of lettuce was possible with mycelia originated from sclerotia on autoclaved lettuce plant fragments. The fungus was pathogenic on 25 plant species in 8 families in artificial inoculation tests. Lettuce seedlings appeared to be infected by airborne ascospore originated from sclerotia on crops and weeds around paddy fields, because sclerotia existing in soil might perish under long flood conditions during rice cultivation.

Growth Characteristics and Physiological Adaptation of Pinus densiflora Seedling in the Canopy Gap (소나무 묘목(苗木)의 Gap내 생장(生長) 및 생리적(生理的) 적응과정(適應過程))

  • This study was to investigate the growth characteristics, physiological adaptation of Pinus densiflora(Japanese Red Pine) seedlings at the artificial canopy gap in the Quercus acutissima plantation and to analyze its natural regeneration mechanism. Photosynthetic and transpiration rates were analyzed by different levels of photosynthetically active radiation and by seedling growth. Comparing to seedlings at the open area, those at the canopy gap showed more growth in height than in diameter with different levels of light quality and low light intensity, and the increase rate of dry weight was higher in the aboveground than in the underground, maintaining relatively high T/R rate. The C/F(the ratio of non-photosynthetic organs to photosynthetic organs in dry weight) of the aboveground at the canopy gap was higher than that at the open area by 0.1~0.2, while light saturation and light compensation points at the canopy gap were lower than that at the open area by $300{\mu}mol\;m^{-2}s^{-1}$ and 40%, respectively. The seedlings appeared to have shade tolerance to a certain extent at the young growth stage despite Pinus densiflora is typically classified shade-intolerant species. With light intensity lower than $400{\sim}450{\mu}mol\;m^{-2}s^{-1}$, photosynthetic rate and water use efficiency relatively increased by effective use of light energy.

Use of Pellet or Cube-type Phenolic Foam as an Artificial Medium for Production of Tomato Plug Seedlings (토마토 플러그 묘 생산을 위한 펠릿 및 큐브형 phenolic foam 인공배지의 이용)

  • Growers in plug seedling production think that root media in which rockwool is a component has given rise to several environmental problems. Therefore, the demand for new materials as a substitute for rockwool has been increased. This study examined the possibility of cultivation of tomato plug seedlings using a newly developed growing medium with phenolic foam. Plug seeds of tomato cultivar 'Madison' were sown in four pellet-type growing media: Grodan rockwool (GRW), UR rockwool (URW), phenolic foam LC (LC) or phenolic foam LC-lite (LC-lite). Then, the seedlings were transplanted to the four cube-type growing media 19 days after sowing. Seeds were germinated in a growth chamber ($25{\pm}2^{\circ}C$, 80% relative humidity, and dark) for 4 days and then the seedlings were grown with a nutrient solution supplied by an overhead irrigation system in a greenhouse. Plant height, number of leaves, leaf area, and fresh or dry weight of tomato seedlings were the greatest for the seedlings transplanted to URW cube media after being grown on LC-lite pellets. Root grade was the greatest for the seedlings transplanted on LC or LC-lite cube media after being grown on LC pellets. Chemical properties of all media tested for tomato growth were maintained within a stable range, while physical properties of URW showed high values in container capacity, air space, and total porosity. These results demonstrated that the phenolic foam media were effective for seedling growth and can substitute for rockwool as a root medium.

Species-specific Growth Responses of Betula costata, Fraxinus rhynchophylla, and Quercus variabilis Seedlings to Open-field Artificial Warming (거제수나무, 물푸레나무, 굴참나무 묘목의 실외 인위적 온난화에 대한 수종 특이적 생장 반응)

  • Evaluation of tree responses to temperature elevation is critical for a development of forest management techniques coping with climate change. We conducted a study on the growth responses of Betula costata, Fraxinus rhynchophylla, and Quercus variabilis seedlings to open-field artificial warming. Artificial warming set-up using infra-red heater was built in 2012 and the temperature in warmed plots was regulated to be consistently $3^{\circ}C$ higher than that of control plots. The seeds of three species were sown, and the responses of growth, biomass allocation, and net photosynthetic rate of newly-germinated seedlings on the open-field artificial warming were determined. As a result, the growth responses of the seedlings differed with the species. B. costata showed decreases in the height to diameter ratio (H/D ratio), biomass, root weight to shoot weight ratio, and net photosynthetic rate. However, root collar diameter (RCD), height, biomass, and net photosynthetic rate of Q. variabilis were increased, while the response of F. rhynchophylla was rather obscure. There was no significant difference between warmed and control plots in seedling growth for 3 species in July, whereas, RCD, height, and H/D ratio of Q. variabilis were increased and H/D ratio of B. costata was decreased in November under warming. Species-specific growth responses to warming were similar to the species-specific responses of net photosynthetic rate and biomass allocation; therefore, net photosynthetic rate and biomass allocation might attribute to growth responses to warming. Besides, a relatively obvious response in autumn compared to summer might be affected by the phenological change following artificial warming. Species-specific responses of three deciduous species to warming in this study could be applied to the development of adaptive forest management policies to climate change.
