Production of Dry-Type Artificial Seeds Using Alginate-Encapsulated Rice Somatic Embryos (벼 체세포배를 알긴산 캡슐에 넣어 제작한 건조형 인공종자)

  • Dry-type artificial seeds were produced by dehydrating alginate-encapsulated somatic embryos of lire. When placed on half- strength MS solid medium 20% of the artificial seeds dehydrated to the level of 80% water loss were capable of germination. Addition of 0.1 mg/L ABA to alginate solution before encapsulation enhanced the germination percentage of those dehydrated to the level of nil to 90% water loss by up to 1.7-folds. The results suggest that ABA may enable somatic embryos to overcome physical and/or physiological restraint by encapsulation and dehydration.

  • The unsaturated zone is a significant pathway of the surface contaminant movement and is a highly heterogeneous medium. Therefore, there are limitations in applying conventional convection-dispersion equation(CDE). Artificial neural network(ANN) is considered to be a versatile tool for approximating complex functions. For evaluating the applicability of ANN, numerical tests using ANN were conducted with training set generated by HYDRUS-2D which is based on CDE. The results represent that ANN can estimate the solute transport and the choice of network parameters and generation of training set patterns are important for efficient estimation.

Micro-propagation Factors Essential for Mass Production of Synthetic Seeds in Banana

  • This work described some essential factors necessary for micro-propagation of banana for mass production of synthetic seeds for germ plasm conservation, and how peroxides activity of conserved tissue was influenced. Shoot tips of field grown plants were used to obtain shoot clusters on shoot proliferation medium (MS medium supplemented with 5 mg/l BAP). Using longitudinally-split shoot tip technique, 18720, 8640, 7488, 2016 plantlets were obtained from one shoot tip of Maghraby, Grand Naine, Balady, and Williams, respectively, in six subculture, one month each, on solid medium. Shoot tips excised from in vitro grown plantlets were encapsulated in calcium-alginate beads and stored at $4^{\circ}C$ for one month on half-strength MS basal medium without growth regulators or sugars. After one month all the viable-conserved synseeds formed shoots when they were transferred to MS basal medium, some of them showed synchronous formation of shoot and root systems in one week. Plants retrieved from encapsulated shoot tips were hardened off and transferred to soil.

  • Optimal culture conditions for efficient in vitro propagation and polysaccharide production of Orostachys japonicus were established. The highest growth yield was achieved in 1/2 MS medium, while the lowest growth yield was obtained in 4 MS medium. The patterns of polysaccharide formation were a little similar in all cases, but on MB5 medium, the po]ysaccharide contents of plant were higher than others. Among the modified nitrate levels, effective growth level were obtained in 1/4 N and 1/2 N. High contents of polysaccharide were obtained in 4 N. Different concentration of potassium and calcium did not improve the growth and polysaccharide production. The micropropagated shoots were successfully acclimatized artificial soils.

  • In this paper, we have designed artificial double negative(DNG) transmission line composed of series inter-digital capacitor and two shunt inductive short stubs. This artificial DNG transmission line has the property of double positive (DPS) transmission line over some frequency ranges due to RF nature. In detail, this transmission line simultaneously has the contrary properties of DNG and DPS transmission line depending on operation frequency. DPS/DNG transmission line at leaky region is utilized to design frequency scanning antenna with backfire-to-endfire. We have simulated and measured the dispersion and for-field radiation beam patterns of symmetrical leaky wave antenna. The results show rough agreement.

Effects of altering medium strength and sucrose concentration on in vitro germination and seedling growth of Cypripedium macranthos Sw.

  • The genus Cypripedium, known as lady's slipper orchid, has been considered to have a considerable marketability as a potted and garden plant with beautiful flowers; however, this species is becoming endangered and, in some places extinct, due to habitat destruction and illegal collections. As such, an optimized artificial propagation system is necessary for its conservation and horticultural cultivation. For the establishment of the in vitro proliferation of the endangered Cypripedium macranthos Sw., native to Korea, the effects of medium strength and sucrose concentrations on germination, protocorm formation and seedling growth were investigated through asymbiotic seed culture. The highest germination rate and protocorm formation rate were obtained with a 1/4 MS medium; higher MS medium strengths did not generate the favorable conditions required for germination and protocorm growth. The overall growth characteristics of roots and buds developed from protocorms were best in cultures of 1/4 MS medium. On this medium containing $10g{\cdot}L^{-1}$ sucrose, both the germination rate and protocorm formation rate increased significantly. The general growth properties of seedlings developed on the medium with $10g{\cdot}L^{-1}$ sucrose were best, showing the highest bud formation rate and root number. Our results demonstrate that the 1/4 MS medium, supplemented with $10g{\cdot}L^{-1}$ sucrose, could improve in vitro germination, and facilitate the growth of seedlings developed from the protocorms of C. macranthos Sw.

  • Purpose: This study was performed to evaluate the feasibility of visualizing soft tissue lesions and vascular structures using contrast-enhanced cone-beam computed tomography (CE-CBCT) after the intravenous administration of a contrast medium in an animal model. Materials and Methods: CBCT was performed on six rabbits after a contrast medium was administered using an injection dose of 2 mL/kg body weight and an injection rate of 1 mL/s via the ear vein or femoral vein under general anesthesia. Artificial soft tissue lesions were created through the transplantation of autologous fatty tissue into the salivary gland. Volume rendering reconstruction, maximum intensity projection, and multiplanar reconstruction images were reconstructed and evaluated in order to visualize soft tissue contrast and vascular structures. Results: The contrast enhancement of soft tissue was possible using all contrast medium injection parameters. An adequate contrast medium injection parameter for facilitating effective CE-CBCT was a 5-mL injection before exposure combined with a continuous 5-mL injection during scanning. Artificial soft tissue lesions were successfully created in the animals. The CE-CBCT images demonstrated adequate opacification of the soft tissues and vascular structures. Conclusion: Despite limited soft tissue resolution, the opacification of vascular structures was observed and artificial soft tissue lesions were visualized with sufficient contrast to the surrounding structures. The vascular structures and soft tissue lesions appeared well delineated in the CE-CBCT images, which was probably due to the superior spatial resolution of CE-CBCT compared to other techniques, such as multislice computed tomography.

  • In this paper case studies of artificial ground freezing, which have not been applied in Korea, have been investigated for the water cut-off in a subsea tunnel under high water pressure and the most commonly used cooling mediums of brine and liquid nitrogen are examined. Since sea water with pressure has the lower freezing point than pure water, the lower temperature cooling medium is required in the application of subsea tunnel. Also, the cooling medium must have refrigeration safety and is able to reduce executing time. Brine freezing system can reuse cooling medium and is safer than liquid nitrogen freezing. But it takes more time to freeze ground and needs complex circulation plants. On the other hand, liquid nitrogen freezing system can't recycle cooling medium and may cause breathing problems or asphyxiation through oxygen deficiency. But, freezing with liquid nitrogen is fast and requires simple refrigeration equipment. Principal elements of design for ground freezing in subsea tunnel have been extracted and these elements are needed further research.

Contrast Media Side Effects Prediction Study using Artificial Intelligence Technique (인공지능 기법을 이용한 조영제 부작용 예측 연구)

  • The purpose of this study is to analyze the factors affecting the classification of the severity of contrast media side effects based on the patient's body information using artificial intelligence techniques to be used as basic data to reduce the degree of contrast medium side effects. The data used in this study were 606 examiners who had no contrast medium side effects in the past history survey among 1,235 cases of contrast medium side effects among 58,000 CT scans performed at a general hospital in Seoul. The total data is 606, of which 70% was used as a training set and the remaining 30% was used as a test set for validation. Age, BMI(Body Mass Index), GFR(Glomerular Filtration Rate), BUN(Blood Urea Nitrogen), GGT(Gamma Glutamyl Transgerase), AST(Aspartate Amino Transferase,), and ALT(Alanine Amiono Transferase) features were used as independent variables, and contrast media severity was used as a target variable. AUC(Area under curve), CA(Classification Accuracy), F1, Precision, and Recall were identified through AdaBoost, Tree, Neural network, SVM, and Random foest algorithm. AdaBoost and Random Forest show the highest evaluation index in the classification prediction algorithm. The largest factors in the predictions of all models were GFR, BMI, and GGT. It was found that the difference in the amount of contrast media injected according to renal filtration function and obesity, and the presence or absence of metabolic syndrome affected the severity of contrast medium side effects.

Construction of Artificial Biosynthetic Pathways for Resveratrol Glucoside Derivatives

  • Resveratrol, which is a polyphenolic antioxidant, is dose-dependent when used to provide health benefits, to enhance stress resistance, and to extend lifespans. However, even though resveratrol has therapeutic benefits, its clinical therapeutic effect is limited owing to its low oral bioavailability. An Escherichia coli system was developed that contains an artificial biosynthetic pathway that produces resveratrol glucoside derivatives, such as resveratrol-3-Oglucoside (piceid) and resveratrol-4'-O-glucoside (resveratroloside), from simple carbon sources. This artificial biosynthetic pathway contains a glycosyltransferase addition (YjiC from Bacillus) with resveratrol biosynthetic genes. The produced glucoside compounds were verified through the presence of a product peak(s) and also through LC/MS analyses. The strategy used in this research demonstrates the first harnessing of E. coli for de novo synthesis of resveratrol glucoside derivatives from a simple sugar medium.
