Cultivation of Nostoc flagelliforme on Solid Medium

  • In order to construct an artificial cultivation of Nostoc flagelliforme on solid medium, we attempted to assess the viability of approaches, which utilized either BG-11 agar or sand medium using both sterile and non-sterile algal segments. In the trial in which the BG -11 agar medium was inoculated with the non-sterile algal segments, the algae exhibited the rapid growth in the initial 4 days of cultivation. However, after 4 days of cultivation, the growth rate of the algae slowed, and the algal growth was completely stopped by 7 days of cultivation. When the BG -11 medium was inoculated with the sterile algal segments, the algae exhibited the rapid growth for a longer period of 8 days, reaching a length of 24.9 mm. The growth rate during this period was measured to be $24.5\%$. After the 8 days of cultivation, the algal growth rate began to slow and had almost stopped by the 13 days of cultivation. On the other hand, when the sterile algal segments were inoculated onto a sand plate, the algal segments decomposed, reaching total decomposition after 11 days of cultivation. By way of contrast, the desiccation treatment samples continued to grow for 14 days of cultivation. After 14 days of cultivation, the algae achieved a length of 26.1 mm, with a growth rate of $30.6\%$. Our results indicate that periodic desiccation may constitute an effective strategy for the prevention of algal decomposition.

Production of Pectolytic enzymes by Alternaria mali Roberts (Alternaria mali Roberts에 의(依)한 Pectin질(質) 분해효소(分解酵素)의 생산(生産))

  • Isoates with changed pathogenicity were selected among iprodione-resistant Alternaria mali to investigate any relationship between their pectolytie enzyme activity and pathogenicity. In artificial medium, strongly pathogenic isolates $S_1$, and $R_3$ showed higher enzyme activity than weakly pathogenic isolate $R_8$.Activity of endo-polymethylgalacturonase and end-o-polygalacturonase was more than 3 times. But activity of pectin methylesterase and pectin Iyase by isolaste $S_1$ was higher than those by $R_3$ and $R_8$ isolates. In apple medium dialyzed against distilled water, activity of enzyme by each isolate was increased but growth of each isolate was reduced. When iprodione was added to the medium, enzyme activity and growth of isolate $S_1$, were reduced but strongly pathogenic isolate .$R_3$ among iprodione-resistant ones showed increased enzyme activity except for exo-polygalacturonase in dialyzed apple medium.

Plantlet Regeneration and PLBs Propagation of Bulbophyllum auricomum Lindl.

  • Bulbophyllum auricomum Lindl. is very popular among orchid growers due to the attractive fragrance of its flowers and has become an endangered orchid in Myanmar. In this study, we carried out an aseptic technique that can be used to conserve this endangered orchid species. The seeds of B. auricomum Lindl. were obtained from artificial pollination and cultured in MS basal medium for seed germination. The effect of coconut water and BAP in MS basal medium on callus induction was investigated. The highest callus induction was found at 2.0 mg/L BAP. The maximum growth of protocorm-like bodies (PLBs) was evaluated, and the best response was observed on MS medium supplemented with 150 mL/L coconut water at pH 5.6. MS basal medium supplemented with 150 mL/L coconut water along with 2.0 mg/L BAP and 1.0 mg/L NAA (MCBN) showed the highest number of plantlets at 15℃ at the second week of culture. At the second and third week of culture, MS medium supplemented with 2.0 mg/L BAP and 1.0 mg/L NAA (MBN) showed the best result in terms of the number of leaves and the longest leaves at 15℃ and 25℃, respectively. The present study showed evidence of successful in vitro propagation of B. auricomum Lindl.

Micropropagation of Juvenile and mature Trees of Sawtooth Oak (Quercus acutissima C.) (상수리나무 유목(幼木)과 성숙목(成熟木)의 기내번식(器內繁殖))

  • Present study describes a method on the application of efficient tissue culture systems for the micro-propagation of juvenile and mature sawtooth oak(Quercus acutissima). Nodal segments with axillary buds were used as initial explant sources. WPM(Woody Plant Medium) was the best in growth and proliferation of shoot among the media tested. Although the single effect of zeatin revealed on two dorminant shoot elongation with normal growth until the elevation of levels up to 3.0mg/l, BAP($N^6$-benzyl amino purine) usually showed better response than zeatin on shoot multiplication and/or elongation. In addition, the incorporation of BAP and zeatin onto the culture media represents more effectiveness in shoot proliferation and its growth. Optimum concentrations of BAP and zeatin were 0.5 and 0.05~1.0mg/l, respectively. Ninety percent of the proliferated shoots was rooted on half-strength GD (Gresshoff and Doy, 1972) medium containing 0.5mg/l IBA(indole butyric acid) in 4 weeks after culture. More than 70% of the rooted plantlets survived after 5 months of transplanting into artificial soil mix containing equal amount of peatmoss and perlite. Among 27 plus tree clones which were grafted twice onto the juvenile rootstocks, only 4 clones revealed the possibility for shoot multiplication through tissue culture system. The capacity for the micropropagation using mature explant sources was highly depended on clonal differences compared with those of octet age. More than 90% of rooting ratio was obtained from the best responding clone. Among the 7 rooting media tested, GD medium was the best far rooting. The most effective rooting was obtained on half-strength GD medium containing 0.2 to 2.0mg/l IBA. More than 60% of rooted plantlets survived after 5 months of transplanting into the artificial soil mix.

  • Ambient Air Vaporizer (AAV) is an essential facility in the process of generating natural gas that uses air in the atmosphere as a medium for heat exchange to vaporize liquid natural gas into gas-state gas. AAV is more economical and eco-friendly in that it uses less energy compared to the previously used Submerged vaporizer (SMV) and Open-rack vaporizer (ORV). However, AAV is not often applied to actual processes because it is heavily affected by external environments such as atmospheric temperature and humidity. With insufficient operational experience and facility operations that rely on the intuition of the operator, the actual operation of AAV is very inefficient. To address these challenges, this paper proposes an artificial intelligence-based model that can intelligent AAV operations based on operational big data. The proposed artificial intelligence model is used deep neural networks, and the superiority of the artificial intelligence model is verified through multiple regression analysis and comparison. In this paper, the proposed model simulates based on data collected from real-world processes and compared to existing data, showing a 48.8% decrease in power usage compared to previous data. The techniques proposed in this paper can be used to improve the energy efficiency of the current natural gas generation process, and can be applied to other processes in the future.

Exploring how to use virtual reality for elementary school students (초등학생 대상 가상현실 활용방안 탐색)

  • The purpose of this study is to analyze elementary school students' interest in virtual reality(VR) technology, usability, and the possibility of learning media. In particular, it is intended to be used for content creation for artificial intelligence(AI) education in the future. The effectiveness of elementary education using virtual reality technology was confirmed through the analysis of overseas research, and the applicability to elementary school students in Korea was analyzed. To proceed with the analysis, various virtual reality contents were provided to 5th grader of elementary school, and then, interest, usability, usefulness, and possibility of use in class and learning were surveyed. As a result of the study, it was confirmed that students' interest in virtual reality contents was very high, and that it could be used sufficiently as a learning medium. It suggests that it can be used in artificial intelligence education and data science education, which have recently been emphasized in importance. In particular, virtual reality can be used to simulate abstract data and artificial intelligence.

Physiological and Genetic Characteristics of Cultivated Mushroom, Hypsizygus marmoreus

  • A edible mushroom, Hypsizygus marmoreus is commercially cultivated in Northeast Asia. Japan's annual production is 110,000ton or more. Since 2002, cultivation is expanded in Korea. To investigate the morphological, cultural and microscopic characteristics of Hypsizygus marmoreus, 109 isolates were collected from Korea and other countries. Clamp connection, chlamydospore and arthrospore were present in all tested isolates of H. marmoreus except HYM-002 and HYM-004. Also pilealtrama, gilltrama, basidia, basidiospore and cystidia of fruiting body were no difference among the isolates in the present investigation. Morphological characteristics of fruiting body was that color of pileus was brown and white, irregular as marble, the average size 12~22mm and stipes was $46{\sim}91{\times}6{\sim}10mm$. Isolates HYM-031, HYM-047 and HYM-109 formed grayish-brown pileus with a faint pattern. Molecular analysis with RAPD and ITS rDNA sequence analysis were also performed to check the genetic relationships among H. marmoreus isolates. Based on the RAPD analysis using the URP-PCR, all isolates of H. marmoreus were clustered into large 3 groups but more than 90% showed high similarity. In addition, morphological and geographical differences have been classified as an independent cluster. The brown and white strains enclosed in same cluster. So genetically no significance difference was observed between these two strains. ITS gene sequences of 16 selected isolates which were 640 bp long, were aligned and compared. The similarity in ITS sequence was 94.8 to 99.1% among tested isolates and the H. marmoreus isolates in GeneBank. In conclusion the tested isolates were H. marmoreus. Morphological and molecular observations proved that all tested isolates were belonging to H. marmoreus. For the stable artificial cultivation, composition of optimum media, mature period and light condition were established. Optimal formula of artificial cultivation medium was Douglas sawdust: corn cob: soybean meal: wheat bran = 40:30:15:15. In addition, 7% rice bran and 3% yellow sucrose was the most effective composition for spawn's liquid medium. For the maturation of the isolates was favorable for growing for 20 to 30 days at $25^{\circ}C$ and the LED lights in mixture of white and blue was good for growth period. For effective growth, the temperature, humidity and aeration control in every step was important.

  • Present experiments were conducted to determine the possibility of cultivation of 9 edible wild species selected among the higher fungi growing in Korea. In the investigation on the mycelial growth according t6o the different media, the mycelial growth of Coprinus comatus was fast on the CSA medium, when malt extract was added to the basal medium PSA, the mycelial of Lapista nuda and Auricularia auricula-judae was fast in growth and density. In the spawning, the mycelial growth of Pholiota squarrosa on the oak tree's sawdust, Pleurotus cornucopiae on the broad-leaves' sawdust, and Coprinus comatus on the compost was respectably fast and also it shown to be possibility of artificial cultivation owing to their carporphore budding when Coprinus comatus and Lepiota alborubescens cultivated on the rice straw, Auricularia auriculajudae and Pleurotus cornucopiae on the sawdust of the popla and Pholiota squarrosaon the sawdust of the oak tree.
