A study on the utilization of potato, sweet potato, and jelusalem artichoke flour as an extender of urea formaldehyde resin for plywood (고구마, 감자 및 돼지감자 분말(粉末)을 이용(利用)한 합판용(合板用) 요소수지(尿素樹脂)의 증량(增量)에 관(關)한 연구(硏究))

  • This study was carried out to utilize sweet potato, and jerusalem artichoke flour as an extender of urea formaldehyde resin for plywood manufacture. The results obtained are summarized as follows: 1) In dry and wet test of plywood, sweet potato flour extended in urea formaldehyde resin was shown better strength than those of wheat flour. 2) Sweet potato flour was shown good results up to 200 percent extension in the dry test, and there was not shown significant difference between 100 percent and 150 percent extension in the wet test. 3) In the case of 100 percent extension, potato flour was shown best dry shear strength(400 psi) and sweet potato flour was shown best wet shear strength (212 psi).

  • Various Saccharomyces cerevisiae strains were transformed with a 2 ${\mu}$-based multicopy expression plasmid, pYIGP, carrying Kluyveromyces marxianus inulinase gene under the control of GAPDH promoter. Among then two strains, SEY2102 and 2805, showed high levels of cell growth and inulinase expression, and were selected to study their fermentation properties on inulin. Jerusalem artichoke inulin was more effective for cell growth (10∼11 g-dry wt./L at 48 hr) and inulinase expression (1.0 units/mL with SEY2102/pYIGP and 2.5 units/mL with 2805/pYIGP) than other inulin sources such as dahlia and chicory. It was also found that maximal ethanol production of 9 g/L was obtained from Jerusalem artichoke inulin at the early stationary phase (around 30 hr), indicating that recombinant S. cerevisiae cells secreting exoinulinase could be used for the simultaneous saccharification of inulin and ethanol fermentation.

  • High-dose extracts from artichoke leaves are traditionally used for treatment of stress related disorder, that is, hepatic disease, dyspeptic disorder, hyperlipidemic disorder and diuretic disorder. The aim of this study was to investigate anti-stress effects of Artichoke extract(Archoke juice produced from Choa company). The experiments were performed with the use of young(6-8 weeks of age) male mice of ICR stain weighing between 20 and 25 g at the time of first treatment with Archoke juice. The were grouped normal, control, Ginseng, diazepam and Arcoke juice group. (omitted)

  • Polyphenol oxidase from Jerusalem artichoke(Helianthus tuberosus L.) tubers was partially purified by precipitation with ammonium sulfate, followed by gel filtration on Sephadex G-100. The enzyme showed maximal activity at pH 6.5 and $4^{\circ}C$. Kinetic studies indicated $K_{m}$ value of 3 mM for catechol and activation energy of 72.6 kcal/mole. As for substrate specificity of polyphenol oxidase the enzyme showed high affinity towards diphenol compounds, but not towards monophenols. The enzamatic browning was completely inhibited at 1 mM concentration of L-ascorbic acid, sodium hydrosulfite and L-cystein(HCl). The activity of polyphenol oxidase in 0.1 M potassium phosphate buffer(pH 6.5) was fairly stable for a week at $4^{\circ}C$, while it decreased remarkably at $25^{\circ}C$.

Production of Inulase Using Jerusalem Artichoke Tuber Extract (돼지감자 추출물을 이용한 이눌라아제 생산)

  • To produce inulase from Kluyveromyces fragilis No.351 using Jerusalem artichoke tuber extract, optimization of the condition was conducted. As results, the optimal concentration of artichoke tuber extract was 3.5 % and bactocasitone showed better production than yeast extract. The optimal temperature and pH were $30^{\circ}C$ and 5.5, respectively. The addition of $NH_4H_2PO_4$ increased the enzyme production. Inulase synthesis was growth-associated and the enzyme production increased as concentration of dissolved oxygen increased. A higher quantity of industrial grade inulase was prepared by the combination of ultrafiltration and ethanol precipitation with 72% recovery.

  • Fructooligosaccharide-inulin(FOSI) separated from Jerusalem artichoke-autolysate was tested to determine its effect on the growth of fecal microorganisms of pig. Total microorganisms in fecal samples averaged $10^{9.83}$ per g of wet feces and the numbers of predominant Bacteroidaceae and Peptococcaceae were $10^{9.3}\;and\;10^{9.2}$, respectively. Lactobacilli, Eubacteria, Clostridia were found out to be the next common bacteria. The addition of FOSI to the 'feces media' and PYF broth increased the numbers of total microorganisms and lactobacillis up to those of glucose-addition media. The number of Bifidobacteria was greater about $50{\sim}500$ times on FOSI-addition media rather than on glucose-addition media. While FOSI showed no different effect on the Clostridia growth compared with glucose, both sugars reduced the number of E. coli to $10^{-1}{\sim}10^{-3}$ level of no sugar media.

Synergistic Effects of Jerusalem Artichoke in Combination with Pegylated Interferon Alfa-2a and Ribavirin Against Hepatic Fibrosis in Rats

  • Background: Complementary and alternative medicine has been highly appreciated as a supportive regimen for classical treatment strategies. Here we offer a nutrition-based adjuvant therapy for liver fibrosis, a major risk factor for cirrhosis and hepatocellular carcinoma. Aim of the study: To evaluate the possible hepatoprotective effects of Jerusalem artichoke tubers (JAT) in combination with interferon and ribavirin. Materials and Methods: Twelve groups of rats were administered JAT, interferon and ribavirin either separately or in combination from day one of $CCL_4$ administration until the end of the study. Animals were killed after 8 weeks of $CCL_4$-induced hepatotoxicity. Results: Hepatocytes from rats treated with triple combination of interferon, ribavirin, and JAT showed more less normal architecture compared to $CCL_4$-treated rats. We also detected significantly higher hepatic protein expression levels of p53, BAX and transforming growth factor-${\beta}$ (TGF-${\beta}$) in the $CCL_4$-intoxicated group compared to normal controls, as evidenced by immunohistochemical staining and western blotting analyses. Addition of JAT as a supportive regimen improved response to ribavirin and interferon and effectively participated in retaining normal histopathological and biochemical criteria and significantly lowered protein expression of p53, BAX, and TGF-${\beta}$. Conclusions: We suggest that addition of JAT as a supportive r egimen to interferon and ribavirin effectively potentiates their anti-fibrotic effects.

Quality Characteristics of Rice Nutritional Bars Containing Different Levels of Chinese Artichoke (Stachys sieboldii Miq.) Powder (초석잠(Stachys sieboldii Miq.) 분말의 첨가량을 달리한 쌀 영양바의 품질특성)

  • Purpose: The aim of this study was to evaluate the quality characteristics of rice nutritional bars prepared by adding Chinese artichoke (CA) powder to the rice powder. Methods: CA rice nutritional bars were prepared with various levels of CA powder (0%, 5%, 10%, 15%, and 20%) and designated as CA0 (without CA powder), CA5, CA10, CA15, and CA20, respectively. The total phenolic content, DPPH free radical scavenging activity, moisture content, pH, baking loss rate, leavening rate, volume, color, texture analysis, and sensory evaluations of rice nutritional bars were performed. Results: The total phenolic content and DPPH free radical scavenging activity exhibited an increase with increase in the substitute amount of CA powder. The results showed that the pH of rice nutritional bars decreased in all sample groups as the content of CA powder increased. The volume of the sample group with CA powder was higher than that of CA0. As the content of CA powder increased, lightness of the rice nutritional bars crumbs decreased while redness and yellowness increased. The texture analysis of rice nutritional bars showed that hardness, gumminess, and chewiness were the highest in CA0 while springiness and cohesiveness were the lowest in CA0. With respect to characteristic intensity rating, the taste, flavor, and softness of rice nutritional bars increased with increasing amounts of CA powder. In terms of consumer acceptability, CA15 and CA20 received the highest scores with respect to texture. Conclusion: Therefore, the results of this study indicate that replacing 15% of the rice powder with CA led to an efficient enhancement in the antioxidant and nutritional values of rice nutritional bars as well as the sensory quality.

Selection of Yeast Strains for Alcohol Production from Jerusalem Artichoke Tubers (Helianthus tuberosus L.) (돼지감자(Helianthus tuberosus L.)로 부터의 알콜 생산을 위한 균주 선발)

  • To investigate the possibility of ethanol production from jerusalem artichoke tubers (Helianthus tuberosus L.), various yeast strains were evaluated for their potential in metabolizing carbohydrate from jerusalem artichoke tubers to ethanol. Among them, Kluyveromyces fragilis CBS 1555 showed the highest inulase activity and ethanol fermentability. On the batch kinetic analysis, K. fragilis also showed the highest in parameters for ethanol production and substrate utilization, although lower than Saccharomyces cerevisiae Y-10 in cell mass yield and ethanol production yield.
