안전한 닭고기 생산을 위한 고병원성 조류인플루엔자 A/H5N1에 항바이러스 효과를 가진 천연 사료첨가제의 탐색 (Screening of a Natural Feed Additive Having Anti-viral Activity against Influenza A/H5N1)

  • To search for anit-H5N1 influenza virus agent, the anti-viral activity of methanol and aqueous extracts from thirty medicinal plants were examined in this study. The plant material (30 g) was extracted with methanol (300 mL) for 24 hr at room temperature. Methanol extracts were filtered and evaporated, then freeze-dried. Aqueous extracts were prepared with dried plant material (30 g) and hot distilled water (300 mL). After 3 hr, the aqueous extracts were filtered and evaporated, then lyophilized. Extracts prepared from different plants were tested the antiviral activity against influenza viruses [A/vietnam/1194/04 (H5N1)-NIBRG-14] using the hemagglutination (HA) assay. Among the test plants, Asarum sieboldii was found to be a potent inhibitor of H5N1 influenza virus in MDCK cell culture. Virus titers were 7 log, whereas with methanol extract of Asarum sieboldii for 48 hr titers were 3 log, indicating that methanol extract of Asarum sieboldii inhibited the H5N1 influenza viruses from the infected cells.

산수유 추출물 및 분획물의 항산화 활성과 활성성분 분석 (Antioxidative Activity and Active Compound Analysis of the Extract and Fractions of Corni Fructus)

  • In this study, analysis of active compounds that are believed to be highly relevant to antioxidant activity was carried out on the methanol extract and its solvent fractions of Corni fructus. The DPPH radical scavenging activity for the comparison of antioxidant activity was higher in order of aqueous fraction > methanol extract > ethyl acetate fraction > n-hexane fraction. It is similar to the order of total polyphenol contents in the samples. As a result of LC-MS analysis, phenolic acid compounds such as caffeic acid, gallic acid and chlorogenic acid and lognin, which is known as a representative active ingredient of Corni fructus, were identified as active compounds. And the antioxidative activity and the total polyphenol content of the extracts and solvent fractions were found to be related to the contents of the compounds. Particularly, it was confirmed that phenolic acid such as caffeic acid contributes to the antioxidative activity of the aqueous fraction of Corni fructus methanol extract.

  • For the fast dry of the aqueous tape, the process which water was replaced by organic solvent was proposed. So-called, it was the solvent washing dry, Three organic solvents (methanol, ethanol, and acetone) were selected for the washing solvent. The weight loss of the washed tapes was measured to evaluate the dry rate of the tapes and dried tapes were examined the generation of the cracks with the variations of the organic solvent and the washing time. Methanol, ethanol, and acetone were all available organic solvents for this method. The tapes washed in methanol, ethanol, and acetone were dried rapidly for twenty minutes. After thirty minutes, the weight losses were not any more. The solvent of the lower surface tension can decreases the crack of dried tape. If solvent substitutes water completely, though it was fast dried, crack can be eliminated. The tape casting system was designed for the solvent washing dry and prepared. An homogeneous tape was established by continuous tape casting process.

  • Rate of solvolysis of dansyl chloride in aqueous binary mixtures of acetone, methanol and ethanol are reported. Kinetic solvent isotope effects in methanol and product selectivities in alcohol-water mixtures are also reported. Kinetic data are interpreted with the Grunwald-Winstein and Kivinen equations. The value of $k_{CH3OH}/k_{CH3OD}=1.76$ suggests that a general have catalyzed and/or an $S_AN$ pathway is operative in methanol, a less polar solvent. Rate-rate profiles for solvolysis of dansyl chloride in the aqueous binary media indicate a change in reaction channel from $S_AN$ (in less polar media) to $S_N2$ (in more polar media) mechanism.

Ellagitannins from Bark of Juglans mandshurica

  • Juglans mandshurica bark was collected and extracted with 70% aqueous acetone. The extracts were concentrated and then sequentially fractionated using n-hexane, $CH_2Cl_2$, EtOAc, and $H_2O$ to be freeze dried. A portion of $H_2O$ fraction was chromatographed on a Sephadex LH-20 column using aqueous methanol to isolate (S)-2,3-HHDP-D-glucopyranoside (1). Some of EtOAc fraction was also treated on a Sephadex LH-20 column using aqueous methanol and EtOH-hexane mixture to purify pedunculagin (2). The structures of these two epimeric ellagitannins were elucidated by spectroscopic tools, NMR and MS.

Phytotoxic Effects of Xanthium occidentale Extracts and Residues on Seedling Growth of Several Plant Species

  • Compositae plants are known to contain biologically active substances that are allelopathic to agricultural crops as well as weed species. Aqueous or methanol extracts and plant residues from leaves of Xanthium occidentale were assayed against alfalfa (Medicago sativa) to determine its allelopathic effects, and the results showed that the extracts applied onto filter paper significantly inhibited seed germination as well as root growth of alfalfa. Aqueous leaf extracts of $40g\;L^{-1}$ were completely inhibitory on root growth of alfalfa, while root growths of barley (Hordeum vulgare L.) and soybean (Glycine max L.) were less sensitive. Leaf residue incorporation at $100g\;kg^{-1}$ into soil on seedling growth of barnyard grass (Echinochloa crus-galli Beauv. var. oryzicola Ohwi) inhibited both shoot and root fresh weights of barnyard grass by 94 and $96\%$, respectively. Methanol extracts from BuOn and EtOAc fractions of X. occidentale reduced alfalfa root growth more than did those from hexane and water fractions. The results based on bioassay of extracts and residues show that X. occidentale had potent an allelopathic activity against other plant species.

Stoichiometric Solvation Effects. Solvolysis of Methanesulfonyl Chloride

  • Solvolyses of methanesulfonyl chloride in water, $D^2O$, $CH^3OD$, and in aqueous binary mixtures of acetone, eth-anol and methanol are investigated at 25, 35 and $45^{\circ}C.$ The Grunwald-Winstein plot of first-order rate con-stants for the solvolytic react ion of methanesulfonyl chloride with YCl (based on 2-adamantyl chloride) shows marked dispersions into three separate lines for three aqueous mixtures with a small m value (m < 0.30), and shows a rate maximum for aqueous alcoholic solvents. Stoichiometric third-order rate constants, kww and kaa were calculated from the observed first-order rate constants and (kaw + kwa) was calculated from the kww and kaa values. The kinetic solvent isotope effects determined in water and methanol are consistent with the proposed mechanism of the general base catalyzed and/or SAN/SN2 reaction mechanism for methanesulfonyl chloride solvolyses based on mass law and stoichiometric solvation effect studies.

Purification of Isoflavone from Soybean Hypocotyls using Various Resins

  • Isoflavone was extracted with various concentration of aqueous methanol using whole hypocotyls as the starting material. Whole hypocotyls were preferred as the raw material because the residue could be easily removed from the solvent after the extraction process. Extraction yield was almost constant at the methanol concentration of $20-80\%$. Most of the isoflavone was extracted within 1 hr, and the extraction yield remained almost constant thereafter. When the concentration of methanol was $80\%$, the content of total solid was reduced due to the reduced extraction of contaminating protein as the result of protein insolubilization. Among resins tested, Diaion HP-20, Amberlite XAD-16, and Amberlite IRC-50 showed the highest capacity to absorb the compound. Open column chromatography with Diaion HP-20 showed that $80\%$ aqueous ethanol was most efficient as the eluting solvent with final recovery of the phytochemical being more than $95\%$. Maximum adsorption of the phytochemical occurred at the acidic pH 2-4. When the spatial velocity was increased to 15 and more, the degree of adsorption was decreased, whereas below spatial velocity of 15, the adsorption capacity of isoflavone to the resin was almost constant. The purity of the isoflavone purified by column chromatography was $78\%$.

향나무 심재 메탄올추출물의 염색성과 기능성 (Dyeing Properties and Functionality of Methanol Extract from Juniperus chinensis Heartwood)

  • In this study, we report dyeing properties and functionality of cotton and silk fabrics dyed with methanol extract from Juniperus chinensis heartwood. Non-aqueous dye of Juniperus chinensis heartwood extracted with methanol was expressed reddish color. It showed relatively good affinity to silk than cotton. Dye uptake of cotton and silk continued to increase according to the increase of the dye concentration. Cotton was not affected by the dyeing temperature and time, however, dye uptake of silk was increased as the dyeing time and temperature were increased. Dye uptake of cotton and silk were the highest at pH 8 and pH 6 respectively. Dyed fabrics showed various colors depending on mordant types and mordant concentration. Washing, light and perspiration fastness were not good. However, rubbing and dry cleaning fastness showed relatively good grade. Dyed fabrics of ultraviolet-cut ability and deodorant ability were improved. Also, dyed fabrics showed very good antimicrobial abilities of 99.9%.

사진유제용 증감색소의 분광증감과 사진특성 (The Spectral Sensitization and the Photographic Characteristics of Sensitizing Dye for Photographic Emulsion)

  • In this study, benzoxazolo carbocyanine dye was used as sensitizer for photographic emulsion, and the photographic characteristics were examined. The basic properties of sensitizer such as stability in various solvents were examined. The sensitizer was very stable in methanol, acetonitrile, acetone, dimethylformamide, and chloroform solution. Absorption spectra of benzoxazolo carbocyanine dye $2.5{\times}10^{-6}M$ and $5{\times}10^{-6}M$ concentrations in 10% aqueous methanol solutions containing $10^{-2}M$ potassium chloride show the monomer-J-aggregation characteristics. Compared to the absorption peak of the monomer in pure methanol solution, the red shifts of the monomer-J-aggregate peaks of benzoxazolo carbocyanine dye of $2.5{\times}10^{-6}M$ and $5{\times}10^{-6}M$ concentrations in 10% aqueous methanol solutions containing $10^{-2}M$ potassium chloride are 34nm respectively, and the sensitizing peak of benzoxazolo carbocyanine dye in photographic emulsion has red shift of 34nm. Therefore, if was concluded that benzoxazolo carbocyanine dye can be used as green sensitizing dye for the spectral sensitization of photographic emulsion.
