수면박탈로 유도한 Hippocampus Dentate gyrus의 산화 스트레스에 대한 백합, 연자육의 신경세포보호효과 (Neuroprotective Effect of Lilii bulbus, Nelumbins semen on the Sleep Deprivation-induced Oxidative Stress in the Hippocampus Dentate Gyrus)

  • Sleep deprivation is an extremely common event in today's society. It has caused learning cognitive skill deterioration and poor concentration, increased disease such as heart disease, diabetes and obesity, sexual function decrease, infertility increase, depression and autonomic nervous system disorder. Sleep deprivation-induced stress caused NADPH oxidase and oxidative stress. And this oxidative stress induces apoptosis. Lilii bulbus and Nelumbins semen are known to mental and physical relaxation effects. In this study, we induced sleep deprivation(SD) in Sprague-Dawley rats in water for 5 days and thereafter administered orally L. bulbus and N. semen for 5 days. Brain tissues were observed by histochemical, immunohistochemical and tunel staining. The immunoreactives of Tumor necrosis factor ${\alpha}$, Neuronal nitric oxide synthases, Phospho-SAPK/JNK and gp91-phox of the L. bulbus administered group and N. semen administered group were weaker than those of sleep deprivation group. In the L. bulbus administered group and N. semen administered group, apoptosis was decreased than that of sleep deprivation group. Proapoptotic p53, Bax, Cleaved caspase 3 immunoreactives of the administered group were weaker than those of sleep deprivation group, whereas anti-apoptotic Bcl-2 immunoreactity was stronger in the L. bulbus administered group and N. semen administered group. Antioxidant mechanism such as DJ-1, superoxide dismutase 1, Nuclear factor-like 2 immunoreactives of the L. bulbus and N. semen administered group were stronger than those of sleep deprivation group. These results demonstrate that L. bulbus, N. semen had the neuroprotective effects on the sleep deprivation-induced oxidative stress in the hippocampus.

Cisplatin 유도 급성신부전에서 Klotho 단백질의 발현 (Localization of Klotho in cisplatin induced acute kidney failure)

  • Klotho deficiency is an early event in acute kidney injury (AKI) that exacerbates acute kidney damage. The present study explored the expression of Klotho and inflammation related factors in cisplatin-induced AKI. Rats (n = 18) were treated with cisplatin intraperitoneal injection (5 mg/kg) or left untreated as controls (n = 6), then sacrificed at 5 (n = 6) and 10 days (n = 6) treatment. Five days after cisplatin injection, the serum kidney enzymes and kidney cell apoptosis were significantly increased. Moreover, the expression of Klotho was decreased when compared to the control group, especially in the cortex and outer medulla regions. In contrast, inflammation related signals including nuclear factor kappa B, tumor necrosis factor-${\alpha}$, and tumor necrosis factor-like weak inducer of apoptosis were enhanced. However, 10 days after cisplatin injection, Klotho expression was enhanced upon both IHC and Western blot analysis, with slightly recovered renal function and decreased apoptosis. Furthermore, inflammation related signals expression was decreased relative to the 5 days group. Overall, this study confirmed the opposite expression patterns between Klotho and inflammation related signals and their localization in cisplatin-induced AKI kidney.

Cell Cycle Arrest and Cytochrome c-mediated Apoptotic Induction in A549 Human Lung Cancer Cells by MCS-C2, an Analog of Sangivamycin

  • In the course of screening for novel modulators of cell cycle progression and apoptosis as anticancer drug candidates, we generated an analog of sangivamycin, MCS-C2, which was elucidated as 4-amino-6-bromo-7-cyclopentyl-7H-pyrrolo[2,3-d]pyrimidine-5-carboxamide. In the present study, we evaluated the molecular mechanisms of MCSC2-induced cell cycle arrest and apoptosis in A549 human lung cancer cells. To investigate the effects of MCS-C2 on cell cycle progression in A549 cells, we measured the DNA content of A549 cells treated with $5\;{\mu}M$ MCS-C2 using flow cytometry. The analysis revealed an appreciable $G_2$ phase arrest in treated cells. This event was associated with significant upregulation of p53 and $p21^{Cip1}$. In addition, the TUNEL assay was used to examine apoptotic induction in treated cells, and the effects of MCS-C2 on the expression of apoptosis-associated proteins were examined by Western blot. Apoptotic induction in MCS-C2-treated A549 cells was associated with cytochrome c release from mitochondria, which in turn resulted in the activation of caspase-9 and -3 and the cleavage of poly(ADP-ribose) polymerase (PARP). Based on these results, we conclude that MCS-C2 is a candidate therapeutic agent for the treatment of human lung cancer via upregulation and activation of p53.

세포사멸 조절 단백질인 IEX-1의 새로운 결합단백질로서의 CATHEPSIN B의 발견 (Identification of CATHEPSIN B as a Novel Binding Protein of the Cell Death Regulating Protein IEX-1)

  • 선행연구에서 본 연구자는 IEX-1 단백질이 난소 암세포에서 세포사멸(apoptosis)을 유도하는 기능을 수행함을 확인하였으나, 세포사멸과 세포생존의 여러 단계에서 어떠한 신호전달 체계로 IEX-1이 작용하는지는 정확히 알지 못하고 있다. 따라서 IEX-1 단백질과 결합하는 새로운 단백질을 찾기 위해 yeast two-hybrid system을 이용하였다. 그 결과 IEX-1이 여러 다양한 인간 암 세포에서 세포사멸을 유도하는 CATHEPSIN B 단백질과 결합함을 밝혔다. 본 연구에서는 리소좀 프로테아제의 일원인 CATHEPSIN B 단백질과 IEX-1 단백질의 결합을 면역침강법과 western blot 분석으로 확인하였다. 그러므로 이 결과들을 통해서 IEX-1과 CATHEPSIN B는 세포 내에서 서로의 기능에 영향을 미칠 것으로 예상되며, 세포사멸을 유도하는 IEX-1 단백질이 lysosome-mediated apoptotic pathway에 관여할 가능성을 시사한다.

위암의 Fas-associated Death Domain Protein 단백질의 발현 (Immunohistochemical Analysis of Fas-associated Death Domain Protein Expression in Stomach Cancers)

  • Purpose: Evidence exists that dysregulation of apoptosis is involved in the pathogenesis of cancer development. Fasassociated death domain (FADD) protein, an adaptor protein of death receptors, is a critical regulatory component of the extrinsic cell- death pathway that exerts its pro-apoptotic effect upon binding with death receptors. Expression of the FADD protein has not been reported in stomach cancer. The aim of this study was to explore the expression status of the FADD protein in stomach cancers. Materials and Methods: In the current study, we analyzed the expression of the FADD protein in 60 advanced stomach cancer by using immunohistochemistry and a tissue microarray approach. Results: Immunopositivity (defined as $\geq\30\%$) was observed for the FADD protein in 23 ($38\%$) of the 60 cancers. Normal gastric mucosal cells showed expression of the FADD protein. Conclusion: Taken together, these results indicate that decreased expression of the FADD protein is a frequent event in stomach cancers and suggest that to avoid apoptosis, stomach cancer cells in vivo may need loss of FADD expression, which might contribute to tumor development.

Activation of Antioxidant-Response Element (ARE), Mitogen- Activated Protein Kinases (MAPKs) and Caspases by Major Green Tea Polyphenol Components during Cell Survival and Death

  • Green tea polyphenols (GTP) have been demonstrated to suppress tumorigenesis in several chemical-induced animal carcinogenesis models, and predicted as promising chemopreventive agents in human. Recent studies of GTP extracts showed the involvement of mitogen-activated protein kinases (MAPKs) in the regulation of Phase II enzymes gene expression and induction of apoptosis. In the current work we compared the biological actions of five green tea catechins: (1) induction of ARE reporter gene, (2) activation of MAP kinases, (3) cytotoxicity in human hepatoma HepG2-C8 cells, and (4) caspase activation in human cervical squamous carcinoma HeLa cells. For the induction of phase IIgene assay, (-)-epigallocatechin-3-gallate (EGCG) and (-)-epicatechin-3-gallate (ECG) potently induced antioxidant response element (ARE)-mediated luciferase activity, with induction observed at 25 $\mu\textrm{m}$with EGCG. The induction of ARE reporter gene appears to be structurally related to the 3-gallate group. Comparing the activation of MAPK by the five polyphenols, only EGCG showed potent activation of all three MAPKs (ERK, JNK and p38) in a dose- and time-dependent manner, whereas EGC activated ERK and p38. In the concentration range of 25 $\mu\textrm{m}$ to 1 mM, EGCG and ECG strongly suppressed HepG2-ARE-C8 cell-growth. To elucidate the mechanisms of green tea polyphenol-induced apoptosis, we measured the activation of an important cell death protein, caspase-3 induced by EGCG, and found that caspase-3 was activated in a dose- and time-dependent manner. Interestingly, the activation of caspase-3 was a relatively late event (peaked at 16 h), whereas activation of MAPKs was much earlier (peaked at 2 h). It is possible, that at low concentrations of EGCG, activation of MAPK leads to ARE-mediated gene expression including phase II detoxifying enzymes. Whereas at higher concentrations of EGCG, sustained activation of MAPKs such as JNK leads to apoptosis. These mechanisms are currently under investigation in our laboratory. As the most abundant catechin in GTP extract, we found that EGCG potently induced ARE-mediated gene expression, activated MAP kinase pathway, stimulated caspase-3 activity, and induced apoptosis. These mechanisms together with others, may contribute to the overall chemopreventive function of EGCG itself as well as the GTP.

Effect of caspases and RANKL induced by heavy force in orthodontic root resorption

  • Objective: Orthodontic root resorption (ORR) due to orthodontic tooth movement is a difficult treatment-related adverse event. Caspases are important effector molecules for apoptosis. At present, little is known about the mechanisms underlying ORR and apoptosis in the cementum. The aim of the present in vivo study was to investigate the expression of tartrate-resistant acid phosphatase (TRAP), caspase 3, caspase 8, and receptor activator of nuclear factor kappa-B ligand (RANKL) in the cementum in response to a heavy or an optimum orthodontic force. Methods: The maxillary molars of male Wistar rats were subjected to an orthodontic force of 10 g or 50 g using a closed coil spring. The rats were sacrificed each experimental period on days 1, 3, 5, and 7 after orthodontic force application. And the rats were subjected to histopathological and immunohistochemical analyses. Results: On day 7 for the 50-g group, hematoxylin and eosin staining revealed numerous root resorption lacunae with odontoclasts on the root, while immunohistochemistry showed increased TRAP- and RANKL-positive cells. Caspase 3- and caspase 8-positive cells were increased on the cementum surfaces in the 50-g group on days 3 and 5. Moreover, the number of caspase 3- and caspase 8-positive cells and RANKL-positive cells was significantly higher in the 50-g group than in the 10-g group. Conclusions: In our rat model, ORR occurred after apoptosis was induced in the cementum by a heavy orthodontic force. These findings suggest that apoptosis of cementoblasts is involved in ORR.

AGS 인체 위암 세포에서 Akt/mTOR/GSK-3β 신호경로 조절을 통한 개똥쑥 추출물의 Apoptosis 유도 효과 (Apoptosis-Induced Effects of Extract from Artemisia annua Linné by Modulating Akt/mTOR/GSK-3β Signal Pathway in AGS Human Gastric Carcinoma Cells)

  • 개똥쑥은 예로부터 항암, 항바이러스 및 항균의 효능을 지니는 것으로 알려져 왔지만 작용 기작에 대한 내용이 많이 알려지지 않았다. 본 연구에서는 AGS 인체 위암 세포를 대상으로 개똥쑥 추출물(AAE)에 의한 apoptosis 효과와 신호경로 연구를 시행하였다. AAE의 암세포 성장에 미치는 영향을 확인하기 위하여 AGS cell에 AAE를 처리하고 MTT assay와 LDH assay를 수행한 결과 AAE 농도 의존적으로 나타난 세포 성장 억제가 세포 손상에 의한 것임을 확인하였다. 또한, AAE에 의한 암세포 증식 억제 효과가 apoptosis에 의한 것인지 확인하기 위하여 Hoechst 33342 staining과 Annexin V-PI staining을 수행한 결과, Hoechst 33342 staining에서 apoptotic body와 세포질 응축이 농도 의존적으로 증가하는 것을 확인하였고, Annexin V-PI staining에서 apoptotic cells의 변화가 농도 의존적으로 증가함을 확인하였다. Western blotting의 결과 AAE가 농도 의존적으로 세포 생장에 관여하는 신호 단백질인 p-Akt, p-TSC2, p-mTOR, p-GSK-$3{\beta}$의 발현이 감소함을 확인하였고, anti-apoptotic 단백질인 Bcl-2의 발현이 억제됨으로써 proapoptotic 단백질인 Bax, Bak의 발현이 증가하는 일련의 신호경로를 조절할 수 있다는 것을 확인하였다. 미토콘드리아 막 전위의 탈분극 유도를 확인하기 위한 JC-1 assay 수행 결과, AAE 농도 의존적으로 미토콘드리아 막 전위의 탈분극이 유도됨을 확인하였다. 탈분극에 의한 caspase 활성을 확인하기 위해 caspase-3/7 activity assay를 수행한 결과, AAE 농도 의존적으로 caspase activity 증가를 확인하였다. 또한, apoptosis가 일어나는 일련의 신호경로를 확인하기 위해 apoptosis 상위 단백질인 Akt, mTOR, GSK-$3{\beta}$의 활성을 억제하는 LY294002, Rapamycin, BIO를 각각 AGS cell에 처리하고 세포증식에 미치는 영향과 신호 단백질의 발현 양상을 알아보기 위해 MTT assay, LDH assay, western blotting을 수행하였다. 그 결과 AAE와 LY294002, Rapamycin 처리군에서 세포증식 억제와 LDH 방출량 증가뿐만 아니라 세포 생장 신호 단백질인 p-mTOR, p-TSC2, p-Akt, p-GSK-$3{\beta}$의 발현이 감소하는 것을 확인하였고, Bcl-2의 발현이 억제됨으로써 Bax와 Bak의 발현을 증가시키는 신호경로를 조절할 수 있다는 것을 확인하였다. 따라서 AGS cell에 개똥쑥 추출물을 처리하였을 때 유도되는 apoptosis 효과는 Akt/mTOR/GSK-$3{\beta}$ 경로 활성 억제를 통해 Bcl-2 발현이 감소함에 따라 Bax, Bak를 활성화해 세포질로의 cytochrome C 유리에 따른 caspase 활성으로 이루어진다는 것을 알 수 있었다.

Role of Citrate Synthase in Acetate Utilization and Protection from Stress-Induced Apoptosis

  • The yeast Saccharomyces cerevisiae has been shown to contain three isoforms of citrate synthase (CS). The mitochondrial CS, Cit1, catalyzes the first reaction of the TCA cycle, i.e., condensation of acetyl-CoA and oxaloacetate to form citrate [1]. The peroxisomal CS, Cit2, participates in the glyoxylate cycle [2]. The third CS is a minor mitochondrial isofunctional enzyme, Cit3, and related to glycerol metabolism. However, the level of its intracellular activity is low and insufficient for metabolic needs of cells [3]. It has been reported that ${\Delta}cit1$ strain is not able to grow with acetate as a sole carbon source on either rich or minimal medium and that it shows a lag in attaining parental growth rates on nonfermentable carbon sources [2, 4, 5]. Cells of ${\Delta}cit2$, on the other hand, have similar growth phenotype as wild-type on various carbon sources. Thus, the biochemical basis of carbon metabolism in the yeast cells with deletion of CIT1 or CIT2 gene has not been clearly addressed yet. In the present study, we focused our efforts on understanding the function of Cit2 in utilizing $C_2$ carbon sources and then found that ${\Delta}cit1$ cells can grow on minimal medium containing $C_2$ carbon sources, such as acetate. We also analyzed that the characteristics of mutant strains defective in each of the genes encoding the enzymes involved in TCA and glyoxylate cycles and membrane carriers for metabolite transport. Our results suggest that citrate produced by peroxisomal CS can be utilized via glyoxylate cycle, and moreover that the glyoxylate cycle by itself functions as a fully competent metabolic pathway for acetate utilization in S. cerevisiae. We also studied the relationship between Cit1 and apoptosis in S. cerevisiae [6]. In multicellular organisms, apoptosis is a highly regulated process of cell death that allows a cell to self-degrade in order for the body to eliminate potentially threatening or undesired cells, and thus is a crucial event for common defense mechanisms and in development [7]. The process of cellular suicide is also present in unicellular organisms such as yeast Saccharomyces cerevisiae [8]. When unicellular organisms are exposed to harsh conditions, apoptosis may serve as a defense mechanism for the preservation of cell populations through the sacrifice of some members of a population to promote the survival of others [9]. Apoptosis in S. cerevisiae shows some typical features of mammalian apoptosis such as flipping of phosphatidylserine, membrane blebbing, chromatin condensation and margination, and DNA cleavage [10]. Yeast cells with ${\Delta}cit1$ deletion showed a temperature-sensitive growth phenotype, and displayed a rapid loss in viability associated with typical apoptotic hallmarks, i.e., ROS accumulation, nuclear fragmentation, DNA breakage, and phosphatidylserine translocation, when exposed to heat stress. Upon long-term cultivation, ${\Delta}cit1$ cells showed increased potentials for both aging-induced apoptosis and adaptive regrowth. Activation of the metacaspase Yca1 was detected during heat- or aging-induced apoptosis in ${\Delta}cit1$ cells, and accordingly, deletion of YCA1 suppressed the apoptotic phenotype caused by ${\Delta}cit1$ mutation. Cells with ${\Delta}cit1$ deletion showed higher tendency toward glutathione (GSH) depletion and subsequent ROS accumulation than the wild-type, which was rescued by exogenous GSH, glutamate, or glutathione disulfide (GSSG). Beside Cit1, other enzymes of TCA cycle and glutamate dehydrogenases (GDHs) were found to be involved in stress-induced apoptosis. Deletion of the genes encoding the TCA cycle enzymes and one of the three GDHs, Gdh3, caused increased sensitivity to heat stress. These results lead us to conclude that GSH deficiency in ${\Delta}cit1$ cells is caused by an insufficient supply of glutamate necessary for biosynthesis of GSH rather than the depletion of reducing power required for reduction of GSSG to GSH.

  • Several micronutrients (vitamins and minerals) are required as co-factors in DNA synthesis, DNA repair, DNA methylation and apoptosis. Some notable examples include (a) folic acid and vitamin B12 required for maintenance methylation of DNA and the synthesis of dTTP from dUTP, thus prevent the misincorporation of uracil into DNA, a highly mutagenic and chromosome-breaking event, (b) niacin, is essential in the form of the coenzymes NAD and NADP which act as a substrate for polyADPribose polymerase (PARP), an enzyme thought to facilitate efficient DNA repair and telomere length regulation and (c) zinc, apart from its antioxidant role as a co-factor in Cu/Zn SOD, it is required in its stabilizing role of the DNA-binding domain of p53 (residues 102-292) and thus is essential for apoptotic response to DNA damage.
