자리공(Phytolacca esculenta van Houtte) 모상근배양에서 항산화효소의 활성에 미치는 광의 영향 (Effects of Light on Activities of Antioxidative Enzymes in Hairy Root Cultures of phytolacca esculenta Houtte)

  • 자리공 모상근에서 광처리에 따른 항산화효소의 활성을 조사하였다. Catalasa superoxide dismutasae, ascorbate oxidase (AO)의 활성은 광도가 2,000 lx까지 증가할수록 감소하였으며, 특히 AO활성은 2,000 lx에서 암상태보다 92% 감소하였다. Glutathione reductase, glutathione peroxidase (GPO), ascorbate peroxidase 그리고 peroxidase의 활성도는 500 lx까지는 광도가 높아질수록 증가하였으나 그 이상의 높은 광도에서는 현저하게 감소하였다. GPO의 활성은 AO처럼 2,000 lx에서 암상태보다 85%감소하였다. 광파장에 따른 항산화효소의 활성은 청색광의 파장에서 가장 많이 억제되었으며, AO의 활성은 25%까지 감소하였다. 청색광 파장의 광도에 따른 항산화효소의 활성도는 30 lx까지는 증가하다가 200 lx에서는 암상태보다 21-71%까지 감소하는 경향을 나타내었다. AO의 활성은 청색 파장의 광도(300-200 lx)가 증가할수록 급격히 감소하여 200 lx에서는 70%까지 억제되었다. 자리공 모상근의 항산화효소 활성은 청색광 파장의 높은 광도에서 주로 생성된 유해산소 의하여 억제되고 있음을 확인하였다. 광상태하에서 모상근의 생장과 betalain 합성을 향상시키기 위해서는 모상근에서 생성되는 산화제의 효율적인 제거가 요구됨이 시사되었다.

산수유 추출물의 항균 및 항산화성 (Antimicrobial and Antioxidative Activities of Corni Fructus Extracts)

  • Corni Fructus extracts were investigated for antimicrobial and antioxidative activities. In the methanol, ethanol, water extracts from Corni Fructus, antimicrobial activity of the water extract was stronger than the others. Antimicrobial activities were strong against microorganisms such as Streptococcus mutans, Bacillus subtilis, Pseudomonas aeruginosa, Salmonella typhimurium, Escherichia coli and Escherichia coli O-157, the activity against Escherichia coli O-157 was the strongest of all the microorganisms. The growth of all the microorganisms were completely inhibited to 48 hours in the 150${\mu}\ell$ per $m\ell$ of culture media. the shape of E. coli O-157 treated with Corni Fructus water extract was expended and its cellular surface was collapsed severely. When Corni Fructus water extract of 200${\mu}\ell$ was treated, hydrogen donating effect was more than 90%, and antioxidative activity using linoleic acid was similar to 0.1% BHT at the concentration of 50${\mu}\ell$.

꾸지뽕나무 열매 추출물의 항산화 활성 (Antioxidative Activity of Extrcts from Fruit of Curdrania tricuspidata)

  • The comparative activities of acetone, ethanol, and aqueous fractions extracted from fruit powder of Cudrania tricuspidata by different temperature were tested by in vitro experimental models; peroxidation of linoleic acid and autooxidation of rat hepatic and renal microsomes by using thiobarbituric acid (TBA) for assay of free malondialdehyde production, and scavenging activities of free radicals by DPPH (α, α'-diphenyl-β-picrylhydrazyl). In DPPH method, acetone fraction extracted at 30℃ showed the highest free radical scavenging activities and acetone fractions extracted at 30℃ and 60℃ and ethanol fraction extracted at 30℃ showed stronger than BHT (butylated hydroxitoluene) although used ten-fold lower concentrations. In thiocyanate method used linoleic acid an inhibitory effects of all fractions showed higher than control treatment. TBA method used linoleic acid showed the highest antioxidative activity in acetone fraction extracted at 30℃ and 60℃. an inhibition activity against lipid peroxidation in hepatic microsomes of rats showed the highest at acetone faction extracted at acetone fraction among extracted fractions was shown to be the most potent antioxidative properties and this action was more potent in fractions extracted at 30℃ than those extracted at 60℃.

Physiological Effects of Hot Water Extract and Solvent Fractions of Carthamus tinctorius L.

  • In this study, we investigated the biological activities of Carthamus tinctorius including antioxidative, fibrinolytic, thrombin inhibitory, ${\alpha}$-glucosidase inhibitory and collagenase inhibitory activities. Carthamus tinctorius, hot water extract was fractionated into hexane, chloroform, ethyl acetate, butanol and water fraction. Each of these was assayed individually. The hot water extract showed high antioxidative activity and thrombin inhibitory activity at 90.17% and 97.10% respectively. In the fraction activity tests, chloroform fraction showed the highest antioxidative activity at 81.85%. The fibrinolytic activity was strong only in the butanol fraction at 0.70 plasmin units/ml. The thrombin inhibitory activities of hexane, ethyl acetate and butanol fractions were 97.35%, 86.74% and 93.18% respectively. In collagenase inhibitory activity test, hexane fraction showed the highest activity at 87.78%. In conclusion, the hot water extract and solvent fractions of Carthamus tinctorius L can be used as a material for the development of biofunctional tea and foods respectively.

인삼엽의 Photobleaching과 항산화효소 활성 (Activities of Antioxidative Enzymes in Photobleaching of Leaves from Panax ginseng C. A. Meyer)

  • This study investigated the relations between the photosynthetic rate and the activities of antioxidatile enzymes, glutathione reductase, ascorbate free radical reductase, ascorbate peroxidase, glutathione peroxidase, and ascorbate oxidase, in the leaves of Panax ginseng. Under the normal cultivation condition, Panax in showed lower g1utathione reductase and ascorbate free radical reductase activities the Glycine max. But P ginseng showed higher 91utathione Peroxidase, ascorbate Peroxidase, and ascorbate oxidase activities than C. Panax. Therefore, P. ginseng showed weak activities of reductases for the reduction of the oxidized antioxidants. Under the light intensity of 25 KLux, the reductases showed a decrease of over 75% after 24 hours. But the peroyoxidases decreased about 40%. These results showed that the decrease of reductases acitivities was consistent with the decrease of photosynthetic rate. Therefore, we consider that the regulation of antioxidative enzymes or the application of exogenous antioxidants will be effective means for the protection of photodamage in p. ginseng.

비타민 E 보강식이가 당뇨 KK 마우스의 췌장에서 지잘과 산화물의 항산화 효소 활성에 미치는 영향 (Effectso fvitamin E Supplementation on the Lipid Perosides and Activities of Antioxidative Enzymes in the Pancreas of diabetic KK Mice)

  • The purpose of this study was to investigate the effect of vitamin E supplementation on the lipid peroxidation and activities of antioxidative enzymes in the pancreas of diabetic KK mice. KK mice were fed high ft diet containing 20% corn oil(wt/wt), and sacrificed at 2 months of diabetes. A hish vitamin E diet consisted of the high fat diet supplemented with an excessive amount of 이-$\alpha$-tocopheryl acetate (2080IU/kg diet). The incidence of diabetes mellitus was 61% when mice were fed the high fat diet, but was 44% when mice were fed the high vitamin E diet, Vitamin E supplementation fhus seems to have the effect of decreasing of decreasing the onset of diaetes. In the diabetic group, we found increases of MDA (malondialdehyde) and antioxidative enzyme activities. Treatment with vitamin E did not modify the level of fasting blood glucose. However, MDA and antiosicative enzyme activities in diabetic mice were decreased by the high vitamin E diet. Increased levels of lipid peroxidation products suggests the occurrence of oxidative damage in the pancreas of diabetic mice. The increased level of antiosicative enzyme activities could be due to an adaptive response to conditions of increased peroxidative stress. Significant normalization on catalase activity was noted in vitamin E supplemented animals.

뇌 조직에서 알코올 투여에 대한 녹차 건분의 항산화 효과 (Antioxidative Effects of Green Tea Powder Diet Against Ethanol-Induced Oxidative Damage in Rat Brain Regions)

  • The present study investigated the protective effects of green tea against acute ethanol-induced lipid peroxidation and the change of antioxidative enzyme activities in various regions of rat brain : cortex, cerebellum, striatum and hippocampus. The following parameters were examined : malondialdehyde(MDA) levels and activities of superoxide dismutase(SOD), catalase and glutathione peroxidase(GSH-Px). Male Sprague-Dawley rats were given the experimental containing 1% green tea powder or control diet for 4 weeks, and at the end of feeding diet group received acute ethanol(5g/kg body weight) or equicaloric sucrose solution intragastrically. Green tea powder significantly decreased MDA levels in the striatum compared to control-non alcohol treated group to 1% green tea-non alcohol treated group without altering the antioxidative enzyme activities. Green tea resulted in a significant increase in GSH-Px activities in the hippocampus compared to either control-non alcohol treated group(0.043units/mg protein) or 1% green tea-non alcohol treated group(0.071units/mg protein). In conclusion, these results suggest that moderate consumption of green tea leaves can exert protective effects against ethanol-induced oxidative stress in brain regions, by reducing MDA concentrations in the striatum and enhancing GSH-Px activities in the hippocampus. 비타민 E 보강식이가 KK마우스에서 간조직의 항산화계 효소 활성도에 미치는 영향 (Effects of Vitamin E Supplementation on Antioxidative Enzyme Activities in Liver KK Mice)

  • The purpose of this study was to investigate the effects of vitamin E supplementation on the activities of antioxidative enzymes in liver of KK mice of various ages and various duration of diabetes. Diabetes was induced by feeding high fat diet containing 20% corn oil(wt/wt). Weaned KK mice were fed high fat diet containing 51 IU or 2080 IU vitamin E per kg diet. Animals were sacrificed at 4, 6, and 9 months of age. In nondiabetic group, we found the decrease of antionxidative enzyme activities with aging. In diabetic group, antioxidative enzyme activities were decreased, and the change of hepatic vitamin E was related to glutathione peroxidase activity (r=0.71, p<0.001). Treatment with vitamin E did not modify the level of fasting blood glucose. However, it was observered that glutathione reductase and glutathione peroxidase activities as well as hepatic glutathione levels were increased by vitamie E supplementation, whereas catalase activity did not changed. The present result suggest that high vitamin E supplementation protects against lipid peroxidative damage in diabetic KK mice.

표고버섯 첨가가 복어육수의 항산화 활성에 미치는 영향 (The Effect of Added Shiitake Mushroom on Antioxidative Activity of Puffer Fish Stock)

  • This research aimed to improve the healthy properties of puffer fish broth, which has been utilized in Korean and Japanese food. Various healthy foods such as garlic, onion, mushroom, and cauliflower were added as ingredients to pufffer fish stock, and the antioxidative activity of each stock was measured by assaying the DPPH and ABTS radical scavenging activities, reducing power and amount of polyphenol. Shiitake was the most effective in increasing the antioxidative activity of puffer fish stock. The high antioxidative activity of shiitake mushroom seems to be correlated with the amount of polyphenol content in puffer fish broth. The antioxidant activities of puffer fish stock increased proportionally with increasing amount of added shiitake, which in turn was due to the increased amount of total polyphenol in the stock.

  • The biological effects of the water extracts of Rhus Verniciflua Stokes (RVS) were evaluated by protection against hydroxyl radicals. Antioxidative activities were measured using both 1,1-diphenyl-2-picrylhydrazyl (DPPH) and thiocyanate method. Also we used the Glucose oxidase (GO) 20 mU/$\textrm{m}{\ell}$ hydroxyl radical generating system in mouse forebrain cell culture. Water was used for ex-traction from RVS as a solvent which has high polarity especially. In DPPH method, the antioxidative activities of the crude water extract were stronger than any other extracts of low polar-solvents. In the antioxidative effects of mouse forebrain culture using 20 mU/$\textrm{m}{\ell}$ GO, cell viabilities were evaluated 65.6%, 68.8% at 1 $\mu\textrm{g}$. 5 $\mu\textrm{g}$ addition of crude water extracts (30 mg/$\textrm{m}{\ell}$) respectively. 10 $\mu\textrm{g}$ addition of crude water extracts had more than 86.1% cell viabilities, P<0.0l significantly, compared with the group treated with GO alone. In comparison with the antioxidative activities of several commercial antioxidants (ascorbic acid, $\alpha$-tocopherol, catalase, serum), 273 $\mu\textrm{g}$/$\textrm{m}{\ell}$ addition of crude water extracts (300 $\mu\textrm{g}$/$\textrm{m}{\ell}$) showed equivalent antioxidative effect to 25 uM ascorbic acid.
