Effect of Bulohwan-Decoction on Antioxidant Enzyme in Rat Brain (불로환(不老丸)을 투여한 흰쥐 뇌의 항산화효과에 관한 연구)

  • Objectives: Resently Oxidative stress of brain was proved the cause of Alzheimer and stroke sequel. It has important significance in prevention and treatment of cerebropathia that Bulnohwan used as formula of senescence delay have antioxidative effect. The purposes of this study is to investigate the effect of Bulnohwan on antioxidant defense systems such as thiobarbituric acid reactive substances(TBARS), Superoxide dismutase (SOD), Catalase (CAT), Glutathione peroxidase (GSH-PX), Glutathione S-transperase (GST), Glutathione (GSH) in rat brain. Method: Sprague - Dawley rats were divided into 3 groups; saline solution administered control group, Bulnohwan extract administered Experimental group I and Bulnohwan adminisrtrated, 40% dietary restricted Experimental group II. Animals were sacrificed at 12 weeks after treatment TBARS, SOD, CAT, GSH-PX, GST and GSH were measured in mts brain. Results: weight of brain was no stastical significance.(p>0.05) TBARS contents were significant decrease in Experimental group I, II. (p<0.001) SOD activity was stastical significance in Experimental group II, whereas it was no stastical significance Experimental group II.(p<0.0001) Catalase activites were significant increase in . (p<0.00l) Glutathione Peroxidase activites were significant increase in Experimental group I,II. (p<0.000l) Glutathione S-transferase activites were significant increase in Experimental group I, II. (p<0.000) However there were no statistical significance each other. Glutathione contents were significant increase in Experimental group I, II. (p<0.00l) Conclusions: According to the above results, it is considered that Bulohwan has antioxidants effect in rat brain. When Bulohwan goes with diet restriction, there has more Antioxidants effect in rat brain. but this study was perfored retrospectively. So more prospective studies about mutual relation of drugs are needed

Effects of Ethanol Extract of Prunus mume on the Antioxidative System and Lipid Peroxidation on Ethanol-Induced Hepatotoxicity in Rat Liver (매실 추출물이 알코올 투여 흰쥐의 항산화계 및 지질과산화에 미치는 영향)

  • To investigate the antioxidative effects of Prunus mume ethanol extract on the ethanol-induced hepatotoxicity in rat liver, Sprague-Dawley rats weighing 120∼160 g were divided into 5 groups; normal group(NOR), Prunus mume ethanol extract 200mg/kg treated group(PME), ethanol(10 mL/kg, 35%) treated group(ETH), Prunus mume ethanol extract 200 mg/kg and ethanol treated group (PML) and Prunus mume ethanol extract 400 mg/kg, and ethanol treated group(PMF), respectively. The antioxidative activity in vitro was reduced in order of EtOAC>n-hexane>water> chloroform fraction. The growth rate and feed efficiency ratio decreased by ethanol administration were gradually increased to the adjacent level of NOR by administering Prunus mume ethanol extract. It was observed that activities of catalase, superoxide dismutase(SOD), xanthine oxidase and glutathione peroxidase(GSH-Px) of liver and alanine aminotransferase(ALT) and asparate aminotransferase(AST) of serum were elevated by ethanol administration. Besides, Prunus mume ethanol extract markedly decreased elevated activites of catalase, GSH-Px, ALT and AST, except in activites of SOD and xanthine oxidase compared to ETH. Also, the depleted content of GSH by ethanol was increased similar to NOR level by administering Prunus mume ethanol extract. These results suggested that Prunus mume ethanol extract has a possible protective effect on the ethanol-induced hepatotoxicity in rat liver.

Comparison of Biological Activites in Sikhae, Traditional Fermented Sea Products (전통 수산발효식품인 식해류의 생리적 기능성 효과 비교)

  • This study investigated and compared chemical properties and biological activities of four commercial sikhae products (three flat-fish, Pleuronectes herzensteini, Sikhae: product A, B, C; one Alaska pollack, Theragra chalcogramma, Sikhae: product D) and Alaska pollack Sikhae (product E). Total acidity, amino nitrogen, salinity and pH in all products were in ranges of $0.47-1.93\;g\%$, $145.94-204.81\;mg\%$, $3.40-4.00\%$ and 4.38-5.55, respectively. All products showed antimicrobial activities against Bacillus ceraus subtilis, B. cereus, Staphylococcus aureus, Listeria monocytogenes, Vibrio parahaemolyticus, except for Salmonella typhimurium. Particularly the activities were more effective with gram positive bacteria than gram negative bacteria. Antioxidative $(EDA_{50};\;14.20\;mg/mL)$ and Fibrinolytic (0.95 plasmin unit/mL) activities of product E were much stronger than commercial Sikhae products (EDA50; 18.87-34.60 mg/mL and 0.69-0.85 plasmin unit/mL).

  • Antioxidative activites and anti skin aging effects of radish(Raphanus sativus L. var. sativus) extracts were studied. The DPPH(1,1-Diphenyl-2-picrylhydrazal) inhibitory effects and CAT(Catalase) activities of hydrophilic radish extracts(DPPH 81.0%, CAT 175units/mg) were significantly higher than lipophilic radish extracts(DPPH 50.0%, CAT 45unitg/mg). But, SOD(superoxide dismutase) activities of hydrophilic radish extracts(SOD 81.3%) were lower than lipophilic radish extracts(SOD 92.3%) In the Bovine Achilles Tendon collagen treated with hydrophilic radish extracts and lipophilic radish extracts, the pyridinoline concentration of collagen were decreased, respectively.

Antioxidant Effect of Kombucha Broth Against Scenescence Induced Normal Human Diploid Fibroblasts with Oxygen Free Radicals (활성 산소로 산화적 스트레스가 유도된 사람 정상 섬유아세포에 대한 콤부차 발효 배양액의 항산화 효능)

  • Kombucha fermentation broth has been used as a popular health beverage and an alternative therapy with prophylactic and therapeutic benefit. We tried to establish optimal culture conditions for Kombucha fermentation in milk and to investigate cytotoxicity and antioxidative enzyme activity of Kombucha broth against normal human fibroblasts. The optimal conditions of Kombuch culture were established to 3$0^{\circ}C$, 20∼23 hours by DPPH radical scavenging test. There were positive effects on cell growth while no cytotoxicity against primary normal human diploid fibroblasts was found. The activites of glutathione peroxide and catalase in the cells treated by hydrogen peroxide (1 mM) alone and by hydrogen peroxide with Kombucha broth (1 mg/mι) were significantly different (p<0.05). These results suggest that Kombucha broth could be developed as an antioxidant agent for a new cosmetic material.

Effect of Dietary Fatty Acid and Vitamin E Supplementation in Antioxidant Systmes of the Second Generation Rat Brain Sections (식이지방산 조성 및 비타민 E의 보충이 제 2 세대 흰 쥐 뇌조직의 항산화 체계에 미치는 영향)

  • In this study, we examined the effects of dietary fatty acids and vitamin E supplementation on antioxidant systems in the rat brain regions. The Sprague Dawley rats were fed the experimental diets 3-4 wks prior to the conception. Experimental diet consisted of 10% fat(wt/wt) which were safflower oil(SO, poor in $\omega$3 fatty acids), mixed oil(MO, P/M/S ratio=1.03:1.45:1,$\omega$6/$\omega$3 ratio=6.3) and mixed oil supplemented with vitamin E(ME:MO+500mg vitamin E/kg diet). At 3 and 9 weeks of age of the newborn rats, frontal cortex(FC), corpus striatum(CS), hippocampus(H) cerebellum(CB) were dissected out from the whole brain. Activities of glutathione peroxidase(GSH-P(sub)x, superoxide dismutase(SOD) concentrations of malondialdehyde(MDA) were mesaured. Dietary fatty acids were not effective in antioxidative system for rat brain. However, when vitamin E was supplemented to the diet(ME), the activities of GSH-P(suh)x tended to increase in comparison to MO group. Therefore, the activites of GSH-P(suh)x of FC and H at the age of 3 weeks showed significant differences(p<0.05). The activities of Total-SOD tended to decrease in ME group compared to MO group. There were significant differences(p<0.05) in FC and CS at the age of 3 weeks. The activities of Mn-SOD tended to increase and Cu, Zn-SOD tended to decrease when vitamin E was supplemented. The activity levels of antioxidative enzymes at the age of 3 weeks and 9 weeks were similar. This suggested that the activity level of antioxidative enzymes reached to the adult level at the age of 3 weeks which is the end point of lactation period. The concentrations of MDA were not altered by experimental diets. When the activities of antioxidant enzymes were compared, the activities of antioxidant enzymes were the lowest in H and FC. In conclusion, the antioxidative system were not altered by dietary fatty acid at the age of 3 weeks and 9 weeks, but the supplementation of vitamin E altered the antioxidative systems. Studies on Biological Activity of Wood Extractives (IX) - Antionxidative Compounds from Heartwood of Robinia pseudo-acacia - (수목추출물의 생리활성에 관한 연구(IX) - 아까시나무 심재의 항산화활성 물질 -)

  • Antimicrobial and antioxidative activites on heartwood extractives of domestic species were investigated to develop a natural fungicide or preservative. One steroid, stilbene derivatives and two flavonoids were isolated from heartwood of Robinia pseudo-acacia which has been selected due to its high antioxidative activity among the tested species. The structures were determinded as: 3-𝛽-stigmast-5-en-3-ol(daucosterol), 3,3'4,5-tetrahydroxystilbene, 3,3'4'5,7-pentahydroxyflavone(robinetin) and 3,3'4'7-tetrahydroxyflavanone(fustin) respectively on the basis of spectroscopic data and chemical correlations. According to the results of free radical scavenging activity, 3,3'4'5,7-pentahydroxyflavone was evaluated as the highest antioxidative compound among the four compounds and showed higher radical scavenging activity than those of 𝛼-tocopherol and butylated hydroxytoluene(BHT), one of the strongest synthetic antioxidants. 3,3'4'5-Tetrahydroxystilbene and 3,3'4'7-tetrahydroxyflavanone showed higher antioxidative activities than that of 𝛼-tocopherol. However, 3-𝛽-stigmast-5-en-3-ol did not show free radical scavenging activity. In this regard, it could inferred that high antioxidative activity of extractives of R. pseudo-acacia was derived from 3,3'4'5'7-pentahydroxyflavone, 3,3'4'5-tetrahydroxystilbene and 3,3'4'7-tetrahydroxyflavanone.

Antioxidative and Biological Activites of Extracts of Sweetpotato Tips (고구마 끝순 추출물의 항산화 및 생리활성)

  • This study was conducted to increase sweetpotato utilization and to determine the vegetative value of sweetpotato tips by investigating the phenolic compounds, antioxidative effect in oil, electron donating ability, nitrite scavenging effect and ACE inhibition activities. The phenolic compounds present in sweetpotato tips are the gallic, chlorogenic, gentisic, caffeic, couramic and ferulic acid, which are 16-122 times higher compared to other vegetables such as spinach, soybean sprout, and perilla leaves. In each solvent extract, the total phenolic compounds (175.8mg/g) was composed of 55% EtOAc extraction and 39% BuOH extract, respectively. The results of induction period using the Rancimat method showed that the antioxidant activity of SP tips was higher than the tocopherol or BHT. The relative levels of each solvent extract in SP tips were as follows: EtOAc>BHT>BuOH>Tocopherol>Water>$CHCl_3$>Hexane. The peroxide value was measured every 5 days for 25 days during storage and results showed that the peroxide value, the tips, tuberous root and tocopherol were lower compared to spinach, soybean sprout and perilla leaves. Nitrite scavenging effects were excellent in sweetpotato tips, perilla leaves and soybean sprout, especially, inhibition rate of perilla leaves (72%) were superior to the others. In process of solvent extraction, activity of BuOH and water extractions were the best. ACE inhibition activity in sweetpotato tips was 1.5 times higher than in tuberous roots and $1.9{\sim}3.7$ times higher than in spinach, soybean sprout, perilla leaves.

Effects of the Soybean Powder with Rich Aglycone Isoflavone on Lipid Metabolism and Antioxidative Activities in Hyperlipidemic Rats (고지혈증 흰쥐에서 비배당체 이소플라본 고함유 대두분말의 혈청 지질 대사 영향과 항산화효과)

  • This study was performed to evaluate the effects of soybean powder with rich aglycone isoflavone through bio-transformation on lipid metabolism and antioxidative activites in diet induced hyperlipidemic rats. The rats were randomly divided into four groups: NO (basal diet normal group), CO (high fat diet control group), BFF10 (soybean powder with rich aglycone isoflavone 10% group), and BFF20 (soybean powder with rich aglycone isoflavone 20% group). After 7 weeks of BFF10 or BFF20 diets consumption, the concentrations in serum triglyceride, total cholesterol, LDL-cholesterol, and atherogenic index ratios were significantly decreased in the BFF10 and BFF20 diets groups compared with those in the CO group. The activities of alanine amino transferase and aspartate amino transferase were significantly decreased in the BFF10 and BFF20 than those in the CO group. Thiobarbituric acid reactive substance levels of serum were decreased in BFF10 and BFF20 groups compared to that of the CO group. The super oxide dismutase activites were increased in BFF10 and BFF20 groups compared to that of the CO group.
