삼출건비탕가미방(蔘出健脾湯加味方)이 흰쥐의 성장에 미치는 영향 (The Effect of Samchoolgunbitanggamibang of the growth of rats)

  • Objectives : The purpose of this study is to find out the effect of antioxidation related to aging of Cheongeumyeonsudan which is written on Dongui-bogam experimentally. Method : 14 weeks aged SD albino rats were separated into uncontrolled group, controlled group and CGY group. As controlled and CGY groups were induced aging by subcutaneous injection of D-galactose, at the same time we administered the extract of Cheongeumyeonsudan to CGY group for 6 weeks. After then we drew blood from each group, and took measurements; the activity of SOD, GSH-px, catalase in erythrocytes, TBARS value, concentration of total lipid, tryglycende, total cholesterol, HDL-cholesterol in blood plasma. Results : The activities of SOD, GSH-px in erythrocytes were significantly increased in the CGY group compared with control group. The activity of catalase showed a tendency to increase, but it was nor remarkable. The concentration of total lipid, the values of TBARS and total cholesterol was significantly decreased in the CGY group compared with control group, and the concentration of plasma HDL-cholesterol was not remarkable. The concentration of tryglycende in plasma showed a tendency to decreased. Conclusions : it is suggested that Cheongeumyeonsudan decreased the activities of free radical, the concentration of lipid in plasma and generate enzyme which form lipid peroxide.

매실추출물이 활성산소종 소거효과와 B16F1 세포에서 멜라닌 생성에 미치는 영향 (Effect of Fruit Extract of Prunus mume on the Scavenging Activity of Reactive Oxygen Species and Melanin Production in B16F1 Cells)

  • 매실나무(Carthamus tinctorius L.)는 한국, 일본과 중국에서 전통적으로 약효가 있는 음식으로 사용되어 왔다. 특히 과일은 위염증 및 위궤양에 도움이 되는 생물학적 효과가 있다고 보고되었다. 그러나 피부미백과 관련된 효능에 대한 연구는 이직까지 미진하다. 따라서, 본 연구에서는 항산화 및 피부미백에 대한 매실추출물의 효능이 조사되었다. MTT assay를 이용한 세포생존에 대한 결과로 매실추출물은 0.1% 이하의 농도에서 세포독성이 없는 것으로 나타났다. 다음에 매실추출물의 환원력 뿐만 아니라 DPPH radical, hydrogen peroxide 및 superoxide에 대한 직접적인 소거효과가 in vitro에서 평가되었다. 그것은 이상의 활성산소종에 대한 소거효과를 발휘하게 하는 우수한 환원력을 가지고 있다. 더욱이 산화적 스트레스와 연관된 genomic DNA 손상에 대한 보호효과가 농도에 따라 증가하는 것이 관찰 되었다. 뿐만 아니라 L-dopa에 의하여 유발되는 멜라닌 생성에 대한 억제효과를 나타내었다. 그것은 또한 Western blot 분석에서 항산화와 괸련된 NRF-2, SOD-1 및 SOD-2 발현수준을 감소시킨다는 것이 발견되었다. 이러한 결과들은 매실추출물이 항산화에 의한 멜라닌생성의 억제를 통하여 피부 미백효과를 발휘할 수 있다는 것을 암시한다.

오미자 물추출물이 나박김치의 저장성에 미치는 영향 (Effects of Water Extract from Omija (Schizandra chinensis Baillon) on Nabak Kimchi Preservation)

  • 나박김치의 저장성 향상을 위한 천연보존제로서 이용 가능성을 검토하기 위해 오미자 물추출물을 제조하여 이들 시료의 농도를 0.5, 1.0, 1.5, 2.0%로 사용하여 항산화력 및 전자공여능을 측정하고 나박김치 발효에 관여하는 젖산균에 대해 항균활성을 조사하였다. 항산호력은 TBA값을 측정하였는데, 오미자 물추출물의 농도가 높을수록 항산화 효과가 뚜렷이 나타났고, 시간이 경과할수록 그 효과는 현저하였다. 전자공여능 또한 오미자 물추출물의 농도가 높을수록 높은 전자공여능을 나타냈고, 1.0% 이상이 높은 전자공여능을 보였다. 나박김치에서 잦산균을 분리한 결과 117균주를 1차 분리하였고, 이 가운데 성장이 우수하고 산 생성력이 강한 4균주를 2차 선별하여 Lactobacillus brevis, Lactococcus faecalis, Leuconostoc mesenteroides, Lactobacillus plantarum을 확인하였다. 이들의 항균활성을 측정한 결과 Lactobacillus brevis를 제외한 세 균주 모두 오미자 물추출물 첨가에 의한 뚜렷한 성장 억제현상을 나타내었다. 따라서 나막김치 담금시에 항산화성과 항균활성이 있는 오미자 물추출물을 이용할 수 있는 가능성이 높을 것으로 생각되었다.

키토산액 침지 처리에 의한 돈육의 저장성 향상 효과 (Effect of Storage Properties of Pork Dipped in Chitosan Solution)

  • 식육의 부패에 관여하는 미생물에 대해 키토산의 항균력을 측정한 결과, 분자량 약 5 kDa의 키토산에서는 0.001~0.5%의 전 농도에서 항균력이 약하게 나타났으나, 분자량 약 30 kDa과 120 kDa의 키토산을 사용한 경우에는 0.01% 이상에서 대부분의 실험균주에 대해서 강한 항균효과가 나타났으나, Lactobacillus종은 키토산에 대한 항균력은 약간 떨어졌다. 분자량 약 5 kDa, 30 kDa 그리고 120 kDa의 키토산을 0.1~1.0%의 농도로 수용액을 제조하여 돈육 등심을 침지한 다음 1$0^{\circ}C$에서 8일간 저장성에 미치는 영향을 조사한 결과 분자량 약 30 kDa과 120 kDa의 키토산 용액의 1%침지 시 저장 효과가 나타났다. 지질의 산화 억제 효과는 분자량 약 5 kDa의 키토산 사용 시에는 약하게 나타났으나 분자량 약 30 kDa과 120 kDa의 키토산을 사용한 경우에는 저장 중 산화억제 효과가 전 농도에서 높았다. 돈육의 저장 중 색 변화에서는 분자량 약 30 kDa과 120 kDa의 키토산 처리 시 적색도가 안정하게 유지되는 것으로 나타났으며, 수분함량의 경우에 있어서도 분자량 약 30 kDa과 120 kDa의 키토산 처리 시 저장 중 수분할량의 감소 정도는 크지 않았다. pH는 돈육에 처리한 키토산의 분자량 및 농도가 낮을수록 급속하게 증가하는 경향을 나타내었다.

Antioxidant and Anti-hyperglycemic Activity of Polysaccharide Isolated from Dendrobium chrysotoxum Lindl

  • Although polysaccharide is believed to play an important role in the medicinal effect of Dendrobium chrysotoxum Lindl (DCL), its role as an antioxidant and in anti-hyperglycemic induction was not reported. In this study, polysaccharide with molecular weight of approximately 150 kDa, herein named DCLP, was isolated from the stem of DCL, and its antioxidative, hypoglycemic and immune stimulating effects were evaluated using various in vitro and in vivo assay systems. DCLP inhibited hydroxyl radicals ($^{\cdot}$OH)-mediated deoxyribose degradation by scavenging hydroxyl radicals directly as well as by chelating iron ions. DCLP also showed dose-dependent scavenging activity on superoxide anions ($O_2^{{\cdot}-}$) and offered significant protection (p < 0.001) against glucose oxidase-mediated cytotoxicity in Jurkat cells. DCLP had immune stimulating effects, as evidenced by the DCLP-mediated increases in the level of DNA synthesis, viability, and cytokine secretion in mouse lymphocytes. Moreover, oral administration of DCLP produced a significant reduction in blood glucose level in alloxan-induced diabetic mice. These findings suggest that DCLP has a potential utility in treating patients who require enhanced antioxidation, immune function and/or hypoglycemic activity.

초임계 이산화탄소를 이용한 돌나물 추출물이 사염화탄소로 유발된 흰쥐의 지질대사, 지질과산화 및 항산화에 미치는 영향 (The Effects of Sedum sarmentosum Bunge Extract using Super Critical Carbon Dioxide on Lipid Metabolism, Lipid Peroxidation and Antioxidation in Carbon Tetrachloride Induced Hepatotoxicity in Rats)

  • Extraction of Sedum sarmentosum Bunge by super critical carbon dioxide was operated under $40-50^{\circ}C$and 200-250 atm, thus, yield of extraction was very low as 4 wt%. Rats were administrated with the extract orally once a day for succesive 6 days, followed by treatment with carbon tetrachloride ($CCl_4$) on the sixth day. The activities of aminotransferase, alkaline phosphatase, ${\gamma}$-glutamyl transpeptidase, lactate dehydrogenase and contents of triglyceride, total cholesterol in the extract-pretreated rats were decreased compared to the $CCl_4$controled rats, whereas content of HDL-cholesterol was increased. Especially content of hepatic malondialdehyde (MDA) and atherogenic index (AI) were decreased and HTR was increased in the extract-pretreated rats, and reduced peroxidative liver damage in the $CCl_4$-induced hepatotoxicity rats. In addition, activities of hepatic superoxide dismutase, catalase, glutathione peroxidase in the extract-pretreated rats were significantly decreased compared to the $CCl_4$ controled rats, but the content of glutathione was significantly increased. These results suggest that extract of Sedum sarmentosum Bunge has hepatoprotective effect in the $CCl_4$-intoxicated rats.

권백 추출물의 인간 피부 섬유아세포에서의 항산화와 Matrix Metalloproteinase-1 발현저해효과 (Effect of the Selaginella tamariscina Extract on Antioxidation and Inhibition of Matrix Metalloproteinase-1 in Human Skin Fibroblasts)

  • The production of matrix metalloproteinases (MMPs) by the UV irradiated skin fibroblast and the degradation of extracellular matrix (ECM) by these enzymes is known as one of the main reasons of photoaging. In this paper, to investigate the relationship between aging and Selaginella tamariscina extract (STE), we investigated the effects of antioxidant and expression of UVA-induced MMP-1 in human dermal fibroblasts. STE was found to show scavenging activities of radicals and reactive oxygen species (ROS) with the $IC_{50}$/ values of 65.1 $\mu\textrm{g}$/$m\ell$ against 1,1-diphenyl-2-picrylhydrazyl(DPPH) radical and 40.9 $\mu\textrm{g}$/$m\ell$ against superoxide radicals in the xanthine/xanthine oxidase system, respectively. UVA induced MMP expression was reduced 75.5% by treatment with STE, and MMP-1 mRNA expression was reduced in a dose-dependent manner. Therefore STE was able to significantly inhibition of MMP expression in protein and mRNA level. All these results suggested that STE may act as an anti-aging agent by antioxidation and reducing UVA-induced MMP-1 production.

현토고본환이 노화유발 백서의 항산화능에 미치는 영향 (Effect of Hyuntogobon-hwan on Antioxidation Activity in Induced Aging Rats)

  • Objectives : This experiment was done to evaluate the effects of Hyuntogobon-hwan(HTG) on antioxidant capability and lipidic concentration in blood, both of which are presumed to be related to aging. Methods : 12 week-old SD rats were divided into controlled group, uncontrolled group and HTG group. As controlled and HTG groups were given subcutaneous injection of D-galactose(50mg/kg/rat), at the same time HTG group was administered extract of Hyuntogobon-hwan(270mg/200g). HTG injections continued for 6 weeks. After initial injections, blood was drawn from each group and the following were measured: the activity of SOD, GSH-px, catalase in erythrocytes, TSARS value, concentration of total lipid, tryglyceride in blood plasma. Results : The activities of SOD and GSH-px in erythrocytes increased significantly in HTG group compared with controlled group. The activity of catalase seemed to increase slightly, but it was barely noticeable. The concentration of total lipid in plasma decreased significantly in HTG group compared with controlled group. The value of TBARS in plasma seemed to decrease slightly, but it was barely noticeable. Conclusions : According to the above results, Hyuntogobon-hwan has an influence on aging by virtue of activation of antioxidative enzyme systems in erythrocytes and concentrations of lipid in blood plasma.

청기화담환(淸氣化痰丸) 추출물이 비만쥐의 지질강하, 항산화효과 및 전염증성 Cytokines 생산에 미치는 영향 (Effects of Cheunggihwadamhwan Extract on Lowering Lipid, Antioxidation and Production of Proinflammatory Cytokines in Rats Fed on High Fat Diet)

  • Objectives The purpose of this study was to investigate the effects of Cheunggihwadamhwan (CGHDH) extract on lowering lipid, antioxidation and production of proinflammatory cytokines in rats fed on high fat diet. Methods 40 Male Sprague-Dawley rats were fed on high fat diet for 8 weeks and 32 rats (above 400 g) were randomly divided into 4 groups (8 mice in each group) : control group, 100 mg/Kg CGHDH group, 200 mg/Kg CGHDH group, 300 mg/Kg CGHDH group. We fed a control group of rats a basal diet and administered normal saline(100 mg/kg, 1 time/1 day) for 4 weeks. And We fed each experimental group of rats basal diet and administered an extract of Cheunggihwadamhwan extracts (100 mg/kg, 200 mg/kg, 300 mg/kg, 1 time/1 day) for 4 weeks. At the end of the experiment, the rats were sacrificed to determine their chemical composition. We measured lipid in plasma and liver, concentration of proinflammatory cytokines, antioxidative activity and gene expression. The gene expression level was investigated by the way of reverse transcription-polymerase chain reaction (RT-PCR). Results 1. Concentration of plasma FFA, plasma TG, plasma total cholesterol and plasma LDL-cholesterol showed a significant decrement in Cheunggihwadamhwan groups. However, concentration of plasma HDL-cholesterol showed a significant increment in 200, 300 mg/kg Cheunggihwadamhwan groups. 2. Concentration of liver total cholesterol and liver TG showed a significant decrement in Cheunggihwadamhwan groups. 3. Concentration of plasma TBARS showed a significant decrement in all Cheunggihwadamhwan groups. Concentration of liver TBARS showed a significant decrement in 200, 300 mg/kg Cheunggihwadamhwan groups. Concentration of liver GSH-Px, SOD and CAT showed a tendency to decrease in all Cheunggihwadamhwan groups. 4. Concentration of plasma IL-$1{\beta}$, plasma IL-6, TNF-$\alpha$ and NO, showed a tendency to decrease in all Cheunggihwadamhwan groups. Concentration of plasma IL-10 showed a tendency to increase in all Cheunggihwadamhwan groups. 5. In the analysis of reverse transcription-polymerase chain reaction (RT-PCR), the gene expression of Apo-B and Apo-E in the Cheunggihwadamhwan groups showed a low expression than that of control group. The ratio of Apo-B expression per $\beta$-actin expression in the showed a significant decrement in all Cheunggihwadamhwan groups. The ratio of Apo-E expression per $\beta$-actin expression in the showed a significant decrement in 300 mg/kg Cheunggihwadamhwan groups. Conclusions According to this study, the extract of Cheunggihwadamhwan showed a positive effect of lowering lipid, antioxidation and a control of producing proinflammatory cytokines.

정전가매이진탕(正傳加味二陳湯)이 과산화지질 급여 비만 쥐의 지질강하, 항산화효과 및 염증매개물질의 생산에 미치는 영향 (Effects of Jengjengamiyijin-tang (zhengzhuanjiaweierchentang) on Lowering Lipid, Antioxidation and Production of Inflammatory Mediators Being Used Rats Fed on High Oxidized Fat)

  • Objectives The purpose of this study was investigating effects of Jengjengamiyijin-tang (zhengzhuanjiaweierchentang) (JGYT) on lowering lipid, antioxidation and production of inflammatory mediators being used rats fed on high oxidized fat. Methods We divided fat Sprague-Dawley rats fed on high oxidized into 4 groups. Each of 8 rats was divided into a control group and experimental groups. We fed a control group of rats a basal diet and administered normal saline (100 mg/kg, 1 time/1 day) for 4 weeks. And We fed each experimental group of rats basal diet and administered an extract of JGYT extracts (100 mg/kg, 200 mg/kg, 300 mg/kg, 1 time/1 day) for 4 weeks. At the end of the experiment, the rats were sacrificed to determine their chemical composition. We measured lipid of plasma and liver, concentration of proinflammatory cytokines, antioxidative activity and plasma tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), plasma interleukin-6 (IL-6), Apo-B, Apo-E and Leptin gene expression. Results 1. Concentration of plasma FFA, LDL-cholesterol, plasma and liver total cholesterol showed a significant decrement in JGYT groups. However, concentration of plasma HDL-cholesterol showed a significant increment in JGYT groups. 2. Concentration of plasma and liver TG, TBARS showed a significant decrement in JGYT groups. However, concentration of liver GSH-Px, SOD and CAT showed a significant increment in JGYT groups. 3. Plasma GPT activity and concentration of plasma IL-6, TNF-${\alpha}$, NO, Ceruloplasmin, ${\alpha}1$-acid glycoprotein showed a significant decrement in JGYT groups. 4. In the analysis of RT-PCR, gene expression of Apo-B and Apo-E in the JGYT groups showed a low expression than that of control group. However, the gene expression of leptin showed no difference in all the treatment groups. 5. The ratio of leptin expression per ${\beta}$-actin expression showed no significant difference among all treatment groups. However, The ratio of Apo-B and Apo-E expression per ${\beta}$-actin expression showed a significant decrement in JGYT groups. Conclusions According to this study, extract of JGYT showed a positive effect in lowering lipid, antioxidation and control of inflammatory mediators production.
