Antioxidatibe Mechanism of Total Saponin of Red Ginseng (홍삼 총 사포닌의 항산화작용 기전)

  • Oxygen free radicals are highly reactive molecules with unpaired electrons, which are produced with in aerobic cells in the course of normal metabolic events. Normally, aerobic cells are protected from the damage of free radicals by antioxidative enzymes such as superoxide dismutase (SOD), catalase, glutathione (GSH) peroxidase, GSH S-transferase and GSH reductase which scabvenge free radicals as well as nonenzymatic antioxidants such as ceruloplasmin, albumin and nontioxidants in order to elucidate antioxidative mechanisms of red ginseng. The treatment with total saponin of red ginseng significantly devreased the contents of malondialdehyde and total free radicals in the liver. On the other hand, total saponin of red ginseng significantly increased the activities of SOD, catalase and GSH reductase and nonprotein-SH level. These results suggest that total saponin of red ginseng exerts an antioxidative effect by increasing endogenous antioxidants.

  • Cold cuts were prepared from the pork and chicken with polyphosphate salt and spices. Microorganism was more proliferated and acid value was more decreased In pork cold cut rather than chicken cold cut. Otherwise TBA value of chicken cold cut was more increased than that of pork cold cut. Textures of springeness and cohesiveness were not significantly different in both cold cuts. From sensory evaluation, color, appearance and odor were significantly different, but texture, flavor and total acceptability were not significantly different. From these results, if we could study some antioxidants furthermore. we think it possible to manufacture cold cut from chicken.

  • The aim of this study was to investigate the efficiency of the pentagalloic acid compound in inhibiting the metal ions and cell lines that mediate in low density lipoprotein (LDL) oxidation. Pentagalloic acid prolonged the lag time preceeding the onset of conjugated diene formation. In chemically induced LDL oxidation by Cu$^2$(sup)+ plus hydrogen peroxide or peroxyl radical generated by 2, 2-azo-vis (2-amidino propane) hydrochloride (AAPH), pentagalloic acid inhibited LDL oxidation as monitored by measuring the thiobarbituric acid reactive substances(TBARS), malondialdehyde(MDA), and gel electrophoretic mobility. The physiological relevance of the antioxidative activity was validated at the cellular level where pentagalloic acid inhibited mouse macrophage J774 and endothelial cell-mediated LDL oxidation. When compared with several other antioxidants, pentagalloic acid showed a much higher ability than naturally occuring antioxidants, ${\alpha}$-tocopherol and ascorbic acid, and the synthetic antioxidant, probucol.

  • Radical formation reaction of phenolic antioxidants were examined MO theoretically. Results show that substitution of electron-donating alkyl groups in phenol increases the rate of phenoxy radical formation when attacked by an electrophilic radical by stabilization of cationic transition state.

  • The alkaline comet assay, employing a single-cell gel electrophoresis(SCGE), is a rapid, simple and sensitive technique for visualizing and measuring DNA damage leading to strand breakage in individual mammalian cells. The protecting effect of pretreatment with vitamin C and cysteine on the DNA damage of gamma ray was investigated in mice splenic lymphocytes. Vitamin C and cysteine were administered orally for five consecutive days before irradiation. Four week old ICR male mice were irradiated wish 3.5Gy of γ-radiation and were sacrificed 3 days later. Spleens were taken for DNA damage examination by Comet assay and the tail moments of DNA single -strand breaks in tole splenic lymphocytes were evaluated. The results show that pretreatment with vitamin C and cysteine were effective in protecting against DNA damage by gamma ray. Administration of antioxidants like vitamin C and cysteine to mice before irradiation was effective in reducing the tail moment of splenic lymphocytes DNA.

  • Monomeric antioxidant 1 was prepared by the reaction of 3,5-di-tert-butyl-4-hydroxybenzyl alcohol with N-[4-(chlorocarbonyl)phenyl]maleimide in the presence of imidazole. Monomeric antioxidant 2, bearing carbamate group, was synthesized from the reaction of 3,5-di-tert-butyl-4-hydroxybenzyl alcohol and azidomaleimide. Antioxidant 3 was prepared by the reaction of N-(4-hydroxyphenyl)maleimide and 3-(3,5-ditert-butyl-4-hydroxyphenyl) propionic chloride in the presence of triethylamine. These reactive antioxidants were grafted onto polypropylene (PP) by melt-processing with free radical initiators in a mini-max moulder. From the infrared spectra of the grafted PP, it was found that the monomeric antioxidants were grafted onto PP. IR spectroscopic methods were used for the quantitative determination of the extent of grafting of monomeric antioxidant. To optimize the reaction conditions, the influences of the concentration of DCP, monomeric antioxidant, reaction time and temperature on the extent of grafting were studied.

  • The purpose of this study is to get comparative results on the combined antioxidant systems comprised of MHPPD with IPPD, MHPPD with PBN and MHIPPD with MB when they are individually added to NR and SBR compound. Vulcanization characterstics and physical properties of the combined antioxidants have been studied by means of the Mooney viscometer, the Monsanto disc rheometer and so on. According to the test results, MHPPD with IPPD system has shown faster cure rate, better optimum cure time and shorter scorch time than other systems. The vulcanizate comprised of the MHPPD with IPPD antioxidant system has taken advantage of heat resistance, fluid resistance and antiozone resistance.

  • Antioxidants have been used in cosmetic industry for treatment of aged skin and recently have been also introduced as additives in photoprotection products. In order to determine among the Brazilian botanical species presenting interesting antioxidant activities we have screened several extracts from plants from Rain and Amazonical Tropical Forests, as well as some endemic species, using both TBARs and DPPH methods. Extracts with antioxidant activities were found with Jacaranda caroba, Veloso DL, Spilanthes oleracea (Spilanthes acmella var. oleracea), Orbignya phalerata, Pothomorphe umbellata, Chiococca brachiata and Polypodium lepidopteris. Other extracts such as Camelia sinensis, Sambucus australis, Rosmarinus officinalis L. were also studied, and showed some antioxidant activity.

Antioxidant Potential and Chlorogenic Acid Level of Lettuce (Lactuca sativa L.) Cultivars

  • Lettuce (Lactuca sativa L.) is known to contain water-soluble substances that improve antioxidant status due to the richness in antioxidants. Greenhouse experiment was carried out under different shading conditions during spring lettuce growing season. Shade significantly reduced shoot weight, number of leaves and chlorophyll content, while it increased shoot length of lettuce plants. The antioxidant potential of the individual fraction was in order of n-butanol > ethyl acetate > water > n-hexane fraction, although was less than that of commonly used antioxidants, BHT and ascorbic acid. Fractions from lettuce plants dose-dependently increased DPPH free radical scavenging activity, in vitro test. By means of HPLC analysis, BuOH fraction of cultivar 'Hwahyang' (57.93 mg $100g^{-1}$) had the highest amount of antioxidant chi orogenic acid. Shading treatment increased average amount of chlorogenic acid of all cultivars in BuoH, EtOAc, hexane and water fractions by 33, 120, 144, and $58\%$, respectively. These results suggest that lettuce plants had potent antioxidant activity, and their activities were differently exhibited depending on cultivar and fraction.

  • Ethanol can affect a wide range of organ and organelle systems. Some of its effects are directly due to the action of either ethanol or its metabolites, whereas others are related to nutritional deficencies associated with ethanol intake. Some of the liver damages occurring in alcohol abusers are mainly due to generation of free radical during the metabolism of ethanol and subsequent lipid peroxidation. Acetthrough the formation of free radicals, or depletion of levels of antioxidant substances. When prevent generation of the activity. Several authors have reproted the ethanol-related variation of antioxidants such as $\alpha$-tocopherol, ascorbic acid, selenium and glutathionem which are important thereby promoting deficiencies as well as toxic interactions with vitamin A and $\beta$-carotene. It has been appeared that ethanol-induced hepatotoxicity may be partially prevented by the administration of antioxidants.
