  • Experiments were carried out in order to compare the natural antioxidants in the acetonitril extracts of raw and baked seeds of Korean sesame and perilla by thin layer and gas chromatography. The sample was dissolved in n-pentane and extracted with acetonitril and the acetonitril extract was separated by thin layer chromatography using silica gel. The spots were detected by spraying with 2, 6 -dichloroquinone -4-chlorimide, phosphomolybdic acid and dimethylamine as chromogenic reagents. Natural antioxidant, such as ${\delta}-tocopherol$ detected in raw and baked sesame and perilla seed oil by TLC and sesamol was detected only in raw and baked sesame seed oil by GC.

Effects on Lipid Peroxidation and Antioxidants of Paraquat in the Liver of Senescence-Accelerated Mouse(SAM) (제초제인 Paraquat가 SAM의 간조직에서 항산화효소의 활성 및 지질과산화에 미치는 영향)

  • This research employed a senescence-accelerated mouse(SAM) to explore the possibility that differences exist among the major antioxidatns, lipid peroxidation in terms of ability to protect such animal treatment PQ, SAM-R/1 and SAM-P/8 were administered with PQ(200ppm/Kg) orally. The toxicity of PQ on SAM was determined as a bioassays of SOD, catalase and lipid peroxidation in the mouse liver. The data show that the SOD activity was induced by paraqwuat terement in both SAM-R/1 and SAM-P/8. The degree of lipid peroxidation was increased with PQ treatment. This means that SOD rather than catalase may protect against oxygen radical toxicity. Finally, over data lead to the toxicity of PQ and its function may efect to the antioxidants including SOD, catalase and lipid peroxidation in both SAM-R/1 and SAM-P/8 .

Effect of Proanthocyanidin-rich Extracts from Pinus radiata Bark on Termite Feeding Deterrence

  • Antioxidants are known to affect the feeding habits of termites and a good source is pine bark which contains high levels of antioxidants which can be extracted with neutral solvents. In this study procyanidins (PCs)-rich MeOH-extract and a hot-water extract (HWE) were prepared from Pinus radiata bark. HWE was further separated into crude PCs (CPCs), polymeric PCs (PPCs), and low-molecular-weight PCs (LMWPCs) fractions. The MeOH extract and these fractions were examined for termite (Reticulitermes flavipes Kollar) antifeedant activities using a no-choice test. Catechin was used as a positive control. The LMWPCs was found to be the most effective deterrent to termite feeding. The comparative efficiency of the compounds tested were LMWPCs > PPCs > CPCs > MeOH extract, with the latter being more dependent on molecular weight than on its antioxidant activity.

  • The tyrosinase inhibition activity and mutagenicity as assessed by the Ames test on phenolic antioxidants (5-Caffeoyl quinic acid, 3,4-Dihydroxy cinnamic acid, Quercetin 3-O-${\beta}$-D-glucopyranose, Kaempferol 3-O-${\beta}$- D-glucopyranose) and the ethyl acetate fraction isolated from mulberry leaves were examined. The ethyl acetate fraction and chlorogenic acid exhibited weaker tyrosinase inhibitory activities than kojic acid. In addition, the ethyl acetate fraction from mulberry leaves, containing phenolic antioxidants, showed no mutagenicity by the Ames test.

  • Fungal metabolite, gliotoxin is an epipolythiodioxopiperazin (ETP) class and has various roles including immunomodulatory and apoptotic effects. This study was designed to evaluate the mechanism by which gliotoxin exerts the apoptosis on human promyelocytic leukemic HL-60 cells. Herein, we demonstrated that the gliotoxin decreased the cell viability in a time-dependent manner Gliotoxin-induced cell death was confirmed us apoptosis characterized by chromatin condensation and ladder-pattern fragmentation of genomic DNA. Gliotoxin increased the catalytic activities of caspase-3 and caspase-9. Activation of caspase-3 was further confirmed by degradation of procaspase-3 and poly(ADP-ribose) polymerase (PARP) by gliotoxin in HL-60 cells. Furthermore, gliotoxin induced the changes of mitochondrial transmembrane potential (MTP). Antioxidants, including GSH and NAC, markedly inhibited apoptosis with conistent suppression of enzymatic activity of caspase-3, caspase-9, and MTP loss in gliotoxin-treated cells. Taken together, we suggest that gliotoxin function as an oxidant and ploys proapoptotic roles in HL-60 cells via activation of intrinsic caspase cascades as well as mitochondrial dysfunction.

  • There are now increasing evidences that free radicals and reactive oxygen species are involved in a variety of pathological events. Reactive Oxygen Species (ROS) are produced during normal cellular function. ROS lead to lipid peroxidation, massive protein oxdiation and degradation. Under normal conditions, antioxidant are substnaces that either directly or indirectly protect cell against adverse effect of ROS. several biologically important compound include ${\beta}$-carotene, taruine and flavonoids reported have antioxidant function. The various antioxidant either scavange superoxide and free radicals or stimulate the detoxification mechanisms within cells resulting in increased detoxification of free radicals formation and thus in prevention of many pathophysiologic processes. This study carried out to investigate the antioxidant activity of flavonoids, myricetin with other antioxidants, ${\beta}$-carotene and taurine on B16Fl0. In order to investigate the efficacy of antioxidant activity, we measured cell viability, antioxidant enzyme activity (SOD, GPX, CAT) and intracellular reactive oxygen intermediate (ROI). In this results, we show that these flavonoids with other antioxidant substrates are increased antioxidant activity level.

Suppressive Effects of Various Antioxidants on Melamine-induced Oxidative DNA Damage in Human Lymphocytes

  • Melamine, which is used to produce melamine resin for various industrial applications, has a high nitrogen content by mass. For this reason, it has been illegally added to foods to increase their apparent protein content. In the present investigation, melamine-induced oxidative damage of human lymphocyte DNA was evaluated by Comet assay. The in vitro oxidative DNA damage caused by melamine increased in a dose-dependent manner. This DNA damage was significantly inhibited by treatment with ascorbate. Moreover, the traditional Korean medicinal herb, named Acanthopanax, red ginseng and green tea markedly reduced the DNA damage. Various edible plant extracts also inhibited melamine-induced oxidative DNA damage in vitro. Melamine enhanced intracellular ROS generation, and this effect was suppressed by treatment with various antioxidants.

Studies on the Antioxidant Action of Antioxidant of Oils and Fats (유지의 항산화제의 항산화능력에 대하여)

  • Many synthetic antioxidants are now used to prevent the oxidation of oils and fats. Among the synthetic and tioxidants, it was known that those of HQ(Hydroquinone), BHA(Butyl hydroxyanisole), IG(isoamyl gallate), and NDGA(Nordihydro guaiaretic acid) are most efficient as the antioxidizing agent. In this work, Degree of antioxidation was investigated on the some such antioxidans BHA, CI(citric acid), AA(Ascoric acid) for several kinds of oils and fats which are most available in Korea by determining Acid value and Iodine Value. The results of experiments were as follows 1. The decreasing order of antioxidizing action by acid value is BHA, AA, CA. 2. The order of antioxidizing action by Iodine value is almost same as the case of Acid Value. 3. It is effective to add the antioxidants to kep oils and fat. 4. Oxidation is accelerated by standing oils and fats after boiling in the from fry pan. 5. Sesame oil is little oxidized without antioxidant.

Improvement of Light Fastness of Berberine Colorant by Natural Antioxidants

  • In order to improve the light fastness of a natural colorant berberine, several natural antioxidants such as gallic acid, L-ascorbic acid, and ${\alpha}-tocopherol$ were applied by aftertreatment method. Even though the increase in light fastness of berberine colorant was not substantial, L-ascorbic acid was the most effective. It was considered that this improvement of the light fastness was due to high antioxidation action of L-ascorbic acid to colorant, which led very highly excited species of the colorant formed by photo-energy to return to the original stable state without being decomposed.

Screening of Antioxidants to Human Low Density Lipoprotein (LDL) from Marine Resources (수산자원으로부터 Human Low Density Lipoprotein (LDL)에 대한 항산화제의 탐색)

  • This study was undertaken to evaluate antioxidative activities of substances isolated from marine resources against human low density lipoprotein (LDL). Methanol-water extract(80 : 20, v/v) of Sargassum ringgoldianum had the highest antioxidant activity and the active substance was purified by silica gel column chromatography by eluting chlorform : methanol mixture (80 : 20 v/v). The active fraction was seperated to several spots on the TLC in chlorofrom : methanol (10 : 1, v/v) mixture. Antioxidative activity of band 4 of fraction 2 on TLC was highest than that of $\alpha$-tocopherol against human LDL oxidation by the method of thiobarbituric acid reactive substance (TBARS). The band 4 of fraction 2 inhibited the copper mediated oxidation of human LDL with almost completely at 1 or 2 mg/ml.
