The antioxidant activity of methanol extracts of plants and marine algae was tested by using 1,1-diphenyl-2-picrylhydrazyl(DPPH). Five plant extracts(Prunus davidiana, Eriobotrya japonica, Artemisia iwayomogi, Stirodella tolyrrhiza and Ulmus davidiana) and two algae (Ecklonia stolonifera and Symphycoladia latiuscula) were found to be the most effective in DPPH radical scavenging activity. The methanol extract obtained from the stems of Prunus davidiana was fractionated with several solvents. The ethylacetate soluble fraction exhibiting the strongest antioxidant activity was further purified by repeated silica gel and Sephadex LH-20 column chromatography. Antioxidant flavonoids and flavonoid glycosides were isolated and the most active ones was identified as (+)-catechin by MS, $^1H-NMR$ and $^{13}C-NMR$. Its antioxidant activity was higher than that oil vitamin C.
BACKGROUND/OBJECTIVES: Vitamin E is a fat-soluble vitamin and functions primarily as a lipid antioxidant. Inadequate vitamin E status may increase risk of several chronic diseases. Thus, the objectives of this study were to estimate intake and plasma concentration of each tocopherol and to evaluate vitamin E status of Korean adults. SUBJECTS/METHODS: Three consecutive 24-h food recalls and fasting blood samples were collected from healthy 20- to 59-y-old adults (33 males and 73 females) living in the Seoul metropolitan area, South Korea. ${\alpha}$-, ${\beta}$-, ${\delta}$-, and ${\gamma}$-tocopherol intakes and plasma concentrations of tocopherols (${\alpha}$-, ${\delta}$-, and ${\gamma}$- tocopherol) were analyzed by gender. RESULTS: Dietary vitamin E and total vitamin E intake (dietary plus supplemental vitamin E) was $17.68{\pm}14.34$ and $19.55{\pm}15.78mg$${\alpha}$-tocopherol equivalents, respectively. The mean daily ${\alpha}$-tocopherol, and ${\gamma}$-tocopherol intakes were $3.07{\pm}2.27mg$ and $5.98{\pm}3.74mg$, respectively. Intakes of total vitamin E and each tocopherol of males were significantly higher than those of females (P < 0.05). Plasma ${\alpha}$-tocopherol concentration was $15.45{\pm}10.16$ of males and $15.00{\pm}4.54{\mu}mol/L$ of females, respectively. There were no significant differences in plasma tocopherol concentrations by gender ($P{\geq}0.05$). Plasma ${\alpha}$-tocopherol was negatively correlated with ${\gamma}$-tocopherol intake (P < 0.05). Twenty-three percent of the subjects had plasma ${\alpha}$-tocopherol concentrations < $12{\mu}mol/L$ indicating a biochemical deficiency of vitamin E. Approximately 8% and 9% of these participants had plasma ${\alpha}$-tocopherol:total lipid ratio less than $1.59{\mu}mol/mmol$ and plasma ${\alpha}$-tocopherol:total cholesterol ratio less than $2.22{\mu}mol/mmol$, respectively, which are also indicative of vitamin E deficiency. CONCLUSIONS: Vitamin E intakes of Korean adults were generally adequate with the Korean Dietary Reference Intakes for vitamin E. However, ${\alpha}$-tocopherol intake was lower than that reported in other countries, and 23% of the subjects in the current study were vitamin E deficient based on plasma ${\alpha}$-tocopherol concentrations.
Status of blood minerals and their absorption by neonate calves as influenced by fat soluble vitamins supplementation in their respective mothers, mineral supplementation in calves themselves has been evaluated. The objective was to know the impact of antioxidant vitamin supplementation to advance pregnant buffaloes, on enhanced acquired immunity during first few hours after birth, in relation to weight gain in buffalo calves. Advance pregnant buffaloes (n = 30) consisting of average body weight of $550{\pm}15$ kg and of 4-6 parity were fed on 25 kg green (green Jawar-Sorghum bicolor), 2-3 kg wheat straw and 3-4 kg concentrate mixture individually per day. Intramuscular injections of vitamin triplex A $D_3$ E consisting of -2,500,000 IU of vit A -Palmitate; 2,500,000 IU of vitamin $D_3$ and 1,000 IU of vit E (dl-alpha tocopherol acetate) were given per dose, a month prior to parturition, twice at 15 days interval to 15 dams. Rest of the 15 pregnant buffaloes served as negative controls. Secretion of immune proteins, immunoglobulin (Ig) enhanced by 80% in colostrum. The blood serum levels of Zn, Cu, Ca, Mg were measured from birth to 90 days in calves. A significant (p<0.05) difference between the blood serum Zn levels of calves born to vitamin supplemented and non-supplemented dams was measured and a positive correlation between blood serum Zn levels and injections of vitamins was identified. Association of Zn and Cu with passive immunity status has been identified in these calves. A significant positive correlation between Zn and Cu was also identified which showed a change under the impact of vitamin supplementation in buffaloes. The study signifies the role of micronutrients supplementation in dams prior to parturition, in calf immunity development. The study indicates significant mineral - vitamins interactions during this process.
This study was performed to compare the vitamin A, E, ${\beta}-carotene$, polyphenol content, and DPPH radical scavenging activity of carrot, apple, and blueberry juices as affected by extraction methods using slow juicer (HJ), no vacuum blending (NVJ) and vacuum blending (VJ). Juice yields of carrot, apple, and blueberry were 100% in NVJ and VJ groups. Vitamin A content of all juices was the highest in VJ group. Vitamin E content in carrot and apple juices increased most in the HJ group, followed by the NVJ and VJ group; Vitamin E content in blueberry juice was higher in the NVJ group than VJ or HJ group. ${\beta}-carotene$ and polyphenol content in all juices were highest in the VJ group. In addition, the differences between vacuum blending and non-vacuum blending were only shown on apple and blueberry juices. Polyphenol content in carrot and blueberry juices increased most in the HJ group, followed by the NVJ, and VJ groups, and in apple juice, polyphenol content was higher in the VJ group than the HJ or NVJ group. DPPH radical scavenging activity in the VJ group was significantly higher than the NVJ group. The results of this study showed the vacuum blending method had outstanding antioxidative nutrients compared to other juicing methods.
The aim of the present study was to evaluate the effects of two well-known natural antioxidants, vitamin C (VC) and vitamin E (VE), on the antifungal activity of honokiol against Candida albicans. The broth microdilution method was employed to test the antifungal activities of honokiol with or without antioxidants in the medium against C. albicans strain. Intracellular reactive oxygen species and lipid peroxidation were determined by fluorescence staining assay. Mitochondrial dysfunction was assessed by detecting the mitochondrial DNA and the mitochondrial membrane potential. We observed that VC could significantly potentiate the antifungal activities of honokiol while VE reduced the effectiveness of honokiol against C. albicans. In addition, VC accelerated honokiol-induced mitochondrial dysfunction and inhibited glycolysis leading to a decrease in cellular ATP. However, VE could protect against mitochondrial membrane lipid peroxidation and rescue mitochondrial function after honokiol treatment. Our research provides new insight into the understanding of the action mechanism of honokiol and VC combination against C. albicans.
Background: Tobacco consumption is one of the leading causes of oral submucous fibrosis, oral cancer and even premature death. The present study was designed to compare the biochemical parameters and non-enzymatic antioxidant status and the lipid peroxidation products in pan masala tobacco users as compared with age-matched non-user controls. Methods: Pan masala and tobacco users of age $33.2{\pm}9.94$ years and age-matched controls ($31.2{\pm}4.73$ years) were enrolled for the study. Plasma levels of vitamin E, vitamin C, albumin, bilirubin, uric acid, glucose, urea, creatinine, aspartate amino transferase (AST), alanine amino transferase (ALT) were measured by standard methods. Serum malondialdehyde (MDA) levels were estimated as a measure of lipid peroxidation. Results: In the pan masala tobacco users, as compared to the controls, the level of vitamin C ($68.5{\pm}5.9$ vs $97.9{\pm}9.03{\mu}mol/L$, $p{\leq}0.05$) vitamin E ($18.4{\pm}5.3$ vs $97.9{\pm}9.03{\mu}mol/L$, $p{\leq}0.001$), albumin ($37.5{\pm}7.01$ vs $44.3{\pm}9.99g/L$, $p{\leq}0.001$), and malondialdehyde ($10.8{\pm}1.29$ vs $1.72{\pm}1.15nmol/ml$, $p{\leq}0.001$) were found to be significantly altered. Malondialdehyde was significantly correlated with vitamin E (r=1.00, p<0.001) and vitamin C (r=1.00, p<0.001) in pan masala tobacco users. Serum levels of AST ($31.0{\pm}16.77$ IU) and ALT ($36.7{\pm}31.3$ IU) in the pan masala tobacco users were significantly raised as compared to the controls (AST, $25.2{\pm}9.51$ IU, p=0.038; ALT, $26.2{\pm}17.9$ IU, p=0.038). Conclusion: These findings suggest that pan masala tobacco users are in a state of oxidative stress promoting cellular damage. Non-enzymatic antioxidants are depleted in pan masala tobacco users with subsequent alteration in the biochemical parameters. Supplementation of antioxidants may prevent oxidative damage in pan masala tobacco users.

The purpose of this study was to investigate the effects of vitamin C supplementation and yoga practice on total antioxidant status (TEAC), lipid peroxidation, and blood lipid profiles of middle-aged women. Thirty two women ($53.34{\pm}5.08$ years old) in B city volunteered and the signed consents were received before the study. Experimental groups were normal (n=8), vitamin C intake group (n=8), yoga group (n=8), and vitamin C intake with yoga group (n=8). Total experimental period was 12 weeks. Yoga group practiced yoga 3 times a week for 12 weeks with intensity of RPE $13{\sim}15$. Each practice was 60 minutes. Vt. C group was instructed to take 1 g of Vt. C every day after dinner meal. Changes in TEAC after 12 weeks were 28.23% for the normal group, 30% for Vt. C group, 26.58% for yoga group, and 43.66% for Vt. C with yoga group; the increases in TEAC among groups were not significantly different. Serum malondialehyde (MDA) concentration of the normal group increased by 10% during 12 weeks of experiment while that for the Vt. C, yoga, and Vt. C with yoga group were significantly decreased by 25%, 32.14%, and 33.87%, respectively (p<0.05) compared with the normal group. Neither yoga, Vt. C supplementation nor combined program for 12 weeks were effective enough to change the serum lipid profiles compared with the normal group. In conclusion, Vitamin C supplementation or regular yoga practice seems to have health promoting effects of retarding the oxidative stress by decreasing lipid peroxidation in middle-aged woman.

This study was carried out to study influence of antioxidants on serum and tissue of rats fed with thermally oxidized oil. The experimental animals used 30male rats of sprague-dawley weighting $150{\pm}25g$. They are divided into 5 groups and fed to experimental diets which are composed of 15% thermally oxidized oil in addition to ascorbic acid, Vitamin E, Ethylendintrioteraaceticacid(EDTA) and none added oxidized oil by heat and fresh soybean oil group. Thermally oxidized oil was prepared from the soybean oil by heating at $180^{\circ}C$ for 30 hours. After feeding for 4 weeks, the result are as follows; 1. Body weight gain were lower B diet group than A diet group. 2. Total cholesterol levels in serum of all experimental diet groups except B diet group were higher than that of A diet group. 3. HDL-cholesterol levels of all experimental diet groups except E diet group were lower than that of A diet group. 4. The activities of GOT, GPT in serum of all experimental diet groups except B diet group and D diet group were higher than that of A diet group. 5. Vitamin E levels in serum of E diet group were highest than that of all experimental diet groups, and Vitamin E levels in liver of A diet group were highest than that of all experimental diet groups. 6. Lipid peroxide in Serum were highest B diet group than that of all experimental diet groups and the other experimental diet group significantly lower than that of the A diet group. 7. Lipid peroxide in liver of all experimental diet group except E diet group were significantly higher than that A diet group and lipid peroxide in kidney of all experimental diet group except B diet group were lower than that of A diet group. Four these results, as Vitamin C, Vitamin E, and EDTA added diets have effect of thermally oil by antioxidants, it could be suggested that thermally soybean oil diet has required to add antioxidant because it has not sufficient Vitamin E for antioxidant and intake and overtake level of thermally soybean oil diet should be studied to go ahead.

Soybean leaves, a Korean edible plant material, have been reported to prevent the development of osteoporosis and breast cancer. Based on this rational, soybean fallen leaves ethanolic extract (SBFL) was used for the experiment of cell invasion related to metastasis and antioxidant activity. The effect of SBFL on matrix metalloproteinases (MMPs) in human fibrosarcoma cells, HT1080 as well as its anti-oxidant activity was investigated in this study. The effect of SBFL on scavenging activity of reactive oxygen species was evaluated in vitro using lipid peroxidation assay,DPPH radical and reducing power assay. SBFL showed the positive effects on antioxidant activity, compared with vitamin C and vitamin E used as positive controls. Furthermore, SBFL showed cytotoxicity above 16 µg/ml in MTT assay. In particular, it was found that SBFL decreased the activation of MMP-9 stimulated by phorbol 12-myristate 13-acetae (PMA) and phenazine methosulfate (PMS). SBFL treatment increased the expression levels of p-FoxO-1 and SOD-1. Moreover, SBFL inhibited cell invasion stimulated by vascular endothelial growth Factor (VEGF). These results indicate that SBFL could inhibit cell invasion related to the activation of MMP-9 and oxidative stress, suggesting that it could be available as a main ingredient for prevention of metastasis.
