가시오가피 열수추출물과 카르니틴 공급이 고콜레스테롤 식이를 공급한 흰쥐의 지질대사와 항산화계에 미치는 영향 (Effect of Rot Water Soluble Extract from Eleutherococcus and Senticosus and Dietary Carnitine on the Lipid Metabolism and Antioxidant Defense System of Rats on Hypercholesterol Diet)

  • This study investigated the synergy effects of hot water soluble extract from Eleutherococcus senticosus and dietarty carnitine on the lipid metabolism and antioxidant defense system of rats on hypercholesterol diets. Sprague-Dawley male rats were fed either a hypercholesterol diet with water or a hot water soluble extract from Eleutherococcus senticosus and dietarty carnitine. The experimental groups consisted of the control group (CO), the group supplied with hot water soluble extract from Eleutherococcus senticosus(ES), and the group supplied with hot water soluble extract from Eleutherococcus senticosus and dietarty carnitine(ESC). Eleutherococcus senticosus was extracted, made into 0.5% solution, and fed to the ES and ESC groups. A 3% carnitine diet was supplied to the ESC group. Hypercholesterol diets contained 18% beef tallow and 5% cholesterol. After 4 weeks of administering these diets, serum and liver were obtained and the level of serum lipid and the activities of GOT and GPT were measured. In addition, the level of liver lipid and TBARS and the activity of GSH-Px were measured. The results were as follows: 1) Weight gain and FER in the group supplied with hot water soluble extract from Eleutherococcus senticosus and dietarty carnitine(ESC) was low significantly(P<0.05). 2) In the groups supplied with hot water soluble extract from Eleutherococcus senticosus(ES) and the group supplied with hot water soluble extract from Eleutherocorcus senticosus and dietarty carnitine(ESC), total serum cholesterol levels were both significantly low (P<0.05,P.0.01). 3) Serum GOT activity was significantly low(P<0.05) in the group supplied with hot water soluble extract from Eleutherococcus senticosus(ES) and in the group supplied with hot water soluble extract from Eleutherococcus senticosus and dietary carnitine(ESC). There was no difference between serum GPT activity in the control group and the groups supplied with hot water soluble extract from Eleutherococcus senticosus or dietary carnitine. 4) Liver triglyceride was low (P.0.05) in the group supplied with hot water soluble extract from Eleutherococcus senticosus and dietarty carnitine(ESC). No difference was observed in other lipid composition, TBARS level, and GSH-px activity in liver between the control group and the experimental groups. In summary, the effects of hot water soluble extract from Eleutherococcus senticosus and dietarty carnitine on the lipid metabolism and antioxidant defense system (such as weight gain, FER, total serum cholesterol and liver triglyceride) were low when Eleutherococcus senticosus and dietarty carnitine were supplied simultaneously.

솔잎즙의 투여가 고지방식이를 급여한 흰쥐의 혈청 지질성분과 간의 항산화계에 미치는 영향 (Effects of the pine needle sap administration on serum lipid composition and liver antioxidant defense system in rats fed high fat diet)

  • This study examined the effect of the pine needle sap administration on serum lipid composition and liver antioxidant defense system in rats fed with high fat diet. Forty eight male Sprague-Dawley rats were divided into six groups. three control groups were fed with 5% lipid diet and three high fat groups were fed with 30% lipid diet. Each group was administered with the following pine needle sap respectively : C-0.0, 1.ml water, C-0.5, 0.ml pine needle sap and 0.ml water C-1.0, 1.ml pine needle sap : H-0.0, 1.ml water, H-0.5, 0.ml pine needle sap and 0.ml water H-1.0, 1.0ml pinus needle sap. After 4 weeks of experimental periods the level of serum obtained and serum lipid was measured respectively. The results were as follows 1. Significant low level was observed for food intake, weight gain, FER in the experimental group administered with the pine needle sap. 2. No difference of the concentration of serum triglyceride was observed out the experimental group with 5% fat diet, however out of the experimental group with 30% fat diet it was low in (H-1.0) group where 1.0ml of the pine needle sap was administrated. 3. In the experimental group with 5% fat diet the concentration of serum total cholesterol and LDL cholesterol was low, especially the hypocholesteromic effect in serum administered by the pine needle sap was significantly high in group C-1.0 where 1.0ml of pine needle sap was administered. In the high fat group provided with 30% fat when the pine needle sap was administered there was a tendency that concentration of total cholesterol and LDL cholesterol decreased however there was no difference significantly. 4. Serum GOT activity was relatively high in high fat diet group. However, when 1$m\ell$ of pine needle sap was administered the activity was lower than that of the control group in H-1.0 group and the activity level was similar with that of the high fat diet group in c-1.0 group. There was no difference of serum GPT activitives followed by the administered of pine needle sap among the experimental group. 5. Liver TBARS levels were high in general in high fat diet group, however it showed no difference when the pine needle sap with different concentration level was administered. The results of this study indicate that the pine needle sap administration was effective in decreasing the food intake and weight gain of the experimental animals in the high fat diet and also effective in decreasing the levels of serum triglyceride, total cholesterol and LDL cholesterol and GOT activities. However it was not effective to change the level of TBARS and GSH -Px activities of liver. Thus, it was found that the pine needle sap administration was effective for the improvement of serum lipid composition condition of the experimental animals in the high fat diet group but it was not effective in the antioxidant defense system of liver.

항산화 방어 시스템에 의한 유기용매 내성세균 Pseudomonas sp. 균주에서의 톨루엔 내성 (Toluene Tolerance in Solvent Tolerant Pseudomonas sp. Strains By Antioxidant Defense Systems)

  • 유기용매 내성세균들이 항산화 방어시스템에 의하여 독성 유기용매로부터 자신을 보호하는지를 밝히기 위해 두 균주의 유기용매 내성 세균에서 항산화 효소 활성과 총 항산화능(T-AOC)을 조사하였다. 톨루엔 농도 100 mg/l에서 유기용매 내성세균 BCNU 106의 슈퍼옥사이드 디스무타아제(SOD) 레벨은 상대적으로 증가하였으며, 유기용매 내성세균 BCNU 171의 경우에는 톨루엔 농도 200 mg/l에서 상대적으로 증가하였다. 톨루엔 농도 200와 300 mg/l에서 유기용매 내성세균 BCNU 106에서 카타라제(CAT) 레벨이 3배 이상 증가함이 확인되었고, BCNU 171에서는 CAT 레벨이 2배 이상 증가함이 확인되었다. 또한 유기용매 내성세균에서 글루타치온 S- 전이효소(GST) 레벨이 높은 것으로 조사되었다. 총 항산화능도 유기용매에 대하여 내성을 보이지 않는 일반 세균인 KACC 10266보다 유기용매 내성세균들에서 높은 것으로 조사되었다. 한편 톨루엔 농도 200 mg/l 존재 하에서 BCNU 171 균주의 SOD 레벨은 1시간 후 정점에 도달하였다. BCNU 106에서 1시간과 14시간 후 CAT 레벨이 정점에 도달하였고, BCNU 171균주에서는 3시간 후 정점에 도달하였다. T-AOC 레벨의 정점은 BCNU 106에서는 톨루엔에 노출 9시간 후에 나타났으며, BCNU 171에서는 1시간과 9시간 후 두 번 T-AOC 레벨의 정점이 나타났다. 모든 유기용매 내성 세균은 왕성한 항산화 방어 반응을 보였으며, 이러한 항산화 방어 시스템을 통하여 유기용매 내성 세균이 유기용매를 비롯한 가혹한 환경하에서 생존할 수 있는 것으로 판단된다.

Effects of Dietary Supplemented Inorganic and Organic Selenium on Antioxidant Defense Systems in the Intestine, Serum, Liver and Muscle of Korean Native Goats

  • The present study was designed to assess whether dietary inorganic and organic selenium (Se) could affect antioxidant enzymes, including superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and glutathione-S-transferase (GST), and the level of malondialdehyde (MDA), a marker of lipid peroxidation, in the intestine, serum, liver, and gastrocnemius muscle of Korean native goats. A total of eighteen Korean native goats was allotted into three dietary groups, consisting of basal diet (CON), or basal diet with either 0.25 ppm inorganic (IOSEL) or 0.25 ppm organic Se (ORSEL), and fed the corresponding diets for 5 wks. Growth performance, including body weight and total gain, and blood biochemical profiles, including GSH-Px, were not significantly different between the three dietary groups. Also, the specific activities of SOD, GSH-Px, and GST, and the level of MDA in the intestinal mucosa and liver from goats were not substantially affected by either inorganic Se or organic Se. However, goats fed the diet containing organic Se showed a significant increase in GSH-Px and GST activities in the gastrocnemius muscle compared with those fed the basal diet. In conclusion, increased muscle GSH-Px and GST activities suggest that dietary organic Se may affect, at least in part, the antioxidant defense system in muscle of Korean native goats under the conditions of our feeding regimen.

한국인 정신분열병 환자와 Catalase 유전자 다형성의 연합 (The Association between Korean Schizophrenics and Catalase Gene Polymorphism)

  • 본 연구에서는 정신분열병과 antioxidant defense system사이의 관련성을 알아보기 위해 항산화 효소(antioxidant enzyme)중의 하나인 CAT의 유전자 다형성을 한국인 정신분열병 환자군과 대조군 사이에서 비교 분석하였다. 환자군과 대조군의 HinfI 다형성에 따른 CAT유전자형과 대립유전자형의 빈도는 통계적으로 유의한 차이는 없었지만, 여성 정신분열병 환자군과 여성 대조군 사이의 유전자형 빈도에서는 통계적으로 유의한 차이를 나타내었다. 연구결과 CAT유전자가 여성 정신분열병과 관련이 있을 수 있다는 가능성이 제시된다.

Acute phase protein mRNA expressions and enhancement of antioxidant defense system in Black-meated Silkie Fowls supplemented with clove (Eugenia caryophyllus) extracts under the influence of chronic heat stress

  • Background: The current study investigates the anti-stress effects of clove (Eugenia caryophyllus) extracts (0, 200, 400, and 600 mg/kg) on serum antioxidant biomarkers, immune response, immunological organ growth index, and expression levels of acute phase proteins (APPs); ovotransferrin (OVT), ceruloplasmin (CP), ceruloplasmin (AGP), C-reactive protein (CRP), and serum amyloid-A (SAA) mRNA in the immunological organs of 63-d-old male black-meated Silkie fowls subjected to 21 d chronic heat stress at $35{\pm}2^{\circ}C$. Results: The results demonstrated that clove extract supplementation in the diet of Silkie fowls subjected to elevated temperature (ET) improve growth performance, immune responses, and suppressed the activities of glutathion peroxidase (GSH-Px), superoxide dismutase (SOD), catalase (CAT), and thioredoxin reductase (TXNRD); reduced serum malonaldehyde (MDA) and glutathione (GSH) concentrations when compared with fowls raised under thermoneutral condition (TC). Upon chronic heat stress and supplementation of clove extracts, the Silkie fowls showed a linear increase in GSH-Px, SOD, CAT, and TXNRD activities (P = 0.01) compared with fowls fed diets without clove extract. ET decreased (P < 0.05) the growth index of the liver, spleen, bursa of Fabricius and thymus. However, the growth index of the liver, spleen, bursa of Fabricius and thymus increased significantly (P < 0.05) which corresponded to an increase in clove supplemented levels. The expression of OVT, CP, AGP, CRP, and SAA mRNA in the liver, spleen, bursa of Fabricius and thymus were elevated (P < 0.01) by ET compared with those maintained at TC. Nevertheless, clove mitigates heat stress-induced overexpression of OVT, CP, AGP, CRP and SAA mRNA in the immune organs of fowls fed 400 mg clove/kg compared to other groups. Conclusions: The results showed that clove extracts supplementation decreased oxidative stress in the heat-stressed black-meated fowls by alleviating negative effects of heat stress via improvement in growth performance, antioxidant defense mechanisms, immunity, and regulate the expression of acute phase genes in the liver and immunological organs.

Dual effects of a mixture of grape pomace (Campbell Early) and Omija fruit ethanol extracts on lipid metabolism and the antioxidant defense system in diet-induced obese mice

  • BACKGROUND/OBJECTIVES: We investigated the effects of a combination of grape pomace (Vitis labrusca, Campbell Early) and Omija fruit (Schizandra chinensis, Baillon) ethanol extracts on lipid metabolism and antioxidant defense system in diet-induced obese mice. MATERIALS/METHODS: Forty male C57BL/6J mice were divided into four groups and fed high-fat diet (control group, CON) or high-fat diet added 0.5% grape pomace extract (GPE), 0.05% Omija fruit extract (OFE) or 0.5% GPE plus 0.05% OFE (GPE+OFE) for 12 weeks. RESULTS: In contrast to the GPE- or OFE-supplemented groups, the GPE+OFE group showed significantly lower body weight and white adipose tissue weights than the CON group. Moreover, GPE+OFE supplementation significantly decreased plasma total cholesterol and increased the plasma HDL-cholesterol/total-cholesterol ratio (HTR) compared to the control diet. The hepatic triglyceride level was significantly lower in the GPE+OFE and GPE groups by increasing ${\beta}$-oxidation and decreasing lipogenic enzyme compared to the CON group. Furthermore, GPE+OFE supplementation significantly increased antioxidant enzyme activities with a simultaneous decrease in liver $H_2O_2$ content compared to the control diet. CONCLUSIONS: Together our results suggest that supplementation with the GPE+OFE mixture may be more effective in improving adiposity, lipid metabolism and oxidative stress in high-fat diet-fed mice than those with GPE and OFE alone.

갈근으로부터 추출한 ${\beta}-sitosterol$이 HEI-OC1 세포의 항산화 효소 활성에 미치는 영향 (Effect of ${\beta}-sitosterol$ from Pueraria thunbergiana on the Antioxidant Enzyme Activities in HEI-OC1 Cells)

  • A mechanism of hair cell damage caused by noise and ototoxic agents is mediated through generation of free radicals and reactive oxygen species(ROS). It is known that most of animals have defense systems of ROS that protect against ROS, and the cochlea of animals also has ROS defense system, which appear efficient in detoxifying ROS generated under normal condition. This system includes several antioxidant enzymes such as superoxide dismutase(SOD), catalase(CAT), glutathione peroxidase (GPX), and glutathione reductase(GR). The radix of Pueraria thunbergiana(P. thunbergiana) is traditionally prescribed to attenuate the clinical manifestation of inner ear dysfunction and various clinical situations including fevers, gastrointestinal disorders, skin problems, migraine headaches, lowering cholesterol, and treating chronic alcoholism in Oriental Medicine. In the present study, to investigate the protection mechanism of ${\beta}-sitosterol$ from P. thunbergiana on cisplatin cytotoxicity toward HEI-OC1, we measured the effects of ${\beta}-sitosterol$ on activities of SOD, CAT, GPX, and GR in cisplatin treated cells. SOD, CAT, GPX, and GR activities were significantly increased in the presence of 0.001-0.1 ${\mu}g/ml$ of ${\beta}-sitosterol$ compared to the control group. These results indicate that ${\beta}-sitosterol$ protects cisplatin-induced HEI-OC1 cell damage through increasing the antioxidant enzyme system such as SOD, CAT, GPX, and GR.

베타아밀로이드로 유도된 신경소교세포 사멸에 대한 루테올린의 보호효과 연구 (Protective Effect of Luteolin against β-Amyloid-induced Cell Death and Damage in BV-2 Microglial Cells)

  • Objectives : The purpose of this study is to investigate neuroprotective effects and molecular mechanisms of luteolin against ${\beta}$-amyloid ($A{\beta}_{25-35}$)-induced oxidative cell death in BV-2 cells. Methods : The protective effects of luteolin against $A{\beta}_{25-35}$-induced cytotoxicity and apoptotic cell death were determined by MTT dye reduction assay and TUNEL staining, respectively. The apoptotic cell death was further analyzed by measuring mitochondrial transmembrane potential and expression of pro- and/or anti-apoptotic proteins. To elucidate the molecular mechanisms underlying the protective effects of luteolin, intracellular accumulation of reactive oxygen species, oxidative damages, and expression of antioxidant enzymes were examined. Results : Luteolin pretreatment effectively attenuated $A{\beta}_{25-35}$-induced apoptotic cell death indices such as DNA fragmentation, dissipation of mitochondrial transmembrane potential, increased Bax/Bcl-2 ratio, and activation of c-Jun N-terminal kinase and caspase-3 in BV-2 cells. Furthermore, $A{\beta}_{25-35}$-induced intracellular formation of reactive oxygen species and subsequent oxidative damages such as lipid peroxidation and depletion of endogenous antioxidant glutathione were suppressed by luteolin treatment. The neuroprotective effects of luteolin might be mediated by up-regulation of cellular antioxidant defense system via up-regulation of ${\gamma}$-glutamylcysteine ligase, a rate-limiting enzyme in the glutathione biosynthesis and superoxide dismutase, an enzyme involved in dismutation of superoxide anion into oxygen and hydrogen peroxide. Conclusions : These findings suggest that luteolin has a potential to protect against $A{\beta}_{25-35}$-induced neuronal cell death and damages thereby exhibiting therapeutic utilization for the prevention and/or treatment of Alzheimer's disease.

Effects of Dietary Lycopene Supplementation on Plasma Lipid Profile, Lipid Peroxidation and Antioxidant Defense System in Feedlot Bamei Lamb

  • Lycopene, a red non-provitamin A carotenoid, mainly presenting in tomato and tomato byproducts, has the highest antioxidant activity among carotenoids because of its high number of conjugated double bonds. The objective of this study was to investigate the effect of lycopene supplementation in the diet on plasma lipid profile, lipid peroxidation and antioxidant defense system in feedlot lamb. Twenty-eight Bamei male lambs (90 days old) were divided into four groups and fed a basal diet (LP0, 40:60 roughage: concentrate) or the basal diet supplemented with 50, 100, and 200 mg/kg lycopene. After 120 days of feeding, all lambs were slaughtered and sampled. Dietary lycopene supplementation significantly reduced the levels of plasma total cholesterol (p<0.05, linearly), total triglycerides (TG, p<0.05) and low-density lipoprotein cholesterol (LDL-C, p<0.05), as well as atherogenic index (p<0.001), whereas no change was observed in high-density lipoprotein cholesterol (p>0.05). The levels of TG (p<0.001) and LDL-C (p<0.001) were decreased with the feeding time extension, and both showed a linear trend (p<0.01). Malondialdehyde level in plasma and liver decreased linearly with the increase of lycopene inclusion levels (p<0.01). Dietary lycopene intake linearly increased the plasma antioxidant vitamin E level (p<0.001), total antioxidant capacity (T-AOC, p<0.05), and activities of catalase (CAT, p<0.01), glutathione peroxidase (GSH-Px, p<0.05) and superoxide dismutase (SOD, p<0.05). The plasma T-AOC and activities of GSH-Px and SOD decreased with the extension of the feeding time. In liver, dietary lycopene inclusion showed similar antioxidant effects with respect to activities of CAT (p<0.05, linearly) and SOD (p<0.001, linearly). Therefore, it was concluded that lycopene supplementation improved the antioxidant status of the lamb and optimized the plasma lipid profile, the dosage of 200 mg lycopene/kg feed might be desirable for growing lambs to prevent environment stress and maintain normal physiological metabolism.
