지방전구세포와 고지방식이비만마우스에서 가미곽향정기산의 전탕액과 발효액의 항비만효과 (The Antiobese Effects of Gamikwakhyangjungkisan and Fermented GamiKwakhyangjungkisan in Preadipocytes and Mice Fed High Fat Diet)

  • Objectives This experimental study was designed to investigate the antiobese effects of Gamikwakhyangjungkisan and Fermented GamiKwakhyangjungkisan. Methods The cellular lipid contents were assessed by Oil-Red-O staining. The expression of $PPAR{\gamma}$ and $C/EBP{\alpha}$ were determined by real time RT-PCR and western blotting. In addition, body weight gain and serum lipid levels were measured in the mice with obesity induced by the high fat-diet for four weeks. Results Gamikwakhyangjungkisan and Fermented GamiKwakhyangjungkisan is reduced 3T3-L1 cells' differentiation and the expressions of $PPAR{\gamma}$ and $C/EBP{\alpha}$ in high concentration group. High-fat diet + Fermented GamiKwakhyangjungkisan group significantly reduced body weight gain. High-fat diet + Fermented GamiKwakhyangjungkisan group significantly increased HDL-cholesterol contents and reduced LDL-cholesterol contents. Furthermore, Fermented GamiKwakhyangjungkisan is excellent antiobese effects than Gamikwakhyangjungkisan. Conclusions These results demonstrate that Gamikwakhyangjungkisan and Fermented GamiKwakhyangjungkisan exerts antiobese effect in 3T3-L1 cells and mice fed high fat diet. Furthermore, Fermented GamiKwakhyangjungkisan is excellent antiobese effects than Gamikwakhyangjungkisan.

황기(黃芪) 목통(木通) 복합 추출물이 고지방 식이로 유도된 비만 흰쥐에 미치는 영향 (Effects of Mixed Medicinal Herbs on Altering Body Weight and Lipid Profiles in Rats Fed High-fat Diet)

  • Obesity has increased dramatically increased in recent days. It has resulted in obesity-associated diseases and metabolic syndromes including hyperlipidemia, diabetes and cardiovascular diseases. Our study was investigated to determine the antiobese effects of mixed 2 medicinal herbs (AR+AC) in rats fed high-fat diet with an abundance of carbohydrates. Two groups of male Sprague-Dawley rats were fed different diets during 8 weeks: normal diet containing 5% corn oil (w/w), high-fat diet containing 10% (w/w) lard plus 5% (w/w) corn oil (HF). And High-fat diet group was treated with saline or 100 mg/kg ARAC or 300 mg/kg ARAC (PO). Antiobese effect was evaluated by measuring the changes of body weight, adipose tissues weight. Also we examined levels of leptin, adiponectin and lipid profiles in serum. Finally we compared morphological change in adipocytes and hepatocytes between HF and ARAC group. Mixed combination of 2 medicinal herbs, but not alone, caused significant decrease in body weight, organ weight (liver, epididymal fat, and BAT) compared with control HF group. The ARAC group had markedly lower serum levels of adiponectin when compared with HF group. Also the epididymal white adipocyte size of ARAC group were diminished comparing to the HF group. These results suggest that the possibility of ARAC combination, as an antiobese formula, by suppression of body weight gain and increased lipid profiles.

육의감비탕(肉薏減肥湯)의 항비만 효능 및 유효성분 규명 (Identification of Anti-obesity Constituents from Yukeuigambitang)

  • 식품의 원료로 사용가능한 안전한 생약자원 유래의 항비만 소재 개발을 위하여 추출물 단계에서 3T3-L1 전지방세포의 지방세포로의 분화 억제 활성 효과를 나타낸 육의감비탕의 생리활성 성분을 규명하고자 하였다. 70% EtOH로 가열추출 후 계통학적인 용매분획법에 의해 분획한 분획물을 대상으로 3T3-L1 세포의 지방분해 억제시험을 실시한 결과 $CH_2Cl_2$ 분획물에서 가장 우수한 활성이 나타났으며, n-Hexane 분획물과 EtOAc 분획물에서도 약한 지방세포 분화억제 효능이 관찰되었다. 이중 $CH_2Cl_2$ 분획물로부터 각종 chromatography 기법을 통하여 분석한 결과 3종의 phenol류 화합물을 GC-mass 화합물사전 검색을 통해 동정 할 수 있었으며 이 중 1종의 화합물을 HPLC를 이용하여 단리하였다. 단리된 화합물은 육의감비탕을 열추출하는 과정에서 구성하고 있는 생약 중 육계로부터 유래된 유도체인 것으로 추측되며 따라서, 육의감비탕에 포함되어 있는 육계로부터 육의감비탕 조제시 생성되는 물질로 판단된다. 육의감비탕은 선행연구에서 처음으로 개발된 처방으로 본 연구를 통해 단리된 화합물외에도 항비만 효과를 지닌 유사한 화합물들이 존재할 것으로 사료되어 향후 기능성식품의 소재로서 안전하게 사용할 수 있을 것으로 기대된다.

한방초콩환이 고지방식이를 급여한 흰쥐의 체중 및 지질 변화에 미치는 영향 (Effects of Mixed Pills of Chokong (Pickled Black Soybeans) with Medicinal Herbs on Body Weight Gain and Lipid Profiles in Rats Fed High-fat Diet)

  • Objectives : In this study we investigated the antiobese effects of mixed pills of pickled black soybeans with herbs(herbal Chokong pill, hereafter HCKP) in rats fed high-fat diet. It was evaluated by measuring the changes of body weight, adipose tissues weight and lipid profiles in serum. Methods : Black soybeans were pickled in vinegar for 15 days to prepare Chokong, at room temperature. For treatment group, HCKP was prepared, which five kinds of medicinal herbs(Rhynchosia nulubilis, Glycyrrhiza uralensis, Zizyphus vulgaris, Atractylodes macrocephala K, Astragalus membranaceus and Cornus officinalis) were added to dried Chokong. Four groups of male Sprague-Dawley rats were fed different diets during 9 weeks: normal diet containing 5%(w/w) com oil, high-fat diet containing 10%(w/w) lard plus 5%(w/w) corn oil (HF), high-fat diet supplemented with 1%(T1) and 5%(T5) HCKP powder, respectively. Results : The T5 group had markedly lower body weight gain and weights of epididymal adipose tissue when compared with HF group. There were significant differences in visceral adipose tissue weights, serum total cholesterol and triglyceride concentrations between the HF and T5 group. Then, the efficacy of powered HCKP on body weight and lipid profiles change in rats fed high-fat diet were induced dose dependantly. Conclusion : These results suggest that the possibility of HCKP, as an antiobese functional formula, by suppression of body weight gain and improved lipid profiles.

Effect of FC-GT Supplement on Body Fat and Lipid Metabolism in Rats

  • The objective of this study was to investigate the effects of supplementation of an antiobese functional formula (FC-GT) on body weight and lipid metabolism in rats fed a high-fat diet. Three groups of male Sprague-Dawley rats were fed different diets for 6 weeks: normal control (NC), high-fat (HF), and high-fat supplemented with powdered antiobese functional formula (FC-GT) (5% wt/wt) groups. Although body weight was not significantly different among the groups, relative weights of epididymal and perirenal white adipose tissues were significantly lower in the FC-GT group than in the HF group. FC-GT supplementation significantly lowered the plasma total cholesterol and triglyceride concentrations, whereas it elevated the ratio of HDL-C/total-C and improved the atherogenic index. Hepatic cholesterol and triglyceride concentrations were significantly lowered in the FC-GT group compared to the HF group. The accumulation of hepatic lipid droplets and the epididymal white adipocyte size of the FC-GT group were diminished compared to the HF group. Hepatic HMG-CoA reductase activity was significantly lower in the FC-GT group than in the HF group. Plasma GPT activity was significantly lowered in the FC-GT group compared to the HF group. Additionally, fecal weight was significantly increased in the FC-GT group than in the HF group. In addition, contents of fecal triglyceride and cholesterol were significantly higher in the FC-GT group compared to the other groups. The antioxidant activities of hepatic SOD, CAT, and GR were significantly increased in the FC-GT group compared to the HF group. Hepatic mARS and plasma mARS levels were significantly lowered in the FC-GT group compared to the NC group. Accordingly, we conclude that supplementation of FC-GT improves plasma and hepatic lipid levels in high-fat fed rats.

육미지황탕가미방(六味地黃湯加味方)이 C57BL/6JHam-ob/ob mice의 비만(肥滿) 및 제2형 당뇨병(糖尿病)에 미치는 영향(影響) (Antiobese and Antidiabetic Effects of Yookmijihwang-tang-gamibang, a Traditional Polyherbal Formula on the Obese and Type II Diabetic C57BL/6JHam-ob/ob Mice)

  • Objectives : The object of this study was to observe the pharmacological effects of Yookmijihwang-tang-gamibang (Liuweidihuang-tang-jiaweifang, YGB) consisted of 17 types of herbs has been traditionally used in Korean medicine for treating various disorders in clinics, aqueous extracts on the genetically obese and type II diabetic C57BL/6JHam-ob/ob(ob/ob) mice. Methods :Three different dosages of YGB were orally administered, once a day for 28 days to ob/ob mice with ob/ob control and C57BL/6JJms normoglycemic intact mice. Four weeks after treatments of YGB: the changes on the body weight, food consumption, blood glucose levels, leptin and adiponectin contents were observed for monitoring the antiobese and antidiabetic effects of YGB. The effects were compared to those of CLA(conjugated linoleic acid) which improve type II diabetes and inhibit related obesity. Results : After end of 28 days of continuous treatments, ob/ob control showed increases of adipocyte hypertrophy, vasodilated atrophic glomerulus which were detected with marked hyperplasia of pancreatic islets, insulin and glucagon producing cells. These obese and related type II diabetes induced in ob/ob mice were markedly and significantly inhibited by 28 days of continuous treatment of three dosages of YGB. The YGB 50mg/kg showed similar favorable effects on the diabetes and related diabetic complications as compared with CLA 750mg/kg in ob/ob mice of the present study. Conclusions : The results obtained in this study suggest that over 25mg/kg of YGB extracts favorably retarded the obese and type II diabetes in genetically obese and type II diabetic ob/ob mice.

Pair Fed 흰쥐에 있어서 녹차의 항증체, 지질개선 및 항산화 효과 (Effects of Green Tea on Weight Gain, Plasma and Liver Lipids and Lipid Peroxidation in Pair Fed Rats)

  • 본 연구에서 pair feeding함으로서 사료 섭취량이 체중이나 지단백 대사에 미칠 요인을 제거한 상태에서 녹차분말이나 녹차추출물이 갖는 항 비만효과나 지질 강하효과를 비교 조사하였다. 녹차분말이나 추출물에서 증체 억제효과를 보였는데 이는 녹차성분이 소장에서의 지방흡수를 억제하거나 지방산화를 촉진하고 대사율을 올려 열량소비를 증가시킨다는 기존 연구결과와 이해를 같이 하는데, 예비실험에서 사료섭취량 자체가 억제됨을 감안 할 때 녹차 추출물이나 분말의 항비만 효과에 대해서 긍정적이다. 본 연구에서 녹차 추출물의 높은 콜레스테롤 흡수 억제력에도 불구하고 녹차의 콜레스테롤 강하효과는 나타나지 않았는데 이는 체 콜레스테롤 pool size에 강한 homeostasis 기질이 작용한 것이라고 생각할 수 있다. 식이 콜레스테롤과 지방의 흡수억제에 있어서 녹차분말과 녹차 추출물의 장내 유용성과 체 콜레스테롤의 합성과 담즙 생성에 있어서 EGCG를 포함한 차 카테킨 작용기전을 이해하기 위해서 보다 많은 연구가 필요하리라 생각된다. 녹차 분말과 추출물은 항산화 지표에도 정도에 다소 차이가 있으나 긍정적인 효과를 보였는데 녹차 카테킨은 수용성으로 bioavailability가 비교적 높은 polyphenol계 항산화 물질로서 녹차 잎의 건조중량 50% 이상이 카테킨이고 이중 80% 이상이 catechin gallate 형태로 강한 항산화 효력을 지닌 것으로 나타나 있다. 노화를 활성산소에 의한 세포의 손상과정이라 볼 때 식품으로서 녹차는 매우 중요한 입지에 있다. 차로 마시는 녹차의 섭취방법에서 분말이나 추출물로 섭취방법을 다양화하고 있는데 분말과 추출물의 영양적 효과를 비교해 볼 때 유리 (free)된 카테킨 형태로 추출물이 유리할 수 있지만 녹차분말이 식품으로서의 높은 선호도와 함께 HDL-콜레스테롤을 증가시키는 효과가 있다면 분말차의 섭취도 좋은 방법이다. 음료로서 따져 볼 때 일회 분량 2 g의 녹차를 3회 우려 마시는 경우 이로부터 섭취할 수 있는 항산화물질은 EGCG기준으로 $109{\sim}147\;mg$정도이고 이를 비타민 C 로 환산하는 경우 $142{\sim}168\;mg$이다. 천연의 항산화 물질을 자연스럽게 섭취하는 방법으로서 차의 음용을 생활화하는 것은 비만이나 퇴행성 또는 노화관련 질환의 예방차원에서 바람직하리라 본다.

약용식물 물 추출물들 혼합식이에 의한 고지방식이로 유도된 마우스의 비만 억제효과 (Antiobese Effects of Diet Containing Medicinal Plant Water Extracts in High Fat Diet-Induced Obese Mice)

  • 우리는 고지방식이에 의해 유도된 비만 마우스의 몸무게, 부고환 지방조직 무게, 부고환 지방조직의 지방세포 크기 및 혈장 지질 농도에 약용식물 물 추출물들(MPWEs) 혼합식이가 어떤 영향들을 미치는지 연구하였다. MPWEs 혼합식이의 항비만 효과를 알아보기 위하여 C57BL/6J 마우스는 11주 동안 고지방식이를 섭취시켰다. 마지막 6주 동안, 계속해서 고지방식이(HFD)만 섭취시키거나 고지방식이에 MPWEs(5 g/kg, HFD+MPWEs)이나 고지방식이에 orlistat[0.5 g/kg, HFD+orlistat(항비만 약)]을 섞어 섭취시켰다. HF-free군은 11주 동안 일반 식이를 섭취시켰다. 11주간 고지방식이를 섭취한 군은 일반식이군과 비교하여 혈장 지질 수준, 몸무게, 간 무게 및 부고환 지방조직 무게가 현저하게 증가하였다. MPWEs를 함유하고 있는 식이는 몸무게, 간 무게 및 부고환 지방조직 무게와 마찬가지로 혈장 총콜레스테롤, LDL 콜레스테롤, 중성지방 그리고 혈당 농도를 현저하게 감소시켰다. 혈장 중성지방 수준은 6주 동안 orlistat를 함유한 고지방식이군(HFD+orlistat)에서 현저히 낮았고 MPWEs를 함유한 고지방식이군(HFD+MPWEs)의 그것과 유사하였다. 고지방식이군 마우스의 지방세포크기가 일반식이군의 그것에 비해 현저하게 증가하였고, MPWEs와 orlistat(positive control)는 부고환 지방세포 크기를 현저하게 감소시켰으나 orlistat가 MPWEs보다 약간 더 영향력이 있었다. 이들 결과들은 MPWEs 섭취는 몸무게 증가, 지방세포 형성 및 지방세포 크기 증가를 저해함으로써 항비만 효과가 있을 것이라는 것을 시사하고 있다.

고지방식이로 유발된 비만마우스에서 산마늘 잎추출물의 항비만효과 (Allium victorialis Leaf Extract Prevents High Fat Diet Induced Obesity in Mice)

  • 본 실험은 고지방식이로 유발된 비만마우스에서 산마늘 잎 메탄올 추출물의 항비만 효과를 알아보고자 체중변화, 사료섭취량변화, 렙틴과 아디포넥틴 수치변화, 난소주위 지방무게 변화 측정과 지방세포의 조직병리학적 검사를 실시하였으며, 메트포민을 체중당 250 mg 투여한 약물대조군과 비교평가 하였다. 비만유발을 위하여 고지방식이를 13주간 공급하였으며, 고지방식이 공급 7일째에 증류수(비만대조군), 메트포민(약물대조군), 산마늘 잎 메탄올 추출물을 투여하였다. 비만대조군은 확연한 비만이 유발되었으며, 산마늘 잎 메탄올 추출물(62.5, 125, 250 mg/kg) 투여군에서는 이러한 비만이 유의적으로 감소하였다. 결론적으로 산마늘 잎 메탄올 추출물은 비만유발 마우스에서 비만개선작용을 나타내었으며, 나아가 인체에서도 유의한 비만개선작용을 할 것으로 생각된다.

Monascus Pilosus로 발효시킨 검정콩 첨가 식이가 고지방식이 마우스의 체중과 혈청 지방함량 및 간 조직 항산화계 효소활성에 미치는 영향 (Dietary effects of black bean fermented by Monascus pilosus on body weight, serum lipid profiles and activities of hepatic antioxidative enzymes in mice fed high fat diets)

  • The anti-obesity effects of fermented black bean were tested with mice fed a high fat diet for seven weeks. Body weight gain and feed efficiency ratio (FER) in the high fat diet control (HC) group were markedly higher, compared with those of the normal control (NC) group, but were significantly lower in the 2% black bean powder supplemented high fat diet (BB) group and 2% black bean powder fermented by M. pilosus supplemented high fat diet (BBM) group, compared with those of the HC group. Food intake in the HC and BB groups was significantly lower than that of the NC and BBM groups. Water intake in the HC group was significantly lower than that of the NC group, but was higher in the BB and BBM groups, compared with that of the HC group. On the other hand, relative liver and kidney weight in the HC group was lower than that of the NC group, but was higher in the BB and BBM groups, compared with that of the HC group. In addition, whereas epididymal fat weight in the HC group was markedly higher than that of the NC group, it was significantly lower in the BB and BBM groups, compared with that of the HC group. Meanwhile, hepatic GSH in the HC group was significantly lower than that of the NC group, but was slightly higher in the BB and BBM groups, compared with that of the HC group. Although hepatic LPO in the HC group was dramatically higher than that of the NC group, it was significantly lower in the BB and BBM groups, compared with that of the HC group. In addition, serum TG, total cholesterol, and LDL-cholesterol in the HC group was significantly higher than that of the NC group, but was significantly lower in the BB and BBM groups, compared with that of the HC group. On the contrary, HDL-cholesterol in the HC group was significantly lower than that of the NC group, but was higher in the BB and BBM groups, compared with that of the HC group. In addition, activity of XOR D type in the HC group was lower than that of the NC group, but was slightly higher in the BB and BBM groups, compared with that of the NC group. Activities of ROS scavenging enzymes, such as SOD, GPX, and GST in the HC group were significantly lower than those of the NC group, but were significantly higher in the BB and BBM groups, compared with those of the HC group. In addition, serum ALT activity in the HC and BB groups was higher than that of the NC group, but was significantly lower in the BB and BBM groups, compared with that of the HC group. In histopathological findings, hepatic fat accumulation in the HC group was higher than that of the NC group, but was lower in the BBM group, compared with that of the HC and BB groups. In particular, antiobese, hypolipidemic, and antifatty liver effect of black bean powder fermented by M. pilosus was specifically higher than that of non-fermented steamed black bean. In conclusion, the constituents of black bean fermented by Monascus pilosus have been proven to not only inhibit obesity and hyperlipidemia but also decrease hepatic fat accumulation in high fat diet-induced obese mice.
