어성초즙 및 추출물의 항돌연변이 효과 (Antimutagenic Effects of the Juice and Boiling Water Extract of houttuynia cordata Thunb)

  • The inhibitory effects of the juice and boiling water extract of Houttuyina cordata Thunb., which has been known as a traditional folk antitumor agent, on the genotxicity induced by mutagens were investigated . For this purpose, Salmonella typhimurium TA98 and TA100, and the transheterozygous (mwh/+) third larvae of Drosophila melanogaster were used. The juice of fresh H. cordata 9JHC) and boiling H. cordata extract (BHC) showed antimutagenicities toared alfatoxin B₁ in S. typhimurium TA 98 and TA100. The JHC's inhibitory effect increased by 76% to 98% as the adding concentrations increased, and the BHC had a 86-97% inhibitory effect. however, in the case of N-methyl-N'-nitro-N nitrosoguanidine (MNNG), the JHC's inhibitory effect was 52-60% irrespectively of its concentrations and the BHC had a low inhibitory effect. The JHC and BHC slightly reduced the somatic chromosomal mutagenicity by MNNG in the wing hair spot test system (mwh/+) of D. melanogaster, which suggests that they can inhibit gene mutation , deletion and mitotic chromosomal recomgination.

  • The antimutagenicities of methanol extracts from traditional and commercial kochujang (Korean Red Pepper Soybean Paste) and their ingredients were evaluated in Salmonella/mammalian microsome assay system. The traditional kochujang showed higher antimutagenic effects than the commercial one against Ν-methyl-Ν'-nitro-Ν-nitrosoguanidine (MNNG) in the Salmonella typhimurium TA100, a base-pair substituted mutant strain.(omitted)

Antimutagenic and Cancer Cell Growth Inhibitory Effects of Seaweeds

  • The antimutagenic and cancer cell growth inhibitory effects of methanol extracts from 9 kinds of seaweed were studied in the Ames assay and cell culture systems, respectively. The methanol extracts from the seaweeds of sea lettuce, chlorella, sea tangle, sea mustard, sporophyll of sea mustard, fusiforme, seaweed papulosa, purple laver and ceylon moss showed antimutagenicities against aflatoxin B₁(AFB₁) and N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) in the Salmonella typhimurium TA100. These extracts revealed relatively higher antimutagenicity against AFB₁(indirect mutagen) than MNNG(direct mutagen). Sporophyll of sea mustard and seaweed papulosa exhibited strong antimutagenic activity against AFB₁, and sporophyll of sea mustard, sea tangle and ceylon moss also reduced the mutagenicity induced by MNNG. The sporophyll fo sea mustard exerted the highest antimutagenic activity among the samples treated. The methanol extracts from 9 kinds of seaweed inhibited the growth of two cancer cell lines, AGS human gastric adenocarcinoma cells and HT-29 human colon carcinoma cells. Sea tangle, sea mustard and sporophyll of sea mustard inhibited the growth of cancer cells significantly. These results suggest that various seaweeds show not only antimutagenic activity but also growth inhibitory effect of some cancer cells.

Antimutagenicity of Phellinus linteus in Salmonella typhimurium

  • The mutagenicities and antimutagenicities of butanol (PL I) and water (PL II) extracts from the filtrate of the cultured broth of Phellinus linteus were examined using the Ames/Salmonella test. No mutagenic activity of PL I and PL II was found in Salmonella typhimurium strains TA98 and TA100, either with or without S9 activation. In contrast, PL I and PL II showed inhibitory effects on the mutagenic activities induced by the directly-acting mutagens, 4-nitro-o-phenylenediamine (NPD) using the tester strain TA98 and sodium azide (NaN3) using the tester strain TA 100 in the absence of S9 mix. PLI and PL II also showed inhibitory effects on the mutagenicities of the indirectly-acting mutagens, 2-aminofluorene (2-AF) using the tester strain TA98 and benzo[a]pyrene (B[a]P) using the tester strain TA 100 in the presence of S9. These results suggest that P. linteus has an antimutagenic activity and may play a role in the prevention of cancer.

Inhibitory Effect of Kochujang Extracts on Chemically Induced Mutagenesis

  • Antimutagenic effects of 5 kinds of kochujang(Korean red pepper soybean paste) samples compared with doenjang(Korean soy paste) were studied using the Ames test with Salmonella typhimurium TA100 and the SOS chromotest, with E. Coli PQ37. Th eantimutatenic effects of methanol extracts from red pepper powder and meju(fermented soybean) powder, the major ingredients of the kochujang,were also evaluated for the mutagenicity of aflatoxin B1 (AFB1) in the Ames assay. The methanol extracts from the kochujang samples showed lower antimutagenicities than those of doenjang against AFB1 and N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) in the Ames assay. Traditional kochujang I and II exhibited strong antimutagenic activity against AFB1 and MNNG. The traditional kochujang samples against MNNG were aslo observed in the SOS chromotest system with the same fashions as shown in the Ames mutagenicity test. The methanol extracts from meju powder had the strongest inhibitory effects on mutabenicity induced by AFB1, however, those form red pepper powder showed lower inhibition rate than kochujang. These results suggest that traditional kochujang exhibit higher antimutagenic acitivity than the commercial variety, and that meju powder seems to be one of the major antimutagenic components in kochujang.

  • The antimutagenic effects of 27 kinds of plant flavonoids on the mutagenicity of aflatoxin $B_1(AFB_1)$ and N-methyl-N'-nitro-N-nitrosoguanidine(MANG) in Salmonella typhimurium TA 100 were investigated. In the mixed applications of $AFB_1\;(1\;\mu{g/plate)}$ with the flavonoids $(300\;\mu{g/plate)}$ in the presence of a mammalian metabolic activation system (S9 mix), chrysin, apigenin, luteolin and its glucoside, kaempferol, fisetin, morin, naringenin, hesperetin, persicogenin, (+)-catechin and (-)epicatechin showed the antimutagenic effect against $AFB_1$ with more than 70% inhibition rate. A little or no antimutagenicities except flavone against MNNG $(0.5\;\mu{g/plate)}$ were observed. For the antimutagencity of the flavonoids on $AFB_1$, the flavonoid structure that contains the free 5, 7-hydroxyl gorup seemed to be essential. However, saturation of the 2, 3-double bond of elimination of the 4-keto group did not affect the activity.

해산 극피동물 중의 당단백질의 특성과 이용 II. 해삼당단백질과 황산콘드로이친의 항돌연변이 및 항암효과 (Antimutagenic and Anticancer Effects of Glycoprotein and Chondroitin Sulfates from Sea Cucumber(Stichopus japonicus))

  • The antimutagenic and anticancer activities of glycoprotein(GP) and chondroitin sulfate(CS) from sea cucumbers were studied using Ames mutagenicity test and human cancer cells culture test. The GP's inhibitory effect toward aflatoxin B1(AFB1) and 3, 2'-dimethyl-4-amino-biphenyl(DMAB) increased with the higher added concentrations up to 5% level(w/w) regardless fractionation methods. The GP from sea cucumbers through DEAE-cellulose column chromatography showed an inhibitory effect ranged from 84 to 98%, and the maximum antimutagenicities resulted in red sea cucumber with 98% (AFB1) and 95% (DMAB). But 5% level of CS from various sea cucumbers had an inhibitory effect toward those both indirect mutagens ranged from 79% to 85%. However, in case of direct mutagens(N-methyl-N'-nitro-N-nitrosoguanidine, MNNG and 4-nitroquinoline-1-oxide, 4-NQO), the GP's inhibitory effect was 55∼78% and the CS had a low inhibitory effect(58∼70%) at the added level of 5%. The GP from sea cucumbers exhibited the strong inhibitory effects with 89∼95% and 82∼92% on the growth of HT-29 human crcinoma cells and AZ-521 human gastric cancer cells (at 5% level).

들깻잎에서 동정된 항돌연변이 물질 (Antimutagenic Compounds Identified front Perilla Leaf)

  • 들깻잎의 메탄을 추출물이 S. typhimurium TA98과 100에서 aflatoxin B$_1$(AFB$_1$)의 돌연변이 유발성을 억제시키는 효과가 있었다. 메탄을 추출물을 silica gel column을 사용하여 더욱 분획하여 항돌연변이 효과를 시험하였으며, 그중 효과가 가장 컸던 fraction 중의 화합물을 분리, 동정한 결과 2-ethoxy acetate, 1, 2, 3, 4-tetramethyl-cis-cyclobutene, 두 개의 methyl 11, 14, 17-eicosatrienoate 이성체, 12-acetyl-9-octadecanoic acid, 그리고 Phytol이 있음이 잠정적으로 밝혀졌다. Phytol과 methyill, 14, 17-eicosatrienoate의 표준품을 사용하여 AFB$_1$및 Trp-P-2에 대한 항돌연변이 효과를 시험한 결과 그 효과가 컸음이 밝혀졌다.

직접 돌연변이원에 대한 가시오갈피 추출물의 항돌연변이 효과 (Inhibitory Effect of Methanol Extract of Eleutherococcus senticosus Maxim. on the Direct Mutagen Mutagenicity)

  • Ames test (S. typhimurium TA98, TA100)에서 6종의 직접변이원에 대한 가시오갈피(뿌리, 줄기, 잎)의 메탄을 추출물 및 용매 분획물의 항돌연변이 효과를 고찰하였다. Salmonella typhimuriun TA98, TA100에서, 6종의 직접변이원으로 유도된 각각의 변이원성에 대한 가시오갈피 뿌리, 줄기 및 잎의 메탄을 추출물의 돌연변이 억제효과를 검색한 결과 1-nitropyrene의 변이원성에 대하여 선택적으로 강한 억제효과를 나타내었다. 따라서 1-nitropyrene 변이원의 돌연변이원성에 대한 용매분획물의 활성을 비교한 결과 클로로포름 분획물이 가장 강한 억제효과를 나타내었다. S. typhimurium TA98, TA100에서, 뿌리의 클로로포름 분획물은 세포내 항돌연변이 효과를 강하게 나타낸 반면 줄기와 잎은 세포내 돌연변이 억제효과를 나타내지 않았다. 이 결과로부터 가시오갈피 뿌리에는 주로 세포내 항돌연변이 효과를, 줄기와 잎에는 세포외 항돌연변이 효과를 강하게 나타내는 활성 성분이 함유되어 있음을 알 수 있었다.
