Knowledge, attitudes and practices toward antimicrobial usage: a cross-sectional study of layer and pig farm owners/managers in Chiang Mai, Lamphun, and Chonburi provinces, Thailand, May 2014 to February 2016

  • This study aimed at determining the current knowledge, attitude, and practices (KAP) of layer and pig farmers to antimicrobial usage and its consequences. Description of such KAP could provide insights useful for promoting the rational use of antimicrobials in livestock. From May 2014 to February 2016, a survey involving 251 respondents in Chiang Mai, Lamphun, and Chonburi provinces, Thailand, was conducted by using a validated questionnaire. More than half (51.0%) of the respondents incorrectly believed that antimicrobial drug efficacy could not be reduced by using sub-recommended dosages, 61.2% had misconceptions about non-therapeutic antimicrobial use, and 66.9% inaccurately felt that antimicrobials were also effective for virus and fungi. Over half (50.6-55.2%) did not see the need to follow instructions or advice of veterinarians. Moreover, only 10.4% regularly relied on responses to drugs sensitivity tests when evaluating the effectiveness of antimicrobials. Overall, assessment of KAP regarding antimicrobial usage indicated that the majority of respondents had low levels of knowledge of antimicrobials, neutral rather than positive attitudes, and employed poor practices in the use of antimicrobials. The results indicate improvements in KAP could be helpful in developing more effective interventions by farmers, reduce antibiotic usage, and slow the growth of antimicrobial resistance.

국내 가축 및 수산에서 항생제 판매 추이(2003-2012) (Trends in antimicrobial sales for livestock and fisheries in Korea during 2003-2012)

  • In this study, trends in the sales of antimicrobials for use in livestock facilities and fisheries from 2003 to 2012 were investigated with regard to antimicrobial group, antimicrobial usage, and animal species. The overall amount of antimicrobials sold each year from 2003 to 2007 was 1,500 tons, after which they decreased, with the lowest sales being 936 tons in 2012. The total volume of antimicrobials used for feed additives decreased markedly by 94% from 2003 to 2012, which was mainly attributed to banning of feed additives. However, antimicrobial consumption through self prescription by farmers for disease prevention and treatment increased by 25% from 2003 to 2012. The largest volume of antimicrobials sold was for use in pigs (48~57%), followed by poultry (18~24%), fisheries (11~25%), and cattle (5~8%). Tetracycline was the highest selling antimicrobial, followed by penicillins and sulfonamides, although the overall sale of all three antimicrobials gradually decreased over the study period. This study demonstrated that the total consumption of antimicrobials has gradually decreased since 2008. Nevertheless, usage by nonprofessionals increased, which can ultimately cause emergence and spread of antimicrobial resistance. Thus, early establishment of veterinary prescription guidelines for prudent use of antimicrobials is urgently needed in Korea.

혐기소화공정에서 항생항균물질이 메탄생성에 미치는 영향 (Effects of Antimicrobials on Methane Production in an Anaerobic Digestion Process)

  • 혐기성 소화 반응조에서 항생항균물질의 영향을 평가하기 위해서 회분식 및 연속식 실험을 통하여 biogas 발생량, 유기산 생성, 유기물 제거 거동 등을 관찰하였다. 회분식 조건에서 단일항생항균물질 1~10 mg/L에서는 저해 영향이 관찰되지 않았으며, 50 mg/L에서 일부 VFAs가 축적되었고 16~30%의 유기물 제거효율이 감소되었지만, 일시적 저해 영향 후 점차 안정되었다. 그러나 항생항균물질 100 mg/L에서 바이오가스 발생량이 48~58% 감소하였으며, VFAs가 메탄으로 전환되지 않고 축적되었다. 연속식 반응기에서 4종의 혼합항생항균물질을 1~10 mg/L 주입시 미생물에 대한 저해영향은 없었으며, 50 mg/L 주입시 30~40%의 메탄가스 발생량이 감소하였고, 이는 이론적 메탄발생량의 60~65% 수준이었다. 항생항균물질 100 mg/L 주입시 85% 감소한 약 2.2 L/day의 바이오가스가 발생하였으며, 서서히 회복하여 7.2 L/day까지 바이오가스가 발생하였으나 대조군 대비 52% 수준이상은 회복되지 않았다. 항생항균물질 50 mg/L에서는 독성 저해 후 반응조 체류시간의 3배인 약 30일 정도 후 유기물 제거효율 및 바이오가스 회수가 가능하였으나, 100 mg/L 주입 후 40일 후에도 유기물 제거효율 및 바이오가스 발생량은 낮은 수준이었다. 따라서 혐기성 소화조에 고농도 항생항균물질이 유입되어 저해영향이 관찰되었을 경우 소화슬러지의 일부 또는 전체를 교체해야 할 것으로 판단된다. 혼합항생항균물질을 10~100 mg/L까지 주입하였을 경우 잔류항생항균물질 농도는 주입 6시간 후 CTC 80~90%, OTC 50~70%, SMZ 80~90%, TLS 20~40%가 반응조내에서 독성으로 작용한 후 입자성 유기물이나 EPS 등에 흡착되거나 소화온도에 의하여 감소한 것으로 판단된다. 따라서 혐기성 처리를 이용한 공정에 고농도 항생항균물질이 포함된 유기성폐수가 유입되었을 경우 공정의 유지관리에 어려움을 줄 수 있으며, 혐기소화조 유출수내 잔류항생항 균물질이 자연수계로 유입될 경우 2차 오염을 야기할 수 있기 때문에 항생항균물질의 사용의 적절한 관리가 요구되어 진다.

유기낙농가 사육환경 중 미생물 및 항생제 내성 분석 (Analysis of Microorganisms and Antibiotic Resistance in Organic Dairy Farm)

  • Recently, antimicrobial resistance of microorganisms has been a major concern of its relation to food safety and national health, therefore, customer's needs to organic animal food is still increasing. In this study, we reviewed the usage of antimicrobials in animal farms and antimicrobial resistance of microorganisms isolated from organic dairy farm environments. The isolates from dairy farms were Acinetobacter sp., A. lwoffi, A. johnsonii, A. towneri, Aerococcus viridans, Aeromonas media, A. veronii, Bacillus pseudofirmus, B. pumilus, B. licheniformis, Corynebacterium glutamicum, Escherichia coli, Enterococcus faecium, Lysinibacillus fusiformis, Paenibacillus illinoisensis, Staphylococcus epidermidis, S. hominis, Streptococcus equinus, S. lutetiensis, and Saccharomyces cerevisiae. Indicator microorganisms isolated from organic dairy farms were tested for susceptibility to 20 types of antimicrobials. E. coli (ATCC 25922) isolated from dairy farm fence showed resistance to 8 types of antimicrobials, such as oxacillin, penicillin, vancomycin, etc., and E. faecium isolated from feces showed resistance to 9 types of antimicrobials, such as cephalothin, oxacillin, streptomycin, etc., respectively. However, these results showed less antimicrobial resistance compare with customary dairy farm.

기능성 음료의 곰팡이 제어를 위한 천연항균제 활성 평가 (Evaluation of Antifungal Activity of Natural Antimicrobials in Fuctional Beverage)

  • 기능성 음료의 저장성을 향상시키고자, 7종류의 천연항균물질($\varepsilon$-Polylysine, 유카추출물, TLS(Vitamin $B_1$ 유도체), BMB-FS(황금추출물), 키토올리고당, KDSP 001, KDSP 002) 곰팡이 성장저해활성을 비교 평가하였다. Aspergillus niger, Penicillium citrimum, Rhizopus oryzae, Fusarium moniliforme, Mucor rouxii, 5 종류의 곰팡이를 상대로 활성을 살펴보았으며, 성장저해활성이 가장 뛰어난 물질의 최소저해농도를 조사하였다. 모든 곰팡이에 대하여 성장 저해활성을 보인 물질은 vitamin $B_1$, 유도체인 TLS 이었으며, 100 ppm 이상의 농도에서는 균사체가 성장하지 못하였다. 황금추출 물 BMB-FS의 경우 F. moniliforme을 상대로 100 ppm이상의 모든 농도에서 성장저해활성을 보였으며 P. citrinum와 M. rouxii의 경우 1,000 ppm이상의 농도가 되어야 균체 성장을 저해하는 것으로 나타났다. 가장 뛰어난 활성을 보여준 TLS의 최소저해농도(MIC)는 A. niger를 제외한 4가지 곰팡이에 대하여 60ppm으로 나타났으며, A. niger에 대한 MIC는 100ppm으로 분석되었다. 이 같은 결과는 기능성 음료의 저장기간을 연장시키는데 중요한 정보를 제공하며, 천연보존료 연구에 도움이 될 것이라 생각된다.

한국 농장동물 수의사의 항생제 신중사용에 대한 인식과 실천 (Korean farm animal veterinarians' perception and practice of prudent use of antimicrobials)

  • Antimicrobial (AM) resistance is a growing threat in human and veterinary medicine, spreading across species and perceived as One-Health issue. Prudent use of AM products is essential in mitigating this risk in both human and veterinary medicine. Farm veterinarians, responsible for prescribing appropriate AM and offering advice to farmers on their proper usage, are regarded as key players in the livestock industry. An online survey of farm veterinarians (n = 1,531) was conducted to assess their educational experience, beliefs, current status of prescription, practical behavior, and self-efficacy regarding the prudent use of antimicrobials (PUA). The data from 170 respondents were analyzed using descriptive statistics, regression and mediation analysis. Participants chose AM based on their experiences, academic resources, and peer veterinarians. Approximately 77% did not routinely conduct antibiotic-susceptibility-test. Most participants believed in the importance of PUA and the role of veterinarians to reduce AM resistance, but they reported a lack of awareness among farmers and insufficient national support interfere with their practical behavior with regard to PUA. Half of the participants had PUA-education, and 78.6% reported that education had a positive impact on their behavior with PUA. Self-efficacy partly mediates the relationship between belief and behavior for PUA, which accounted for 23.77% of the total effect that beliefs have on behavior for PUA. The findings of the study show that enhancing veterinarians' abilities to practice and their self-efficacy through education tailored to the current status of farms can contribute to the reduction of AMs in the livestock sector.

Effect of High Vanillin Treatment on Storage Quality of Fresh-cut Apples

  • The effect of post-cut vanillin treatment at high concentrations on changes of quality and microorganism in fresh-cut apples was studied. Apples (Malus domestica Borkh. cv. Fuji) were sliced, treated by dipping in different vanillin solution, 0, 40, 80, and 120 mM, packed in polyethylene bag, and then stored for up to 3 weeks at $4^{\circ}C$. Changes in total aerobic bacteria, yeast and molds, browning, soluble solids, and titratable acidity during storage were investigated. Growth of total aerobic bacteria throughout storage was strongly inhibited by vanillin regardless of treatment concentrations. Growth of yeast and molds was inhibited by vanillin of all concentrations until 2 weeks of storage. Levels of browning index, soluble solids, and titratable acidity were not significant difference among the treatment conditions until 2 weeks of storage. However, when stored for 3 weeks, browning index increased more at 80 or 120 mM vanillin, while soluble solids and titratable acidity more be decreased by 120 mM vanillin as compared with other treatment conditions. These results show that the usage of vanillin in processing of fresh-cut apples had a limitation for maintaining quality.
