식품보존제를 이용한 항균지 제조 (Manufacture of Antimicrobial Paper Using Food Preservative)

  • The functions of food packaging are not only prevention from physical damage and loss during carrying and transportation, but also extension of shelf-life by adding antimicrobial substrate in packaging materials. Consumption of active packaging is gradually increasing. With different dosage of potassium sorbate(P.S.), the food preservative agent, antimicrobial papers were made by internal and external application of starch. The antimicrobial action of the paper was analyzed by the halo test and the shake flask method. The mechanical properties and strength were also measured. Antimicrobial papers adding P.S. showed higher values in tensile index than adding starch. The antimicrobial paper using starch showed similar microbe decreasing rate as that using P.S. Though microbe decreasing rate was 21.9%, it showed possibility to make antimicrobial paper using food preservative.

국내 말에서 분리된 Staphylococcus aureus의 특성 및 약제 내성 양상 (Characteristics and Antimicrobial Resistance Patterns of Staphylococcus aureus Isolated from Horse)

  • 국내에서 사육중인 말에서 채취한 시료로부터 Stapylococcus aureus를 분리 동정한 결과 39.2%(58/148)의 분리율을 나타내었다. S. aureus의 혈장응고효소 생산능 시험에서는 98.3%가 양성반응을 나타내었고 신선 혈장의 희석농도는 1:2에서 가장 높은 양성반응을 나타내었다. 본 연구에서 최종 동정된 S. aureus의 기본적인 생화학 성상은 일반적으로 알려져 있는 S. aureus의 특성과 유사한 결과를 얻을 수 있었으나, urease test의 경우 전체 균주의 73% 만이 양성 결과를 나타내어 균주별로 다양한 반응 양상을 관찰 할 수 있었다. 또한 34종의 substrate에 대한 분해 양상에 대한 검사 결과 fructose, maltose, ${\alpha}$-hydroxy butyric acid, thymidine-5'-mono phosphate, uridine-5'-mono phosphate 등의 substrate에 대해서는 모든 균주가 이용능력을 가지고 있었지만 ribose, sorbitol, xylose, xylitol 등의 substrate에 대해서는 모든 균주가 기질 이용능력을 가지고 있지 않는 것으로 확인되었다. 그리고 PCR에 의한 S. aureus를 동정한 결과 108bp 크기의 band를 확인하였다. 국내 말에서 분리한 58주의 S. aureus에 대한 약제 내성 양상을 조사한 결과 spectinomycin, sulfonamides, erythromycin, tetracyclin, ciprofloxacin, neomycin, enofloxacin, penicillin 등의 약제에 높은 저항성을 나타내었다.

  • This paper reviews about synthesis of nanoparticles by spark discharge and applications. A method of catalytic activation with Pd and Pt aerosol nanoparticles produced by spark discharge was introduced. After annealing, the catalytically activated substrate placed into a solution for electroless silver deposition. The silver was then formed only on the activated regions of the substrate. Silver line patterns having a width of $18{\mu}m$ and a height of $1{\mu}m$ were created with the ability to be effectively reproduced. Antimicrobial nanoparticles such as silver were used for removal of bioaerosols. Silver nanoparticles deposited air filters such as ACF filters were evaluated by antimicrobial test.

Extraction and Application of Bulk Enzymes and Antimicrobial Substance from Spent Mushroom Substrates

  • Pleurotus ostreatus, P. eryngii, and Flammulina velutipes are major edible mushrooms that account for over 89% of total mushroom production in Korea. Recently, Agrocybe cylindracea, Hypsizygus marmoreus, and Hericium erinaceu are increasingly being cultivated in mushroom farms. In Korea, the production of edible mushrooms was estimated to be 614,224 ton in 2013. Generally, about 5 kg of mushroom substrate is needed to produce 1 kg of mushroom, and consequently about 25 million tons of spent mushroom substrate (SMS) is produced each year in Korea. Because this massive amount of SMC is unsuitable for reuse in mushroom production, it is either used as garden fertilizer or deposited in landfills, which pollutes the environment. It is reasonably assumed that SMS includes different secondary metabolites and extracellular enzymes produced from mycelia on substrate. Three major groups of enzymes such as cellulases, xylanases, and lignin degrading enzymes are involved in breaking down mushroom substrates. Cellulase and xylanase have been used as the industrial enzymes involving the saccharification of biomass to produce biofuel. In addition, lignin degrading enzymes such as laccases have been used to decolorize the industrial synthetic dyes and remove environmental pollutions such as phenolic compounds. Basidiomycetes produce a large number of biologically active compounds that show antibacterial, antifungal, antiviral, cytotoxic or hallucinogenic activities. However, most previous researches have focused on therapeutics and less on the control of plant diseases. SMS can be considered as an easily available source of active compounds to protect plants from fungal and bacterial infections, helping alleviate the waste disposal problem in the mushroom industry and creating an environmentally friendly method to reduce plant pathogens. We describe extraction of lignocellulytic enzymes and antimicrobial substance from SMSs of different edible mushrooms and their potential applications.

버섯 수확 후 배지의 산업적 활용 (Industrial utilization of spent mushroom substrate)

  • Over a million tons of spent mushroom substrate (SMS) are generated as by-products of mushroom cultivation every year in Korea. Disposal of SMS by mushroom farmers is difficult, therefore, recycling solutions that do not harm the environment are necessary. SMS consists of mushroom mycelia and residues of fruiting bodies, containing a variety of bioactive substances, such as extracellular enzymes, antimicrobial compounds, and secondary metabolites. This paper reviews utility of SMS for bioremediation, controlling plant disease, and production of lignocellulytic enzymes, organic fertilizer, and animal feed.

IBAD를 이용하여 알루미나 위에 HAp를 Coating하는 연구와 이의 항균력 시험 (Ag Impregnated HAp Coatings on Alumina Substrate by IBAD and Its Biological Test)

  • Hydroxyapatite($Ca_10(PO_4)_6(OH)_2$)는 생체친화성이 뛰어나고 인체의 뼈와 성분이 비슷하여 생체이식재료로 많이 사용되고 있다. 한편 HAp는 소결후 낮은 강도 때문에 HAp자체만으로는 생체이식재료로서의 강도를 기대할 수가 없다. 그러나 알루미나는 생리화학적인 환경에서 높은 강도와 매우 안정한 성질을 가지고 있기 때문에 알루미나 위에 HAp를 coating시켜 사용하려는 시도가 있었다. 본 연구는 알루미나와 HAp의 장점만을 이용하고, HAp에 항균성 물질을 첨가하여 항균효과를 나타내도록 하였다. 항균test는 생체재료이식시 가장 많은 감염을 일으키는 bacteria를 사용하여 실시하였다. 먼저 HAp에서 $Ag^+$$Ca^{2+}$이 이온교환을 시키기 위하여 20ppm, 100ppm의 $AgNO_3$용액에 48시간동안 처리하고, 이 재료를 사용하여 항균test를 하였다. 항균test에 사용된 bacteria는 gram negative bacteria인 Escherichia coli와 Pseudomonas aeruginosa, gram positive bacteria인 Staphylococcus epidermidis를 사용하였고, 항균test결과는 매우 좋은 것으로 나타났다. $AgNO_3$용액에서 처리한 sample들은 SEM과 XRD를 사용하여 표면구조와 성분을 조사하고, 유사생체용액(Simulated Body Fluid)에서 Ag release curve를 조사하였다.

  • To develop natural antimicrobial agents for keeping qualities of postharvested greenhouse produce the antimiocrobial actions of Polygonum cuspidatum Sieb. et Zucc. extract , which showed remarkable antimicrobial effects against microorganism causing the postharvest decay of greenhouse produce, were investigate. In the inhibitory experiment of enzymes related to energy production metabolism hexokinase activities decreased to 73% and 68% by treating with Polygonum cuspidatum Sieb. et.Zucc. extract and Eugenia caryophyllata Thumnberg extract in comparison with control, respectively. Direct visualization of microbial cells by using both transmission electron microscope and scanning electron microscope showed that microbial cell membrane was destroyed by treating with the dilute extract solution. this change of celluloar membrane permeability could be identified in the experiment that 0-nitrophenyl-${\beta}$-D-galactopyrano-side(ONPG), the artificial substrate of ${\beta}$-galactosidase, was hydrolyzed in the presence of the extract, indicating that the membrane was perturbed. The separation and identification of the most antimicrobialo substances isolated from Polygonum cuspidatum Sieb et. Zucc. extract and Eugenia caryophyllata Thunberg extract were carried out by using gas chromatography and mass spectrometry 9GC/MSD), which were identified as eugenol. As a result, the functionality of Polygonum cuspidatum Sieb. et Zucc. extract and Eugenia caryophyllata Thunberg extract as antimicrobial agents for keeping qualities of postharvested greenhouse produce may be recommended.

시설채소산물의 선도유지를 위한 한국산 약용식물추출물의 항균특성 (Antimicrobial Activities of Korean Medicinal Herb Extracts for Preserving Greenhouse Fresh Produce)

  • Korean medicinal herb extracts(KMHE) were applied to the preservation of greenhouse produce in order to prove their effectiveness. KMHE showed remarkable antimicrobial effects against Bacillus cereus, Peudomonas syringae, and Corynebacterium xerosis causing the postharvest decay of greenhouse produce. Among KMHE the extracts of Rheum palmatum L. and Coptis chinensis Franch most obviously inhibited the growth of microorganims causing the Postharvest decay of greenhouse produce, which destroyed to undetectable levels when treated with more than 500ppm of KMHE. The activities of KMHE were stable in the wide spectrum of pH and temperature. Direct visualization of microbial cells by using both transmission electron microscope and scanning electron microscope showed microbial cell membrane the function of which was destroyed by treating with the dilute solutions of KMHE. This change of cellular membrane permeability could be identified in the experiment that O-nitrophenyl-$\beta$-D-galactopyranoside(ONPG), the artificial substrate of $\beta$-galactosidase, was hydrolyzed in the presence of KMHE, indicating that the membrane was perturbed by KMHE.

  • Sophorolipid, a glycolipid type biosurfactant produced from Candida Bombicola, inhibited the growth of bacteria and fungi. Between two types of sophorolipid, acid and lactone form, the latter had more strong antimicrobial activity. Culturing parameters including oil substrate, initial glucose concentration, aeration, culture mode, affected the ratio of two forms. The fatty acid moety varied depending on the oil having different fatty acid chains. Minimum inhibitory concentration of lactonic sophorolipid toward Propionibacterium acne was below 0.5ppm. This result indicated the potentials of sophorolipid as antimicrobial agent in various area including cosmetics.

Isolation of Antimicrobial Substances from Hericium erinaceum

  • Mycelium of Hericium erinaceum isolate KU-1 was cultured in liquid medium (HL medium) and solid medium (Ko medium) at pH 4.0 in $28^{\circ}C$. 1.0% glucose or fructose was the most favorable carbon source, and 0.2% amonium acetate or $NaNO_3$ was an exellent nitrogen source for mycelial growth as well as production of antimicrobial substances. The mixture of saw dust 70% with rice bran 30% (SR medium) was the substrate for formation of sporophores. The active substrates in extracts from mycelium, culture filtrate and fruiting body were separated by TLC. The solvent for TLC was EtOAc: Chloroform: MeOH (10 : 5 : 10). Phenol-like substances appeared at Rf $0.5{\sim}0.9$, and fatty acid-like substances appeared at Rf $0.1{\sim}0.2$. The purified materials from the extracts showed antimicrobial effects to Escherichia coli, Bacillus subtilis, Staphylococcus aureus, Aspergillus niger, Candida albicans and Microsporum gypseum. The S. aureus was the most inhibited. Minimal inhibitory concentration (MIC) of purified white powder and the Hercenone derivatives against S. aureus were $5.65\;{\mu}g/ml$ and $1.85\;{\mu}g/ml$, respectively.
