N-(fluorodichlomethylthio) phthalimide-silver complexes were prepared and investigated the antimicrobial activity on rubber mat manufactured with waste rubber. We are exposed to harmful bacteria and fungi all the time. We manufactured antimicrobial mat to be imposed to mats that it can prevent generation of bacteria and microorganisms, and restrict their reproduction. Infection of medical devices causes significant morbidity and mortality. For aim of this study, we measured the antimicrobial mat manufactured with N-(fluorodichlomethylthio) phthalimide-Ag complex by CCD, FT-IR and NMR. The effect of mole ratio of N-(fluorodichlomethylthio) phthalimide-Ag complex on antibacterial activity to bacteria and fungi is investigated. Reduction rate is evaluated using the Quinn method. Escherichia Coli is effectively inhibited than Staphylococcus aureus by antimicrobial mat.
A bacterium strain called Bacillus coagulans was isolated from an industrial wastewater drainage and selected for its antimicrobial activities against bacteria and fungi. Characterization studies strongly suggested that this strain is Bacillus coagulans. Antimicrobial activity was found against gram-positive, gram-negative bacteria and yeast strain. Maximal activity was observed after 24 h when incubated at $30^{\circ}C$ and pH 8. The activity was found to be stable at $75^{\circ}C$ for 30 min and at pH range of 2-12. Analysis of the antimicrobial compound by SDS-PAGE suggested a molecular mass of approximately 7.5 KDa. The substance was characterized as a bacteriocin, because of its proteinaceous nature and low molecular weight. Our bacteriocin could potentially be used as a food preservative, because of its thermostable property and broad antimicrobial spectrum.
The solvent extracts of Ulmi cortex, which were extracted by using several solvents with different polarities, were prepared for utility as a natural preservatives. The antimicrobial activities and cell growth inhibitions were investigated to each strain with the different concentrations of Ulmi cortex extracts. Methanol extract showed the highest antimicrobial activity. The methanol extract was represented the broad antimicrobial activities for the gram positive and negative strains. Minimum inhibitory concentrations (MIC) for each strains were appeared to around 0.3mg/ml at each of Bacillus cereus, Bacillus subtilis, and Staphylococcus aureus. The cell growth inhibitions were not shown on Lactobacillus bulgaricus, Lac tobacillus plantarum, and Bifidobacterium bifidum, but greatly on the Clostridium butyricum. The meth anol extracts were further reextracted sequentially with hexane, chloroform, ethyl acetate, and butanol for purifying crude methanol extracts. The extract, which was reextracted by butanol, showed the highest antimicrobial activity.
Lactic acid bacteria in Kimchi fermentation were tested for inhibitory activity against Gram positive bacteria and Gram negative bacteria. The Lactobacillus brevis (KCCM 35464) was found to produce a antimicrobial substance. It showed relatively wide range of inhibition spectrum against gram positive and gram negative bacteria and maintained the inhibitory activity between pH 4.0 and pH 9.0. The antimicrobial substance was obtained in the stationary growth phase and was purified by gel chromatography. The inhibitory effect of the antimicrobial substance on sensitive bacterial strains was determined by filter paper test. The activity of antimicrobial substa- nce was stable at 75$\circ$C. On the basis of its electrophoretic pattern is SDS-PAGE, antimicrobial substance appeared as a single band of 59 KDalton.
Various kinds of water soluble N,N,N-trimethylchitosan ammonium chloride(TMC) with different molecular weights were synthesized to examine the antimicrobial activity against Methicillin Sensitive Staphylococcus aureus(MSSA) and Methicillin Resistant Staphylococcus aureus(MRSA), which causes serious hospital infection, and to apply them to antimicrobial finishing agents for textiles. Chitosan samples were highly deacetylated with sodium hydroxide solution and degraded with hydrogen peroxide to control the molecular weight. TMC has the antimicrobial activities against MRSA and MSSA. TMC showed an excellent antimicrobial activity below the molecular weight of 70,000, especially at 40,000. The minimum inhibitory concentration (MIC) of TMC with optimum molecular weight against MRSA and MSSA was 250ppm. Because MRSA did not resist TMC in the subculture test of bouillon medium, it was expected that the successive use of TMC against MRSA was possible.
As part of our search for less toxic antimicrobial substances from natural resources, naringin was isolated from feels of Citri fructus and then hydrolyzed to naringenin. The antimicrobial activity of naringenin was examined by measuring the minimal inhibitory concentration(MIC) against fourteen species of bacteria. The antimicrobial activity of narngenin in combination with rutin or hesperetin was evaluated by checkerboard method. Among fourteen species tested, the antimicrobial activity of naringenin was the most prominant against Staphylococcus aureus and Shigella boydii showing MIC of $100\;{\mu}g/ml$ for both species. Combinations of naringenin with rutin or hesperetin showed synergism against several species of bacteria, but no antagonism was observed.
To developed natural food preservatives, ethanol and water extracts were prepared from the defatted camellia(Camellia japonica L.) seeds. Antimicrobial activities were examined against 14 microorganisms which were food borne pathogens and/or food poisoning microorganisms and food-related bacteria and yeasts. Ethanol extract showed antimicrobial activities against several microorganisms tested, but not on lactic acid bactria. Especially, minimum inhibitory concentrations(MIC) for yeasts were as low as 1mg/ml. Water extract exhibited antimicrobial activities for the yeasts tested, but almost bacteria were not observed. The ethanol extract was fractionated in the order of hexane, chloroform, ethylaetate and butanol fractions to test antimicrobial activity. The strongest activity for the tested yeasts were found in the butanol fractions, but bacteria were chloroform fraction. Hansenula anomala treated with ethanol were observed by scanning electron micrograph, and the results exhibited morphological changes, including the irregularly contracted cell surface and expanded ellipsoidal shape.
This study was performed to investigate the antimicrobial effects of Portulaca oleracea extracts against food-borne pathogens. First, the Portulaca oleracea was extracted with methanol at room temperature, and then further fractionated by using petroleum ether, chloroform, ethyl acetate and methanol, respectively. The antimicrobial activity of the Portulaca oleracea extracts was determined using a paper disc method against food-borne pathogens and food spoilage bacteria. The ethyl acetate extracts of Portulaca oleracea showed the highest antimicrobial activity against Staphylococcus aureus and Shigella dysenteriae. There was also a synergistic effect of the combined extracts of Portulaca oleracea and Indigofera kirilowii as compared to each extract alone. Finally, the growth inhibition curve of ethyl acetate extracts of Portulaca oleracea against Staphylococcus aureus and Shigella dysenteriae was determined The ethyl acetate extract of Portulaca oleracea showed strong antimicrobial activity against Staphylococcus aureus at the concentration of 4,000 ppm. The 4,000 ppm of ethyl acetate extract from Portulaca oleracea, retarded the growth of S. aureus by more than 24 hand Shigella dysenteriae up to 12 h at $37^{\circ}C$.
Most people use cosmetics to protect their skin. Preservatives are often used to prevent their contamination upon use. There has been a great demand for natural preservatives due to recent reports on the side effects of parabens. Therefore, we evaluated the antimicrobial activities of Lonicera japonica and Magnolia obovata extracts and determined their potential as natural preservatives. We found that the 50% ethanol extract from L. japonica had antibacterial activity only against S. aureus and P. aeruginosa, while the ethyl acetate fraction showed antimicrobial activity against all six microbial strains tested. On the other hand, the 70% ethanol extract and the ethyl acetate fraction from M. obovata showed antimicrobial activity against all six strains. A synergistic effect against S. aureus, B. subtilis, and C. albicans was confirmed when two ethyl acetate fractions having antimicrobial activity against all six strains were used in combination. Synergistic activity against B. subtilis was also confirmed through kill-time analysis. High-performance liquid chromatography was performed to identify the components of each extract. Based on the minimum inhibitory concentration and the results of a disc diffusion assay, we confirmed that caffeic acid and luteolin influenced the antimicrobial activity of L. japonica and that the antimicrobial activity of M. obovata was influenced by the interaction of magnolol and honokiol with other components. Therefore, this study suggests that the combination of L. japonica and M. obovata extracts may be used as a plant-derived natural preservative.
Objectives : The present study was designed to investigate effects of mixed extracts (9:1, v/v) of Hwangryunhaedoktang, Chamaecyparis obtusa essential oil. We evaluated the antimicrobial and antioxidant activity by manufacturing mixed extracts as the materials for functional medicinal herb cosmetics. Methods : We performed antimicrobial were tested microbes (Staphylococcus aureus, Staphylococcus epidermidis, Escherichia coli, Candida albicans, Propionibacterium acnes) by disc diffusion method and measure clear zone. Antioxidant activities were measured by DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity. Results : Antimicrobial activities of mixed extracts against S. epidermideis, P. acnes, S. aureus, E. coli, and Candida. A were $10.9{\pm}3.0mm$, $16.9{\pm}2.0mm$, $9.2{\pm}2.0mm$, $10.3{\pm}1.0mm$, $11.8{\pm}3.0mm$ respectively. The Chamaecyparis obtusa essential oil had the highest antimicrobial activities against S. epidermideis, P. acnes, S. aureus, E. coli, and Candia. A and clear zone of microbes ware $16.7{\pm}3.0mm$, $28.8{\pm}0.2mm$, $15.9{\pm}2.0mm$, $11.5{\pm}1.0mm$, $16.3{\pm}3.0mm$. Hwangryunhaedoktang extract showed antimicrobial activity but only P. acnes and S. aureus The antioxidant activities of the mixed extracts were tested through the evaluation of DPPH radical scavenging activity. The 100 % mixed extracts were found to have 90 % DPPH radical scavenging activity. The mixed extracts was presented similar antioxidant activities compared with that of ascorbic acid. As a result, A mixture extract is expected to have antimicrobial effects and free radical scavenging activity was found. Conclusions : Accordingly, It can be concluded that mixed extracts has the potential to cosmetic material.
