Characterization of Bacteriocin-Like Substances Produced by Bacillus subtilis MJP1 (Bacillus subtilis MJP1이 생산하는 Bacteriocin-Like Substances)

  • The MJP1 bacterial strain, which possesses antifungal activity, was isolated from meju and identified as Bacillus subtilis based on its morphological and biochemical properties, as well as its 16S rRNA sequence. Antimicrobial activity was found against various species of Gram-positive bacteria, yeasts, and molds, including food-spoilage microorganisms. The antifungal activity was found to be stable after heat and proteolytic enzyme treatment, and in the pH range of $6.0{\sim}10.0$. The antibacterial activity was stable in the pH range of $6.0{\sim}10.0$, but about 50% of the activity was lost after 24 hr at $30^{\circ}C$. The antibacterial compound was also inactivated by proteolytic enzyme treatment, indicating its proteinaceous nature. The apparent molecular masses of the partially purified antifungal and antibacterial compounds, as indicated by using the direct detection method in Tricine-SDS-PAGE, were approximately 2.4 kDa and 4.5 kDa, respectively. These studies suggest that B. subtilis MJP1 produces two bacteriocin-like substances with antifungal and antibacterial activities.

  • Selective culture of actinomycetes from soil microbes and their antibiotic producing characters by agar-disk method were examined. Some of the organisms which produced antibiotics on agar disk did not produce antibiotics in liquid culture. Further examination indicated that production of antibiotic was dependent on the composition of medium. Many streptomycestes produced antibacterial substances in tryptic soy broth but others produced antifungal antibiotics in V-8 broth. Production of antibacterial substances by Streptomyces sp. was also dependent on the medium composition.

  • The report explores the possibility that the single extract of natural substances and the mixture of single extracts of natural substances create a synergy effect to increase the antimicrobial activity. It also compares the previous researches to find out if the natural medicinal herbs’ extract has antimicrobial activity on dandruff causative organism, Malassezia furfur. For the experiment on the mixture of single extracts of natural substances, the results are like following: 1. Staphylococcus aures’s antibacterial activity is resistant to mixture of three natural substances. 2. Escherichia coil’s antibacterial activity is resistant to mixture with Coptidis rhizoma. 3. Candida albicans’ antifungal activity is resistant to mixture with Chinese galls, which means the different results may be expected when tested with each germ. 4. On the other hand, Malassezia furfur has no antifungal activity on the single extract of natural substances and shows weak antifungal activity, whose diameter is 3.20 mm when tested with the mixture of 50% of Coptidis rhizoma and 50% of Phytoncide. The result is totally different from the one on the same eumycetes, C. albicans. That is because M. furfur has more lipophilic chemicaled cell walls than C. albicans does and it also consists of lamella layer, inner plasma membrane and intermediate multiple layers.

  • Most reports demonstrated the substrate specificity-based kinetic properties of chitin or chitosan degrading enzymes. However, there is virtually less information on the high quality and quantity production of chitin or chitosan hydrolysates having a larger than (GlcN)7 from the hydrolysis of high molecular weight chitosan using specific enzymes and their biological activity. Therefore, the production of such molecules and the discovery of such enzyme sources are very important. Fortunately, the author has established a mass production method of chitosan hydrolysates (GlcN)n, n=2-13 that have been characterized as a potent antioxidant substance, as well as antifungal and antibacterial activities against Penicillium species and highly selective pathogenic bacteria. In addition, preclinical studies using (GlcN)n, n=5-25 demonstrated that these molecules played a very important role in maintaining biometric balance. Collectively, it is implicated that the application of these mixed substances to foods with significant biological activity is very encouraging.

  • 토마토의 엽권에서 분리한 Fusariym graminearum이 분비하는 물질은 벼 도열병균(Pyricularia oryzae)의 여러 종의 식물병원 진균에 대한 항균활성을 나타내었으며, 이러한 활성물질을 PDA에서 본 균을 배양 한 후 chloroform으로 추출하여 분리정제 하였다. HPLC에 의하여 5종류의 활성 물질을 분획하였으며, 그중 1번(F402) 화합물을 벼 도열병균(P. oryzae)을 포함한 22개 식물 병원 진균에 대하여 살균 활성범위를 조사한 결과, 이 화합물은 50$\mu\textrm{g}$/ml 농도에서 Pythium ultimum, Rhizoctonia solani, Sclerotinia sclerotiorum은 전혀 억제하지 못하였으며, Phytophthora spp., Cladosporium fulvum, Fusarium spp., Corynespora cassicola에는 어느 정도의 활성이 있었지만 낮게 나타났고, P. oryzae, Cochliobolus miyabeanus, Alternaria solani는 100% 억제하여 활성이 높게 나타났다. 또한 장내 세균에 대한 활성을 MIC로 비교할 때 Streptococcus pyogenes, Streptococcus faecium에 대하여는 각각 12.5, 25 $\mu\textrm{g}$/ml였고 Staphylococcus aureus는 25-50$\mu\textrm{g}$/ml으로 나타났으며, Pseudomonas aeruginosa, Salmonella typhimurium, Klebsiella aerogenes, Enterobacter cloacae에서는 100$\mu\textrm{g}$/ml 이상으로 활성이 나타나지 않았다. F402를 200$\mu\textrm{g}$/ml의 농도로 직접 살포한 식물체에서의 방제효과는 벼도열병, 벼 깨씨무늬병, 보리 흰 가루병에 대하여는 80%이상이었으나, 벼 잎집무늬마름병, 오이 잿빛곰팡이병, 토마토 역병, 밀 녹병에서는 낮았다.

Production of Antimicrobial Substances by Strains of Myxobacteria Corallococcus and Myxococcus (Corallococcus와 Myxococcus 속 점액세균 균주들에 의한 항균 물질의 생산)

  • We prepared culture extracts of 174 Corallococcus and 207 Myxococcus strains isolated in Korea, and compared their antimicrobial activity against Candida albicans, Pseudomonas aeruginosa, and Staphylococcus aureus. The percentage of strains showing antifungal activity was lower in Corallococcus (7.5% [13 of the 174 strains]) than in Myxococcus (51.7% [107 of the 207 strains]). However, the percentage of strains exhibiting antibacterial activity was higher in Corallococcus (12.1% [21 strains]) than in Myxococcus (1% [2 strains]). The culture extracts of 6 Corallococcus strains inhibited both P. aeruginosa and S. aureus and displayed similar high-performance liquid chromatography chromatograms, although the shapes of their fruiting bodies were dissimilar. The rate of production of antibacterial substances was the highest when the strains were cultured in CYS medium for more than 6 days.

  • Marine sponges have been considered as a drug treasure house with respect to great potential regarding their secondary metabolites. Most of the studies have been conducted on sponge's derived compounds to examine its pharmacological properties. Such compounds proved to have antibacterial, antiviral, antifungal, antimalarial, antitumor, immunosuppressive, and cardiovascular activity. Although, the mode of action of many compounds by which they interfere with human pathogenesis have not been clear till now, in this review not only the capability of the medicinal substances have been examined in vitro and in vivo against serious pathogenic microbes but, the mode of actions of medicinal compounds were explained with diagrammatic illustrations. This knowledge is one of the basic components to be known especially for transforming medicinal molecules to medicines. Sponges produce a different kind of chemical substances with numerous carbon skeletons, which have been found to be the main component interfering with human pathogenesis at different sites. The fact that different diseases have the capability to fight at different sites inside the body can increase the chances to produce targeted medicines.

  • Green tea has been shown to have multifunctional health-promoting properties including cholesterol level control an antidiabetic effect and anticancer, antioxidant, and antimicrobial properties, inboth in vivo and in vitro experiments.We earlier reported antifungal and antibacterial effects of green tea extract by investigating cell membrane functions. The purpose of the present study was to purify and identify antimicrobial substances from green tea extract. Such materials were extracted from green tea (Camellia sinensis. var. sinensis) and purified by high-performance liquid chromatography. The antimicrobial substances in the extract were identified as epicatechin gallate and epigallocatechin gallate by nuclear magnetic resonance spectrophotometry.

Searching of Antimicrobial Active compounds from Microalgae (해양 미세조류로부터 항균성 물질의 탐색)

  • As a part of the investigation on useful compounds from microalgae, and its recently that marine planktonic algae have been recognized as potial sources of antibacterial and antifungal substances, we searched for antimicrobial active compounds from the extracts of six microalgae-Lyngbya sp., Tetraselmis sp., Microcystis sp., Chlorella sp., Navicula sp. and Rhalassiosira sp.-treated with several solvents. There were two active species-Lyngbya ap., Teraselmis sp.- in the antimicrobial activity test to bacteria, yeast and molds, especially the activity existed in the extracts by ethyl acetate of supernatants to the microalgae incubatio. and there won’t any activity in two diatoms to the test microorganism.

  • Antimicrobial substances were screened by paper disk plate method in marine sponges, Halichondria okadai, Halichendria sp., H iaponica and Haliclona Pemollis, collected from the south coast of Korea. Antibacterial components were detected in two species, H okadai and Halichondria sp.. Three components such as benzoic acid, okadaic acid(OA) and dinophysistoxin-1(DTX1) were identified from these sponges as the antimicrobial compounds by MS and NMR spectral data. OA$(550{\~}600{\mu}g/kg)$ and $(400{\~}490{\mu}g/kg)$ were determined from the wet H okadai and Halichondria sp., respectively, by using fluorometric HPLC analysis with 9-anthryldiazomethane(ADAM) as fluorescent labelling reagent.
