Induction of ROS-dependent apoptosis by ethanol extract of Hizikia fusiforme in HT29 colon carcinoma cells (톳 에탄올 추출물에 의한 HT29 결장암 세포의 ROS 의존적 세포사멸 유도)

  • Hizikia fusiforme, a type of brown algae, is widely used in Asian cuisine. It has been reported to have various pharmacological effects. In this study, the effects of the ethanol extract from H. fusiforme (EAHF) on the proliferation of human colon carcinoma cells were investigated. The effect on the survival of human hepatocarcinoma and colon carcinoma cells was examined, and results revealed that the anti-proliferative effects of EAHF were higher in colon carcinoma cells than in hepatocarcinoma cells. The inhibition of proliferation of HT29 colon carcinoma cells by EAHF treatment was closely related to the induction of apoptosis. EAHF treatment also increased caspase activity and poly(ADP-ribose) polymerase degradation, induced mitochondrial dysfunction, altered Bcl-2 family protein expression, and increased the rate of cytochrome c released from the mitochondria into the cytoplasm. Furthermore, the production of reactive oxygen species (ROS) was markedly stimulated by EAHF treatment, and when ROS production was blocked, EAHF-induced cytotoxicity was significantly attenuated. These results indicate that the anticancer activity of EAHF in HT29 colon carcinoma cells was induced by ROS-dependent mitochondrial impairment. While EAHF exhibited potent anticancer activity in colon carcinoma cells in this study, further studies on the active components of EAHF and their efficacy should be performed.

PD-L1 Aptamer-functionalized Liposome Containing SAHA for Anti-lung Cancer Immunotherapy

  • Liposomes are one of the most actively studied and promising drug delivery systems for the treatment of various diseases. In this study, an aptamer-conjugated liposome called "aptamosome" was used, in which an anti-PD-L1 aptamer targeting cancer cells was conjugated to the liposome. These aptamosomes showed remarkable cellular uptake and efficient delivery to Lewis lung carcinoma 2 (LL/2) cancer cells. In addition, suberoylanilide hydroxamic acid (SAHA), a histone deacetylase inhibitor (HDACi), was delivered through this aptamer to induce a strong anticancer immunotherapeutic effect. The results of this study showed that when LL/2 cells were treated with SAHA-entrapped aptamosome [SAHA] and liposome [SAHA] and free SAHA, aptamosome [SAHA] improved cell death compared with that of liposomes [SAHA] or free SAHA, and it has demonstrated anticancer efficacy. Moreover, aptamosome [SAHA] induce the secretion of chemokines that promote the migration of activated T cells into tumor tissues. Finally, in vivo experiments showed that aptamosome [SAHA] significantly inhibited the growth rate of LL/2 tumors. Therefore, liposomes combined with an anti-PD-L1 aptamer for efficient SAHA delivery are suggested as an excellent model for drug delivery systems suitable for targeting cancer cells.

Effect of CP-2 Extracted from Coptis and Croton tiglium L. on the Growth and Steroidgenesis of Follicular Granulosa Cells (콥티스속 근경과 탈지된 클로톤 종자의 혼합 추출물(CP-2)이 난포 Granulosa Cell에 미치는 영향)

  • We investigated the effects of CP-2 extracted from the mixture of Copis and Croton tiglium L, which showed very high cytotoxic effect to the various tumor cells, on the growth and steroidenesis of primary and transformed cell lines PA-GS6 and PO-GRS1 by cotransfectionwith SV40 and Ha ras oncogenes. CP-2 inhibited the growth of PA-GS6 and PO-GRS1 in a dose dependent manner when the growth of them was measured by cell number and by protein content, while CP-2 did not affect the growth of primary granulose cells. Productions of progesterone ofprimary and transformed granulosa cells were stimulated by forskolin, but this stimulatory effect was blocked by treatment of CP-2. Clinical application of CP-2 asa new anti-cancer drug and utilization of transformed granulosa cells as a model system for the screening of anti-cancer drug were also discussed.

Effect of AC-264, a Novel Indole Derivative, on Apoptosis in HL-60 Cells

  • The anticancer effect and apoptotic mechanism of a novel indole derivative AC-264, a lead derived from a chemical library, were investigated in human promyelocytic leukemia HL-60 cells. HL-60 cells treated with AC-264 at various concentrations showed the morphological features of apoptosis, such as plasma membrane blebbing and cell shrinkage. AC-264 exhibited cytotoxic effect in various cancer cell lines with different degrees of potency. Especially, AC-264 was effective on increasing the population of apoptotic cells in HL-60 cells, as detected by the number of cells stained with Annexin V and PI. Furthermore, AC-264 activated caspase-3 enzyme activity and induced internucleosomal DNA fragmentation. These results indicated that AC-264 produces anti-cancer effect via apoptotic cell death by activating caspase-3 and inducing internucleosomal DNA fragmentation in HL-60 cells.

  • The purpose of this study was to characterise the subacute toxic potency of i.v. administered KHPC-005 and 006. Few test compounds-related toxic effects were observed in body weight gain, clinical signs, urinalysis, hematological parameters and serum biochemical values. Gross necropsy and histopathology revealed no evidance specific toxicity. Our data indicated that no-observed effect level of KI-IPC-005 and 006 were estimated to be 10mg/kg and 4mg/kg in male rats, and 10mg/kg and 1.33mg/kg in female rats, respectively.
