The in vitro inhibitory activities of essential oils from Thymus magnus and T. quinquecostatus as well as their main constituents were evaluated against susceptible and resistant species of Streptococcus pneumoniae, Staphylococcus aureus, Salmonella enteritidis, and Salmonella typhimurium. Notably, the essential oil fraction of T. magnus and its main components displayed significant inhibitory action against both antibiotic-susceptible and resistant strains of S. pneumoniae, S. aureus, and S. typhimurium with minimum inhibitory concentrations (MICs) ranging from 0.125 to 8 mg/mL. The differential MIC values imply that the oil fraction and its main components exhibit distinct patterns of activity against the tested bacterial species. Moreover, the disk diffusion test revealed that the inhibitory activities of oil fraction and components were dose-dependent. Data from the checkerboard titer test confirmed synergism between the antibiotic, norfloxacin, and T. magnus oil or thymol, particularly against the resistant strains of S. aureus.
The in visto inhibitory activities of essential oils of the Rosmarinus officinalis as well as its main constituents were evaluated against antibiotic-susceptible and -resistant strains of Staphylococcus aureus, streptococcus pneumoniae, Salmonella enteritidis and S. typhimurium. The essential oil fraction of R. officinalis and its main components, 1,8-cineole and camphor, exhibited significant inhibitory activities against most of the tested strains in this study, with MICs(minimum inhibitory concentrations) racing from 0.5mg/ml to 16mg/ml. The total oil fraction showed higher activity than its main components, 1,8-cineole and camphor against S. aureus strains. No remarkable differences were evident between MICs of the susceptible and resistant strains of S. aureus. Among the tested strains, S. pneumoniae CCARM 3523, the resistant strain to norfloxacin, oxacillin and erythromycin exhibited significantly lower sensitivity to the tested oils than antibiotic-susceptible strain. The oils revealed mostly higher inhibitory activity against S. typhimurium than against S. enteritidis.
This study purposes to find the best one for dyeing with artemisia in order to develop a more efficient method of dyeing with artemisia and investigates the antibiotic activities of artemisia. After dyeing cotton, rayon, silk, wool and nylon fabrics with dyebath extracted from artemisia, observation on dyeability by temperature, concentration and time has been carried out. And I took an observation on color change, color fastness to washing and color fastness to light after mordanting treatment. Also observation on the antibiotic properties of non-dyed fabrics, artemisia-dyed fabrics and artemisia-dyed fabrics with post-mordant has been performed to investigate the antibiotic activities of artemisia. And the following results have been obtained. 1. The dyeability of artemisia was better for protein fabrics such as silk or woo and amide-based synthetic fabrics like nylon than for cellulose fabrics such as cotton and rayon. 2. For all tested fabrics, the dyeability was increased as the concentration of artemisia dyebath, dyeing time and dyeing temperature was increased. Especially the dye ability for protein fabrics was most affected by the temperature, and high dyeability was obtained at high temperatures. 3. For the color fastness of artemisia-dyed fabrics with mordanting treatment, the color fastness to washing was good with grade of 4-5 while the color fastness to light ranged from grade 2 to grade 3, which requires improvement for practical use. 4. The artemisia-dyed fabrics were shown to have much higher antibiosis than the non-dyed fabrics, and the artemisia-dyed fabrics with mortanting treatment showed the following result; Cu 99.9%, Al 64% and bittern 64.7%.
To investigate natural antibiotics from plant essential oils and to evaluate their synergism with current antimicrobial drugs in inhibiting antibiotic-resistant strains of Streptococcus pneumoniae. The minimal inhibitory concentrations (MICs) of eleven plant essential oils and their main components were established for two antibiotic-susceptible and two antibiotic-resistant strains of S. pneumoniae, using broth microdilution tests. Potential synergism with oxacillin, norfloxacin, or erythromycin was evaluated using a checkerboard microtitre assay. Among the tested oils, Mentha piperita oil and its main component, menthol, exhibited the strongest inhibitory activities against all of the tested strains. The activity of antibiotics against antibiotic-resistant strains of S. pneumoniae was enhanced significantly by combination with Mentha piperita oils and its main component, menthol. In conclusion, the combination Mentha piperita essential oil or menthol with antibiotics could be used to reduce the effective dose of antibiotic and to modulate the resistance of S. pneumoniae strains.
Antitumor antibiotic material was produced by Streptomyces sp. YBE-316 which was isolated from soil, and the optimal culture conditions for the antitumor antibiotic material production were as follows; 2.0% (w/v) sucrose, 0.8% (w/v) polypeptone, 0.4% (w/v) yeast extract, 0.2% (w/v) K$_{2}$HPO$_{4}$, pH 7.0, at 30$\circ$C, 150 rpm and for 100 hours culture. The antitumor antibiotic material had strong antitumor antibiotic activities against most testing tumor cell lines, gram positive and negative bacteria, yeasts, and, especially, Penicillium chrysogenum in fungi.
A synthetic cecropin A(1-13)-melittin(1-13) [CA-ME] hybrid peptide was known to be an antimicrobial peptide having strong antibacterial, antifungal and antitumor activity with minimal cytotoxic effect against human erythrocyte. Analogues were synthesized to investigate the influences of the flexible hinge region of CA-ME on the antibiotic activity. Antibiotic activity of the peptides was measured by the growth inhibition against bac-terial, fungal and tumor cells and vesicle-aggregating or disrupting activity. The deletion of Gln-Gly-Ile (P1) or Gly-Gln-Gly-Ile-Gly (P3) from CA-ME brought about a significant decrease on the antibiotic activities. In contrast, Gly-Ile-Gly deletion (P2) from CA-ME or Pro insertion (P5) instead of Gly-Gln-Gly-Ile-Gly of CA-ME retained antibiotic activity. This result indicated that the flexible hinge or β-bend structure provided by Gly-Gln-Gly-Ile-Gly, Gln-Gly, or Pro in the central region of the peptides is requisite for its effective antibiotic activity and may facilitate easily the hydrophobic C-terminal region of the peptide to penetrate the lipid bilayers of the target cell membrane. In contrast, P4 and P6 with Gly-Gln-Gly-Pro-Gly or Gly-Gln-Pro in the central region of the peptide caused a drastic reduction on the antibiotic activities. This result suggested that the con-secutive β-bend structure provided by Gly-Gln-Gly-Pro-Gly or Gly-Gln-Pro in the central hinge region of the peptide seems to interrupt the ion channel/pore formation on the target cell membranes.
Objectives : Since cosmetics have been one of the help of life, unlike medicine, natural products have been used for cosmetics, generally giving the image of safety and relief compared to synthetic products. Among them, Houttuynia cordata has been known as a useful herbal medicine with antibiotic, anti-inflammatory and anti-oxidative activities. In this study, we were to use Houttuynia cordata extract on formulation of cosmetic emulsions. Methods : The effects of Houttuynia cordata extract on emulsion stability and viscoelastic properties of emulsion were measured using turbiscan and rheometer. And we assessed the anti-oxidative and antibiotic activities of Houttuynia cordata extract. Results : 1. The results of this investigation for emulsion stability showed that the stability emulsion containing Houttuynia cordata extract was decreased depending on concentration of Houttuynia cordata extract. 2. Super oxide dismutase activity was strongly dependents on concentration of Houttuynia cordata extract. 3. Houttuynia cordata extract showed good anti-microbial activity against Staphylococcus aureus. Conclusions : Houttuynia cordata extract can be effectively used in cosmetic emulsions when the relation between natural product extracts and formulation of cosmetics is elucidated.
To investigate the antibiotic constituents of Korean basidiomycetes the carpophores of the wood-rotting basidiomycetes were collected from several locations of Korea, and from them 17 mycelial strains were isolated on potato-dextrose-agar plates supplemented with tetracycline ($20\;{\mu}g/m{\ell}$. The isolated strains were shake-cultured in glucosepeptone-yeast extract medium and then the antibacterial activities of the culture filtrates were assessed by disc-plate method. Among them, 12 strains (70.6%) were active, and basidiomycete strain LMCB-109 (Daedalea quercina) and LMCB-116 showed potent activities against all the six bacterial target organisms including Serratia marcescens.
BACKGROUND: Although antibiotics have contributed to treatment of bacterial infection, the antibiotic abuse can lead to antibiotic resistant bacteria. Impact of human activities on distribution of antibiotic resistance has been intensively issued and occurrence of antibiotic resistant bacteria in contaminated environments would not be a surprise. Nonetheless, anthropogenic contamination with the dissemination of antibiotic resistance along uncontaminated environments has been less considered. The aim of this study is to investigate antibiotic resistant bacteria across Ulleungdo, known as antibiotic resistance free and anthropogenic pollution free environment in Rep. of Korea. METHODS AND RESULTS: Antibiotic resistant bacteria in coastal seawater of Ulleungdo were investigated in July 2021. Antibiotic susceptibility test using the disk diffusion method was applied with six drugs according to the Clinical and Laboratory Standards Institute (CLSI) guideline. Total 43 bacterial isolates were tested and 20 isolates among of them showed multidrug resistance. Particularly, the number and ratio of resistant bacteria were relatively high in a densely populated area of Ulleungdo. The bacterial communities were investigated using 16S rRNA gene metabarcoding approach in the coastal seawater and soils of Ulleungdo. In the bacterial communities, Firmicutes were selectively distributed only in seawater, suggesting the possibility of anthropogenic contamination in coastal seawater of Ulleungdo. CONCLUSION(S): We found antibiotic resistant bacteria in a populated area of Ulleungdo. The occurrence of antibiotic resistant bacteria in Ulleungdo seems to result from the recent anthropogenic impact. Consistent monitoring of antibiotic resistant bacteria in the uncontaminated environment needs to considered for future risk assessment of antibiotics.
The antifungal activities of 153 extracts from 51 species of Korean mushroom on six phytopathogenic fungi were investigated. The powder of fruit-body of each mushroom was extracted with petroleum ether, 80% ethanol and distilled water. The water extracts of four mushrooms including Amanita virosa showed antifungal activities on Alternaria alternata, Colletotrichum graminicola, Fusarium solani, Idriella bolleyie. The 80% ethanol extracts of seven mushrooms including Boletus auripes showed antibiotic activities against A. alternata, C. gramincola, Cylindrocarpon destructans, F. solani, F. oxysporum cucumerinum, and I. bolleyie. The petroleum ether extracts of six mushrooms including Amanita citrina showed antibiotic activities against A. alternata, C. destructans, and C. graminicola. The 102 extracts of 34 mushrooms including Agaricus arvensis didn't show antibiotic activities.
