In vitro antimicrobial properties of Bacillus subtilis KCTC 1326 for fish bacterial disease management

  • This study evaluated in vitro antimicrobial properties of Bacillus subtilis KCTC 1326 as an environmentally friendly alternative to antibiotics. B. subtilis KCTC 1326 was characterized on biochemical properties and antibiotics susceptibility. It exhibited antimicrobial effects against all 12 species of fish bacteria used in this experiment. Among them, the largest antibacterial zone was observed for Streptococcus parauberis (34 mm), while the smallest antibacterial zone was observed for Citrobacter freundii (8 mm). Additionally, in the co-culture inhibitory assay of B. subtilis and Edwardsiella piscicida, the growth of E. piscicida was suppressed with increasing concentrations of B. subtilis KCTC 1326, with complete inhibition observed at 107 and 108 CFU/mL of B. subtilis KCTC 1326 after 24 hours of incubation. Moreover, at 48 hours of incubation, the growth of E. piscicida was inhibited across all concentration ranges of B. subtilis KCTC 1326. Therefore, this study indicated the utilizing of B. subtilis KCTC 1326 as an antimicrobial for controlling fish bacterial diseases.

Species and Antimicrobial Susceptibility of Nonfermentative Gram-Negative Bacilli Isolated from Clinical Materials (임상 검사물에서 분리된 비발효성 그람음성 간균의 균종과 항생제 감수성)

  • Species and antimicrobial susceptibility of nonfermenting gram-negative bacilli(NFB) isolated from clinical materials at Yonsei Medical Center during the period of September 1980 to August 1981 were analyzed and the following results were obtained. 1. NFB were isolated from 17.1% of sputum, 14.8% of pus, 5.0% of urine, 3.3% of throat and 1.4% of blood specimens. 2. Among the isolates 57.6% were Pseudomonas aeruginosa and 32.7% were Acinetobacter calcoaceticus. P. maltophilia and P. cepacia were 3.2% and 2.8% respectively. Other bacteria were rarely isolated. 3. The monthly isolation rates were high during the June to November period(8.8-12.0%), and low during the December to May period(4.2-8.4%). 4. Many of the isolates showed resistance to various antimicrobials. Although there were variations depending on the bacterial species, amikacin, colistin, gentamicin, tobramycin and co-trimoxazole showed more antibacterial activities than other antimicrobials.

Determination of Semen Quality and Antibacterial Susceptibility Pattern of Bacteria Isolated from Semen of Iraqi Subjects

  • Infertility is a key issue affecting mood and behavior in men. Microorganisms are one of the primary etiological agents that may be associated with infertility. The objective of the present study was to identify bacterial causative agents from the semen of infertile subjects and determine the effect of bacterial infection on sperm quality, as well as determine the susceptibility of these bacteria to drugs. Forty semen samples from 30 infertile patients and 10 fertile individuals were collected. The pH, volume, motility, and concentration of semen were analyzed. The samples were processed and identified by biochemical testing using API identification kits. The antibiotic susceptibility pattern was determined using the disc diffusion method. Abnormal sperm quality was observed. The mean age of the individual and their sperm morphology, concentration, progressive motility, pH level, and pus cell content were 31.9 years, 2.7%, 10.4 million/ml, 27.3%, 8.3, and 5.7, respectively. Among the tested samples, oligoasthenozoospermia was found to show the highest occurrence, at 27/30 samples, followed by teratozoospermia, at 25/30 samples, and asthenozoospermia, at 22/30 samples. Of the tested infertile patients' sperm, 19, 6, and 5 isolates were identified as Escherichia coli, Klebsiella pneumonia, and Staphylococcus epidermidis, respectively. The results also revealed multi-drug resistance in the bacteria. Compared to that shown by the other tested antibiotics, amikacin showed higher activity against all isolated bacteria. However, the bacteria exhibited maximum resistance against gentamicin, cefotaxime, levofloxacin, and ampicillin. In conclusion, leukocytospermia and bacterial infections are possibly responsible for sperm abnormalities. Multi-drug resistant bacteria were detected. Gentamicin, cefotaxime, levofloxacin and ampicillin were shown the highest resistance, while amikacin was the most effective antimicrobial agent against the isolated bacteria.

Resistance Patterns of Frequently Applied Antimicrobials and Occurrence of Antibiotic Resistance Genes in Edwardsiella tarda Detected in Edwardsiellosis-Infected Tilapia Species of Fish Farms of Punjab in Pakistan

  • Edwardsiella tarda is one of the most significant fish pathogens, causes edwardsiellosis in a variety of freshwater fish species, and its antibiotic resistance against multiple drugs has made it a health risk worldwide. In this study, we aimed to investigate the antibiotic resistance (ABR) genes of E. tarda and establish its antibiotic susceptibility. Thus, 540 fish (299 Oreochromis niloticus, 138 O. mossambicus, and 103 O. aureus) were collected randomly from twelve fish farms in three districts of Punjab in Pakistan. E. tarda was recovered from 147 fish showing symptoms of exophthalmia, hemorrhages, skin depigmentation, ascites, and bacteria-filled nodules in enlarged liver and kidney. Antimicrobial susceptibility testing proved chloramphenicol, ciprofloxacin, and streptomycin effective, but amoxicillin, erythromycin, and flumequine ineffective in controlling edwardsiellosis. Maximum occurrence of qnrA, blaTEM, and sul3 genes of E. tarda was detected in 45% in the liver, 58%, and 42% respectively in the intestine; 46.5%, 67.2%, and 55.9% respectively in O. niloticus; 24%, 36%, and 23% respectively in summer with respect to fish organs, species, and season, respectively. Motility, H2S, indole, methyl red, and glucose tests gave positive results. Overall, E. tarda infected 27.2% of fish, which ultimately caused 7.69% mortality. The Chi-squared test of independence showed a significant difference in the occurrence of ABR genes of E. tarda with respect to sampling sites. In conclusion, the misuse of antibacterial agents has led to the emergence of ABR genes in E. tarda, which in association with high temperatures cause multiple abnormalities in infected fish and ultimately resulting in massive mortality.

Antibacterial activity of isothiocyanates from cruciferous vegetables against pathogenic bacteria in olive flounder (십자화과 채소 유래 isothiocyanates의 넙치 어병세균에 대한 항균활성)

  • The antimicrobial effects of ten isothiocyanates (ITCs) present in cruciferous vegetables and radish root hydrolysate were investigated against pathogenic bacteria from olive flounder. Minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) were measured against two gram-positive bacterial strains (Streptococcus parauberis, S. iniae) and four gram-negative bacterial strains (Edwardsiella tarda, Vibrio ichthyoenteri, V. harveyi, Photobacterium damselae) by using a broth microdilution technique. The antibacterial activity of ITCs was in the order sulforaphane > sulforaphene > phenylethyl ITC > erucin > benzyl ITC > iberin > I3C > allyl ITC > phenyl ITC > hexyl ITC. The susceptibility of fish pathogens to ITCs was in the order of V. harveyi > E. tarda > P. damselae > S. parauberis > S. iniae > V. ichthyoenteri. Antimicrobial activity (MIC) of radish root hydrolysate was 0.250 mg/mL against S. iniae, 0.438 mg/mL against S. parauberis, and 0.500 mg/mL against both E. tarda and V. harveyi. The aliphatic ITCs were potent inhibitors of the growth of fish pathogens, followed by aromatic ITCs and indolyl ITC. The presence of a double bond in the chemical structure of ITCs decreased antibacterial activity, while ITCs with a thiol (-S-) group and a longer carbon chain increased antibacterial activity. These results suggest that ITCs have strong antibacterial activities and may be useful in the prevention of fish pathogens.

The Antibacterial Activity Against Fish Pathogen of Paenibacillus sp. MK-11 Isolated from Jeju Coast (제주연안으로부터 분리한 Paenibacillus sp. MK-11의 어류 질병 세균에 대한 항균활성 탐색)

  • In this study, we isolate and identify bacteria from seawater collected from Jeju coast, to evaluate the antimicrobial activity against the fish pathogenic bacteria. 14 bacterial strains were isolated and identified using physiological, biochemical and molecular tools. Antibacterial activity of all the 14 isolates were screened against four major fish pathogens namely, two Gram-positive: Streptococcus iniae, Streptococcus parauberis and two Gram-negative: Vibrio anguillarum, Edwardsiella tarda. Results revealed that among the 14 isolates, MK-11 was found to have antibacterial activity against S. iniae, S. parauberis, V. anguillarum Particularly, S. iniae was susceptibility with the MIC value of $250{\mu}g/ml$. The biochemical and physio-chemical results reveal that MK-11 had the sugar-alcohol disassemble ability of the D-sorbitol and D-mannitol. Also the utilization of the yeast extract, sorbitol and di-potassium phosphate were noted to be high. The optimum culture condition such as pH and temperature was recorded as pH 6.0, $25^{\circ}C$ and along with 1% NaCl which differs from the previous reports particularly in nutrient resolutions. As results of the analysis of 16S rDNA sequences, MK-11 show the high similarity with Paenibacillus polymyxa, P. jamilae, P. brasilensis 99.78%, 99.43%, 99.39%, repectively. Hence, in the present study, the isolated Paemibacillus sp. MK-11 from Jeju seawater possesses the antibacterial activity against fish pathogens and it could be used as a new antibiotic agents against the gram positive fish pathogens.

  • 새로 합성된 6-exomethylene penamsulfone derivatives 7종에 대해 In Vitro antibacterial activities를 Broth Microdilution법으로 Sulbactam-Na(SAM), Clavulanic acid-K(CLA)와 Cefotaxime-Na(CFT), Ampicillin Trihydrates(AMP), Amoxicillin-Na(AMX), Cefoperazone(CFZ)을 병용투여하여 $\beta$-lactamase 생성 균주(30 여종)에 실시, MIC를 구하였다. 균주에 따라 SAM, CLA에 비해 CH1145, CH1155 등이 우수한 효과를, CH1140, CH2150등이 유사한 효과를 보였다.

Effect of Salicylate on Antibacterial Activity of Different Antibiotics

  • Susceptibility of Psudomonas aeruginosa, Staphylococcus aureus, and Bacillus subtilis to gentimicin and ceforaxime was affected by salicylaye. In presence of salicylaye (15 mM) and gentamicin (1.0 .mu.g/ml), log efficiency of plating (log E. O. P. s) for the tested bacteria were -1.24, -2.17 and -1.66 respectively. The activity of cefotaxime against Bacillus subtilis was reduced (log E. O. P. = 1.33). The highest potentiating effects of salicylaye were shown when using gentamicin against Staphylococcus aureus, cefotaxime against Ps. aeruginosa, log E. O. P.s were -3.0, and -2.4 respectively. On the other hand, no significant effects were detected with cefotaxime against Staphylococcus aureus (log E. O. P. = -0.04). No significant killing was shown in presence of gentamicin or salicylaye alone. There was no significant effect for salicylaye on MICs (By broth dilution) could be observed except in case of gentamicin against Staphyloccus aureus, which was reduced from 0.02 .mu.g/ml to 0.0012 .mu.g/ml. These results raise the concern that high concentrations of salicylaye in patients might interfere with antibiotic therapies.

Cultural characterization of probiotic Lactobacillus sakei BK19

  • We have selected an valuable pmbiotic strain; Lactobacillus sakei BK19 which has wide antagonic spectrum against fish pathogens . Present study investigated cultural characterization of L. sakei BK19 including pH tolerance , susceptibility of antibacterial agents and growth pattern with different environment such as nutritions, temperature and salinity. L. sakei BK19 showed Significantly higher resistance at low pH(around pH 4) environment and relative high antibiotic tolerance . In the study of optimal culture condition, maltose and saccharose provided the optimal nutritional culture condition while lactose and mannitol were unable to supply its carbon source for the fermentation of L. sakei BK19. Moreover. L. sakei BK19 showed good growth at the temperature range of 15 to $45^{o}C$ und the NaCl concentration of 0 to 7%. Hence, this particular probiotic strain may be benificial both in seawater and fresh weter conditions.
