An In Vitro Study on the Activity of Abelmoschus manihot L. Flower Extract on Skin Anti-wrinkle and Skin Whitening (금화규(Abelmoschus manihot ) 꽃 추출물의 In Vitro 피부 미백 및 주름 개선 효능 연구)

  • In this study, the skin whitening and anti-wrinkle activity of Abelmoschus manihot (A. manihot) flowers were evaluated as a cosmetic material. A. manihot flowers were extracted using water and 70% ethanol. Tyrosinase inhibitory activity was evaluated to increase concentration-dependent inhibitory activity in both water (AMW) and 70% ethanol extracts (AME), and was found to inhibit melanin biosynthesis at concentrations that did not affect survival in B16F10 cell lines at 10, 25, 50, 75, and 100 ㎍/mL. As a result of measuring the anti-wrinkle effectiveness of A. manihot flowers, 70% ethanol extract (AME) showed higher anti-wrinkle activity than water extract (AMW). Through the results of this study, it is considered that the A. manihot flower extract can be used as a functional cosmetic material with whitening and anti-wrinkle activity.

Whitening Effect and Mechanism of Aerial Part of Pueraria lobata (갈만 추출물의 미백활성 및 기전 연구)

  • The purpose of this study was to evaluate the whitening effect of aerial part of Pueraria lobata and mechanisms. Aerial part of Pueraria lobata, dose-dependently reduced the melanin content. Aerial part of Pueraria lobata, significantly decreased cellular tyrosinase activity, while there was not any effect on tyrosinase in cell-free conditions. To elucidate the mechanisms behind the aerial part of Pueraria lobata, treated melanogenesis regulation, the expressions of melanogensis related genes, proteins, and the activity of ${\alpha}-glucosidase$ were determined. Aerial part of Pueraria lobata, significantly inhibited gene and protein levels of MITF, tyrosinase and TRP-1. It suppressed the ${\alpha}-glucosidase$, leading to inhibition on the maturation of tyrosinase. Also aerial part of Pueraria lobata, was observed to have the high antioxidant activity. These results suggested that whitening effect of aerial part of Pueraria lobata, should be due to the down-regulation of MITF, tyrosinase and TRP-1 expression and the intercepting maturation of tyrosinase through suppressing ${\alpha}-glucosidase$. Another should be the high anti-oxidant activity. The findings show the possibility that aerial part of Pueraria lobata, can be used as a potential skin-whitening agent.

UV Protection and Whitening Effects of Collagen Isolated from Outer Layer of the Squid Todarodes pacificus (동해 오징어 (Todarodes pacificus) 껍질 유래 콜라겐의 UV 보호능 및 미백효과)

  • To develop a whitening agent, cytotoxicity of the soluble collagen isolated from Todarodes pacificus (CIT) was evaluated. CIT tested for cytotoxicity on human dermal fibroblast (CCD-986sk) was exhibited very low cytotoxicity. Because tyrosinase is the key enzyme for melanin biosynthesis, the use of various tyrosinase inhibitors is a common practice for whitening purpose in cosmetics. Tyrosinase inhibitory activity and melanin production assay were measured to confirm the whitening effect. The inhibitory effect of MMP-1 in UV-irradiated human dermal fibroblast was also performed. CIT showed strong inhibition potency on tyrosinase by 51.5% at 0.2 mg/mL which increased the inhibition by increasing the concentration of CIT, and showed 69.1% inhibition at 1.0 mg/mL. CIT showed strong inhibition effect on melanin production with 82% at 1.0 mg/mL. The CIT also reduced about 76% expression of MMP-1 in UV-irradiated CCD-986sk cell at 1.0 mg/mL. From the preliminary observations, we suggest that the collagen isolated from CIT could be a potential source of natural skin-whitening and anti-aging agents for the photo-damaged skin.

Cosmetic Application Using Skin Whitening and Anti-microbial effects of Ethyl Acetate and n-Butanol Fractions from Eruca sativa (Eruca sativa 에칠아세테이트와 부탄올 분획물의 미백 및 항균효과를 이용한 화장품 응용연구)

  • Eruca sativa, called arugula, is a perennial plant in the Brassicaceae family, an edible plant commonly used in Italian cuisine. To study as a cosmetic material application E. sativa was extracted with 70% ethanol (ES). Then ES was fractionated using n-hexane, chloroform, ethyl acetate, n-butyl alcohol and water (EHex, EEA, ECHCl3, EBuOH and EDW). EEA showed mushroom tyrosinase inhibitory activity. ES, EEA and EBuOH showed inhibition of tyrosinase activity. As a result, ES is expected to have skin whitening efficacy. ES was applied to 0.05, 0.1% the toner and emulsion formulation to test the stability. The anti-microbial activity of eight bacteria and fungi including Staphylococcus aureus and Propionibacterium acnes which cause dermatitis and acne was evaluated. EEA showed effects in all of microorganisms. The toner and emulsion containing ES with 0.05, 0.1% were passed in the challenge test. At -20, 4, 25, 55 ℃ and daylight, there was no significant change on pH, viscosity for 4 months. However, emulsions had phase separation phenomenon at 55 ℃, so the base formulation needs improvement. In addition, through the skin penetration test, EEA penetrated 0.058% in 6 hr, predicting the clinical efficacy. This means that E. sativa can contribute whitening agent and the synergistic effect of preservatives.

Inhibitory Effects of Saposhnikoviae Radix Extracts on the Melanin Production and Elastase Activity in B16F10 cells (흑색종 세포주에서 멜라닌 생성과 엘라스타제 활성 억제에 미치는 방풍의 효과)

  • Saposhmikoviae Radix can treat various skin disease by anti-pruitus and anti-inflammatory effects. This study was designed to investigate effects of Saposhmikoviae Radix Extracts(SRE) on skin elasticity and whitening using B16F10 cell lines. In this experiment, We observed effect of SRE on cell viability, inhibition of melanin synthesis and inhibitory effect on tyrosinase and elastase. In results, SRE treated group showed lowered proliferation rates significantly compared to non-treated group. More than SRE $125{\mu}g/m{\ell}$ of treated groups were lower levels of melanin synthesis respectively. SRE did not show inhibitory effect on tyrosinase activities in vitro and in B16F10 cells. Finally, SRE suppressed elastse type I and IV activities in dose-dependent manner in vitro. And SRE also slightly suppressed elastase activities in B16F10 cells. In conclusion, these results suggest that SRE can inhibit melanin synthesis and inhibt elastase activity. So, We suggest that SRE can be maintained skin elasticity or whitening.

The Antioxidation Effect of Mutimo cylindricus Extract and Its Influence on Cell Bioactivity (회초리말(Mutimo cylindricus)의 항산화, 항염 및 미백 활성)

  • The objective of this study was to evaluate functionality of Mutimo cylindricus as a cosmetic ingredient, one of the brown algae. The M. cylindricus 70% ethanol extract was manufactured for antioxidant measurement. DPPH and ABTS methods were used to measure antioxidants, and its EC50 values were 2.040 and 2.182 mg/mL in each experiment. The measurement of total polyphenol contents and reducing power showed total polyphenol content of 103 mg gallic acid/g extract and reducing power of 134 mg ascorbic acid/g extract. To measure cell toxicity, MTT method was used, and its result showed that the extract was not cytotoxic. And it has anti-inflammatory and whitening activity at concentrations of 100 ㎍/mL. The result confirmed that M. cylindricus extract is available as a cosmetic material with whitening and anti-inflammatory properties.

Effect of Dendrobium loddigesii Rolfe Methanol Extract on Melanogenesis in α-MSH Stimulated B16F10 Cells (환초석곡 메탄올 추출물의 흑색종세포주에서 멜라닌 생성 억제 효능)

  • Dendrobium loddigesii (DL) is a valuable and versatile herbal medicine with the anecdotal claims of anti-oxidant and anti-inflammation. In the present study, we investigated the whitening effects of DL under various conditions with B16F10 melanoma cells. The DL extract inhibited melanin contents and tyrosinase activity in a dose-dependent manner, compared with untreated group. Treatment of the DL extract effectively suppressed the ${\alpha}$-MSH-stimulated melanin formation, tyrosinase activity and dendrite outgrowth. Moreover, the ${\alpha}$-MSH-induced mRNA expressions of tyrosinase-related protein-1 (TRP-1), tyrosinase-related protein-2 (TRP-2), microphthalmia-associated transcription factor (MITF) and protein expression of tyrosinase were significantly attenuated by DL treatment. These results indicate that DL may be a great cosmeceutical ingredient for its whitening effects.

Antioxidant Activity and Whitening Effect of Forsythiae Fructus Extracts (연교 추출물의 항산화활성 및 미백 효과)

  • The Forsythiae Fructus is an oriental medicine containing various lignans. In this study, the Forsythiae Fructus were extracted by hot water (Sample 1), hot water after bio-conversion using Lactobacillus strain (Sample 2-LP2, 2-LA, 2-LC, 2-LL, 2-BL and 2-LM) and 70% ethanol (Sample 3). Total polyphenol and flavonoid contents were improved by bio-conversion process using Lactobacillus strain, compared to water extract. Especially, sample 2-LL and 2-LA which had shown the high total polyphenol and flavonoid content in antioxidant activity. Also, sample 2-LL and 2-LA showed higher melanin generation inhibitory activity as of 55%, 53% in maximum extract concentration of $100{\mu}g/m{\ell}$. In the anti-inflammation test of the Forsythiae Fructus extracts, nitric oxide (NO) synthesis was inhibited. Specially, both 70% Forsythiae Fructus ethanol extract and sample 2-BL which have shown the relatively higher 1,1-diphenyl-2-picryl-hydrazyl (DPPH) radical scavenging and superoxide dismutase (SOD) like activities. In conclusion, the Forsythiae Fructus extracts with bio-conversion process has effect of skin whitening and anti-inflammation activity than other extracts. It could be used as a valuable materials for functional cosmetics.

Nitric Oxide Production Inhibitory and Scavenging Activity and Tyrosinase Inhibitory Activity of Extracts from Taraxacum officinale and Taraxacum coreanum (서양민들레와 흰민들레 추출물의 Nitric Oxide 생성억제 및 소거 활성과 Tyrosinase 저해 활성)

  • The study was conducted to investigate functional materials as skin whitening and anti-inflammatory agent from Taraxacum officinale and Taraxacum coreanum. The total polyphenol and flavonoid content in the ethanol extract of Taraxacum officinale were found to be 64.07mg/g and 32.46mg/g, respectively. In tyrosinase inhibitory activity, the hot water extract of Taraxacum coreanum was higher than the other extracts. However, in nitric oxide (NO) scavenging ability, the ethanol extract of Taraxacum coreanum was higher than the other extracts. the ethanol extract of Taraxacum coreanum showed strong NO production inhibitory effect in lipopolysaccharide (LPS)-stimulated Raw 264.7 cell. In the cell viability measurement by MTT assay and the lactate dehydrogenase (LDH) assay against L929 cell, the extracts were exhibited fine cell viabilities and normal LDH release levels as nontoxic result in sample concentration of $250{\sim}1000{\mu}g/m{\ell}$. As a result, the ethanol extract and the hot water extract of Taraxacum coreanum could be applicable to functional materials for anti-inflammatory and skin whitening related fields, respectively.
