Effect of Citrus macroptera Fruit Pulp Juice on Alteration of Caspase Pathway Rendering Anti-Proliferative Activity against Ehrlich's Ascites Carcinoma in Mice

  • Citrus macroptera (Rutaceae) has long been used in folk medicine in Bangladesh. Considering the folkloric context, this study was aimed to scrutinize anti-proliferative activity of C. macroptera fruit pulp juice (CMFPJ) against Ehrlich's ascites carcinoma (EAC). The anti-proliferative capacity of CMFPJ was investigated and confirmed primarily using MTT assay. In vivo anti-proliferative aptitude of CMFPJ was investigated with 25, 50, and 100 mg/kg/day intraperitoneal (i.p.) treatment. Anti-proliferative efficacy of CMFPJ was assessed based on EAC growth inhibition. CMFPJ inhibited EAC growth in vitro in a dose-dependent manner. And the percentages of in vivo EAC growth inhibition were 19.53, 49.2, and 68.9% at 25, 50, and 100 mg/kg CMFPJ respectively. CMFPJ significantly induced expression of apoptosis regulatory genes caspase-8, caspase-9, cytochrome-c, and caspase-3. This considerable anti-cancer activity was perhaps due to combinatorial effect of lectin, polyphenols, and flavonoids present in CMFPJ.

Anti-proliferative and Anti-telomerase Activity of Curcuma Rhizome Extract on Oral Squamous Cell Carcinoma and Osteosarcoma Cells

  • Anti-proliferation of methanol extract of Curcuma rhizome on oral squamous cell carcinoma (KB) and osteosarcoma (HOS) cells were investigated. In order to elucidate the involvement of telomerase inhibitory activity as a part of anti-proliferative effect of Curcuma rhizome on cancer cells, we measured telomerase activity in Curcuma rhizome extract-treated cancer cells. The concentration inhibited cell proliferation to 50% $(IC_{50})$ of the methanol extract of Curcuma rhizome against oral squamous cell carcinoma (KB) cells and osteosarcoma (HOS) cells were 21.30 ${\mu}g/ml$ and 39.3${\mu}g/ml$, respectively. The methanol extract of Curcuma rhizome showed inhibitory telomerase inhibitory effect which is required for cancer cell immortality. Therefore, it seems that the anticancer effect of methanol extract of Curcuma rhizome is at least partially due to telomerase inhibitory effect. Five fraction samples were prepared according to its polarity differences and analyzed anti-proliferative effects of each fraction samples on oral squamous cell carcinoma and osteosarcoma cells. Anticancer effect was observed in dichloromethane, and ethylacetate fractions. The highest anticancer effect was found in dichloromethane fraction which had $IC_{50}$ value of 23.3 ${\mu}g/ml$ and 10.5${\mu}g/ml$ against oral squamous cell carcinoma (KB) cells and osteosarcoma (HOS) cells, respectively.

목화 부위별 추출물의 암 세포주 증식 억제 효과 (Inhibitive Effects of Cotton Plant Sectional Extracts in Cancer Cell Lines)

  • Objectives : This study was carried out to investigate the inhibitive effects of cotton plant sectional extracts in cancer cell lines, Calu-6(human, Caucasian, lung, adenocarcinoma) and MCF-7(human, Caucasian, breast, adenocarcinoma). The incidence of cancer has been increasing even in korea due to the change of dietary life and westernization and becoming conspicuous as the disease threatening health. But cancer treatment have not been fully effective against the high incidence or low survival rate of most cancer. Methods : Calu-6 and MCF-7 cells were cultured and seeded in cell culture plates, respectively. And sectional extracts of cotton plant were treated to MCF-7 cells. Results and Conclusion : Sectional extracts of cotton plant showed no anti-proliferative effect on MCF-7 cells, but root and stem extracts showed strong anti-proliferative effects on Calu-6 cells. Fruit, leaf and flower extracts also showed anti-proliferative effects on Calu-6 cells but not so much like root and stem extracts. But seed extract showed no anti-proliferative effect on Calu-6 cells.

  • We investigated the anti-proliferative effects of lxeris sonchifolia H. (godulbaegi) root extracts. luteolin(3'. 4', 5. 7-Q-glucoside and 3'. 4', 5, 7-tetrahydoxyflavone) and apigenin (3', 4'. 5. 7-O-gluconic acid) on HepG2 (p53 wild type) cells. Hep3B (p53 null) cells, and Chang liver cells. In MTT assay 3', 4'.5. 7-tetrahydoxyflavone showed the most efficient anti-proliferative effects on these three cell lines. However, there was no significant anti-proliferative effect on Chang liver cell line in MTT results. (omitted)

Anti-proliferative and Apoptosis Inducing Effect of Momordin I on Oral Carcinoma (KB) Cells

  • Treatment of oral cancers with chemotherapeutic agents become evaluated as an effective method to reduce cancer cell proliferation. Anti-proliferative and anti-oral cancer activities of momordin I on oral cancer cells were evaluated in this study. Momordin I was originally purified from a natural product, Ampelopsis radix and showed the antiproliferative activity against oral carcinoma, KB cells. Obtained $IC_{50}$ value was approximately $10.4{\mu}g/ml$. Time-and dose-dependent chromosomal DNA fragmentations were observed in momordin I-treated KB cells. Flow cytometry analysis showed time-dependent apoptotic cell appearance after treatment of momordin I. Approximately 18.6% apoptotic cells were observed at 72 hours after $20{\mu}g/ml$ of momordin I treatment. These observation were consistent with the results obtained in DNA fragmentation analysis. These data suggest that momordin I has anti-proliferative effect and induces cell death in KB cells through apoptosis.

Growth Inhibitory Activity of Honokiol through Cell-cycle Arrest, Apoptosis and Suppression of Akt/mTOR Signaling in Human Hepatocellular Carcinoma Cells

  • Honokiol, a naturally occurring neolignan mainly found in Magnolia species, has exhibited a potential anti-proliferative activity in human cancer cells. However, the growth inhibitory activity against hepatocellular carcinoma cells and the underlying molecular mechanisms has been poorly determined. The present study was designed to examine the anti-proliferative effect of honokiol in SK-HEP-1 human hepatocellular cancer cells. Honokiol exerted anti-proliferative activity with cell-cycle arrest at the G0/G1 phase and sequential induction of apoptotic cell death. The cell-cycle arrest was well correlated with the down-regulation of checkpoint proteins including cyclin D1, cyclin A, cyclin E, CDK4, PCNA, retinoblastoma protein (Rb), and c-Myc. The increase of sub-G1 peak by the higher concentration of honokiol ($75{\mu}M$) was closely related to the induction of apoptosis, which was evidenced by decreased expression of Bcl-2, Bid, and caspase-9. Hohokiol was also found to attenuate the activation of signaling proteins in the Akt/mTOR and ERK pathways. These findings suggest that the anti-proliferative effect of honokiol was associated in part with the induction of cell-cycle arrest, apoptosis, and dow-nregulation of Akt/mTOR signaling pathways in human hepatocellular cancer cells.

유근피(楡根皮) 추출물의 유방암 세포주 MCF-7 생장 억제 효과 (Anti-proliferative Effect of Ulmi Pumilae Cortex Extracts on MCF-7 cells)

  • Purpose: This study was conducted to investigate the anti-proliferative effects of Ulmi Pumilae Cortex Extracts(UPCE) on MCF-7(human, breast, adenocaecinoma) and NIH3T3 (human, murine, fibroblast). Methods: MCF-7 cells and NIH3T3 cells were cultured and seeded in cell culture plates, respectively. UPC was extracted with hot water and then further fractionated it into five types: hexane, chloroform, ethyl acetate, butanol, and water soluable fractions. These five different fractions from UPCE were tested for their anti-proliferative effects on MCF-7 cells and NIH3T3 cells by MMT assay. Results: Among the five solvent-fractions of UPCE, n-hexane fraction and ethyl acetate fraction showed a strong anti-proliferative effects on MCF-7 cells but they displayed significant cytotoxicity on NIH3T3 cells, too. On the other hand, chloroform fraction showed a marked anti-proliferative effects on MCF-7 cells and low cytotoxicity on NIH3T3 cells. Conclusion: Chloloform fraction from UPCE showed selective anti-cancer activities on human breast cancer cell MCF-7 relatively to the other fractions.

Anti-proliferative Effect of a Novel Anti-oxidative Peptide in Hanwoo Beef on Human Colorectal Carcinoma Cells

  • The present study aimed to characterise anti-oxidant peptides from water-soluble protein extracts of Hanwoo beef and evaluate their anti-proliferative effect on human colorectal carcinoma cells (HCT116). Antioxidant peptides were purified from the low-molecular-weight fraction (<3 kDa) of Hanwoo beef extract. Antioxidant activity of peptide fractions was determined using the oxygen radical absorbance capacity (ORAC) assay. Purified peptide (P3) displayed higher ORAC activity than the low-molecular-weight fraction ($202.66{\mu}M\;TE/g$ vs $167.38{\mu}M\;TE/g$ of dry matter, respectively) (p<0.05). The peptide sequence of P3 was Cys-Cys-Cys-Cys-Ser-Val-Gln-Lys (888.30 Da). The novel peptide P3, at $250{\mu}g/mL$, also significantly inhibited HCT116 cell proliferation up to 25.24% through phosphorylation of ERK, JNK, and p38 kinase (p<0.05). Hence, antioxidant peptide P3 from Hanwoo beef extract can be used as an antioxidative and anticancer agent in the functional food industry.

Anti-Proliferative Effect of Polysaccharides from Salicornia herbacea on Induction of G2/M Arrest and Apoptosis in Human Colon Cancer Cells

  • In this study, we investigated the anti-proliferative effect of polysaccharides from Salicornia herbacea on HT-29 human colon cancer cells. Crude polysaccharides from S. herbacea (CS) were prepared by extraction with hot steam water, and fine polysaccharides from S. herbacea (PS) were obtained through further size exclusion chromatography. The anti-proliferative effect of CS and PS were measured using the MTS assay, apoptosis analysis, cell cycle analysis, and RT-PCR. HT-29 cells were treated with CS or PS at different dosages (0.5, 1, 2, 4 mg $ml^{-1}$) for 24 or 48 h. CS and PS inhibited proliferation and stimulated apoptosis of cells in a dose-dependent manner. Flow cytometric analysis after Annexin V-FITC and PI staining revealed that treatment with CS or PS increased total apoptotic death of cells to 24.99% or 91.59%, respectively, in comparison with the control (13.51 %). PS increased early apoptotic death substantially - up to 12 times more than the control. Treatment with CS or PS resulted in a concentration-dependent increase of the G2/M cell population of the cell cycle as determined by flow cytometry. G2/M arrest was induced significantly with the highest concentration (4 mg $ml^{-1}$) of PS. RT-PCR was performed to study the correlation between G2/M arrest and transcription of cell cycle control genes. The anti-proliferative activity of CS and PS was accompanied by inhibition of cyclin B1, and Cdc 2 mRNA. Moreover, both CS and PS induced expression of the p53 tumor suppressor gene and the Cdk inhibitor p21. These results suggest that polysaccharides from S. herbacea have anti-cancer activity in human colon cancer cells.

유자(Citrus junos)씨와 유자씨 유지의 메탄올 추출물에 의한 HT-29 대장암 세포 생장 억제 효과 및 유효 성분 분석 (Anti-proliferative effect of methanolic extracts from Citrus junos seeds and seed oils on HT-29 human colon cancer cells and identification of their major bioactive compounds)

  • 본 연구의 목적은 유자씨와 유자씨 유지로부터 HT-29 암세포 생장 억제효과를 확인하고 주요 원인 물질을 확인하는 것이다. 유자씨, 헥산 추출 유자씨 유지, 냉압착 유자씨 유지로부터 60% 메탄올을 이용하여 추출물(각각 ES, EHO, ECO)을 얻었다. 추출물의 성분은 HPLC-MS를 이용하여 확인하였다. ES, EHO, ECO를 HT-29 세포에 처리하여 생장 억제 효과를 확인한 결과, EHO와 ECO가 유의적인 효과가 있었다(p<0.05). 반면, ES와 리모닌, 노밀린은 24시간과 48시간 처리 후에 암세포 생장 억제 효과가 없었다(p>0.05). 유자씨 유지 추출물의 암세포 생장 억제효과에 주요 역할을 하는 성분을 탐색하기 위해 제조용 LC로 EHO와 ECO를 분획하여 이 분획물의 암세포 생장 억제 효과와 조성을 확인하였다. 분획물 중에서 EHO의 3개 분획물과 ECO의 2개 분획물이 유의적인 HT-29 세포 생장 억제 효과가 있었다(p<0.05). 이 5개 분획물의 HPLC-MS 분석 결과, 아이소핌피넬린, 버갑텐, 이찬젠신이 주요 성분일 것으로 추정되었다. 아이소핌피넬린, 버갑텐, 이찬젠신을 HT-29 세포에 처리한 결과, 이찬젠신이 유의적인 생장 억제 효과가 있었고(p<0.05), 아이소핌피넬린과 버갑텐은 약간의 생장 억제 효과가 있었으나 유의적이지는 않았다(p>0.05). 따라서 아이소핌피넬린, 버갑텐, 이찬젠신이 유자씨 유지 내의 주요 암세포 생장 억제 물질이며, 이 중에서 이찬젠신이 그 활성이 가장 높은 물질인 것으로 추정한다.
