Background: Different types of cancer exhibit abnormalities in cell cycle regulators. The murine double minute-2(MDM2) cell cycle regulator is a proto-oncogene that negatively regulates the P53 tumour suppressor gene. Surface epithelial tumours constitute approximately two thirds of ovarian neoplasms. Each histologic type can be classified as benign, borderline and malignant. This study aimed to examine immunohistochemical expression of the MDM2 protein in ovarian serous and mucinous epithelial tumours (benign, borderline and malignant). Materials and Methods: This study included forty five ovarian tumours, subdivided into fifteen cystadenomas (5 serous and 10 mucinous), fifteen borderline tumours (11 serous and 4 mucinous) and fifteen cystadenocarcinomas (9 serous and 6 mucinous). Paraffin sections were stained with haematoxylin and eosin for histopathologic study, and with mouse monoclonal anti-MDM2 antibody for immunohistochemistry. Results: MDM2 positivity was detected in 28.9% of the studied ovarian tumours. All benign tumours were negative and positivity was significantly higher in malignant than borderline tumours (P value of chi-square test =0.000). Significantly, all MDM2 positive mucinous tumours were malignant with no positive mucinous borderline tumours. Malignant tumours showed positive MDM2 expression in 83.3% of mucinous type and in 55.6% of serous type. Borderline serous tumours showed negative MDM2 in 72.7% of cases (P value of Z test =0.04). Conclusions: Alterations in the expression of the cell cycle regulator (MDM2) occur early in the process of tumourigenesis in serous and mucinous ovarian tumours. We suggest that MDM2 may be used in those tumours as a marker for risk stratification and identification of cases with cancer development and progression. We recommend further studies on MDM2 immunohistochemistry, in conjunction with adjuvant methods as DNA ploidy and FISH gene amplification, focusing on the mucinous tumours and differentiating between the three tumour categories, benign, borderline and malignant.
This experiment was focused on the characterization of anti- Toxoplasma monoclonal antibodies (mAbs) and the effect of mAbs on the parasite invasion of mouse peritoneal macrophages. Twenty eight mAbs including M110, M556, R7A6 and M62l were characterized by Ab titer, immunoglobulin isotyping and western blot pattern. Antibody titer (optical density) of 4 mAbs. Ml 10. M556. R7A6 and M62l. were 0.53,0.67, 0.45 and 0.39 (normal mouse serum; 0.19) with the same IgGl isotypes shown by Enzyme-linked immunosorbent assay (ELISA). Western blot analysis showed that Ml 10. M556. R7A6 and M62l reacted with the 33 kDa (p30),31 kDa (p28),43 kDa and 36 kDa protein. Immuno-gold labelling of mAbs M110, M556, R7A6 and M621 reacted with the surface membrane, dense granules and parasitophorous vacuolar membrane (PVM) , rhoptries and cytoplasm of tachyzoite, respectively. For in vitro assay, preincubation of tachyzoties with four mAbs, Ml 10, M556, R7A6 and M62l resulted in the decrease of the number of infected macrophages (p < 0.05) and the suppression of parasite multiplication at 18 h post-infection. Four monoclonal antibodies including Ml 10 (SAGI) were found to have an important role in the inhibition of macrophage invasion and T. gondii multiplication in vitro, and these mAbs may be suitable for vaccine candidates, diagnostic kit and for chemotherapy.
Sialic acid containing glycosphingolipids (gangliosides) have been implicated in the regulation of various biological phenomena such as atherosclerosis. Recent report suggeststhat exogenously supplied disialoganglioside (GD3) serves a dual role in vascular smooth muscle cells (VSMC) proliferation and apoptosis. However, the role of the GD3 synthase gene in VSMC responses has not yet been elucidated. To determine whether a ganglioside is able to modulate VSMC growth. the effect of overexpression of the GD3 synthase gene on DNA synthesis was examined. The results show that the overexpression of this gene has a potent inhibitory effect on DNA synthesis and ERK phosphorylation in cultured VSMC in the presence of PDGF. The suppression of the GD3 synthase gene was correlated with the down-regulation of cyclinE/CDK2. the up-regulation of the CDK inhibitor p21 and blocking of the p27 inhibition,whereas up-regulation of p53 as the result of GD3 synthase gene expression was not observed. Consistently, blockade of GD3 function with anti-GD3 antibody reversed VSMC proliferation and cell cycle proteins. The expression of the CD3 synthase gene also led to the inhibition of TNF--induced matrix metalloproteinase-9 (MMP-9) expression in VSMC as determined by zymography and immunoblot. Furthermore, GD3 synthase gene expression strongly decreased MMP-9 promoteractivlty in response to TNF-. This inhibition was characterized by the down-regulation of MMP-9,which was Iranscriptionally regulated at NF-B and activation protein-1 (AP-1) sites in the MMP-9promoter Finally, the overexpression of MMP-9 in GD3 synthase transfectant cells rescued VSMC proliferation. However MMP-2 overexpression was not affected the cell proliferation. These findings suggest that the fl13 synthase gene represents a physiological modulator of VSMC responses that may contribute to plaque instability in atherosclerosis.
연구배경 : 기관지내시경에서 관찰되는 anthracofibrosis가 활동성결핵과 연관이 있다는 보고가 있으나, 관찰 당시에 결핵의 활동성 여부가 확실하지 않아서 항결핵치료 시행여부를 결정하기 어려운 경우가 있다. 본 연구에서는 결핵의 활동성과 연관이 높은 IL-2 $sR{\alpha}$, IFN-${\gamma}$의 혈청 농도와 결핵의 혈청진단방법인 TBGL 항체의 농도가 anthracofibrosis 환자에서 결핵의 활동성 여부를 진단하는데 도움이 되는지를 알아보고자 하였다. 방 법 : 건강한 자원자(16명), 활동성결핵으로 진단되어 투약하기 직전의 환자(22명), 활동성결핵으로 진단되고 6개월간 항결핵제 투여를 마친 환자(13명), 그리고 anthracofibrosis 환자(15명)에서 혈청을 채취한 뒤, IL-2 $sR{\alpha}$, IFN-${\gamma}$ 그리고 TBGL 항체의 농도를 ELISA 방법으로 측정하였다. 결 과 : 건강한 자원자, 결핵치료전 환자, 결핵치료후 환자, 그리고 anthracofibrosis 환자의 IL-2 $sR{\alpha}$는 각각 $640{\pm}174$, $1,611{\pm}2,423$, $953{\pm}562$, $863{\pm}401$ pg/ml, IFN-${\gamma}$의 농도는 각각 0, $8.16{\pm}17.34$, $0.70{\pm}2.53$, $2.33{\pm}6.67$ pg/ml, 그리고 TBGL 항체 농도는 각각 $0.83{\pm}0.80$, $5.91{\pm}6.71$, $6.86{\pm}6.85$, $3.22{\pm}2.59$ U/ml였다. TBGL 항체 농도만이 건강한 자원자에서 다른 군에 비해 낮았을 뿐(p<0.05) IL-2 $sR{\alpha}$와 IFN-${\gamma}$ 농도는 네 군 사이에 차이가 없었다. Anthracofibrosis 환자 중에서 나중에 활동성 결핵으로 진단된 6명과 나머지 9명 사이에서도 상기한 지표의 농도 차이는 관찰되지 않았다. 결 론 : 1) Anthracofibrosis 환자의 40%에서 활동성 결핵이 동반되었다. 2) 혈청 IL-2 $sR{\alpha}$, IFN-${\gamma}$ 그리고 TBGL 항체 농도는 anthracofibrosis에서 결핵의 활동성 지표로 유용하지 않았다. 3) 혈청 TBGL 항체 농도는 건강한 자원자와 나머지 군 사이에 유의한 차이를 보였다.
Objective: The purpose of this study was to evaluate the efficacy and effect of various cryopreservation method on the survival and the cytoskeletal stability of metaphase II mouse oocyte. Methods: Mouse ovulated oocytes were collected and cryopreserved by a modified slow-freezing method with 1.5 M 1, 2-propanediol (PrOH)+0.1 M sucrose or by vitrification using cryo loop and EM grid with 40% ethylene glycol+0.6 M sucrose. Four hours after thawing, intact oocytes were fixed and stained with fluorescein isothiocyanate (FITC)-conjugated monoclonal anti-$\beta$-tubulin antibody to visualize spindle and propidium iodide (PI) to visualize chromosome. Spindle morphology was classified as follows: normal (barrel-shaped), slightly and absolute abnormal (multipolar or absent). Results: Survival rate of the frozen-thawed oocytes in vitrification group was significantly higher than that of slow-freezing group (62.7% vs. 24.4%, p<0.01). Vitrification with cryo loop showed significantly higher survival rate than that with EM grid (67.7% vs. 53.5%, p<0.05). On the other hand, proportion of normal spindle and chromosome configurations of the frozen-thawed oocytes between two vitrification group was not significantly different. Conclusion: For mouse ovulated oocytes, vitrification with cryo loop may be a preferable procedure compared to slow-freezing method. Further study should be needed to investigate developmental competency of frozen-thawed mouse oocytes.
목적: 최근들어 비호지킨 림프종 환자군에서 $^{131}I$ 표지 rituximab을 이용한 반복적인 방사면역치료가 효과적인 치료방법임이 보고되고 있다. 이 연구에서는 $^{131}I$ 표지 rituximab을 이용하여 반복적인 방사면역치료를 시행받은 비호지킨 림프종 환자군에서 치료전 FDG-PET과 치료후 감마카메라를 이용한 전신 평면방출영상을 이용하여 종양의 흡수선량을 평가하였다. 대상 및 방법: 모두 4명의 환자들에게 치료용량의 $^{131}I$ 표지 rituximab ($7270.5{\pm}276.5MBq$)을 정맥주사하였다. 방사면역치료의 반복휫수는 3명에서는 3회, 1명에서는 4회였으며, 7개의 측정가능한 종양에 대해 총 21회의 평가가 이루어졌다. 감마카메라를 이용한 전신 평면방출영상을 방사면역치료 후 5분, 5시간, 24시간, 48시간 및 72시간에 연속적으로 촬영하였고, FDG PET/CT를 방사면역치료 전 1주일 이내와 방사면역치료 후 30일째 되는 날에 각각 촬영하였다. 방사면역치료 전 PET/CT로부터 관상면의 최대강도투사영상을 얻었고, 이 영상에 AMIDE소프트웨어를 이용하여 연속적인 전신 평면방출영상을 중첩시켰는데, 이 과정에서 4개의 해부학적 표지자(양측 어깨와 엉덩이)를 이용하였다. 중첩된 영상에서 종양부위와 왼쪽 안쪽 넓적다리(배경영역)의 관심영역을 그리고, 종양자체의 시간-방사능곡선을 구하였다. PET/CT에 포함된 CT영상으로부터 종양의 부피를 측정했으며, 각각의 종양부위의 SUVmax를 구하여 종양부피와 SUVmax의 방사면역 치료 전후 변화율을 평가하였다. 종양의 흡수선량과 부피변화율 간의 상관관계를 분석하였고, 방사면역치료 횟수에 따라서 종양의 흡수선량 및 부피 변화율에 유의한 차이가 있는지 알아보았다. 결과: 전체 환자군의 종양부위 흡수선량은 $397.7{\pm}646.2\;cGy$ ($53.0{\sim}2853.0\;cGy$)이었다. 방사면역 치료 전 종양의 부피는 $11.3{\pm}9.1\;cc$ ($2.9{\sim}34.2\;cc$)이었고, 치료 후 종양의 부피 변화율은 $-29.8{\pm}44.3%$($-100.0%{\sim}+42.5%$)이었다. 종양의 흡수선량과 부피 변화율 간에는 유의한 상관관계가 관찰되지 않았으며(p>0.05), 방사면역치료 횟수에 따라서 종양의 흡수선량 및 부피 변화율에도 유의한 차이가 관찰되지 않았다(p>0.05). 결론: FDG-PET의 최대강도투사영상을 이용하여 종양의 위치와 경계를 보다 명확하게 파악할 수 있었고, 감마카메라 영상과의 중첩을 통해서 효과적인 종양의 흡수선량평가가 가능하였다.
