Comparison of anti-oxidant activities of seventy herbs that have been used in Korean traditional medicine

  • Many herbs have been used as therapeutics in Korean traditional medicine. In view of their clinical indications, anti-oxidant activity may contribute to their pharmacological effects. However, anti-oxidant information on these plants has not been available. In this study, seventy herbs which have been used in Korean traditional medicine were selected and screened for anti-oxidant activity using their water extracts. The anti-oxidant activity was assessed by their ability to inhibit three oxidation reactions; luminol/Fenton reagent, 2, 7-dichlorodihydrofluorescein(DCHF)/Fenton reagent and DCHF/peroxynitrite. In each assay, 70 herbs were divided into two groups; anti-oxidant group which inhibited the respective oxidation reaction and was majority(about 60 herbs), and pro-oxidant group which enhanced the oxidation reaction but was minority(more or less 10 herbs). When the herbs were listed in the order of their anti-oxidant strength, the orders obtained from each assay were found to be quite similar. The upper top rankers(more or less 10 herbs) in each assay showed strong activity compared to the others. The uppermost rankers in each assay were Rubus coreanus Miquel/Rubus schizostylus(覆盆子), Schisandra chinensis Baillon/Schizandra chinensis(五味子) and Terminalia chebula Retziusl Terminalia chebula(訶子). Of the pro-oxidant herbs, about 4-5 herbs were strongly pro-oxidant, which enhanced the control oxidation reactions to 150-300%. But the meaning of this observation is not known since few of them in one assay were also anti-oxidant in other assays. The results obtained in the present study may serve as information for understanding pharmacological effects of these herbs and developing new drugs from them.

A Verification of Cosmetic Effect about Anti-oxidant and Anti-wrinkle of 11 Native Plants (자생식물 11종의 항산화 및 항주름에 관한 향장효능 검증)

  • To check the possibility of using natural extracts for cosmetic substances, we tested the bioactive effects of 11 crude extracts originating from native plants grown in the Republic of Korea on anti-oxidant and anti-wrinkle activities. When DPPH scavenging activities were measured, the crude extracts of Vitex rotundifolia, Scirpus triangulatus, S. wichurae, S. tabernaemontani, and Aeschynomene indica assays had more than 70% of DPPH scavenging activities at 100 μg/ml concentration. The data from the hydrogen peroxide scavenging assay revealed that the crude extracts from the 10 species, except for Eleocharis mamillata var. cyclocarpa, could scavenge more than 90% at 1,000 μg/ml concentration. The crude extract of V. rotundifolia had the highest xanthine oxidase scavenging activity at a 1,000 μg/ml concentration. Based on the four anti-oxidant assays, the crude extracts of V. rotundifolia, S. triangulatus, S. wichurae, S. tabernaemontani, and A. indica were shown to have excellent anti-oxidant capacities overall. In addition to the anti-oxidant capacities, the crude extract from S. wichurae exhibited high collagenase inhibitory activity, therefore, it could be a promising candidate for a cosmetic ingredient with anti-oxidant and anti-wrinkle qualities. The results of this study indicate that many wild plants native to the Korea Peninsula can be used as excellent natural substances for cosmetics based on their bioactive effects.

  • The anti-oxidant activities of tectochrysin, a major compound of propolis, were investigated. Tectochrysin exhibited a significant decrease in serum transaminase activities elevated by hepatic damage induced by $CCl_4$-intoxication in rats. Tectochrysin tested exhibited a lipid peroxidation causing a significant decrease in MDA production in TBA-reactant assay. Tectochrysin was strong in the increase in the anti-oxidant enzymes such as hepatic cytosolic superoxide dismutase, catalase and glutathione peroxidase activities in $CCl_4$-intoxicated rats. These results suggest that tectochrysin possess not only the anti-oxidant, but also the activities in $CCl_4$-intoxicated rats. Especially, tectochrysin was found to cause significant increases in the rat liver cytosolic SOD, catalase, GSH-px activities as well as a significant decrease in the MDA production.

Anti-oxidant and Anti-tumor Activities of Crude Extracts by Gastrodia elata Blume (천마추출물의 항산화 및 항암 활성)

  • Gastrodia elata Blume is a major imp0l1ant medicinal resource in Korea. In order to confirm the biological activities of Gastrodia elata Blume, we carried out various in vitro assays. Of them, anti-oxidant and anti-tumor activities were detected from assays. The prototype of Gastrodia elata Blume extracts was used for 1he evaluation of DPPH, FRAP, hydroxyradical scavenging assay as anti-oxidant assays, as well as anti-tumor asctivities as wound assay and invasion assay. As a result, the prototype of Gastrodia elata Blume extracts showed potent anti-oxidative activity and anti-tumor activity in vitro. These above results suggest that 1he Gastrodia elata Blume extracts could have potential to alleviate oxidation process, cell motility activity, and tumorigenesis.

Anti-oxidant Activity and Whitening Activity of Bamboo Extracts (대나무 추출액의 항산화 작용과 미백작용)

  • To investigate the possibility of development as a whitening agent using bamboo extracts (Phyllostachys nigra var. henonis), we measured DPPH assay, NBT/XO assay, intracellular ROS scavenging assay, tyrosinase assay and MSH-induced melanin production in B-16 cells. Bamboo extracts had dose-dependently anti-oxidant activity in DPPH, NBT/XO and intracellular ROS assay. Bamboo extracts appear to inhibit xanthine oxidase directly. Bamboo extracts inhibited not only purified tyrosinase activity but also inhibited tyrosinase activity and melanin production in B16 melanoma cells stimulated by $1{\mu}M\;{\alpha}$-MSH. Anti-oxidant activity and cytotoxicity of ethyl acetate fraction was more potent than those of water fraction, whereas whitening effect of water fraction was stronger than ethyl acetate fraction. Therefore, these results suggest that water fraction of bamboo extracts may be useful for the development as whitening agents reducing cytotoxicity.

  • Lepidoptera (butterflies) extracts, traditionally employed as medicines, have various biological activities. Five species of Lepidoptera (Papilio maackii, Papilio xuthus, Pieris rapae, Eurema hecabe, and Sasakia charonda) were extracted with distilled water (DW), dimethyl sulfoxide (DMSO), ethanol (EtOH), and methanol (MeOH). Each extract was analyzed for anti-oxidant and anti-inflammatory activities using the 1,1-diphenyl-2-picrylhydrazyl (DPPH) assay method, the ferric reducing ability of plasma (FRAP) test, and a cyclooxygenase-2 (COX-2) promoter assay. The results suggest that Lepidoptera extracts have valuable anti-oxidant and anti-inflammatory properties, supporting the idea that the extracts may serve as a food biomaterial(s) preventing oxidative processes and inflammatory damage.

Antioxidant and Anti-inflammatory Effects of Fractions from Pruni persicae Flos (도화(桃花, Pruni persicae Flos) 분획물의 항산화 및 항염증 효과)

  • Objectives : The solvent extracts of Pruni persicae Flos were investigated for the activities of anti-oxidant and anti-inflammatory effects to apply as a functional ingredient for cosmetic products. Methods : In this study, the fractions of P. persicae Flos were extracted with 70.0% acetone and purified using Sephadex LH-20 column chromatography. As a result, eight fractions were isolated. We performed MTT assay, total polyphenol contents, DPPH free radical scavenging assay, SOD-like activity, xanthine oxidase inhibition assay, astringent activity assay, hyaluronidase inhibition assay and the production of nitric oxide. Results : For anti-oxidant effects, the electron donating ability of fraction (Fr.) 2-5, Fr.-8 isolated from P. persicae Flos was above 90.0% at 100 ppm respectively. The superoxide dismutase (SOD) - like activity of Fr.-5 isolated from P. persicae Flos was 92.1% at 1,000 ppm. The xanthine oxidase inhibitory effect of Fr.-6 isolated from P. persicae Flos was about 83.3% at 1,000 ppm. Hyaluronidase inhibition activity related to the anti-inflammation effect was 94.0% for Fr.-4 isolated from P. persicae Flos at 500 ppm. In the anti-inflammation effect, the Fr.-4 isolated from P. persicae Flos inhibited the generation of nitric oxide. Conclusions : All these findings suggested that the fractions of P. persicae Flos has a great potential as a cosmeceutical ingredient with a anti-oxidant and anti-inflammatory effects.

Anti-inflammatory mechanism and Anti-oxidant Effects of Naesohwangryun-tang in LPS-Stimulated RAW 264.7 Macrophage Cells (LPS로 유도된 RAW 264.7 대식세포주에서 내소황련탕(內疎黃連湯)의 항염증 기전 및 항산화 효능 연구)

  • Objectives : The aim of experiment is to examine anti-inflammatory effect and anti-oxidant effect of Naesohwangryun-tang (NSHRT) in LPS-stimulated RAW264.7 macrophage cells. Methods : In the present study, The cell viability was performed by MTT assay. Nitric oxide (NO) production and prostaglandin E2 (PGE2) synthesis were performed by NO assay and ELISA KIT. The anti-oxidant effect was performed by DPPH and ABTS radical scavenging activity. The inhibitory effects of pro-inflammatory mediators and cytokines were confirmed by realtime PCR and western blotting. Results : NSHRT was no cytotoxicity at treated group. NO and PGE2 production were inhibited compared to the LPS treated group and also mRNA and protein expressions were significantly decreased compared to the LPS treated group. Conclusions : According to the above experiments, we confirmed that NSHRT has anti-inflammatory and anti-oxidant effects. It is suggested that NSHRT is potential ingredient of skin diseases.

Anti-Oxidant and Anti-Inflammatory Complex Health Functional Analysis of Chinese Resource Plant Boehmeria, Carlemannia and Clausena (중국 자원식물 보메리아, 칼라마니아, 클로세나의 항노화 항염 복합 건강관리 기능성 연구)

  • Ageing has been known to be deeply related to oxidative activities. Furthermore, inflammation is a response initiated by conditions such as infection and injury. It has been known that complex connections occur between oxidative stress and the inflammatory response, however underlying mechanism needs to be investigated. Chinese resource plants have been widely used as functional food and medicine for a long time, however it is not studied well how many of these resource plants work. We first decided testing anti-oxidant and anti-inflammatory activities with 95% ethanol extracts of the chinese resource plants, Boehmeria pilosinscula (Blume) Hassk., Carlemannia tetragona Hook f. and Clausena emarginata C.C.Huang. To measure anti-oxidant activity, we performed DPPH assay in Raw264.7 cells with 95% ethanol extracts. Clausena extracts showed significantly higher anti-oxidant activity than those of Boehmeria and Carlemannia. We then performed Nitric Oxide assay to measure the inflammation suppression levels with 95% ethanol extracts of these plants. Clausena extracts showed significantly higher suppression of nitric oxide production than those of Boehmeria and Carlemannia. This indicates that inflammation levels are significantly reduced by Clausena. After measuring anti-oxidant and anti-inflammatory activities, we then performed MTT assay to measure the cell survival rate of Raw264.7 cells treated with these extracts. Boehmeria showed much more cell survival rate than Carlemannia and Clausena. Taken together, this result suggests that Clausena extracts have more anti-oxidant and antiinflammatory activities than Boehmeria and Carlemannia, while Boehmeria extracts have more cell survival rate than Carlemannia and Clausena.

Study on Anti-Skin Aging Effect of Sanguisorba officinalis L. (지유(地楡)의 피부 노화(老化)에 대한 연구(硏究))

  • To develop a new anti-skin aging cosmetics or functional foods by using antioxidative activity and collagenase inhibitor, a potent collagenase or elastase inhibitor was screened from various extracts of medicinal plants and its optimal extraction condition was investigated. And antioxidative activity, antimicrobial activity and inhibitory of effect against collagenase activity were investigated. In the these results, we selected the Sanguisorba (Sanguisorba officinalis L.) that presents a potential biological activities. Sanguisorba which is very rich in triterpenoid saponin and tannins was recently reported its anti-oxidant activities and phytoestogenic activities in vivo test and many clinical studies. The experiments were carried out in vitro to determine anti-oxidant activities of Sanguisorba extracts on DPPH radical scavenging activity assay, Superoxidase scavenging activity assay, Elastase and collagenase activity assay. It show that the Sanguisorba extracts have the most significant anti-oxidant on free radical scavenging activity assay, and also inhibited significantly activities of elastase, collagenase. Further, Sanguisorba extracts are activated Type I collagen protein expression in CCD-986sk cells. These result suggest that the Sanguisorba extracts on DPPH radical scavenging activity assay, Superoxidase scavenging activity assay, elastase and collagenase activity assay, Type I collagen protein expression in CCD-986sk cells effected could be developed cosmetic ingredients for anti-aging.
