도화(桃花, Prunus persica Flos)의 항산화 및 항염증 활성 (Anti-oxidant and Anti-inflammation Activities of Prunus persica Flos)

  • 도화 추출물의 항산화능 및 항염증 효과를 검증하여 기능성 화장품 소재로서의 가능성을 검증하였다. 도화의 항산화능 측정으로 전자공여능을 측정한 결과 도화 에탄올 및 아세톤 추출물 500ppm에서 90.0% 이상의 효과를 나타내어, 합성 항산화제로 알려진 BHT와 유사한 효과를 나타내었으며, SOD 유사활성능 측정 결과 도화 에탄올 및 아세톤 추출물 1,000ppm에서 40.0% 이상의 활성을 나타내었다. Xanthine oxidase 저해활성능 측정 결과 도화 에탄올 및 아세톤 추출물 1,000 ppm에서 vitamin C와 유사한 30.0%의 효과를 나타내어, 도화 열수 추출물에 비해 도화 에탄올 및 아세톤 추출물의 효과가 높게 나타났다. 항염증 효과 측정으로 hyaluronidase 저해활성을 측정한 결과 도화 열수, 에탄올 및 아세톤 추출물에서 35.0%의 저해활성을 나타내었으며, macrophage 세포를 이용한 nitric oxide (NO) 저해활성을 측정한 결과 도화 추출물 모든 농도에서 12시간 이후부터 NO 생성량이 억제됨을 확인할 수 있었다. 또한 피부염증 유발균인 Staphylococcus epidermidis와 Propionibacterium acnes의 항균효과를 측정한 결과 도화 에탄올 추출물에서 항균 효과를 나타내었다.

한약재 추출물에 의한 Helicobacter pylori의 생장 및 Urease 활성 억제 (Innibition of Cell Growth and Urease Activity of Helicobacter pylori by Medicinal plant Extracts)

  • 14종 한약재의 70% 에탄을 추출물의 Helicobacter pylori에 대한 항균활성은 황련 (Coptis japonica Makino)이 가장 뛰어 났으며, 소엽 (Perilla frutescens var. acuta KUDO), 소목 (Caesalpinia sappan L.) 및 형개 (Schizonepeta tenuifolia Briq.)의 경우에도 높은 항균활성을 나타내었다. 그러나 Helicobacter pylori의 위내 서식을 도와주는 것으로 알려진 urease 활성억제능은 연교 (Forsythiae Fructus)의 메탄올 (80%) 추출물에서 가장 높았으며, 소목 및 형개의 추출물도 높았다. 특히 연교의 경우 한약재 추출물을 넣지 않은 대조군과 비교하여 80% 이상의 활성이 억제되었다. 연교의 메탄올 추출물을 물, 에틸아세테이트 (ethyl acetate), 부탄을 순으로 분획하여 각 분획물에 대한 urease 활성억제능을 검색한 결과 에틸아세테이트 분획에서 92%의 urease 활성이 억제되었다.

  • Flavonoid are large group of the polyphenolic compounds which are distinguished by an aromatic or phenolic ring structure and the phenolic compounds are induced by microbial infection, ultraviolet radiation, temperature and chemical stress. They are known for their antioxidant activity, anti-allergic, anti-inflammatory, anti-microbial and anti-cancer activities. In this study, changes in flavonoid content were investigated using heterologous chalcone isomerase (CHI) expression system. Also, phenolic compounds level was measured to examine the relation between flavonoids and phenols contents. Explants of lettuce (Lactuca sativa L.) were transformed with Agrobacterium tumefaciens LBA 4404 strain containing pFLH-CHI (derived from pPZP2Ha3) vector constructed with CHI gene from Brassica rapa. The putative transgenic plants were confirmed by genomic DNA PCR analysis. Also the transcription levels of the gene were analyzed by semi-quantitative RT-PCR with gene specific primers. The total flavonoid contents were increased at $T_0$ and $T_1$ generations over 1.4 and 4.0 fold, respectively. Total phenol contents also increased at $T_1$ generation. These results indicate that CHI gene plays an important role to regulate the accumulation of flavonoids and its component changes.

In vitro control of plant pathogens by using anti-fungal hydroxy fatty acid obtained from microbial bioconversion of linolenic acid

  • Bioconverted linolenic acid (bLNA) obtained from linolenic acid by Pseudomonas aeruginosa PR3, showed anti-fungal activity against plant pathogens such as B. cinerea, F. oxysporum, F. solani, P. capsici and C. capsici. The oil sample also showed anti-fungal activity with MIC values, ranging from >250 to >1,000 ${\mu}g/ml$. Varied concentrations of bLNA had a great potential effect on spore germination of different fungi.

Microbial Strains and Bioactive Exopolysaccharide Producers from Thai Water Kefir

  • The aims of this novel work were to determine the microbial strains and exopolysaccharide (EPS) producers in water kefir from Nakhon Ratchasima Province, Thailand. Thirty-three microbial strains were identified using 16S rRNA gene analysis consisting of 18 bacterial strains, as 9 strains of acetic acid bacteria (AAB), 9 strains of lactic acid bacteria (LAB), and 15 yeast strains. All bacteria were able to produce EPS with a diverse appearance on agar media containing different sugars at a concentration of 8%. Culture supernatants from AAB and LAB showed 31-64% 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging activity with the highest antioxidant activity of 64% from Acetobacter pasteurianus WS3 and WS6. Crude EPS from A. pasteurianus WS3 displayed the highest ferric reducing antioxidant power at 280 mM $FeSO_4/g$ EPS, greatest anti-tyrosinase activity at 20.35%, and highest EPS production of 1,505 mg EPS/L from 8% sucrose. These microbes offer beneficial health implications and their EPSs can be used as food additives and cosmetic ingredients.

Effect of Lactobacillus plantarum FH185 on the Reduction of Adipocyte Size and Gut Microbial Changes in Mice with Diet-induced Obesity

  • This study aimed to investigate the effects of Lactobacillus plantarum FH185 on the reduction of adipocyte size and gut microbial changes in mice with diet-induced obesity. The strain was found to have a lipase inhibitory activity of 70.09±2.04% and inhibited adipocyte differentiation of 3T3-L1 cells (18.63±0.98%) at a concentration of 100 µg/mL. To examine the effect of the strain supplementation on gut microbial changes in mice with diet-induced obesity, male C57BL/6J mice were fed on four different diets (i.e., A, normal diet (ND); B, high-fat diet (HFD); C, HFD with ABT-3 (109 CFU/day); and D, HFD with L. plantarum FH185 (109 CFU/day)) for 6 wk. According to the results of fecal pyrosequencing, the ratio of Firmicutes to Bacteroidetes in groups C and D was lower than in the control groups at the phylum level. At the family level, Lactobacillaceae in groups C and D was observed to dominate, while Lachnospiraceae in groups A and B was observed to dominate. At the genus level, Lactobacillus in groups C and D was comparatively higher than in groups A and B. To examine the effects of strain supplementation on the reduction of adipocyte size, the left and right epididymal fat pads were quickly isolated after the animals were sacrificed, and the adipocyte sizes were measured. In groups A, C and D, the percentage of 2,000 m2 of adipocyte was higher than in the other size of adipocyte, while the percentage of over 5,000 m2 of adipocyte was highest in group B. The mean adipocyte size of group D was significantly larger than that of group A, but smaller than that of group B.

Propolis 추출물의 병원성 미생물에 대한 항균 효과 (Antibacterial Effects of Propolis Extracts on Pathogenic Bacteria)

  • The optimal concentration of ethanol to separate a high quantity of propolis was $60\%$ but that for the best flavonoids extraction was $80\%$ We compared the yields of propolis from different countries. In this study we used $60\%$ ethanol concentration as a standard. The yield of propolis was proportional to the contents of flavonoids. Namely, Polish propolis which showed the highest yield with $56\%$ by the extraction with $60\%$ ethanol revealed also the highest flavonoids content with $3.49\%$ among all the samples tested The major constituents of propolis differed from country to country. It has been suggested that the different geographical origin influenced the efficacy and the constituents of propolis. Antibacterial activity of ethanol extracted propolis from different countries was tested against 6 microbial strains of type cultures including Gram-positive (Staphylococcus aureus, Streptococcus uberis, Streptococcus agalactiae) and Gram- negative bacteria (Klebsiella pneumoniae, Proteus vulgaris and E coli) in vitro. Propolis extract showed anti-microbial activity against all the tested bacterial strains. In addition, propolis was sensitive to E coli which was resistant to broad spectrum antibiotics like ampicillin. These results showed that propolis may substituted for commercial antibiotics. The efficiency of anti-microbial activity of the propolis was slightly higher in $80\%$ than $97\%$ ethanol extract.

항균 효능이 있는 비타민 B1 유도체(Thiamine Dilauryl Sulfate:TDS)의 나노입자화를 통한 기능성 향장 활성 증진 (Enhancement of the Cosmeceutical Activity by Nano-encapsulation of Thiamine Di-lauryl Sulfate (TDS) with antimicrobial efficacy)

  • 본 연구는 레시틴으로 나노입자화 시킨 티아민 디라우릴 설페이트의 향장활성 증진에 관한 것이다. TDS를 포집시킨 나노입자는 150 ~ 200 nm의 크기를 나타내는 구형이며, 또한 제타포텐셜을 측정하여 여러 pH 범위에서 안정한 것을 확인하였다. TDS 나노입자는 인간 섬유아세포(CCD-986sk)에 높은 농도를 처리하여도 85%의 세포생존률을 보였다. 자유라디칼소거활성 실험을 진행한 결과 나노입자화하지 않은 TDS 희석액(1.0 mg/mL)은 81.6%의 활성을 나타내었고, 나노입자화한 TDS 용액은 이보다 더 높은 88.1%의 높은 라디칼 소거활성을 보였다. TDS 나노입자는 자외선을 조사시킨 CCD-986sk에서 MMP-1의 발현을 41.4% 감소시켰다. TDS 용액과 TDS 나노입자를 가지고 salmonella typhimurium, listeria monocytogenes에 대하여 항균활성을 측정하였다. TDS 나노입자의 경우 양성대조군의 항균활성과 비슷한 결과를 나타내었다. 이러한 결과들로 TDS 나노입자가 항산화, 미백, 주름개선 효능같은 향장 소재로서의 적용이 가능할 것이라 생각된다.

Spectral, Electrochemical, Fluorescence, Kinetic and Anti-microbial Studies of Acyclic Schiff-base Gadolinium(III) Complexes

  • A new series of acyclic mononuclear gadolinium(III) complexes have been prepared by Schiff-base condensation derived from 5-methylsalicylaldehyde, diethylenetriamine, tris(2-aminoethyl) amine, triethylenetetramine, N,N-bis(3-aminopropyl)ethylene diamine, N,N-bis(aminopropyl) piperazine, and gadolinium nitrate. All the complexes were characterized by elemental and spectral analyses. Electronic spectra of the complexes show azomethine (CH=N) within the range of 410-420 nm. The fluorescence efficiency of Gd(III) ion in the cavity was completely quenched by the higher chain length ligands. Electrochemical studies of the complexes show irreversible one electron reduction process around -2.15 to -1.60 V The reduction potential of gadolinium(III) complexes shifts towards anodic directions respectively upon increasing the chain length. The catalytic activity of the gadolinium(III) complexes on the hydrolysis of 4-nitrophenylphosphate was determined. All gadolinium(III) complexes were screened for antibacterial activity.
