Anti-lipid Peroxidation and Liver Protective Effects of Polygonum aviculare L. (마디풀(Polygonum aviculare L.)의 전초가 지질과산화(脂質過酸化) 및 간기능(肝機能)에 미치는 영향)

  • Polygonum aviculare L. was extracted by methanol and then fractionated systematically with solvents and column chromatographic method in order to isolate ingredients with anti-lipid peroxidation and liver protectective effects. Among these fractions, ethylacetate soluble part(MWE) showed the strongest in the anti-lipid peroxidation. Furthermore, the crude subtraction(MWE 4) with Rf value near 0.42, which is separated from MWE by column chromatography using a solvent system, inhibited the lipid peroxidation of rat liver in vitro. Moreover, MWE 4 decreased GOT, GPT and TBA value compared with control and suggested high protective effects against hepatotoxicity induced by carbon tetrachloride in mice. The active compounds in MWE 4 were assumed to be flavonoid glucosides.

  • Isatis indigotica is commonly used as traditional chinese medicine for antipyretic, antiviral, and detoxifying purpose in China. In order to examine whether it prevents lipid peroxidation, root of Isatis indigotica was extracted with methanol and fractionated with hexane, ethyl acetate, butanol and water. The activity was evaluated by DPPH method and lipid peroxidation in rat liver homogenate and C $u^{++}$ -induced oxidated LDL. The results showed that ethyl acetate fraction and indigo, which is known as its constituent, had significantly anti-lipid peroxidative effects.s.

  • The root of Astragalus membranaceus (Leguminosae) has been used in traditional chinese prescriptions for strengthning the superficial resistance, promoting pus discharge and tissue regeneration, diuretis and alleviating edema. Lipid peroxidation has been suggested as a major cause of atherosclerosis, cancer, liver disease, and the aging process. In order to investigate the anti-lipid peroxidation activity, Astragali Radix was extracted with $80\%$ MeOH and fractionated with hexane, $CH_{2}Cl_2$, EtOAc, BuOH and Water. The antilipid peroxidation activities of them were determined by human erythrocyte ghost and $CCl_4$-induced lipid peroxidation. $CH_{2}Cl_2$ and EtOAc fractions, especially, isoflavonoids, 7,2'-dihydroxy-3',4'-dimethoxyisoflavan-7-O-${\beta}$-D-glucoside and calyco sin-7-O-${\beta}$-D-glucoside from EtOAc fraction showed anti-lipid peroxidation activities.

Effect of Glutamine on the Diclofenac Induced Bacterial Translocation and Lipid Peroxidation (Diclofenac에 의해 유발된 장내세균전위와 지질과산화에 대한 글루타민의 효과)

  • The aim of this study was to examine whether administration of glutamine are able to prevent the NSAID induced bacterial translocation and lipid peroxidation in the rats. The an imals with glutamine were fed with L-glutamine for 5 days before diclofenac administration (100 mg/kg orally). 48 hour after diclofenac administration, intestinal permeability, serum biochemical profiles, and malondialdehyde levels of ileum were measured for evaluation of gut damage. Also, enteric aerobic bacterial counts, number of gram-negatives in mesenteric Iymph nodes, liver, spleen and kidney and malondialdehyde levels in liver, spleen, kidney and plasma were measured. Diclofenac caused the gut damage, enteric bacterial overgrowth, increased bacterial translocation and increased lipid peroxidation. Co-administration of glutamine reduced the gut damage, enteric bacterial overgrowth, bacterial translocation and lipid peroxidation induced by diclofenac. This study suggested that glutamine might effectively prevent non-steroidal anti-inflammatory drug induced bacterial translocation and lipid peroxidation in the rat.

  • The ethylacetate fraction of the overground portion of Polygonum aviculare L. exhibited the anti-lipid peroxidation and the liver protective effect in intoxicated rats. Through silica gel chromatography of the ethylacetate fraction monitered by bioassay, two flavonoids, avicularin and juglanin were isolated as active components. Avicularin and juglanin remarkablely inhibited the lipid peroxidation of rat liver induced by 50% ethanol. Especially avicularin exhibited the stronger anti-lipid peroxidation effect than juglanin. Avicularin as a main principle of Polygonum aviculare L. significantly exhibited liver protective activities by decreasing s-GOT and s-LDH levels which represent for the hepatotoxicity induced by $CCl_4$ in rats. In addition, avicularin significantly decreased not only s-LDH but also s-bilirubin levels in intoxicated rat induced by ${\alpha}-naphthylisothiocyanate\;(ANIT)$. These results suggest that avicularin has the protective effects against the hepatoxicity induced by $CCl_4$ and ANIT in rats.

The Effect of Cichorium intybus Extract on Hepatotoxicity in Rats

  • Cichorium intybus (Compositae) has been used for fevers, dyspepsia, headache and jaundice, as a demulcent. Also, it has relaxation effects and relief effects against coffee and teas, and is widely used as food. We investigated anti-lipid peroxidative effects and liver protective activity on $CCl_4$ induced lipid peroxidation and hepatotoxicity in rats. MeOH Ex. enhanced the inhibition of anti-lipid peroxidative effects in liver lipid. In chemical parameters obtained from serum analysis, MeOH Ex. revealed significant decrease on hepatotoxicity. The results were as follows; 1. The inhibitory effects of lipid peroxidation were shown in accordance with the increase of samples' concentration level. 2. In chemical parameters obtained from serum analysis, the activities of GOT, GPT, AlP were restored to near the normal level. The contents of cholesterol and BUN showed inhibitory effects with valence. 3. The weights of liver and spleen were not able to restore to the normal level. But on a general level, they were reduced more than the control group.

Anti-lipid Peroxidation Activity of Acanthopanax var, albeofructus (흰털오갈피나무 추출물의 지질과산화 억제작용)

  • Acanthopanax species (Araliaceae) has been traditionally used as analgesics, stimulants of immune system, and replenishment of body functions. Acanthopanax divaricatus var. albeofructus is a indigenous plant in Korea. The anti-lipid peroxidation activities of the loaves and root bark extracts of A divaricatus var. albeofructus were determined. Activities of MeOH extracts were examined in vitro and in vivo with control group intoxicated by carbon tetrachloride. Both extracts showed significant inhibitory effects on the lipid peroxidation.

Effects of Polyacetylene Compounds from Panax Ginseng C.A. Meyer on $CCl_4$-Induced Lipid Peroxidation in Mouse Liver

  • The inhibitory effect of three polyacetylene compounds, panaxydol, panaxynol and panaxytriol isolated from Panax ginseng C.A. Meyer on $CCl_4$induced lipid peroxidation in vivo and in vitro hepatic microsomal lipid peroxidation induced by ADP-$Fe^{3+}$, NADPH and NADPH-cytochrome P-450 reductase were investigated. Their effects on lowering the lipid peroxide levels both in serum and liver and lowering the serum enzyme (GOT, GPT, LDH) activities without the $CCl_4$-induction were also determined. Male ICR mice were pretreated i.p. with polyacetylene compounds or DL-${\alpha}$-tocopherol before administration of $CCl_4$ i.p. and 20 hr after the administration of $CCl_4,$ serum and liver were analyzed. Hepatic microsome was isolated and used for the in vitro NADPH-dependent lipid peroxidation system. Except for panaxynol, treatment with polyacetylenes to control mice did not reduce the levels of lipid peroxides and serum enzyme activities. Panaxynol itself inhibited lipid peroxidation in the liver of normal mice. Polyacetylene compounds protected from the $CCl_4$-induced hepatic lipid peroxidation and lowered serum lipid peroxide levels. Polyacetylenes also inhibited the in virto hepatic microsomal lipid peroxidation in a dose-dependent manner. The results suggest that panaxydol, panaxynol and panaxytriol seem to be the antioxidant components which contribute the anti-aging activities of Panax ginseng C.A. Meyer.

Effects of Butanol Extract of Aralia elata on Lipid Peroxidation (두릅나무 부탄올 추출물이 지질 과산화에 미치는 영향)

  • It is well known that lipidperoxide, formed in vivo, induced the denaturation of enzyme and destruction of cell membrane to acute injury of tissue. Aralia elata have physiological activates, the improvement of lipid metabolism, antidiabetic activity etc., which was thought to have the relationship to lipid peroxidation. The anti-lipidperoxidative effect of Aralia elata have not yet established. In this study, we examined the anti-lipidperoxidative effects of Aralia elata (Butanol fraction) on CCI$_{4}$ induced lipidperoxidation in rats, and elucidated the anti-lipidperoxidative mechanism. In rat liver homogenate intoxicated with CCI$_{4}$ (0.5 ml/100g), BuOH fraction of Aralia elata (80 mg/Kg/day) exhibited 85.41% anti-lipidperoxidative effect but in serum 69.63% inhibitory effects, respectively. In mitochondrial and microsomal fraction showed inhibition of 55.85% and 69.30%, respectively. In order to elucidate the mechanism of anti-lipidperoxidation effects of Aralia elata, enzymatic (NADPH dependent) and non-enzymatic (Ascorbic acid catalyzed) reaction, in vitro, were performed. In enzymatic reation, Aralia elata exhibited 59.43% anti-lipidperoxidation effects, but in non-enzymatic reaction exhibited 43.27% inhibition. Therefore, it is noteworthy that antioxidative power of them may mainly results from the inhibition by enzymatic reaction.

Evaluation of phlorofucofuroeckol-A isolated from Ecklonia cava (Phaeophyta) on anti-lipid peroxidation in vitro and in vivo

  • Lipid peroxidation means the oxidative degradation of lipids. The process from the cell membrane lipids in an organism is generated by free radicals, and result in cell damage. Phlorotannins, well-known marine brown algal polyphenols, have been utilized in functional food supplements as well as in medicine supplements to serve a variety of purposes. In this study, we assessed the potential anti-lipid peroxidation activity of phlorofucofuroeckol-A (PFF-A), one of the phlorotannins, isolated from Ecklonia cava by centrifugal partition chromatography in 2,2-azobis (2-amidinopropane) dihydrochloride (AAPH)-stimulated Vero cells and zebrafish system. PFF-A showed the strongest scavenging activity against alkyl radicals of all other reactive oxygen species (ROS) and exhibited a strong protective effect against ROS and a significantly strong inhibited of malondialdehyde in AAPH-stimulated Vero cells. The apoptotic bodies and pro-apoptotic proteins Bax and caspase-3, which were induced by AAPH, were strongly inhibited by PFF-A in a dose-dependent manner and expression of Bcl-xL, an anti-apoptotic protein, was induced. In the AAPH-stimulated zebrafish model, additionally PFF-A significantly inhibited ROS and cell death, as well as exhibited a strong protective effect against lipid peroxidation. Therefore, these results suggest that PFF-A has excellent protective effects against ROS and lipid peroxidation induced by AAPH in both an in vitro Vero cell model and an in vivo zebrafish model.
