Development of Plant Anti-inflammatory Agents : Comparison of Anti-inflammatory and Analgesic Activities of Extracts from Lonicera japonica (식물성 항염증제의 개발 : 인동추출물에 대한 항염증 및 진통작용의 비교)

  • For developing plant anti-inflammatory agents, extracts from Lonicera japonica were obtained and evaluated for their anti-inflammatory and analgesic activities using acute/chronic inflammatory models and writhing tests. It was found that the extracts with a modified extraction method showed higher anti-inflammatory and analgesic activities than those of the extracts based on the ancient literatures.

Anti-Inflammatory Components of Platycodi Radix Butanol Fractions (길경(Platycodi Radix) Butanol 분획물의 항염증 활성 성분)

  • This study was conducted to examine the development of a new anti-inflammatory substance with potent anti-inflammatory activities that was derived from the Platycodi Radix butanol fraction. To accomplish this, the chemical structures and anti-inflammatory activities of the components were elucidated. Upon column chromatography of the tertiary subfraction, fractions 8-4-1 and 8-4-2 were identified as platycodin D and D3, respectively, following recrystallization, based on melting point (MP), infrared (IR), and positive fast atom bombardment (FAB)-mass and nuclear magnetic resonance (NMR) spectral data. Platycodin D and D3 exhibited strong anti-inflammatory activities in rats when administerd at oral doses of 12 mg/kg and 36 mg/kg, p.o., respectively. Platycodin D and D3 induced inhibitory effects on capillary vascular permeability in rats at oral doses of 16 mg/kg and 24 mg/kg, p.o., respectively, and potent inhibition of leukocyte emigration in a carboxymethyl cellulose (CMC)-pouch when administered at doses of 3 mg/rat and 7 mg/rat, s.c., respectively. These results verified the high antiinflammatory potency of the platycodin D and D3 components in Platycodi Radix.

Evaluation of Antioxidant, Anti-cholinesterase, and Anti-inflammatory Effects of Culinary Mushroom Pleurotus pulmonarius

  • Culinary mushroom Pleurotus pulmonarius has been popular in Asian countries. In this study, the anti-oxidant, cholinesterase, and inflammation inhibitory activities of methanol extract (ME) of fruiting bodies of P. pulmonarius were evaluted. The 1,1-diphenyl-2-picryl-hydrazy free radical scavenging activity of ME at 2.0 mg/mL was comparable to that of butylated hydroxytoluene, the standard reference. The ME exhibited significantly higher hydroxyl radical scavenging activity than butylated hydroxytoluene. ME showed slightly lower but moderate inhibitory activity against acetylcholinesterase (AChE) and butyrylcholinesterase than galantamine, a standard AChE inhibitor. It also exhibited protective effect against cytotoxicity to PC-12 cells induced by glutamate ($10{\sim}100{\mu}g/mL$), inhibitory effect on nitric oxide (NO) production and inducible nitric oxide synthase protein expression in lipopolysaccharide-stimulated RAW 264.7 macrophages, and carrageenan-induced paw edema in a rat model. High-performance liquid chromatography analysis revealed the ME of P. pulmonarius contained at least 10 phenolic compounds and some of them were identified by the comparison with known standard phenolics. Taken together, our results demonstrate that fruiting bodies of P. pulmonarius possess antioxidant, anti-cholinesterase, and inflammation inhibitory activities.

Drug Interactions of Ibuprofen and Prednisolone in Antiinflammatory and Antipyretic Effects (항염 및 해열효과에 미치는 Ibuprofen과 Prednisolone의 약 상호 작용)

  • The authors investigated drug interaction of ibuprofen and prednisolone in antiinflammatory and antipyretic activities. We have found significant differences of the antiinflammatory and antipyretic activities between single and concurrent administration of ibuprofen and prednisolone, using Sprague-Dawley Strain rats, carrageenin as a phlogistic agent and brewer's yeast as a fever inducing agent. 1) Ibuprofen(20mg/kg) was administered to the rats orally and resulted in significant reduction of (31.70 %) the swelling of rat paw induced by carrageenin, 2) prednisolone (9mg/kg) showed significant reduction of (45.76%) the swelling, 3) concurrent administration of ibuprofen (20mg/kg) and prednisolone (9mg/kg) also reduced (57.40%) the swelling. In ibuprofen (125mg/kg) administration, the inhibition rate of edema was 39.32% and in prednisolone (1mg/kg) administration, the rate was 39.04%. In concurrent administration of ibuprofen (125mg/kg) and prednisolone (1mg/kg), the inhibition rate of edema was 63.09%. Concurrent administration of ibuprofen and prednisolone showed more anti-inflammatory effects than single administration of ibuprofen and prednisolone respectively. Prednisolone itself did not show antipyretic effect, but concurrent administration of ibuprofen and prednisolone showed more antipyretic effects than ibuprofen single administratron.

Anti-inflammatory Activity of Veronica peregrina

  • Veronica peregrina (Scrophylariaceae) has been widely used as a Korean traditional medicine for the treatment of various pathological conditions including infection, hemorrhage and gastric ulcer. In the current study, we investigated the inhibitory effect of methanolic extracts of V. Peregrina (VPM) on the LPS-mediated nitric oxide (NO) production in mouse (C57BL/6) peritoneal macrophages. NO production was significantly down-regulated by the treatment of VPM dose dependently. To evaluate the mechanism of the inhibitory action of VPM on NO production, we performed iNOS enzyme activity assay and checked the change of inducible nitric oxide synthase (iNOS) levels by Western blotting. Although VPM did not affect iNOS enzyme activity, iNOS protein expression was attenuated by VPM indicating VPM inhibits NO production via suppression of iNOS enzyme expression. In addition, VPM attenuated the expression of another pro-inflammatory mediator such as cyclooxygenase-2 (COX-2) in a dose dependent manner. We also found that VPM can reduce trypsin-induced paw edema in mice. Based on this study, we suggest that V. peregrina may be beneficial in diseases which related to macrophage-mediated inflammatory disorders.

Antioxidant, anti-inflammatory and anti-acetylcholinesterase activities of fruiting bodies of Phellinus xeranticus (기와층버섯 자실체의 메탄올 및 열수추출물의 항산화, 항염증 및 아세틸콜린에스테라제 저해 활성)

  • Phellinus xeranticus is an medicinal mushroom belongs to Family Hymenochaetaceae of Polyporales, Basidiomycota. The purpose of this study was to investigate the antioxidant, anti-inflammatory and anti-acetylcholinesterase activities of methanol and hot water extracts prepared from fruiting bodies of Phellinus xeranticus. Besides measuring of 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging activity, a reducing power and a chelating activity on ferrous ions were also measured to evaluate the antioxidant activity of the extracts. To measure the anti-inflammatory activities of the extracts, nitric oxide(NO) production from lipopolysaccharide (LPS) stimulated RAW 264.7 macrophage cells and carrageenan-induced acute hind paw edema of rats were investigated. The results showed that the extracts have excellent DPPH scavenging and chelating activity on the ferrous ions compared with positive controls. The nitric oxide (NO) production in LPS-induced RAW 264.7 macrophage cells were decreased as the concentration of the mushroom extracts increased. Significant reduction of paw edema of rats were observed at 2~6 h after treatment of methanol and hot-water extracts with 50 mg/kg concentration to the rats which are induced acute hind paw edema by carrageenan administration. The anti-acetylcholinesterase activity of the methanol extract of the mushroom showed 83.34% inhibition on AcHE which is lower than that of positive control galanthamine. The experimental results suggested that methanol and hot-water extracts of Phellinus xeranticus fruiting bodies might be used for good sources of antioxidant, anti-inflammatory and anti-acetylcholinesterase agents.

  • The purpose of this research was to investigate the effect of Yonggak-san (YGS) on the anti-allergic reaction in vivo and in vitro. Administration of YGS(500 mg/kg) enhanced hemaggutination(HA)titer against SRBC. On the while, YGS inhibited hyaluronidase activity in vitro and passive cutaneous anaphylaxis reaction, lethal anaphylaxis and mortality induced by compound 48/80 in mice, YGS decreased Arthus reaction, acute hind paw edema induced by histamine. But YGS did not affect delayed type hypersensitivity induced by SRBC. These results suggest that YGS have anti-allergic action

  • The anti-inflammatory and analgesic action of the components isolated from some edible plants carried out. We isolated the quercitrin, compound 3 and o-counmaric acid from Cedrela sinensis, Oenanthe javanica and Artemisia princeps var. Orientalis respectively. O-coumaric acid showed the inhibitory effect on carrageenin-induced edema as well as vascular permeability in mice. And we also found the analgesic activity in all compounds isolated from these plants.

  • Studies were undertaken to develop for the anti-inflammatory analgesic activities of 'Insam-Tang' extract, which composed of four crude drugs; Ginseng Radix, Glycyrrhizae Radix, Atractylodis Rhizoma, and Zingiberis Rhizoma, was known to be applied in edema, convulsion, excremental blood, and cooling pain etc, in oriental herb remedies. It was exhibited significant anti-inflammatory effect on carrageenin inflammation, and preventive and therapeutic effects to chronic inflammation on Freund' complete adjuvant in rats, Especially, therapeutic effect of extract 500mg/kg was observed more effective than that of phenylbutazone 50mg/kg. Also, it was shown significant effect on leakage of dye into the peritoneal cavity and analgesic effect on squirm symptom induced by acetic acid in mice.

  • This study was attempted to examine the acute toxicity and the effect of Sopunghwalhul-Tang water extract(SHTE) on analgesic action in mice, anti- inflammatory action in rats and blood pressure in rabbits. The results were as follows; SHTE showed 10% mortality at 2,000mg/kg(p.o.), had significant analgesic activity in the pain caused by 0.7% acetic acid and was shown to have significant anti-inflammatory activity in the edema-induced by 5% acetic acid at 450mg/kg. SHTE given into a ear vein of rabbits produced a dose-related vasodepressur responses. SHTE-induced hypotension was significantly inhibited by pretreatment with atropine, but was not affected by chlorisondamine, phentolamine, propranolol, and dephenhydramine.
