To explore the anti-allergic and anti-inflammatory effects of extracts of Petasites genus, we studied the effects of s-petasin, a major sesquiterpene from Petasites formosanus (a butterbur species) on asthma and peritonitis models. In an ovalbumin-induced mouse asthma model, s-petasin significantly inhibited the accumulations of eosinophils, macrophages, and lymphocytes in bronchoalveolar fluids. S-petasin inhibited the antigen-induced degranulation of ${\beta}$-hexosamidase but did not inhibit intracellular $Ca^{2+}$ increase in RBL-2H3 mast cells. S-petasin inhibited the LPS induction of iNOS at the RNA and protein levels in mouse peritoneal macrophages. Furthermore, s-petasin inhibited the production of NO (the product of iNOS) in a concentration-dependent manner in the macrophages. Furthermore, in an LPS-induced mouse model of peritonitis, s-petasin significantly inhibited the accumulation of polymorpho nuclear and mononuclear leukocytes in peritoneal cavity. This study shows that s-petasin in Petasites genus has therapeutic effects on allergic and inflammatory diseases, such as, asthma and peritonitis through degranulation inhibition in mast cells, suppression of iNOS induction and production of NO in macrophages, and suppression of inflammatory cell accumulation.
Objectives : In this study, we investigate the anti-allergic effects of Ullmus macrocarpa Hance (Ulmus) on RBL-2H3 mast cell (basophilic leukemia cell line), which are mediated by FcεRIs. Methods : We evaluated the effect of the ethanol extract of Ulmus on the allergic inflammatory response in IgE-antigen-mediated RBL-2H3 cells. Cell toxicity was determined by MTT assay and the markers of degranulation such as beta-hexosaminidase, histamine, PGD2, TNF-α, IL-4, IL-6 production of inflammatory mediators and FcεRI-mediated protein expression by western blot. Results : Ulmus inhibited degranulation and production of allergic mediators (e.g., TNF-α, IL-4, and IL-6) in them. Ulmus reduced histamine levels, expression of FcεRI signaling-related genes such as Lyn, Syk, and Fyn, and extracellular signal-regulated kinase phosphorylation in mast cells. Also, Ulmus reduced PGD2 release and cyclooxygenase-2 expression, and cytosolic phospholipase A2 phosphorylation in FcεRI-mediated RBL-2H3 mast cells. Conclusions : These results indicate that Ulmus exhibits anti-allergic activity through inhibition of degranulation and inflammatory mediators and cytokine release. These findings suggest that Ulmus may have potential as a prophylactic and therapeutic agent for the treatment of various allergic diseases.
Objectives : This study was carried out to investigate the effects of Phellinus igniarius Quel extract on the Anti-inflammatory, Anti allergy, Anti-oxidant and Anti-wrinkle. Methods : To investigate in vitro anti-oxidant activity assay, ethanol extracts of medicinal plants tested by DPPH method. In the next experiment, to investigate anti-inflammatory test, the RAW 264.7 macrophage cells was cultured using DMEM including the 10% FBS. To study anti-allergic effect, we blended cultured Human Mast Cells(HMC-1), and then observe TNF-$\alpha$, IL-8 by ELISA Results : Phellinus igniarius Quel extract has the effects of anti-inflammation and anti-allergy, which may be due to its inhibitory potential on the macrophage activation. Furthermore, Phellinus igniarius Quel extract has the anti-wrinkle effects through the inhibitory potential on the collagnease, elastase and gelatinase activities. Conclusions : The above results suggest that Phellinus igniarius Quel extract could be applicable for improvement of several skin functions.
Generally, berry fruits have various pharmacological activities such as anti-inflammation, anti-oxidation and anti-cancer effects. The effects of gooseberry, a berry fruits, on atopic dermatitis (AD) have not been widely examined. The aim of this present study is to investigate whether gooseberry modulates AD. We examined the pharmacological effects of gooseberry on 2, 4-dinitrochlorobenzene (DNCB)-induced AD symptoms in mice. To determine the anti-atopic mechanism of gooseberry, we investigated its effects on the production of inflammatory cytokines and activation of nuclear factor-${\kappa}B$ in PMA + ionophore -stimulated human mast cells (HMC-1). The results demonstrated that gooseberry attenuated AD clinical symptoms such as erythema, edema and dryness as well as histamine and IgE serum levels in DNCB-induced AD model mice. Additionally, gooseberry suppressed the expression of inflammatory cytokines and activation of nuclear factor-${\kappa}B$ in stimulated HMC-1. These findings demonstrate that gooseberry is potential agent for treating AD and allergic inflammation.
Objective : Allergic Inflammation is related with secretion of Cytokine. This study was performed to examine the effects of Woobangja on anti-allergic inflammation. Method : While macrophage 264.7cells was chosen as a normal group a control group was classified into three groups. One was stimulated with LPS. and another was pretreated with Woobangja for 1 hour. The third was pretreated with gydrocortisone for 1 hour. After the pretreatment, macrophage were incubated with lipopolysaccharide(LPS) 100 ng/ml for 12h and media collected and $TNF-{\alpha}$, IL-6, $IL-1{\beta}$, IL-10 concentration in supernatants were measured each by Enzyme linked immuno-sorbent assay. Woobangja were used $50\;{\mu}g/ml$, $100\;{\mu}g/ml$, $250\;{\mu}g/ml$, $500\;{\mu}g/ml$, 1 mg/ml. Hydrocortisones were used respectively $10^{-8}\;M$,$10^{-7}\;M$,$10^{-6}\;M$,$10^{-5}\;M$,$10^{-4}\;M$. Results : Woobangja showed inhibitory effect on $TNF-{\alpha}$ by LPS-stimulated macrophage 264.7. The inhibitory effect was most significant in 1mg/ml(p<0.01), and has increased according to the number of doses. Woobangja also showed inhibitory effect on IL-10 by LPS-stimulated macrophasg 264.7. The inhibitory effect was most significant in $100\;{\mu}g/ml$, and was not in a dose-dependent manner as Hydrocortisone group. Woobangja and Hydrocortison showed contrary effect on $IL-1{\beta}$ in al five concentration(p<0.01), and at the lowest concentration ($50\;{\mu}g/ml$) the level of $IL-1{\beta}$ was the lowest. On the other hand hydrocortison was observed to have inhibitory effect on $IL-1{\beta}$ in all five concentration(p<0.01). IL-6 was inhibited by hydrocortison in a roughly dose-dependent manner, but was not inhibited by Woobangja. On the contrary Woobangja obviously increased the expression of $IL-1{\beta}$ in all five concentration(p<0.01), but it was not related with concentrations. Conclusion : 1. Woobangja does significantly inhibit the expression of $TNF-{\alpha}$ by LPS-stimulated macrophage 264.7. 2. Woobangja does significantly increse the expression of IL-6 by LPS-stimulated macrophage 264.7. 3. Woobangja does significantly increse the expression of $IL-1{\beta}$ by LPS-stimulated macrophage 264.7. 4. Woobangja does significantly inhibit the expression of IL-10 by LPS-stimulated macrophage 264.7. 5. Woobangja is observer to have anti-allergic inflammatory effect through inhibiting inflammatory cytokine.
유기농 및 일반재배 대두를 다양한 용매에서 추출하여 그 추출물에 대한 항산화 효과, 항염증 효과, 대식세포의 세포사멸 보호 효과, 그리고 알레르기 저해효과를 비교하였다. DPPH 법과 SOD 효소의 활성 측정으로 항산화 능력을 평가하였으며, 항염증 효과는 LPS로 유도시킨 Raw 264.7 세포주에서 염증 매개 단백질인 Cox-2와 iNOS의 발현량을 western blotting으로 조사하여 분석하였다. 또한 대식세포 보호 효과를 세포사멸 조절 단백질인 Bcl-xl의 발현량 조사로 평가하였고, 1 mM $H_2O_2$ 처리로 유도되는 세포사멸의 보호효과를 세포 수 측정법으로 조사하였다. 항알레르기 효과는 흰쥐의 비만세포인 RBL-2H3 세포주를 사용하여 IgE로 매개되는 비만세포의 탈과립화를 OPT assay로 측정하여 분석하였다. 그 결과, 일반 재배보다 유기 재배 대두 추출물이 더 좋은 항산화, 세포사멸 보호, 항염증 효과를 나타내었다. 특히 유기농 대두의 열수 추출물은 LPS로 유도시킨 염증반응을 효과적으로 억제하는 것으로 나타났다. 그러나 항알레르기 효과는 모든 추출물에서 히스타민 유리량이 약간 감소하는 것으로 나타나 유기농과 일반재배의 차이를 확인할 수 없었다. 대두에는 많은 유용한 생리활성물질 뿐 만 아니라 알레르기 유발 물질도 존재하는 것으로 알려져 있지만 조추출물 상태에서는 특이적인 알레르기 유발보다 오히려 항알레르기 효과가 나타났다. 결론적으로, 만성 염증 및 천식, 그리고 아토피피부염과 같은 염증관련 질환에는 유기재배된 대두가 일반 대두보다 좋은 억제 효과를 가지고 있다고 판단된다.
The aim of this study is to evaluate the anti-inflammatory effect of Hataedock treatment using Coptidis Rhizome and Glycyrrhiza Uralensis (CG) mixed extract in allergic rhinitis induced NC/Nga mice. We divided NC/Nga mice into 3 groups as follows; allergic rhinitis-induced group after CG Hataedock treatment (CGT, n=10), no treatment group (Ctrl), allergic rhinitis elicited group (ARE). To induce allergic rhinitis, NC/Nga mice of 3 weeks age were sensitized on 7, 8 and 9week by Ovalbumin (OVA) antigen in intranasal space. Hataedock using CG extract was administered on week 3 in allergic rhinitis-induced group (CGT) after Hataedock treatment. To identify distribution of Interlukin (IL)-4, Cluster of differentiation 40 (CD40), high-affinity IgE receptor ($Fc{\varepsilon}RI$), substance P, Matrix metallopeptidase 9 (MMP-9), Nuclear $factor-{\kappa}B$ ($NF-{\kappa}B$) p65, Inducible nitric oxide synthase (iNOS) and Cycloxygenase-2 (COX-2), we used histological examination. CGT significantly inhibited IL-4 and CD40 response compared with ARE. The reduction of Th2 cytokine expression decreased inflammatory mediators such as $Fc{\varepsilon}RI$, substance P, MMP-9, $NF-{\kappa}B$ p65, iNOS and COX-2. Such immunological improvement induced reduction of respiratory epithelial damage and mucin secretion in goblet cell. These results indicate that Hataedock treatment suppresses allergic rhinitis through modulating of Th2 responses and diminishing various inflammatory mediators in nasal mucosal tissue. It might have potential applications for prevention and treatment of allergic rhinitis.
Objective : Allergic disease, a very common chronic illness that affects all ages of patients, has been well characterized as an IgE-dependent immunologic response. Recently, interest has grown about the late inflammatory reaction as well as the early one characterized by IgE and mast cells in allergic disease. The purpose of this study was to find the anti-inflammatory effect of Taeeumjowui-tang (Taiyintiaowei-tang) in allergic reaction. Methods : The experiment was performed using Raw 264.7 cells pretreated with Taeeumjowui-tang contents extracts. The results were measured for different concentrations of Taeeumjowui-tang extracts (100, 200, $300{\mu}g$/ml): the toxicity and proliferation of cells by MTT analysis, LPS-induced NO production using Griess reagent and IL-6/ TNF-${\alpha}$ production, which is a significant criteria for diagnosing allergic reaction. Results : No toxicity of Taeeumjowui-tang (100, 200, $300{\mu}g$/ml) on Raw 264.7 cells was found after 24 hours incubation. LPS-induced NO production was reduced after treatment with Taeeumjowui-tang (100, 200, $300{\mu}g$/ml)(P<0.001). IL-6 decreased only at $100{\mu}g$/ml(P<0.05). TNF-${\alpha}$ production decreased only at $300{\mu}g$/ml, but still statistically insignificant. There was no relationship between any components of Taeeumjowui-tang alone and inhibition of NO production. Conclusions : These data suggest that Taeeumjowui-tang has anti-inflammatory effects in allergic reaction.
Purpose : This study aimed to investigate the anti-allergic effect of Persicaria perfoliata water extract (PPWE) on IgE stimulated rat basophilic leukemia (RBL-2H3) cell line. Methods : P. perfoliata (L.) H. Gross has been used in traditional medicine as an anti-allergic agent, antipyretic, and diuretic and for respiratory disorders. To analyze the anti-allergic activity of PPWE, release of β-hexosaminidase in RBL-2H3 cells was estimated by enzyme linked immunosorbant assay (ELISA). Also, the cytotoxic effect of PPWE was identified by WST assay, and nuclear factor (NF)-κB and its upstream signaling molecules were assessed by western blot analysis. Results : PPWE treatment significantly attenuated β-hexosaminidase release in a dose dependent manner without any cytotoxicity. PPWE inhibited β-hexosaminidase activity by 38.4±1.2, 36.6±0.6, 32.5±2.2 and 26.5±1.2 at 500, 250, 100, and 50 ㎍/㎖ of PPWE, respectively, compared with the control group. In addition, an analysis of the expression level of NF-κB, an inflammation transcription factor, in RBL-2H3 cells upon IgE stimulation provided reults consistent with the results of β-hexosaminidase release. The phosphorylated status of upstream signaling molecules for transcription factor, mitogen activated protein kinases (MAPKs), was also analyzed. The results showed that PPWE treatment dose-dependently inhibited phosphorylation of extracellular regulatory kinase (ERK) and c-Jun N-terminal kinase (JNK). These results show that PPWE had a strong IgE-mediated degranulation inhibitory effect on RBL-2H3 cells. Conclusion : P. perfoliata ameliorated IgE-mediated allergic reaction via the modulation of MAPK and NF-κB signaling pathway in RBL-2H3 cells. These results indicate that P. perfoliata could be a potential candidate for a treatment strategy against various allergic disorders.
$CheongGiGeoYangTang$ has been used for anti-allergenic purpose. However there was no experimental study about its effect. Therefore, this study was designed to investigate the anti-allergenic effect of $CheongGiGeoYangTang$. Methods Modifiability of RBL-2H3 mast cells' IL-4, IL-13 was analyzed by qRT-PCR and ELISA. Also, the suppressive effect of GATA-1, GATA-2, NF-AT1, NF-AT2, AP-1 and NF-${\kappa}B$ p65 transcription factors was observed by western blotting. OVA-specific IgE, IL-4 and IL-13 production in ovalbumin allergy model was examined as well. Results It was showed that the RBL-2H3 mast cells treated with $CheongGiGeoYangTang$ extract(CGGYT) was significantly suppressed mRNA expression, production of IL-4 and IL-13, and prominently inhibited the expression of transcription factors including GATA-1, GATA-2, NFAT-1, NFAT-2, c-Fos and NF-${\kappa}B$ p65 but not c-Jun. The administration of CGGYT was suppressed the amount of OVA-specific IgE, IL-4 and IL-13 in OVA/alum-sensitized mice. Conclusions We considered CGGYT would regulate the allergic inflammation as inhibition of IL-4 and IL-13 production in activated mast cells and Th2 cells.
