  • Nitrate-selective ion exchange resin which have bulky tertiary amine as functional group have been synthesized by the reaction of chloromethylated polystyrene-divinylbenzene copolymer and the corresponding tertiary amine [$NR_3=NE_{t3} 1, N{(C_2 H_4 H_3)}_32]$in ethanol, while commercial resin has $NMe_3$ as functional group. The fundamental properties such as bulk density, water content, appearance index, exchange capacity, effective size, uniformity coefficient of synthesized anion exchange resin (1) have been measured. The ion exchange resin (1) and (2) exhibited the better selectivity for nitrate than sulfate in both batch and continuous column experiments.

  • Ion exchange is the most reliable process to remove the ionic impurities and the economic operation. ion exchange is widely used in water and wastewater treatment, especially softening and demineralization. ion selectivity depends on the hydrated radius, charge of ions and concentration. The objective of this study was to determine the selectivity order of cations with equilibrium and column ion exchanges and to investigate the effect of the background anion on selectivity. Cation selectivity increases with decreasing concentration and increasing charge ( $H^+$ < $K^+$ << $Cu^{2+}$ < $Co^{2+}$ < TEX>$Ca^{2+}$ << $Ce^{3+}$)in equilibrium and column cation adsorptions.

  • Crude cytoplasmic fraction of phototrophic purple sulfur bacterium, Thiocapsa roseopersicina NCIB 8347, were initially prepared and purified by sonication, ultracentrifugation, ammonium sulfate fractionation and heat-treatment and it has been previously reported. Using various applications of chromatography far the purification of membrane-bound and soluble hydrogenases from heat-treated enzyme fraction were studied at present report. When the heat-treated enzyme preparation was applied to the anion column chromatography using Q-sepharose, Fraction I and II, which were extracted with the KCl 0-0.5 M gradient, showed the specific evolution hydrogenase activity 3.86 and 2.27 U/mg-protein respectively. Specific hydrogenase activitys of Fraction I and II were further increased to 4.35 and 7.46 U/mg-protein for Fraction I and to 2.49 and 4.41 U/mg-protein fur Fraction II respectively, when hydrophobic interaction column, Phenyl superose, and anion exchange column, Mono-Q, were applied. Size exclusion chromatography using superdex 200 concentrated the hydrogenase Fraction I and II to 9.19 and 7.84 U/mg-protein respectively at the final step of purification.

Investigation of Adsorption Mechanism and Selective Adsorption of Carbonyl Compounds of Mainstream Tobacco Smoke by Ion Exchangers (이온교환체에 의한 담배 주류연 중 카보닐 화합물의 선택적 흡착 특성 및 흡착 메카니즘 구명)

  • This work has been conducted to select appropriate filter materials for removing carbonyl compounds in mainstream tobacco smoke. To investigate of the usability of this filter materials, two types of bead ion exchangers were synthesized and their adsorption characteristics for carbonyl compounds were investigated. Sulfonic acid group-containing cation exchanger and ammonium group-containing anion exchanger were synthesized by the suspension polymerization of glycidylmethacrylate(GMA) and divinylbenzene(DVB) followed by the subsequent functionalization, respectively. The removal efficiency of carbonyl compounds by these two ion exchangers increased in the presence of moisture. However, the amount of carbonyl compounds adsorbed on the anion exchanger was larger than that on the cation exchanger under two levels of water contents tested. This phenomenon seems to arise from the electron delocalization in carbonyl group of the anion exchangers. There was not any significant relationship between the amount of carbonyl compounds adsorbed on ion exchangers and the length of adsorption column. From the large ion exchange capacity and rapid ion exchange reaction rate of the anion exchanger, it is suggested that the anion exchanger may be a good filter material for removing carbonyl compounds in the mainstream tobacco smoke.

  • Destructive methods are used for the turnup determination of an irradiated PWR fuel. One of the methods includes U, Pu, Nd-148 and Nd-(145+146) determination by an isotope dilution mass spectrometry using triple spikes (U-233, Pu-242 and Nd-150). The method involves two sequential ion exchange resin separation procedures. Pu is eluted from the first anion exchange resin column (Dowex AG 1$\times$8) with 12 M HCl-0.1 M HI mixed solution, followed by U elution with 0.1 M HCl. Nd is isolated from other fission products on the second anion exchange resin column (Dowex AG 1$\times$4) with a nitric acid-methanol eluent. Each fraction is analysed by thermal ionization mass spectrometry. The difference between Nd-148 and Nd-(145+146) method is found with an average 2.07%. The results are compared with those by the heavy element method using U and Pu isotopes and by the destructive y-spectrometric measurement of Cs-137. The dependences of isotope composition of U and Pu on burn-up, and correlation between those isotopes are illustrated graphically.

  • This article provides comprehensive guidance on the maintenance, cleaning, regeneration, and storage of silica-based HPLC (High-Performance Liquid Chromatography) columns. The general considerations emphasize the importance of using in-line filters and guard cartridges to protect columns from blockage and irreversible sample adsorption. While these measures help, contamination by strongly adsorbed sample components can still occur over time, leading to an increase in back pressure, loss of efficiency, and other issues. To maximize column lifetime, especially with UHPLC (Ultra-High Performance Liquid Chromatography) columns, it is advisable to use ultra-pure solvents, freshly prepared aqueous mobile phases, and to filter all samples, standards, and mobile phases. Additionally, an in-line filter system and sample clean-up on dirty samples are recommended. However, in cases of irreversible compound adsorption or column voiding, regeneration may not be possible. The document also provides specific recommendations for column cleaning procedures, including the flushing procedures for various types of columns such as reversed phase, unbonded silica, bonded normal phase, anion exchange, cation exchange, and size exclusion columns for proteins.   • The methods of separation and recovery of rare earth elements in monazite sand have been studied by the ion exchange chromatography. Both of cation and anion exchange resin were used as ion exchange resins and the solutions of EDTA, DTPA, IMDA and Ln-EDTA were used as eluents. The H+, Zn2+, Fe3+, Al3+, Cu2+, and NH4+ forms of cation exchange resin were used as retaining ions. Ln-EDTA solution was loaded on the EDTA form of anion exchange resin and separated. The Ln-EDTA solution was also used as an eluent for a selective separation of one element from the rare earth mixture solution. The size effects of resin column, the elution mechanism for the various elution types and the separation of a large amount of rare earths were studied.

  • The purity of CeO$_2$, Sm$_2$O$^{3+}$ and La2O$^{3+}$ have been determinated by separating the impurity of rare earth-elements in those oxides with anion exchange resin and measuring the absorbance of the separated element with MTB. 1 ml of the mixture solution containing each 1 mg/ml of Y$^{3+}$, Yb$^{3+}$, Eu$^{3+}$, Sm$^{3+}$, Nd$^{3+}$, Pr$^{3+}$, Ce$^{4+}$and La$^{3+}$ is loaded in top layer of anion exchange resin column, Amberlite CG400 and separated with the mixture solution of the various concentration of methyl alcohol and nitric acid as an eluent. Using the result, the impure rare earth elements in pure CeO$_2$, Sm$_2$O$^{3+}$ and La$_2$O$^{3+}$ are determinated.

  • The steroid 9${alpha}$-hydroxylase activity has been detected in cytosol fraction, $100,00{\times}g$ supernatant of cell free extract of Mycobacterium fortuitum. The activity was not linear with protein concentration in the assay suggesting 9${alpha}$-hydroxylase is a multicomponent enzyme. The 9${alpha}$-hydroxylase system was partially purified through fractional saturation of ammonium sulfate, strong anion exchange (Mono Q) column chromatography, gel filtration (Superose 12) column chromatography, and testosterone affinity gel chromatography. Ammonium sulfate 50~60% saturated fraction of the cytosol gave 9${alpha}$-hydroxylase activity. For further purification, the half-saturated ammonium sulfate fraction was applied to Mono Q, Superose 12, or affinity gel column. The purification factors of 9${alpha}$-hydroxylase containing fraction after Mono Q, Superose 12, and affinity gel chromatography was 13, 11, and 17 respectively.
