Effects of Structural Difference of Ionic Liquids on the Catalysis of Horseradish Peroxidase

  • The dependence of the catalytic properties of horseradish peroxidase on the structural changes of ionic liquids was investigated with two water-miscible ionic liquids, N-butyl-3methypyridinium tetraftuoroborate ([$BMP_y$][$BF_4$]) and 1-butyl-3-methylimidazolium methylsulfate ([BMIM][$MeSO_4$]), each of which shares an anion ($BF_4^-$) or a cation ($BMIM^+$) with 1-butyl-3-methylimidazolium tetraftuoroborate ([BMIM][$BF_4$]), respectively. The oxidation of guaiacol (2-methoxyphenol) with $H_2O_2$was used as a model reaction. In order to minimize the effect of solution viscosity on the kinetic constants of the enzymatic catalysis, the enzymatic reactions for the kinetic study were performed in water-ionic liquid mixtures containing 25% (v/v) ionic liquid at maximum. Similarly to the previously reported results for [BMIM][$BF_4$], as the concentration of [$BMP_y$][$BF_4$] increased, the $K_m$value increased with a decrease in the $k_{cat}$value: the $K_m$value increased markedly from 2.8 mM in 100% water to 12.6 mM in 25% (v/v) ionic liquid, indicating that ionic liquid significantly weakens the binding affinity of guaiacol to the enzyme. On the contrary, [BMIM][$MeSO_4$] decreased the Km value to 1.4 mM in 25% (v/v) ionic liquid. [BMIM][$MeSO_4$] also decreased $k_{cat}$more than 3-folds [from 13.8 $s^{-1}$in 100% water to 4.1 $s^{-1}$in 25% (v/v) ionic liquid]. These results indicate that the ionic liquids interact with the enzyme at the molecular level as well as at a macroscopic thermodynamic scale. Specifically, the anionic component of the ionic liquids influenced the catalysis of horseradish peroxidase in different ways.

  • In this study, we synthesized bead-type GMA-DVB copolymer using glycidylmethacrylate (GMA) with high reactivity and hydrophilicity. Macrorecticular anion exchanger containing the trimethylammonium group were then prepared by amination with trimethylammonium chloride. We observed that the size of $NO_3^-$ is smaller than that of $SO_4^{2-}$ which disturb $NO_3^-$ removal in most of coexistent anions in ground water. Thus we investigated selective affinity for $NO_3^-$ and properties of individual ion exchangers with various DVB content. For each resins, we confirmed formation of copolymer by FT-IR spectrometer and investigated ion exchange capacity, swelling ratio, the amination yield and the effect with degree of crosslinking on adsorbability for nitrate. When amount of DVB is 4 wt%, amination yield, ion exchange capacity and swelling ratio was 384.3%, 3.25 meq/g and 77.1%, respectively. In these result, it can found that synthetic optimal condition is 4 wt% DVB content for monomer.

  • In this study, sulfonated poly(ether ether ketone) (SPEEK) as cation exchange membrane and blended and crosslinked poly(vinyl amine) (PVAm) with poly(vinyl alcohol) (PVA) membrane as anion exchange membrane were used and then the performance experiments of the membrane capacitive deionization (MCDI) installed with both membranes were carried out. The newly prepared anion exchange membrane were characterized through water content, ion exchange capacity and FT-IR. The crosslinking time of 3 h to 5 h indicated that the salt removal was reduced from 81.3, 65.7% to 53.8%. The effect of PVAm contents from 40, 60, to 80% on the salt removal was shown 81.3, 75.2 and 37.7%, respectively. As a result, it was concluded that the crosslinking time and the content of PVAm had an influence on the salt removal efficiency.

Antioxidant potential of aerial part of Asclepias curassavica. Linn (Family-Asclepiadaceae)

  • Asclepias curassavica. Linn, an erect, simple (or) much branched perennial herb with a somewhat woody base, belonging to the family Asclepiadaceae. It has been reported to have multiple pharmacological effect of anti-inflammatory, cardiotonic, anticancer, anthelmintic and to treat piles and gonorrhoea. It is to be expected that several activities might be related to a possible antioxidant action from this plant. The hydro alcoholic extract of Asclepias curassavica was tested in vitro for its antioxidant activities, such as DPPH radical, nitric oxide radical, superoxide anion radical, lipid peroxidation assay, hydroxyl radical, reducing power, and total phenol content. The extract exhibited scavenging potential with $IC_{50}$ value of $8.7\;{\mu}g/ml,\;198.4\;{\mu}g/ml\;and\;21.7\;{\mu}g/ml$ for DPPH, nitric oxide and superoxide anion radicals. The values were found to higher than those of Vitamin-C, rutin, and curcumin, as standards. The extract showed 50% protection at the dose of $134.2\;{\mu}g/ml\;and\;41.4\;{\mu}g/ml$ in lipid peroxidation as well as deoxyribose degradation, those values more to that of standard, vitamin E $(IC50\;values,\;119.2\;{\mu}g/ml\;and\;32.5\;{\mu}g/ml,\;respectively)$. The reducing power of the extract depends on the concentration and amount of extract. Since a significant amount of polyphenol could be detected by the equivalent to $0.0495\;{\mu}g$ of pyrocatechol from 1 mg of extract. It can be concluded that hydro alcoholic extract of aerial part of Asclepias curassavica could be considered as potent antioxidant, which makes it suitable for the prevention of human disease.

  • The solubility of carbon dioxide in ionic liquids and amine solvents has been investigated in gas-liquid absorption equilibrium reactor. Absorption capacity and kinetics of $CO_2$ with $CO_2$ pressure and absorption time in 9 different ionic liquids and 2 kinds of amine solvents were evaluated. In order to understand the effect of ionic type, we changed the cation or anion of ionic liquids. $CO_2$ absorption capacity and absorption rate of amine solvents were higher than those of ionic liquids. $CO_2$ absorption capacity of [emim][$Tf_2N$], $0.14molCO_2/mol\;IL$ at 1 bar, was the highest among the ionic liquids. $CO_2$ absorption capacity of ionic liquid steeply decreased with increasing temperature. Anion of ionic liquid dominates interaction with $CO_2$ and cation plays secondary role.

  • The rate constants for the nucleophilic addition reactions of thioglycolic acid to vinylsulfilimine(VSI) derivatives(p-OCH3, H, p-Cl and p-Br) were determined by an ultraviolet spectrophotometric method, and rate equations which can be applied over a wide pH range were obtained. On the basis of rate equation, general base catalysis and substituent effect, a plausible addition reaction mechanism was proposed: Below pH 3.0, the reaction was proceeded via the addition of neutral molecule to carbon-carbon double bond after protonation at the nitrogen atom of the sulfilimine, and in the pH range of 3.0 to 9.0, the neutral molecule and its anion attacked to carbon-carbon double bond competitively. Above pH 9.0, sulfide anion added to the double bond (Michael type addition).

Effect of Dietary Cation-Anion Difference during Prepartum and Postpartum Periods on Performance, Blood and Urine Minerals Status of Holstein Dairy Cow

  • Twenty four periparturient cows were used to determine the effects of DCAD on acid-base balance, plasma and urine mineral concentrations, health status, and subsequent lactation performance. Each group of 12 cows received either a diet containing -100 DCAD or +100 DCAD for 21 d prepartum. Both anionic and cationic groups were divided into two groups, one received a +200 DCAD and the other +400 DCAD diet for 60 d postpartum. Prepartum reduction of DCAD decreased DMI, urinary and blood pH, urinary concentrations of Na or K and increased plasma and urinary Ca, Mg, Cl and S. Also cows fed -100 DCAD diet consumed the most dry matter in the first 60 d after calving. Postpartum +400 DCAD increased milk fat and total solid percentages, urinary and blood pH and urinary Na and K concentrations, but urinary Ca, P, Cl and S contents decreased. Greater DMI, FCM yields were observed in cows fed a diet of +400 DCAD than +200 DCAD. No case of milk fever occurred for any diets but feeding with a negative DCAD diet reduced placenta expulsion time. In conclusion, feeding negative DCAD in late gestation period and high DCAD in early lactation improves performance and productivity of dairy cows.

Evaluation of Antioxidant Activity of Solvent Fractions of Angelica gigas Root Using TOSC Assay (TOSC 방법을 이용한 참당귀 뿌리 용매분획(Angelica gigas)의 항산화 활성 평가)

  • Objectives: The aim of this study was to elucidate the antioxidant effect of solvent fractions of Angelica gigas root. Methods: The ethanol extract of Angelica gigas root was suspended in water and then partitioned with dichloromethane (MC Fr.), ethyl acetate (EA Fr.) and butanol (BuOH Fr.), sequentially. The antioxidant activities of solvent fractions of Angelica gigas root were evaluated for radical scavenging activity against stable free radicals (2,2-diphenyl-1-picrylhydrazyl) DPPH, hydrogen peroxide and superoxide anion radicals. In addition the antioxidant activities of solvent fractions of Angelica gigas root against peroxyl radicals, hydroxyl radicals and peroxynitrites were determined by the total oxy-radical scavenging capacity (TOSC) assay. Results: Among the solvent fractions of MC Fr., EA Fr., and BuOH Fr., BuOH Fr. was found to have stronger antioxidant activity with $IC_{50}$ values of 59.72, 14.36, 30.96 and $44.75\;{\mu}g/m{\ell}$ on the DPPH radical, nitrite, superoxide anion and hydrogen peroxide, respectively, than BHA used as a positive control. Moreover, specific TOSC values(564.8, 276.4 and 405.5 TOSC/mM) of BuOH fr. against peroxyl radicals, hydroxyl radical and peroxynitrite were 4 times higher than GSH (136.5, 67.4 102.6 TOSC/mM) used as a positive control. Conclusions: These results suggest that the BuOH fr. of Angelica gigas root has a high antioxidant activity and can be useful to develop functional food against oxidative stress conditions.

Antioxidative Effect of Rhus javanica Linne Extract Against Hydrogen Peroxide or Menadione Induced Oxidative Stress and DNA Damage in HepG2 Cells

  • The free radical scavenging activities and the protective effects of Rhus javanica extracts against oxidative damage induced by reactive oxygen species (ROS) were investigated. n-Hexane, ethyl acetate and water fractions were prepared from a methanol extract. DPPH radical, superoxide anion and hydroxyl radical scavenging activities were estimated. Intracellular ROS formation was quantified using fluorescent probes, 2', 7'-dichlorofluorescin diacetate (DCFH-DA) for hydroxyl radical and dihydroethidium (DHE) for superoxide anion. The oxidative DNA damage was investigated by the comet assay in HepG$_2$ cells exposed either to $H_2O$$_2$ or to menadione. The highest $IC_{50}$/ values for DPPH radical scavenging activity was found in the ethyl acetate fraction with a value of 5.38 $\mu\textrm{g}$/mL. Cells pretreated with $\geq$ 1 $\mu\textrm{g}$/mL of the ethyl acetate extract had significantly increased cell viability compared to control cells, which were not pretreated with the extract. Intracellular ROS formation and DNA damage in HepG$_2$ cells, which were pretreated with the various concentrations of Rhus javanica ethyl acetate extract and then incubated either with $H_2O$$_2$ or with menadione, reduced in a dose-dependent manner. These findings suggest that Rhus javanica might have biologically active components which have strong protective effects against ROS induced oxidative damages to the biomolecules, such as cell membranes and DNA.

A Study on the Anti-Oxidation Effects of Ulmi Cortex and Hemipteleae Cortex (유피(楡皮)와 자유피(刺楡皮)의 항산화(抗酸化) 효능에 대한 연구)

  • Objectives : This studies were carried out to evaluate the effects on anti-oxidation activities in different parts as trunk bark and root bark of Ulmus devidiana Planchon. var. japonica Nakai and Hemipteleae davidii (Hance) Planchon. Methods : For evaluation of antioxidative effects, scavenging activity on superoxide anion radical and DPPH radical were measured. Also, inhibitory activity on LDL oxidation and linoleic acid peroxidation were measured in each samples. In the in vivo test, inhibitory activity on TBARS production, GSH contents in rat liver were measured. SOD, Catalase, GSH-px and ALDH activity were analysed in ethanol extracts of Ulmi Radicis Cortex. Result : 1. Scavenging activity on superoxide anion radical was higher in water extract than in ethanol extract even in low concentration of 50ppm as over 90%. 2. There was no difference between water extract and ethanol extract in the scavenging activity on DPPH radical but, Ulmi Raclicis Cortex and Ulmi Trunci Cortex showed high effect even in low concentration of 10ppm. 3. GSH reduction was prevented and antioxidative activity such as Mn-SOD, GSH-px in the rat liver recovered in the treatment of ethanol extracts of Ulmi Radicis Cortex. Conclusion : Ulmi Radicis Cortex recorded as Ulmi Trunci Cortex in official regulation book. But, it was known that Ulmi Radicis Cortex was more effective than Ulmi Trunci Cortex in most physical activities.
