랫드의 혈림프절 구조 (Structure of hemolymph node in rats)

  • It has been known that the hemolymph node is one of the lymphoid organs found normally in ruminants and some rodents, and this organ shares morphological and functional characteristics of lymph node and spleen. To clarify the rigorous definition of morphological structures and functions of the hemolymph node in Sprague-Dawley(SD) rats, we examined these organs of SD rats gross anatomically and light microscopically. The hemolymph nodes were normally found in the abdominal cavity and in the neck of SD rats. This organ was surrounded by a thin connective tissue capsule and there was a hilus. The parenchyma comprised a cortex of lymphatic nodules and diffuse lymphatic tissues, and a medulla of diffuse lymphatic tissues arranged in cords. Afferent and efferent lymph vessels were observed but there was no extensive subcapsular and medullary sinuses. These sinuses were filled with erythrocytes. The stroma of hemolymph nodes was composed of reticular cells and fibers, and many lymphocytes, granulocytes, erythrocytes, plasma cells, macrophages and megakaryocytes were supported by the reticular network. The above findings suggest that the hemolymph nodes of SD rats may take part in blood formation, blood filtration and immune reaction.

Adverse effect of IL-6 on the in vitro maturation of porcine oocytes

  • Cytokines are protein mediators that possess the ability to assist cell-to-cell communication in immune system-related activities. In general, pathogen endotoxins activate the release of inflammatory mediators, and with time, there is an increase in the cytokine levels in the body. Interleukin (IL)-6 mediates the acute-phase inflammatory response, and elevated IL-6 levels have been reported in peritoneal fluids of women with pelvic inflammation and endometriosis, thereby associating it with oocyte quality and infertility. To overcome subfertility or infertility in humans and animals, the present study was done to examine the effect of recombinant IL-6 on porcine oocytes matured in vitro and subsequently to determine the fertilization rate and embryo development. Porcine oocytes were incubated with varying concentrations of IL-6 (0 - 2 ㎍·mL-1) for 44 h followed by in vitro fertilization and culturing of the oocytes. The oocytes or embryos were fixed with 3.7% paraformaldehyde (PFA) and stained with fluorescence dyes, and the meiotic spindle, chromosome organization, fertilization status and embryo development were subsequently assessed under a fluorescence microscope. We observed induction of an abnormal meiotic spindle alignment in the oocytes incubated with IL-6 compared to the control oocytes incubated without IL-6. Moreover, significantly decreased fertilization rates and embryo development were observed for oocytes incubated with IL-6 (p < 0.05). Thus, an increased IL-6 level during oocyte maturation could be associated with fertilization failure due to an aberrant chromosomal alignment and a disruption of the cortical granules. Taken together, our results indicate that successful assisted reproduction can be achieved by controlling the levels of inflammatory cytokines.

Full-Length Enriched cDNA Library Construction from Tissues Related to Energy Metabolism in Pigs

  • Genome sequencing of the pig is being accelerated because of its importance as an evolutionary and biomedical model animal as well as a major livestock animal. However, information on expressed porcine genes is insufficient to allow annotation and use of the genomic information. A series of expressed sequence tags of 5' ends of five full-length enriched cDNA libraries (SUSFLECKs) were functionally characterized. SUSFLECKs were constructed from porcine abdominal fat, induced fat cells, loin muscle, liver, and pituitary gland, and were composed of non-normalized and normalized libraries. A total of 55,658 ESTs that were sequenced once from the 5′ ends of clones were produced and assembled into 17,684 unique sequences with 7,736 contigs and 9,948 singletons. In Gene Ontology analysis, two significant biological process leaf nodes were found: gluconeogenesis and translation elongation. In functional domain analysis based on the Pfam database, the beta transducin repeat domain of WD40 protein was the most frequently occurring domain. Twelve genes, including SLC25A6, EEF1G, EEF1A1, COX1, ACTA1, SLA, and ANXA2, were significantly more abundant in fat tissues than in loin muscle, liver, and pituitary gland in the SUSFLECKs. These characteristics of SUSFLECKs determined by EST analysis can provide important insight to discover the functional pathways in gene networks and to expand our understanding of energy metabolism in the pig.

Lmbr1 Expression in Early Embryo Development Stages in White Leghorn and Chinese Silky

  • Lmbr1 is regarded as a key gene that controls the digital model formation in early developmental stages of the chicken. However, there are few reports of lmbr1 expression levels and tendencies in 4-toe and 5-toe chicken species. Therefore, the objective of this study was to compare the lmbr1 expression in White Leghorn (4-toe) and Chinese Silky (5-toe). Firstly, total RNA was extracted from 14 different embryonic development stages (HH3 to HH31) in White Leghorn and Chinese Silky. Secondly, dramatic gene expression changes of lmbr1 were monitored by RT-PCR, which indicated a general up-down-up tendency with subtle differences between these two species. Moreover, Q-PCR reactions were performed to quantitate the expression level of lmbr1 in the 14 selected developmental stages. These data demonstrated a first lmbr1 expression peak of 18.68 and 15.32, a lmbr1 expression trough of 6.61 and 1.80, and a second lmbr1 expression peak of 22.33 and 12.48 in White Leghorn and Chinese Silky, respectively. Finally, embryonic in situ hybridization analysis identified that lmbr1 expressed in the ectoderm in HH21, HH23 and HH24 developmental stages in both species.

Glucose Kinetics for Milk Synthesis in Etawah Crossbred Goats Fed King Grass Silage Prepared with Manure

  • A study was carried out to determine glucose kinetics, nutrient balance and milk production of lactating Etawah crossbred goats. The animals (27.2 to 29.1 kg BW) were randomly divided into four levels of dietary treatment groups: the first group R1 received 100% (3 kg) fresh king grass (Penisetum purpuroides), the second group R2 received 75% king grass and 25% king grass silage prepared with chicken manure, the third group R3 received 50% king grass and 50% silage, and the fourth group R4 received 100% silage. In addition to the roughage, each group received 800 g of concentrate (CP 14.77% of DM; 17.26 MJ/kg). Animals fed king grass silage made with chicken manure were found to be superior to the group fed king grass alone. Glucose kinetics and retained energy were significantly affected. Calculations showed that glucose requirements for maintenance and milk production can be met for the groups with high levels of silage (R3 and R4). The values of glucose flux were in the range of 2.52 to 4.50 mg/min.kg $BW^{0.807}$ which are lower, but close to, the values for the temperate lactating dairy cow. The present glucose flux value for the lactating Etawah crossbred goat is higher than the previous value published from this laboratory.

  • A large number of investigations have shown that changes in nutritional condition affect endocrine status in avian species. Herein, recent findings including novel peptides discovered by the development of the techniques in the field of molecular biology have been reviewed. The insulin-like growth factors (IGF-I and IGF-II) found in chickens have been characterized and shown to be 70 and 66 amino acid polypeptides, respectively. Plasma IGF-I level is very responsive to nutrition, Le. varying dietary proteins and energy intakes, and food restriction. Plasma IGF-II concentration is altered by nutritional deprivation to a much smaller extent than plasma IGF-I concentration. Almost all of the serum and tissue IGFs are found in a complex composed of IGF and IGF-binding protein (IGFBP). In the chicken plasma, the major IGFBP differs from that in mammalian plasma. The proglucagon mRNA encodes glucagon and two glucagon-like peptides (GLP-I and GLP-2). The intracerebroventricular administration of GLP-l strongly decreased food intake of chicks, and it was indicated that the inhibition of food intake by GLP-l was associated with neuropeptide Y, which is one of the neurotransmitters reported to enhance food intake.

Rapid Nondestructive Prediction of Multiple Quality Attributes for Different Commercial Meat Cut Types Using Optical System

  • There are differences of spectral characteristics between different types of meat cut, which means the model established using only one type of meat cut for meat quality prediction is not suitable for other meat cut types. A novel portable visible and near-infrared (Vis/NIR) optical system was used to simultaneously predict multiple quality indicators for different commercial meat cut types (silverside, back strap, oyster, fillet, thick flank, and tenderloin) from Small-tailed Han sheep. The correlation coefficients of the calibration set (Rc) and prediction set (Rp) of the optimal prediction models were 0.82 and 0.81 for pH, 0.88 and 0.84 for L*, 0.83 and 0.78 for a*, 0.83 and 0.82 for b*, 0.94 and 0.86 for cooking loss, 0.90 and 0.88 for shear force, 0.84 and 0.83 for protein, 0.93 and 0.83 for fat, 0.92 and 0.87 for moisture contents, respectively. This study demonstrates that Vis/NIR spectroscopy is a promising tool to achieve the predictions of multiple quality parameters for different commercial meat cut types.

Characterizations of the bovine subtype Interferon-tau Genes : Sequences of Genes and Biological Activity of Transcription Factors in JEG3 Cell

  • Multiple interferon tau (IFNT) genes exist in bovine. An antiluteolytic substance secreted by the bovine conceptus and primarily responsible for maternal recognition of pregnancy is bovine trophoblast protein 1 (bIFNT1), a new type I interferon tau (IFNT) genes. The objectives of this research were to investigate whether multiple, distinct gene encode bIFNT1 and other type I bIFNT gene in the bovine genome and to examine expression of bIFNT1 and other bIFNTc1 mRNAs during conceptus development. These transcrips could be regulated through caudal-related homeobox-2 (CDX2) and ETS2 and/or AP1 (JUN) expression, a transcription factor implicated in the control of cell differentiation in the trophectoderm. The presence of mRNAs encoded by bIFNT1 and type I bIFNTc1 genes were examined quantitatively via reverse transcription-polymerase chain reaction (RT-PCR) analysis of total cellular RNA (tcRNA) extracted from on day 17, 20 and 22 bovine conceptuses. The expression level of bIFNT1 was higher on day 17 transcripts were gradually weakly detectable on day 20 and 22. However, the other bIFNTc1 gene examined transcripts was highly expressed on day 20 and transcripts were weakly detectable on day 17 and 22 bovine conceptuses. Furthermore, human choriocarcinoma JEG3 was co-transfected with an -1kb-bIFNT1/c1-Luc constructs and several transcription factor expression plasmids. Compared to each -1kb-bIFNT1/c1-Luc increased when this constructs were co-transfected with, ETS2, AP1(JUN), CREBBP and/or CDX2. Also, bIFNTc1 gene was had very effect on activity by alone ETS2, and AP1 (JUN) expression factors in choriocarcinoma JEG3 cell. However, bIFNT1 gene expression of the upstream region was not identified. We demonstrated that the activities of bIFN genes are regulated by differential, tissue-specific and developmental competence during pregnancy.

Nutritive Evaluation of Some Browse Tree Legume Foliages Native to Semi-arid Areas in Western Tanzania

  • Browse tree legume leaves from Acacia spp (A. nilotica, A. tortilis, A. polyacantha), Dichrostachys sp, Flagea villosa, Piliostigma thonningii, Harrisonia sp were evaluated for nutritive potential (chemical compositions and degradability characteristics) compared to Gliricidia sepium. Effect of tannins anti-nutritive activity on digestibility was also assessed by polyethylene glycol (PEG) tannin bioassay. Crude protein (CP), ash, neutral detergent fiber (NDF), acid detergent fiber (ADF) and acid detergent lignin (ADL) differed (p<0.05) between legume foliages. Mean CP, ash, NDF, ADF and ADL for fodder species tested were 158, 92, 385, 145, and 100 g/kg DM, respectively. CP ranged from 115 (P. thonningii) to 205 g/kg DM (G. sepium). Acacia spp had moderate CP values (g/kg DM) of 144 (A. nilotica), to high CP in A. tortilis (188) and A. polyacantha (194) comparable to G. sepium. The forages had relatively lower fiber compositions. A. nilotica had (p<0.05) lowest NDF, ADF and ADL (182, 68 and 44) compared to P. thonningii (619, 196 and 130) g/kg DM, respectively. Except G. sepium, all fodder species had detectable high phenolic and tannin contents greater than 5% DM, an upper beneficial level in animal feeding and nutrition. Mean total phenolics (TP), total tannins (TT) and condensed tannins (CT) (or proanthocyanidins) for fodder species tested were 139, 113 and 43 mg/g DM, respectively. F. villosa had (p<0.05) lowest TP and TT of 65 and 56 mg/g DM, respectively, compared to A. nilotica (237 and 236 mg/g DM, respectively). The CT varied (p<0.05) from 6 (F. villosa) to 74 mg/g DM (Dichrostachys sp). In vitro organic matter (OM) degradability (OMD) differed (p<0.05) between fodder species. G. sepium had (p<0.05) high degradability potential compared to A. polyacantha that had (p<0.05) the lowest OMD values. Forage degradability ranked: G. sepium>A. nilotica>P. thonningi>F. villosa>Dichrostachys sp>A. tortilis>A. polyacantha. Addition of PEG resulted to (p<0.05) improvement in in vitro OM digestibility (IVD). Increase in IVD was mainly due to binding action of PEG on tannins; and represents potential nutritive values previously depressed by tannins anti-nutritive activity. Browse fodder has potential as sources of ruminal nitrogen especially for ruminants consuming low quality roughages due to high protein, lower fiber compositions and high potential digestibility. However, utilization of browse supplements in ruminants is hampered by high phenolic and tannin contents. Deactivation of tannin anti-nutritive activity, possibly by feeding tanniniferous browse with other readily available nitrogen sources to dilute tannin anti-nutritive activity could improve utilization of browse fodder supplements. Further studies are needed to assess browse fodder palatability and intake, and their effect on growth performance in ruminants.

Historical Introduction of Japanese Wild Mice, Mus musculus, from South China and the Korean Peninsula

  • In Japan, the wild house mouse Mus musculus consists of two lineages, one from Southeast Asia (Mus musculus castaneus; CAS) and one from northern Eurasia (Mus musculus musculus; MUS). However, the exact origins of the parental lineages are unclear. A recent work using mitochondrial sequences revealed that Japanese CAS and MUS are closely related to haplotypes from South China and the Korean Peninsula, respectively. Recent phylogeographic analyses using nuclear gene sequences have also confirmed a close relationship between Japan and Korea in the MUS component. However, the Japanese CAS components in the nuclear genome are likely to be unique and to differ from those of other CAS territories, including South China. Although the origins are still unresolved, these results allow us to conclude that two areas of the continent, South China and the Korean Peninsula, are the primary source areas of Japanese wild mice and suggest pre-historical introductions associated with certain historical agricultural developments in East Asia.
