고등어 유래 항고혈압 peptide의 분리 정제 (Separation and Purification of Angiotensin I-converting Enzyme Inhibitory peptide from Mackerel)

  • 고등어 유래의 peptide를 제조하고, Sephadex G-25 column, ODS AQ column, Vydac column, Superdex peptide column을 이용하여 순차적으로 분리, 정제하였으며 그 결과를 요약하면 다음과 같다. 고등어 선어로부터 두부, 내장 및 뼈를 제거하여 얻은 고등어육의 수율은 $65.5{\%}$였으며, 고등어육의 단백질 함량은 $63.3{\%}$, 그리고 지질함량은 $32.6{\%}$로 나타났다. 지질을 제거하기 위하여 알칼리 처리한 후 동결 건조한 고등어 단백질의 수율은 고등어 육에 대하여 $18.2{\%}$로 나타났고, 고등어 단백질을 효소가수분해하여 제조한 고등어 가수분해물의 수율은 $50.6{\%}$로 나타났다. 고등어 가수분해물 cut-off 1만의 membrane으로 ultrafiltration한 결과, MW 10,000이하가 $79.6{\%}$, 10,000 이상이 $20.4{\%}$였으며, ACE저해효과는 10,000이하 획분이 10,000이상 획분보다 높은 것으로 나타났다. (Table 1). 1만 이하의 획분을 Sephadex G-25 column으로 분획하여, 7개의 획분을 얻었고 (Fig. 1), 이 가운데 MS2획분의 ACE 저해효과가 가장 높았다(Table 2). ACE 저해효과가 가장 좋은 MS2획분을 ODS AQ column으로 분획하석 4개의 획분으로 분획하였고 (Fig.2), 이 가운데 Ms2O3획분의 ACE저해효과가 가장 좋았다 (Table 3). Ms2O3획분을 Vydac column으로 재분획하여 8개의 획분으로 분취하였다(Fig.3), 이 가운데 MS2O3V5획분의 ACE저해효과가 가장 좋았다(Table 4). Superdex peptide column으로 재정제하여 얻은 peptide의 N-말단으로부터 아미노산 배열은 Tyr-Val-Ala으로 ACE저해활성은 IC-(50)이 $1.4{\mu}M$로 나타났다. Matsumura등 (1993)은 가다랭이 가수분해물로부터 분리한 6개의 peptides 가운데 tripeptide의 ACE저해활성이 높았다고 하였으며, 본 연구 결과에서도 유사한 경향을 나타내었다.

Physiological Functionalities of Vitis hybrid (Sheridan)-Rubus coreanus Red Wine Made by Saccharomyces cerevisiae

  • Vitis hybrid (Sheridan)-Robus coreanus red wine was vinified by fermentation of a mixture of Vitis hybrid and Robus coreanus must at $25^{\circ}C$ for 10 days. The Vitis hybrid-Robus coreanus red wine had ethanol contents of 10.9%. It had high antihypertensive angiotensin I-converting enzyme (ACE) inhibitory activity of 57.8% and antioxidant activity of 64.8%. Changes in the physicochemical properties and functionality of the Vitis hybrid-Robus coreanus red wine was investigated during a post-fermentation period of three months. The ACE inhibitory activity of the red wine increased as the post-fermentation period prolonged, and showed the highest ACE inhibitory activity of 70.4% 60 days post-fermentation. However, the antioxidant activity declined significantly to 47.2% during the post-fermentation period of 60 days. In terms of sensory evaluation, the Vitis hybrid-Robus coreanus red wine had the best acceptability 60 days post-fermentation.

Partial Purification of Mussel Adhesive Protein for Mytilus Edulis and Preparation of Mussel Protein Hydrolysates

  • Mussel adhesive protein (MAP) was extracted from Korean Mytilus edulis and then partially purified using Sephacryl S-300 gel permeation chromatography and reversed-phase high performance liquid chromatography. As an indicator of adhesiveness, is 3,4-dihydroxyphenylalanine (DOPA) content was determined. Its DOPA/protein ratio of 0.19 was higher than those of other reports, indicating a good adhesive. The partially purified MAP was confirmed by acid-urea polyacrylamide gel electrophoresis using cetylpiridinium bromide as a cationic detergent. Sea mussel hydrolysates were prepared using three commercial proteases to provide value-added functional materials and their angiotensin converting enzyme (ACE) inhibitory activities were determined. Among hydrolysates of sea mussel, Protamex was the best and further purification would improved ACE inhibitory activity.

저장기간 중 돌산갓피클에서 Glucosinolates의 ACE 및 α-glucosidase 저해활성 (ACE and α-glucosidase Inhibitory Activity of the Glucosinolates in Dolsan Leaf Mustard Pickle during Storage)

  • This study was carried out for investigate that physiological activity, quantification and qualitative were sinigrin of Dolsan leaf mustard pickle (DLMP) during storage. sinigrin contains high amounts of nutritional and medicinal compounds, which are important for maintaining optimum health. ACE inhibitory activity was ranged between 43.2 and 79.4%. DLMP methanol extracts demonstrated highest ACE inhibitory activity at 79.4% on day 14, whereas DLMP ethanol extracts demonstrated highest Angiotensin I-converting Enzyme (ACE) inhibitory activity of 43.2% at day 0. The ${\alpha}-glucosidase$ inhibitory activity of positive control 0.02% (v/v) acarbose was 78%. The DLMP methanol extracts had the highest ${\alpha}-glucosidase$ inhibitory activity at 64.0% on day 14, whereas DLMP ethanol extracts had the lowest ${\alpha}-glucosidase$ inhibitory activity of 42.8% at day 28. Sinigrin was high in DLMP methanol extracts at $49.55{\mu}g/ml$ on day 14 of storage. Sinigrin standard was eluted at 2.73 min and MS analysis was m/z 283.03 along with fragment ions at m/z 204 and 149.06. These data show that sinigrin formed desulfo-glucosinolates $[M-SO_3-2H_2O+K+2H]^+$. Sinigrin concentration increased until day 14 and then decreased after that. DLMP methanol extracts had consistently higher sinigrin concentration than DLMP acetonitrile extracts during 28 days of storage.

닭 쓸개 methanol 추출물의 생리기능적 특성에 관한 연구 (Studies on Physiological and Functional Properties of Methanol Extract from Chicken Bile)

  • 닭 쓸개의 생리기능적 특성을 연구하기 위하여 TLC 및 HPLC를 이용하여 쓸개내 성분을 정성 및 정량 분석을 하였고, 항균활성, 전자공여능, 아질산염 소거능 및 ACE 저해능을 비교 검토하였다. TLC 및 HPLC에 의해서 분석한 결과 닭 쓸개의 담즙산 중에는 taurochenodeoxycholic acid와 taurocholic acid로 추정되는 성분이 검출되었으며, 이들의 함량은 taurochenodeoxycholic acid 가 5,893mg/100g, taurocholic acid는 385mg/100g이었다. 이들 추출물은 Bacillus cereus에 대하여 강한 항균활성을 보였으며 Salmonella typhimurium, Staphylococcus acreus 및 Vibrio parahaemolyticus에 대해서도 항균 활성을 나타내었다. 그러나 Escherichia coli에 대해서는 항균활성을 보이지 않았다. 쓸개의 냉동 건조물 및 methanol 추출물의 전자공여능은 시료의 농도가 증가할수록 유의적으로 증가하였다. 또한 pH 1,2에서 측정한 아질산염 소거작용에 있어서도 농도가 높을수록 유의적으로 증가하였으며, methanol 추출물이 냉동 건조물 보다 높게 나타났다. Angiotensin I-converting enzyme(ACE) 저해 작용에서도 methanol 추출물과 냉동건조물 모두에서 나타났으며, 첨가량을 증가시켰을 때 methanol 추출물의 작용이 냉동건조물의 경우보다 상승폭이 큰 것으로 확인되었다. 이상의 결과로부터 닭 쓸개는 항균활성, 전자공여능 및 아질산염 소거능 그리고 ACE 저해능 등의 다양한 생리기능적 특성을 지니고 있어 기능성 식품 등의 소재로서의 활용을 기대할 수 있을 것으로 사료된다.

Isoflavone-Free 검은콩펩타이드의 항고혈압 및 ACE 활성 억제효과 (Antihypertensive and ACE Inhibitory Effects of Novel Isoflavone-free Black Soy Peptide Mixture)

  • Antiobesity, hypotriglyceridemic and antihypertensive activities of isoflavone-free peptide mixture (black soybean peptide, BSP) were reported in our previous experiments. In the present study, angiotensin converting enzyme inhibitory (ACEi) activity was decreased in the aorta tissues of spontaneously hypertensive rats (SHRs) treated with BSP (1% in drink water) for 4 weeks, but not in serum. BSP administration significantly decreased ACE activity by 17.5% (from $33.2{\pm}4.5$ to $27.4{\pm}1.96$ mUnit/mg, p=0.0013) in aorta tissue hydrolysate. BSP treatment also decreased significantly mean blood pressure (BP) (from $213.0{\pm}16.96$ to $184.0{\pm}6.53$ mmHg, p<0.0001) as expected. These results indicate that BSP has antihypertensive activity as well as ACEi activity.

소나무 새순 에탄올 추출물의 angiotensin converting enzyme, xanthine oxidase 및 nitrix oxide synthase 활성 (Inhibitory Effects of Ethanol Extracts from Pine Buds (Pinus densiflora) on Angiotensin Converting Enzyme, Xanthine Oxidase and Nitric Oxide Synthesis)

  • 본 연구에서는 솔순의 기능성에 관한 연구를 위하여 에탄올 추출물로 여러 가지 생리활성과 아질산염 소거작용에 대하여 분석하였다. 우선, 솔순의 항산화 활성을 측정하기 위하여 DPPH, SOD 유사활성, XO 저해활성을 측정하였다. 그 결과 DPPH법을 통해 측정한 솔순의 항산화력은 1 mg/ml에서 88.9%의 radical 소거능을 나타내었으며, 동량의 BHA와 비교했을 때 유사한 항산화력이 측정되었다. 이것은 솔잎보다 항산화 활성이 높은 것으로 솔순의 항산화력에 관한 높은 이용가치를 의미한다. SOD 유사활성은 10 mg/ml 농도에서 47.9%로 비교적 높은 SOD 유사활성을 나타내었다. Xanthine oxidase 저해활성 측정 실험에서는 1 mg/ml에서 71.9%의 높은 xanthine oxidase 저해 활성을 나타내었다. Tyrosinase의 저해 효과는 시료의 농도가 증가함에 따라 유의적으로 증가하는 경향을 나타내었으나 전체적인 활성은 4 mg/ml의 솔순 에탄올 추출물에서 15.2%로 나타났다. 솔순 추출물에 대한 항균력을 조사하기 위해 식중독균주인 S. aureus, E. coli, V. paraheamolyticus에 대한 성장저해 효과를 측정하였다. 그 결과 E. coli와 V. paraheamolyticus에 대해 높은 항균력을 나타냈다. 항고혈압 측정실험에서는 시판되는 항고혈압제와 동일한 농도($1{\mu}g/ml$)에서 captopril은 93.0%, 솔순은 50.6%의 저해률을 나타냈으며, $100{\mu}g/ml$에서는 60.8%의 저해률을 나타내어 ACE 저해 활성이 뛰어난 것으로 나타났다. 또한 LPS에 의하여 유도된 NO 합성은 $200{\mu}g/ml$ 농도의 솔순 에탄올 추출물을 처리함으로서 NO 합성률이 54.0% 정도 감소하였다. 이러한 결과는 솔순 에탄올 추출물이 면역기능과 밀접한 관계가 있음을 나타내고 있다. 이상의 결과로 보아 솔순 에탄올 추출물의 우수한 생리활성을 증명하고 있고, 또한 솔잎 추출물에 비해 항산화력, 자유레디칼 소거활성능, 미백효과, 면역활성 및 항고혈압 효과가 높은 것으로 나타나 기능성식품의 소재로서 그 활용도가 높을 것으로 판단된다.

2단 가수분해에 의한 굴 가수분해물의 기능성 개선 (Improving the Functional Properties of Oyster Hydrolysates by Two-step Enzymatic Hydrolysis)

  • This study prepared functional oyster hydrolysates using two-step enzymatic hydrolysis and investigated their functional properties. To prepare two-step enzymatic hydrolysates (TSEH), oysters were hydrolyzed using 1% Protamex (PR) at $40^{\circ}C$ and pH 6.0 for 1 hr before sequential treatment with one of the following enzymes for 1 hr: Alcalase (AL), Flavourzyme (FL), Neutrase (NE), pepsin (PE), and trypsin (TR). The PRAL, PRNE and PRTR hydrolysates had significantly greater angiotensin I converting enzyme (ACE) inhibitory activity than did PR and the other TSEHs. Only the antioxidant activity of the PRNE hydrolysate was significantly different (p<0.05), while none of the TSEHs had antimicrobial activity. The oyster hydrolysate prepared by sequential treatment with Protamex and Neutrase (PRNE) had the best ACE inhibitory activity and antioxidant activity, with $IC_{50}$ values of 0.40 and 0.94 mg/mL, respectively. The PRNE hydrolysate was processed through an ultrafiltration (UF) series with molecular weight cut-off (MWCO) membranes of 3, 5, 10, and 30 kDa, and the ACE inhibitory, antioxidant, and antimicrobial activities of the permeates were determined. The permeate through the 3-kDa MWCO membrane had greater ACE inhibitory activity and antioxidant activity than did the other PRNE permeates, with $IC_{50}$ values of 0.11 and 0.40 mg/mL, respectively.

해조류로부터 항고혈압성분의 분리정제 (Separation and Purification of Antihypertensive Substances from Edible Seaweeds)

  • To isolate natural antihypertensive substances from edible seaweeds, we screened for and separated active compounds contained in natural Underia pinnatifida, cultural Underia pinnatifida, Laminaria japonica, Sporophylls and Agarum cribrosum. They were extracted using room temperature water, boiling water, acetone, and methanol in turn or using room temperature water, ether, acetone, methanol and boiling water in order. The in vitro antihypertensive activity was quantified as inhibitory efficacy against angiotensin-I converting enzyme (ACE), which is a factor inducing hypertension. For all of the seaweeds tested, the fractions soluble in room temperature water and in boiling water showed the strongest ACE inhibitory effect among the extracted fractions. Conversely, the methanol-extracted fractions for all of the seaweeds tested showed no antihypertensive activity. While the ether and acetone fractions had slight antihypertensive effects. The compounds in the aqueous extracts that had antihypertensive activity were presumed to be polysaccharides, such as fucoidan and alginate.

프로바이오틱 유산균 발효조건 탐색을 위한 다반응 최적화의 활용 (Applying Multi-Response Optimization to Explore Fermentation Conditions of Probiotics)

  • This review serves two purposes: first, to promote the use of improved optimization techniques in response surface methodology (RSM); and second, to enhance the optimum conditions for the fermentation of probiotics. According to research in dairy science, Lactiplantibacillus plantarum K79 is a candidate probiotic that has beneficial health effects, such as lowering blood pressure. The optimum conditions for L. plantarumK79 to produce peptides with angiotensin-converting enzyme (ACE) inhibitory activity were proposed, through modeling each of ACE inhibitory activity and pH as a function of the four factors that are skim milk concentration (%), incubation temperature (℃), incubation time (hours), and starter added amount (%). To estimate optimum conditions, the researchers employed a desirability-based multi-response optimization approach, utilizing third-order models with a nonsignificant lack of fit. The estimated optimum fermentation conditions for L. plantarum K79 were as follows: a skim milk concentration of 10.76%, an incubation temperature of 36.9℃, an incubation time of 23.76 hours, and a starter added amount of 0.098%. Under these conditions, the predicted ACE inhibitory activity was 91.047%, and the predicted pH was 4.6. These predicted values achieved the objectives of the multi-response optimization in this study.
