  • The solubilities of caffeine in supercritical ammonia were measured at various temperatures and pressures to represent the relationship between the solubility and the density of ammonia at desired temperature and pressure by means of a simple equation. Using the equation, the interaction virial coefficient between ammonia and caffeine has been determined to give the parameters of Lennard-Jones potential function for the system. Furthermore, there are comparisons and discussions of the interaction virial coefficients and Lennard-Jones potential functions determined by the same techniques from the solubility data of naphthalene in supercritical ammonia and of caffeine and naphthalene in supercritical carbon dioxide from existing data sources others.

  • The quantitative analysis of ethambutol.2HCl as well as commercial ehambutol preparations was undertaken by gas-liquid chromatography by finding optimum conditions, such as the use of internal standard, stability of an ethambutol-2HCl-caffeine standard solution, and the effect of column temperature, N, O-bis-(trimethylsily) acetamide [B.S.A.] concentrations and other substances present in the preparations. Under the chromatiographic conditions, an ethambutol-2HCl. caffeine standard, 9 min, 30sec. The relative molar response of ethambutol.2HCl and caffeine studied was 2.08. Ethambutol.2HCl could be quantitated up to 1$\times$10$^{-8}$ moles. the possible decomposition of B.S.A. due to the moisture when tested and the incoplete reaction for silylation could be minimized.

  • 완충액(緩衝液)에서의 caffeine, niacinamide, pyridoxine hydrochloride, thiamine hydrochloride의 여과조제(濾過助劑)에 의(依)한 흡착(吸着)현상을 비교검토(比較檢討)하였다. 실험(實驗)에 사용(使用)한 vitamin 류(類)의 흡착(吸着)은 Langmvir 흡착식(吸着式)의 pattern 에 따랐으며 caffeine 은 거의 흡착(吸着)되지 않았다. 여과조제중(濾過助劑中) Dawsonite는 Celite 545 보다 큰 흡착(吸着)현상을 나타냈으며 pH변화(變化)에 따른 영향(影響)은 niacinamide가 가장 예민하였고 pH의 증가(增加)는 흡착량(吸着量)을 현저히 감소시켰다.

Effect of Glucose on Fertilization In Vitro of Bovine Follicular Oocytes by Frozen-thawed Spermatozoa (우란포란의 체외수정시 Glucose가 정자침입에 미치는 영향)

  • Frozen semen obtained from 5 different Korean Native Cattle were used for in vitro fertilization of oocytes. The proportions(3∼11%) of oocytes penetrated in the basic medium with 13.9 mM glucose were very low in different semen. However, the penetration rates of oocytes in the presence of caffeine and heparin increased to 68∼84%. The proportions(10∼52%) of oocytes penetrated in the presence of caffeine alone were low at various glucose concentrations(0∼27.8 mM). In the medium with heparin, significantly(P<0.05 at least) higher penetration rate was obtained in the absence(94%) than in the presence(28∼45%) of 0∼27.8 mM glucose concentrations. The medium with caffeine and heparin, however, produced similar penetration rates(84∼97%) regardless of the various glucose concentrations.

  • This experiment was undertaken to investigate the effect of caffeine and vitamin E on the lipid composition of serum and liver in cholesterol-fed rats. Thirty-five male rats of Sprague-Dawley strain weighing about $120{\pm}10\;g$ were divided into 5 groups, each group receiving a different diet for 6 weeks. i.e. basal diet (Basal group), basal diet supplemented with 1.0% cholesterol (Basal+Chol. group), basal diet supplemented with 1.0% cholesterol and 0.25% caffeine (CF group), basal diet supplemented with 1.0% cholesterol and 400ppm vitamin E (VE group), and basal diet supplemented with 1.0% cholesterol and caffeine plus vitamin E (CF+VE group). Net weight gain was significantly depressed only by caffeine supplementation. Serum total lipid was significantly decreased in VE and CF+VE groups. Serum total cholesterol and HDL-cholesterol were slightly increased by caffeine supplementation, whereas triglyceride (TG) was significantly decreased in CF, VE and CF+VE group. Contents of crude lipid and TG in liver were significantly reduced by supplementation of vitamin E. No differences in liver cholesterol were found among experimental groups. Histologically, Basal+Chol. group showed a spotty necrosis, including a profuse fine microvaculoar lipid accumnlation. CF group exhibited a mild inflammatory infiltration, as well as macro- and microvacuolar lipid accumulation. But those were regenerated by supplementation of vitamin E.

Effect of Cryodiluents, Cryoprotectants, Pre-freezing Method and Total Time Required for Freezing on Post-thaw Viability of Boar Spermatozoa (돼지정자의 동결융해 후 활력 및 생존성에 대한 보존액, 동해보호제, 예비동결 및 동결처리시간의 영향)

  • Boar semen can be frozen successfully. However, there is a large variability in the extent of damage boar semen samples experiences during cryopreservation. This experiment was undertaken to find out factors that affect a post-thaw viability of boar spermatozoa. For this purpose, cryodiluents(BF5, LEY, Soejima and M-Soejima), cryoprotectants(glycerol. ethylene glycol, and propylene glycol), pre-freezing method(dryice-pellet, dryice-straw and L$N_2$vapour-st-raw) and total time required for freezing(2. 5, and 7 h) were compared as a factors. To investigate quality of semen during freezing process, motility(%), normal apical ridges(%, NAR), and proportion of living sperm(%) by flow cytometic analysis were assessed after collection, cooled, pre-frozen and post-thawing. Post-thaw motility of semen diluted with M-Soejima was 52.0%, respectively. When heparin, caffeine or heparin+caffeine was added to 2nd cryodiluent of M-Soejima during freezing process, the highest motility after thawing was shown at the addition of caffeine (2mM), with 61.7$\pm$2.9% of motility. M-Soejima with heparin or caffeine was significantly higher than that of controI(p<0.05). The result using glycerol(Gly), ethylene glycol(EG), propylene glycol(PG), and their mixture (Gly+EG and Gly+PG) as cryoprotectants, the highest motility was shown at the mixture treatment with Gly plus PG. However, the highest proportion of live spermatozoa was shown at Gly+EG, there was no significantly difference among treatments(p>0.05). When semen was pre-frozen with three manners(dryice-pellet, dryice-straw, and L$N_2$ vapor-straw), motility(%) of post-thaw spermatozoa was the highest in the L$N_2$ vapor-straw pre-freezing method of M-Soejima cryodiluent with 57.5% of motility, For a simple, economical and timesaving approach to freezing boar semen, total time required for freezing were 2, 5, and 7 hours, post-thaw motility were 43.8, 45.0 and 38.8%, NAR were 19.5, 22.7 and 28.5%, and viability were 20.8, 19.9 and 22.1%, respectively. This data suggests that boar semen diluted with M-Soejima cryodiluent contained caffeine, using mixture of glycerol and propylene glycol or ethylene glycol as cryoprotectants, frozen with 2 hours, can be taken better motility, NAR, and proportion of live spermatozoa.

  • The technique of SCNT is now well established but still remains inefficient. The in vitro development of SCNT embryos is dependent upon numerous factors including the recipient cytoplast and karyoplast. Above all, the metaphase of the second meiotic division (MII) oocytes have typically become the recipient of choice. Generally high level of MPF present in MII oocytes induces the transferred nucleus to enter mitotic division precociously and causes NEBD and PCC, which may be the critical role for nuclear reprogramming. In the present study we investigated the in vitro development and pregnancy of White-Hanwoo SCNT embryos treated with caffeine (a protein kinase phosphatase inhibitor). As results, the treatment of 10 mM caffeine for 6 h significantly increased MPF activity in bovine oocytes but does not affect the developmental competence to the blastocyst stage in bovine SCNT embryos. However, a significant increase in the mean cell number of blastocysts and the frequency of pregnant on 150 days of White-Hanwoo SCNT embryos produced using caffeine treated cytoplasts was observed. These results indicated that the recipient cytoplast treated with caffeine for a short period prior to reconstruction of SCNT embryos is able to increase the frequency of pregnancy in cow.

Removal Characteristics of Tetracycline, Oxytetracycline, Trimethoprime and Caffeine in Biological Activated Carbon Process (생물활성탄 공정에서 Tetracycline, Oxytetracycline, Trimethoprime 및 Caffeine 제거특성)

  • In this study, The effects of three different activated carbon materials (each coal, coconut and wood based activated carbons), empty bed contact time (EBCT) and water temperature on the removal of pharmaceutical 4 species (oxytetracycline, tetracycline, trimethoprime and caffeine) in BAC filters were investigated. Experiments were conducted at three water temperature (5, 15 and $25^{\circ}C$) and four EBCTs (5, 10, 15 and 20 min). The results indicated that coal based BAC retained more attached bacterial biomass on the surface of the activated carbon than the other BAC, increasing EBCT or increasing water temperature increased the pharmaceutical 4 species removal in BAC columns. In the coal-based BAC columns, removal efficiencies of oxytetracycline and tetracycline were 87~100% and removal efficiencies of trimethoprime and caffeine were 72~99% for EBCT 5~20 min at $25^{\circ}C$. The kinetic analysis suggested a firstorder reaction model for pharmaceutical 4 species removal at various water temperatures (5~$25^{\circ}C$). The pseudo-first-order reaction rate constants and half-lives were also calculated for pharmaceutical 4 species removal at 5~$25^{\circ}C$. The reaction rate and half-lives of pharmaceutical 4 species ranging from 0.0360~0.3954 $min^{-1}$ and 1.75 to 19.25 min various water temperatures and EBCTs, could be used to assist water utilities in designing and operating BAC filters.

A Study of the Characteristics of Different Coffee Beans by Roasting and Extracting Condition (종류별 커피의 볶음 및 추출조건에 따른 품질 특성에 관한 연구)

  • Caffeine is one of the most well known stimulants which can potentially increase mental performance, release fatigue and decrease depression. Green beans from different soils and climates contain different levels of caffeine, and as well as extracted coffee with different roasting and extracting methods. An investigation looking at pH, acidity, extractable solid and caffeine contents was assessed according to roasting and extracting conditions of various coffee beans. Brazilian coffee beans did not show much variation in pH with respect to roasting and extracting temperature, however, acidity increased in low roasting and extracting temperatures. This was however most prominently observed in Ethiopian and Indonesian coffee beans. The large expansion of coffee bean cells renders them highly porous to the passage of water, consequently extracted solids were found to increase with increasing temperature. This was especially apparent in Columbian coffee which had the highest extracted solids. The amount of caffeine extracted from coffee beans also increased with the higher temperature extraction. The Indonesian and Vietnam robusta coffee varieties showed the highest caffeine content.

Catechins, Theaflavins and Methylxanthins Contents of Commercial Teas (시판 차류의 Catechins, Theaflavins 및 Methylxanthins 함량에 관한 연구)

  • CThis study used HPLC to analyze the contents of 7 kinds of catechins, 4 kinds of theaflavins, and 2 kinds of methylxanthines in the following 6 kinds of commercial Korean tea: 2 green, 2 black, 1 jasmine and loolong. The following ranges in the 13 tea components of the 6 samples by ethanol extract were evaluated in mg/g: (-)-epigallocatechin, 0(black tea and jasmine tea) to 14.19(green tea); (-)-catechin 0; (+)-epicatechin, 0.62(bran rice-green tea) to 2.91(black tea); (-)-epigallocatechin gallate, 4.59(black tea) to 43.96(jasmine tea); (-)-gallocatechin gallate, 0.58(black tea) to 5.80(jasmine tea); (-)-epicatechin gallate, 5.63(bran rice-ueen tea) to 48.06(jasmine tea): (-)-catechin gallate, 0.26(black tea): theaflavif 0 to 3.66(black tea): theaflavin-3-gallate, 0 to 6.94(black tea): theaflavin-3'-gallate, 0 to 4.01(black tea); theaflavin-3,3-digallte, 0 to 10.25(black tea); caffeine, 4.60(bran rice-peen tea) to 26.44(black tea); and theobromine, 0.10(bran rice-green tea) to 1.81(jasmine tea). The contents of all components were lower by water extract than by ethanol extract. Therefore, total catechin (100.55, 45.88 mg/g) and theobromine (1.81, 0.86 mg/g) contents in jasmine tea, and theaflavin content (24.88, 1.36 mg/g) in black tea by ethanol and water extract were the highest. Caffeine content was the highest in black tea(96.48 mg/g) for the ethanol extract, and in jasmine tea (12.38 mg/g) for the water extract.
