  • In most mammals, metaphase II (MII) oocytes having high maturation promoting factor (MPF) activity have been considered as good oocytes and then used for assisted reproductive technologies including somatic cell nuclear transfer (SCNT). Caffeine increases MPF activity in mammalian oocytes by inhibiting p34cdc2 phosphorylation. The objective of this study was to investigate the effects of caffeine treatment during in vitro maturation (IVM) on oocyte maturation and embryonic development after SCNT in pigs. To this end, morphologically good (MGCOCs) and poor oocytes (MPCOCs) based on the thickness of cumulus cell layer were untreated or treated with 2.5 mM caffeine during 22-42, 34-42, or 38-42 h of IVM according to the experimental design. Caffeine treatment for 20 h during 22-42 h of IVM significantly inhibited nuclear maturation compared to no treatment. Blastocyst formation of SCNT embryos was not influenced by the caffeine treatment during 38-42 h of IVM in MGCOCs (41.1-42.1%) but was significantly improved in MPCOCs compared to no treatment (43.4 vs. 30.1%, P<0.05). No significant effects of caffeine treatment was observed in embryo cleavage (78.7-88.0%) and mean cell number in blastocyst (38.7-43.5 cells). The MPF activity of MII oocytes in terms of p34cdc2 kinase activity was not influenced by the caffeine treatment in MGCOCs (160.4 vs. 194.3 pg/ml) but significantly increased in MPCOCs (133.9 vs. 204.8 pg/ml). Our results demonstrate that caffeine treatment during 38-42 h of IVM improves developmental competence of SCNT embryos derived from MPCOCs by influencing cytoplasmic maturation including increased MPF activity in IVM oocytes in pigs.

  • Catechol and caffeine were simultaneously analyzed with a bismuth-immobilized carbon nanotube paste electrode (BPE) using square wave (SW) stripping voltammetry. Optimum analytical conditions were determined. Simultaneous working ranges of 100-1,500 $mgL^{-1}$ for caffeine and 5-75 $mgL^{-1}$ for catechol were obtained. In the separated cell systems, a working range of 0.1-2.1 $mgL^{-1}$ catechol with a correlation coefficient of 0.9935, and a working range of 10-210 $mgL^{-1}$ caffeine with a correlation coefficient of 0.9921 were obtained. A detection limit (S/N) of 0.15 $mgL^{-1}$ (7.7 ${\times}$ $10^{-7}$ M) and a detection limit of 0.02 $mgL^{-1}$ (1.82 ${\times}$ $10^{-7}$ M), respectively, manifested for catechol and caffeine. It was found that three macro-type electrode systems could be implanted in fish and rat neuro cells. For both ions, the ion currents were observed. The physiological impulse conditions and the neuronal thinking current were also obtained.

  • The purpose of this study was to describe the effects of caffeine ingestion on the metabolic responses during submaximal exercise. Ten male of rowing player aged 18-22yrs. participated in the study. No subjects had any remarkable medical history and none were taking medications. According to the administration of dehydrated caffeine(CA) (6mg/kg) or placebo(PA), they were classified into two groups such as caffeine group and placebo group. A randomized, double-blind, crossover protocol was employed using either CA or PA. Subjects underwent a submaximal bicycle ergometer. Blood was drawn intravenously prior to 60 min., at rest, at 30, and 45 min. of exercise, and recovery period. Plasma concentrations of glucose, free fatty acid and lactate were obtained using enzymatic method. Followings were obtained by the tests and analyses; 1) Blood glucose(BG) of 45 minute of exercise was significantly decreased in CA group of 76.3$\pm$14.8 mg/100ml compared with PA group of 94.9$\pm$11.2 mg/100mg(p<0.5). 2) Free fatty acid(FFA) of 30minute of exercise was significantly increased in CA group of 720$\pm$80 $\mu$Eq/1 compared with PA group of 360$\pm$120 $\mu$Eq/1(p<0.5). After exercise, FFA was significantly higher in CA group than those in PA group(p<0.1). 3) Blood lactate(BL) was not significantly different between the two. After exercise, BL was significantly different in 30 minute(p<05).

  • The Ca uptake by the fragmented sarcoplasmic reticulum of the rabbit skeletal muscle was measured under various concentrations of K, Mg, Caffeine, procaine and quinine. The ATPase activity of this reticular membrane was measured under the same conditions simultaneously. The saturation of Ca uptake was almost completed within 1 minute. The Ca uptake was inhibited by high concentrations of K (above 50 mM) and Mg (above 1 mM)in the absence of ATP. When ATP is present, however, the Ca uptake did not reflect the concentration of K, while it increased greatly as the concentration of Mg was increased. Caffeine and procaine caused the inhibition of Ca uptake in the presence of ATP, but quinine did not. The ATPase activity of the membrane was little affected by the concentration of K, while it was enhanced in the presence of Mg. Caffeine, procaine and quinine did not influence the ATPase activity.

  • Background: Current evidence suggests that intravenous magnesium sulfate might be effective for reducing migraine pain. In a recent pilot study, we showed that intravenous caffeine citrate could reduce the severity of migraine headache. The objective of this study is to investigate the efficacy of intravenous caffeine citrate vs. magnesium sulfate for management of acute migraine headache. Methods: We conducted a prospective quasi-experimental study from January until May 2016 in two educational medical centers of Shahid Beheshti University of Medical Sciences (Shoahadaye Tajrish Hospital and Imam Hossein Hospital), Tehran, Iran. The study included patients who were referred to the emergency department and met the migraine diagnosis criteria of the International Headache Society. Patients were allocated into 2 groups receiving either 60 mg intravenous caffeine or 2 g intravenous magnesium sulfate. The pain scores, based on the visual analog scale, were recorded on admission, as well as one and two hours after receiving the drug. A Chi-Square test and student t-test were used for analysis of baseline characteristics. A Mann-Whitney U test and Wilcoxon singed rank test were used to analyze differences in the visual analogue scale (VAS) score between and within the groups respectively. Results: In total, 70 patients (35 patients in each group) with the mean age of $33.1{\pm}11.3years$ were included (64.3% female). For the Caffeine citrate group, the median pain score decreased from 9.0 (2.0) to 5.0 (4.0) after one hour and to 3.0 (4.0) after two hours. For the magnesium sulfate group, the pain score decreased from 8.0 (2.0) to 2.0 (2.0) after one hour and to 0.0 (1.0) after two hours. Both intravenous caffeine citrate and intravenous magnesium sulfate reduced pain scores significantly but the magnesium sulfate group showed more improvement than the Caffeine citrate group after one hour (P < 0.001) and after two hours (P < 0.001). Conclusions: It is likely that both intravenous caffeine and intravenous magnesium sulfate can reduce the severity of migraine headache. Moreover, intravenous magnesium sulfate at a dose of 2 g might be superior to intravenous caffeine citrate 60 mg for the short term management of migraine headache in emergency departments.

  • Caffeine is a xanthine alkaloid derivative found in many foods and beverages. Dietary caffeine may interact with commonly-consumed over-the-counter (OTC) drugs in body. In this study, cytotoxic effects on the intestinal cells by combined treatment of caffeine with several OTC drugs, including ibuprofen, aspirin, and acetaminophen. Cytotoxic effect of caffeine was more potent in normal intestinal INT 407 cells than in colon cancer HCT 116 cells. Relative toxicity of caffeine and the OTC drugs was significantly enhanced in INT 407 cells when treated together. Intracellular thiol levels of the cells treated with the OTC drugs increased in the presence of caffeine. When HCT 116 cells were incubated with each OTC drug after or before caffeine treatment, the relative cytotoxicity of the OTC drugs increased. The present study may provide basic information about possible health effects through the interactions between caffeine and OTC drugs in the intestinal cells.

  • The purpose of this study was to evaluate the effects of caffeine and calcium on the activities of the osteoblastic cell from mouse calvaria. The author cultured osteoblastic cells obtained from the mouse calvaria and were divided into three groups : the caffeine-treated, the calcium-treated and the combine-treated group. In caffeine-treated group, the cell toxicity was measured by MTT assay at 1, 2 and 4 days after treatment of caffeine. In all groups, the densities of the mineralized bone nodules were measured by imaging analyzer after Von Kossa staining. The alkaline phosphotase (ALP) activities were measured at 2, 7, 14, 21 and 28 days and the interleukin-1 ${\beta}$ activities at 48 hours after treatment of caffeine and calcium. The measurements were statistically executed with ANOVA test and the results were as follows. 1. The cellular toxicity of the caffeine increased with the concentration of caffeine during the incubation period. 2. The maximum densities of mineralization were observed at 0.2 mM caffeine-treated group, 1.2 mM calcium-treated group, 0.1 mM caffeine and 1.8 mM calcium-treated group. 3. The activities of ALP were peaked at 14 days at calcium-treated group as no-treated. But, the activities of ALP increased with concentrations of caffeine at caffeine-treated group. At combine-treated group, the act of ALP were peaked at 24 days at 1.2 mM, 1.8 mM calcium-treated group, But decreased at 2.5 mM calcium-treated group. 4. The activites of the IL-1 ${\beta}$ were increased significantly at 0.2 mM caffeine-treated group, 1.8 mM calcium-treated group and 0.1 mM caffeine and 1.8 mM calcium-treated group. But, they were decreased at all groups of high concentration.

  • This study was performed to investigate the effects of dietary protein levels(5% and 15% PEP), caffeine or green tea powder on body fat deposition in rats. Male weanling Wistar rats weighting the average of 90g were allotted into 6 experimental groups, each of which was fed two different levels of dietary protein with or without caffeine or green tea powder(5p+0.15% caffeine ; 5p+6.1% green tea powder ; 15p+0.15% caffeine ; 15p+6.1% green tea powder) during 8 weeks of the experimental period. Caffeine and green tea powder were supplemented at the levels of 0.15% and 6.1% of experimental diets. The rats fed 5% PEP diet which had received caffeine or green tea powder showed significantly(p<0.01) reduced gain in body weight. The food efficiency of which rats fed both 5% and 15% PEP diet supplemented with 6.1% green tea powder was significantly low(p<0.05, respectively) compared with the control group. Rats fed diets containing 0.15% caffeine and 6.1% green tea powder showed the significant reduction(p<0.01) of hite adipose tissue weight, triglycerides levels of liver and plasma. The addition of 0.15% caffeine or 6.1% green tea powder to 5% PEP diet resulted in significantly(p<0.01) higher levels of plasma total cholesterol, free cholesterol, cholesterol ester, but HDL-cholesterol levels were significantly(p<0.01, in 5% PEP group) high. The atherogenic index(Tchol-HDLchol/HDLchol) in rats fed 6.1% green tea powder diets decreased especially compared with the control group.

  • This study examined the levels of recognition, knowledge, and intake behavior of foods containing caffeine and analyzed the factors affecting amount of caffeine intake to get basic materials for development of educational program for proper intake behavior of foods containing caffeine of high school students in Gyeongbuk region. Of 300 questionnaires that were distributed on December 27, 2013 to February 14, 2014, 255 responses were used for analysis. Frequency distributions, Cronbach's ${\alpha}$, t tests, one-way ANOVA, Pearson's correlation analysis, and regression analysis were conducted by SPSS Windows V.21.0. The mean of the level of concern about foods containing caffeine was 2.76/5.00 points; male and second and third grade students showed lower concern level than female and first grade students. The respondents answered that intake of foods containing caffeine had somewhat harmful effect on health, but it did not much help to refresh and increase concentration. The mean for the percentage of correct answers about knowledge of caffeine was 54.3. Small number of the respondents were advised to reduce intake of caffeine from school teachers or parents. The average of caffeine intake amount was 71.67mg/day; the average for male was 78.98mg/day, whereas for female 61.23mg/day. Almost thirteen percentage of the respondents took caffeine over RDA(Recommended Daily Allowance). According to the result of regression analysis for amount of caffeine intake, grade, BMI, concern about health, perception of whether foods containing caffeine help with refreshment and increase of concentration or not, and experiences of having advice from school teachers to decrease caffeine intake were significant. These results imply that it is necessary for high school students to educate how to take caffeine properly.
