Mammalian target of rapamycin inhibitors for treatment in tuberous sclerosis

  • Tuberous sclerosis complex (TSC) is a genetic multisystem disorder that results from mutations in the TSC1 or TSC2 genes, and is associated with hamartomas in several organs, including subependymal giant cell tumors. The neurological manifestations of TSC are particularly challenging and include infantile spasms, intractable epilepsy, cognitive disabilities, and autism. The TSC1- and TSC2-encoded proteins modulate cell function via the mammalian target of rapamycin (mTOR) signaling cascade, and are key factors in the regulation of cell growth and proliferation. The mTOR pathway provides an intersection for an intricate network of protein cascades that respond to cellular nutrition, energy levels, and growth factor stimulation. In the brain, TSC1 and TSC2 have been implicated in cell body size, dendritic arborization, axonal outgrowth and targeting, neuronal migration, cortical lamination, and spine formation. The mTOR pathway represents a logical candidate for drug targeting, because mTOR regulates multiple cellular functions that may contribute to epileptogenesis, including protein synthesis, cell growth and proliferation, and synaptic plasticity. Antagonism of the mTOR pathway with rapamycin and related compounds may provide new therapeutic options for TSC patients.

Plant Growth Promotion and Suppression of Root Disease Complex due to Meloidogyne incognita and Fusarium oxysporum by Fluorescent Pseudomonads in Tomato

  • While screening for nematicidal activity of bacterial origins, various pseudomonads strains were inhabited in tomato rhizosphere. One isolate designated as $PE_{10}$ was selected for studies on nematicidal properties and plant growth-promoting (PGP) activity and was identified as Pseudomonas aeruginosa based on morphological features, biochemical and physiological tests, and carbohydrate utilization. To investigate nematicidal activity, Meloidogyne incognita juvenile mortality was determined using $PE_{10}$ culture filtrate. Inhibition of strain $PE_{10}$ against Fusarium oxysporum was observed using dual culture technique. Strain $PE_{10}$ showed good siderophore activity, HCN and IAA production abilities, and growth and development enhancement of tomato.

Thyroid Hormones Receptor/Reporter Gene Transcription Assay for Food Additives and Contaminants

  • Many of thyroid hormones disrupting chemicals induce effects via interaction with thyroid hormone and retinoic acid receptors and responsive elements intrinsic in target cells. We studied thyroid hormones disrupting effects of food additives and contaminants including BHA, BHT, ethoxyquin, propionic acid, sorbic acid, benzoic acid, CPM, aflatoxin B1, cadmium chloride, genistein, TCDD, PCBs and TDBE in recombinant HeLa cells containing plasmid construct for thyroxin responsive elements. The limit of response of the recombinant cells to T3 and T4 was $1\times10^{-12}\;M$. BHA. genistein, cadmium and TBDE were interacted with thyroid receptors with dose-responsive pattern. In addition, BHA, BHT, ethoxyquin, propionic acid, benzoic acid, sorbic acid, and TBDE showed synergism while cadmium chloride antagonism for T3-induced activity. This study elucidates that recombinant HeLa cell is sensitive and high-throughput system for the detection of chemicals that induce thyroid hormonal disruption via thyroid hormone receptors and responsive elements. Also this study raised suspect of BHA. BHT, ethoxyquin, propionic acid, benzoic acid, sorbic acid, TBDE, genisteine and cadmium chloride as thyroid hormonal system disruptors.

  • The Fourth Industrial Revolution is currently leading humanity into a super-connected, super-intelligent, and super-converged society through key technologies such as artificial intelligence, the Internet of Things (IoT), and big data. Blockchain technology has the potential to lead social innovation that will improve the mutual harmony and understanding of the complex structures of human society as well as various phenomena (e.g., antagonism, confrontation, and ideological conflict). With such progress in social innovation, blockchain technology will result in a fairer and more transparent human society. In addition, blockchain technology is emerging as a core infrastructure technology, and its growth and expansion are expected to bring about revolutionary improvements to nearly all fields of development and research. In this paper, we briefly outline the main features of blockchain technology. Through a further analysis of its ecosystem, we intend to offer suggestions for a more robust and efficient development of blockchain-related industries.

  • DMZ is came from the antagonism of ideology of the cold war and will of peace embodiment of Gangwon province, the only divided province in the world, at the time of opening physical distribution transportation time on ground between south and north Korea. Analyze the central plan of Gangwon province that includes Mt. Geumgang sightseeing, the Gyeongwon line and the Donghae line. As regaining the overland route with Eurasian continent that lost because of the division of Korea into north and south, We have lived as people in a island country actually for the last 60 years. we should extend the racial imagination that lean toward the ocean to the continent again. As we present the use of efficient overland distribution transportation that is endowed with the fitness of the Gangwon province's plan to make a new developing opportunity physically and mentally. we would seek the revitalization of Gangwon economy and inter Korean trade.

Precision Cardiology: Phenotype-targeted Therapies for HFmrEF and HFpEF

  • Heart failure with mid-range ejection fraction (HFmrEF) and preserved ejection fraction (HFpEF) represent over half of heart failure cases but lack proven effective therapies beyond sodium-glucose cotransporter 2 inhibitor and diuretics. HFmrEF and HFpEF are heterogeneous conditions requiring precision phenotyping to enable tailored therapies. This review covers concepts on precision medicine approaches for HFmrEF and HFpEF. Areas discussed include HFmrEF mechanisms, anti-inflammatory and antifibrotic treatments for obesity-related HFpEF, If inhibition for HFpEF with atrial fibrillation, and mineralocorticoid receptor antagonism for chronic kidney disease-HFpEF. Incorporating precision phenotyping and matched interventions in HFmrEF and HFpEF trials will further advance therapy compared to blanket approaches.

Functional Characterization of Antagonistic Fluorescent Pseudomonads Associated with Rhizospheric Soil of Rice (Oryza sativa L.)

  • Antagonistic fluorescent pseudomonads isolated from rhizospheric soil of rice were characterized by 16S rRNA amplicon and fatty acid methyl ester (FAME) analyses. Antagonistic isolates were grown in the fermentation media, and production of antibiotics was confirmed by thin-layer chromatography (TLC) and high-performance liquid chromatography (HPLC). Production of fungal cell-wall-degrading enzymes such as protease, cellulase, pectinase, and chitinase was determined. Dendrogram based on the major and differentiating fatty acids resulted into 5 clusters, viz., cluster I (P. pseudoalcaligenes group), cluster II (P. plecoglossicida group), cluster III (P. fluorescens group), cluster IV (P. aeruginosa group), and cluster V (P. putida group). Characteristic presence of high relative proportions of cyclopropane (17:0 CYCLO w7c) was observed in antagonistic bacteria. Data revealed biodiversity among antagonistic fluorescent pseudomonads associated with the rice rhizosphere. Results presented in this study will help to identify the antagonistic isolates and to determine their mechanisms that mediate antagonism against fungal pathogens of rice.

청소년의 자세 교정을 위한 감성 공학적 셔츠 디자인에 관한 연구 (A study on shirts design for postural stability of teenagers considering human sensibility ergonomics)

  • For design research of teenagers' posture correcting shirt, postures of teenagers were analyzed and became the base of inventing suitable patterns. The subjects of this study were 198 of both female and male middle school students from the city of Seoul. Students' postures in classroom and length change of body surface were observed and analyzed; based on this, total of 5 postures was selected, and length changes of body surface were applied. The patterns in this study were designed with stretch material to help antagonism by muscle movements. Each fabric that was used in this study had 110% of stretch, 120~140% of stretch, and 150~170% of stretch. Both of one-way-fabric and duplex-fabric were used in this study, depending on application site.

Acute toxicity of four alkylphenols (3-tert-butyl-, 2-isopropyl-, 3-propropyl-, and 4-isopropyl-phenol) and their binary mixtures to Microtox, with comparisons to Ceriodaphnia dubia and Pimephales promelas

  • Toxicity evaluations of 3-tert-butyl-, 2-isopropyl-, 3-isopropyl- and 4-propyl-phenol and their binary mixtures were performed with the Microtox$\^$(R) / assay and compared to invertebrates and fish. The single chemical, 4-isopropylphenol, exhibited the greatest relative toxicity to the Microtox organism (Vibrio fischeri). The relative electrophilicity (LUMO) of the phenols, in contrast to the lipophilicity (Log P), was strongly correlated with toxicity to V fischeri (r$^2$=0.96, p<0.01). In contrast, relative electrophilicity alone could not explain variances in toxicity of the phenols to Ceriodaphnia dubia. Results suggest that electrophilicity in conjunction with lipophilicity provide better correlation with toxicity to C. dubia and Pimephales promelas. Microtox results from the binary mixture toxicity tests of selected phenolics indicate a mechanism of interaction governed by suppression/antagonism.

Assessment of the Contribution of Antagonistic Secondary Metabolites to the Antifungal and Biocontrol Activities of Pseudomonas fluorescens NBC275

  • An understanding of the contribution of secondary metabolites (SMs) to the antagonistic and biocontrol activities of bacterial biocontrol agents serves to improve biocontrol potential of the strain. In this study, to evaluate the contribution of each SM produced by Pseudomonas fluorescens NBC275 (Pf275) to its antifungal and biocontrol activity, we combined in silico analysis of the genome with our previous study of transposon (Tn) mutants. Thirteen Tn mutants, which belonged to 6 biosynthetic gene clusters (BGCs) of a total 14 BGCs predicted by the antiSMASH tool were identified by the reduction of antifungal activity. The biocontrol performance of Pf275 was significantly dependent on 2,4-diacetylphloroglucinol and pyoverdine. The clusters that encode for arylpolyene and an unidentified small linear lipopeptide influenced antifungal and biocontrol activities. To our knowledge, our study identified the contribution of SMs, such as a small linear lipopeptide and arylpolyene, to biocontrol efficacy for the first time.
