The purpose of this study was to compare the ability of an engineered Escherichia coli to degrade chlorpyrifos (Cp) using an organophosphorus hydrolase enzyme, encoded in both Flavobacterium sp. ATCC 27551 or Pseudomonas diminuta, by employing the Lpp-OmpA chimera and the N-terminal domain of the ice nucleation protein as anchoring motifs. Tracing of the expression location of the recombinant protein using SDS-PAGE showed the presentation of OPH by both anchors on the outer membrane. This is the first report on the presentation of OPH on the cell surface by Lpp-OmpA under the control of the T7 promoter. The results showed cell growth in the presence of Cp as the sole source of energy, without growth inhibition, and with higher whole-cell activity for both cells harboring plasmids pENVO and pELMO, at approximately 10,342.85 and 10,857.14 U/mg, respectively. Noticeably, the protein displayed by pELMO was lower than the protein displayed by pENVO. It can be concluded that Lpp-OmpA can display less protein, but more functional OPH protein. These results highlight the high potential, of both engineered bacteria, for use in the bioremediation of pesticide-contaminated sources in the environment.

Adenovirus (Ad) precursor to the terminal protein (pTP) plays an essential roles in the viral DNA replication. Ad pTP serves as a primer for the synthesis of a new DNA strand during the initiation step of replication. In addition, Ad pTP forms organized spherical replication foci on the nuclear matrix (NM) and anchors the viral genome to the NM. Here we identified the interferon inducible gene product 1-8D (Inid) as a pTP binding protein by using a two-hybrid screen of a HeLa cDNA library. Of the clones obtained in this assay, nine were identical to the Inid, a 13-kDa polypeptide that shares homology with genes 1-8U and Leu-13/9-27, most of which have little known functions. The entire open reading frame (ORF) of Inid was cloned into the tetracycline inducible expression vector in order to determine the biological functions related with adenoviral infection. When Inid was introduced to the cells along with adenoviruses, fifty to sixty percent of Ad-infected cells expressing Inid had rounded morphology, which was suggestive of apoptosis. Results from the terminal deoxynucleotidyl transferase (TdT) and DNA fragmentation assays confirmed that Inid induces apoptosis in Ad-infected or in uninfected cells. The Inid binding to pTP may target the cell for apoptotic destruction as a host defense mechanism against the viral infection.

A microgyroscope, which vibrates in two orthogonal axes on the substrate plane, is designed and fabricated. The shuttle mass of the vibrating gyroscope consists of two parts. The one is outer shuttle mass which vibrates in driving mode guided by four folded springs attached to anchors. And the other is inner shuttle mass which vibrates in driving mode as the outer frame does and also can vibrate in sensing mode guided by four folded springs attached to the outer shuttle mass. Due to the directions of vibrating mode, it is possible to fabricate the gyroscope with simplified process by using polysilicon on insulator structure. Fabrication processes of the microgyroscope are composed of anisotropic silicon etching by RIE, gas-phase etching (GPE) of the buried sacrificial oxide layer, metal electrode formation. An eletromechanical model of the vibrating microgyroscope was modeled and bandwidth characteristics of the gyroscope operates at DC 4V and AC 0.1V in a vacuum chamber of 100mtorr. The detection circuit consists of a discrete sense amplifier and a noise canceling circuit. Using the evaluated electromechanical model, an operating condition for high performance of the gyroscope is obtained.

Thiol-based self-assembled anchor linked to glucose oxidase (GOx) and gold nanoparticle (GNP) cluster is suggested to enhance the performance of glucose biosensor. By the adoption of thiol-based anchors, the activity of biocatalyst consisting of GOx, GNP, polyethyleneimine (PEI) and carbon nanotube (CNT) is improved because they play a crucial role in preventing the leaching out of GOx. They also promote electron collection and transfer, and this is due to a strong hydrophobic interaction between the active site of GOx and the aromatic ring of anchor, while the effect is optimized with the use of thiophenol anchor due to its simple configuration. Based on that, it is quantified that by the adoption of thiophenol as anchor, the current density of flavin adenine dinucleotide (FAD) redox reaction increases about 42%, electron transfer rate constant ($k_s$) is $9.1{\pm}0.1s^{-1}$ and the value is 26% higher than that of catalyst that does not use the anchor structure.

This study performed lateral load testing on seventeen wood wall frames in two sections. Section one included eight tests studying structural foam sheathing of shear walls subjected to monotonic loads following the ASTM E564 test method. In this section, the wood frame was sheathed with four different types of structural foam sheathing on one side and gypsum wallboard (GWB) on the opposite side of the wall frame, with Simpson HDQ8 hold down anchors at the terminal studs. Section two included nine tests studying wall constructed with oriented strand board (OSB) only on one side of the wall frame subjected to gradually applied monotonic loads. Three of the OSB walls were tied to the baseplate with Simpson LSTA 9 tie on each stud. From the test results for Section one; the monotonic tests showed an 11 to 27 percent reduction in capacity from the published design values and for Section two; doubling baseplates, reducing anchor bolt spacing, using bearing plate washers and LSTA 9 ties effectively improved the OSB wall capacity. In comparison of sections one and two, it is expected the walls with structural foam sheathing without hold downs and GWB have a lower wall capacity as hold down and GWB improved the capacity.
This study investigated the ultimate lateral load capacity of shear walls constructed with several types of structural foam sheathing. Sixteen tests were conducted and the results were compared to the published design values commutated by the manufactures for each test series. The sheathing products included 12.7 mm (1/2 in) SI-Strong, 25.4 mm (1 in) SI-Strong, 12.7 mm (1/2 in) R-Max Thermasheath, and 2 mm (0.078 in) ThermoPly Green. The structural foam sheathing was attached per the manufacturers' specification to one side of the wood frame for each wall tested. Standard 12.7 mm (1/2 in) gypsum wallboard was screwed to the opposite side of the frame. Simpson HDQ8 tie-down anchors were screwed to the terminal studs at each end of the wall and anchored to the base of the testing apparatus. Both monotonic and cyclic testing following ASTM E564 and ASTM E2126, respectively, were considered. Results from the monotonic tests showed an 11 to 27 percent smaller capacity when compared to the published design values. Likewise, the test results from the cyclic tests showed a 24 to 45 percent smaller capacity than the published design values and did not meet the seismic performance design criteria computation.
In this study, an innovative anchoring approach has been developed dealing with all relevant aspects in consideration of previous works. An ultimate pulling force calculation of anchor is presented from a geotechnical point of view. The proposed umbrella anchor focuses not only on the friction resistance capacity, but also on the axial capacity of the composite end structure and the friction capacity occurring around the wedge. Even though the theoretical background is proposed, in-situ application requires high-level mechanical design. Hence, the required parts have been carefully improved and are composed of anchor body, anchor cap, connection brackets, cutter vanes, open-close ring, support elements and grouting system. Besides, stretcher element made of aramid fabric, interior grouting system, guide tube and cable-locking apparatus are the unique parts of this design. The production and placement steps of real sized anchors are explained in detail. Experimental results of 52 pullout tests on the weak dry soils and 12 in-situ tests inside natural soil indicate that the proposed approach is conservative and its peak pullout value is directly limited by a maximum strength of anchored soil layer if other failure possibilities are eliminated. Umbrella anchor is an alternative to conventional anchor applications used in all types of soils. It not only provides time and workmanship benefits, but also a high level of economic gain and safe design.

Guyed steel lattice towers (or guyed masts) are widely used for supporting antennas for telecommunications and broadcasting. This paper presents a numerical study on the static and dynamic response of guyed towers. Three-dimensional nonlinear finite-element models are used to simulate the response. Through performing static pushover analyses and free-vibration (modal) analyses, the effect of different bracing configurations is investigated. In addition, seismic analyses are performed on towers of different heights to study the influence of earthquake excitation time-lag (or the earthquake travel distance between tower anchors) and antenna weight on the seismic response of guyed towers. The results show that the inclusion of time lag in the seismic analysis of guyed towers can influence shear and moment distribution along the height of the mast. Moreover, it is found that the lateral response is insensitive to bracing configurations. The results also show that, depending on the mast height, an increased antenna weight can reduce the tower maximum base shear while other response quantities, such as cables tension force are found to be insensitive to variation in the antenna weight.

Suture anchors are commonly used in shoulder surgeries, especially for rotator cuff tears. Peri-anchor cyst formation, however, is sometimes detected on follow-up radiologic image after surgery. The purpose of this report is to discuss the case of a patient who presented with regression of extensive peri-anchor cyst on postoperative 4-year follow-up magnetic resonance imaging and had good clinical outcome despite peri-anchor cyst formation after arthroscopic rotator cuff repair.
Non-monolithic concrete structural connections are commonly used both in new constructions and retrofitted structures where anchors are used for connections. Often, flaws are present in anchor system due to poor workmanship and deterioration; and methods available to check the quality of the composite system afterward are very limited. In case of presence of flaw, load transfer mechanism inside the anchor system is severely disturbed, and the load carrying capacity drops drastically. This raises the question of safety of the entire structural system. The present study proposes a wave propagation technique to assess the integrity of the anchor system. A chemical anchor (embedded in concrete) composite system comprising of three materials viz., steel (anchor), polymer (adhesive) and concrete (base) is considered for carrying out the wave propagation studies. Piezoelectric transducers (PZTs) affixed to the anchor head is used for actuation and the PZTs affixed to the surrounding concrete surface of the concrete-anchor system are used for sensing the propagated wave through the anchor interface to concrete. Experimentally validated finite element model is used to investigate three types of composite chemical anchor systems. Studies on the influence of geometry, material properties of the medium and their distribution, and the flaw types on the wave signals are carried out. Temporal energy of through time domain differentiation is found as a promising technique for identifying the flaws in the multi-layered composite system. The present study shows a unique procedure for monitoring of inaccessible but crucial locations of structures by using wave signals without baseline information.
