Genetic Relationship between Populations and Analysis of Genetic Structure in the Korean Native Chicken and the Endemic Chicken Breeds (한국재래닭 및 토착화 품종간의 유연 관계 및 유전 특성 분석)

  • The purpose of this study was to assess the genetic variation and establish the relationship amongst breeds and strains using 7 chicken specific microsatellite markers. A total of 317 DNA samples from four Korean native chicken (KNC) strains (KR: Korean Native Red chicken strain, KY: Korean Native Yellow chicken strain, KL: Korean Native Black chicken strain, KO: Ogol chicken strain) and three introduced endemic chicken breeds (LE: Leghorn chicken breed, RI: Rhode Island Red chicken breed, CO: Cornish chicken breed). The size of microsatellite markers was decided using GeneMapper Software (v.4.0) after being analyzed using an ABI 3130 Genetic Analyzer. Frequencies of microsatellites markers were used to estimate heterozygosities and genetic distances. The lowest distance (0.074) was observed between the KY and KL breeds and the highest distance (0.779) between the KL and LE breeds. The KNC strains (KR, KY, KL) have comparatively near genetic distance each other. On the other side, each individual was not ramified to different groups and were spread evenly in phylogenetic dendrogram about all the KNC of each strain populations. But the endemic breed populations (LE, RI, CO) were ramified to different groups. The microsatellite polymorphism data were shown to be useful for assessing the genetic relationship between Korean native strains and other foreign breeds.

Sports Biomechanical Analysis of Physical Movements on the Basis of the Patterns of the Ready Poses (준비동작의 형태 변화에 따른 신체 움직임의 운동역학적 분석)

  • The purpose of this research is to provide a proper model by analyzing the sports biomechanical of physical movements on the basis of the two patterns(open-stance and cross-stance) at the ready-to-start pose. The subjects for this study are composed of five male handball players from P university and five female shooting players from S university. Three-way moving actions at start(right, left, and forward) are recorded with two high-speed video cameras and measured with two Force platforms and a EMG system. Three-dimensional action analyzer, GRF system, and Whole body reaction movement system are used to figure out the moving mechanisms at the start pose. The analytic results of the moving mechanism at the start pose were as follows. 1. Through examining the three-way moving actions at start, I have found the cross-stance pose is better for the moving speed of body weight balance than the open-stance one. 175 degree of knee joint angle at "take-off" and 172 degree of hip joint angle were best for the start pose. 2. The Support time and GRF data shows that the quickest center of gravity shift was occurred when cross-stanced male subjects started to move toward his lefthand side. The quickest male's average supporting time of left and right foot is 0.19${\pm}$0.07 sec., 0.26${\pm}$0.06sec. respectively. The supporting time difference between two feet is 0.07sec. 3. Through analyzing GRF of moving actions at start pose, I have concluded that more than 1550N are overloaded on one foot at the open-stance start, and the overloaded force may cause physical injury. However, at the cross-stance pose, The GRF are properly dispersed on both feet, and maximum 1350N are loaded on one foot.

A Study on the Contents of Trace Metals in the Crops cultivated in Han-riverside (한강둔치에서 재배된 농작물 중의 미중금속 함량에 관한 연구)

  • This study was conducted to determine the content of trace metals in crops cultivated in Han-riverside, Anyang stream, Tan stream, and Jungryang stream. Trace metals (Pb, Cr, Cd, Fe, Zn, Cu, Hg) were detected in 36 crops, 285 samples by Atomic Absorption Spectrophotometer and Mercury Analyzer. The average concentration of trace metals in crop samples was in the order of Fe (34.592 ppm)>Mn (11.071 ppm)$\geq$Zn(8.853 ppm)>Cu(1.795 ppm)>Cr(0.237 ppm)>Pb(0.100 ppm)>Cd(0.011 ppm)>Hg(0.003 ppm). In crop samples, the contents of Fe, Cu, Mn, and Zn, which ranges were 0.004-203.083 ppm, 0.017~22.727 ppm, 0.000~74.373 ppm, 0.080~37.166 ppm, respectively, were relatively higher than those of Pb, Cd, Cr, and Hg, which ranges were 0.000~0.654 ppm, 0.000~0.270 ppm, 0.000~l.229 ppm, 0.000~0.037 ppm, respectively. The concentration of trace metals in crop samples was in the order of leafy vegetables > stem vegetables > root vegetables > fruity vegetables. In the root vegetables, such as radish, the content of Pb was leaf(0.055 ppm)>root(0.035 ppm), that of Cr, leaf (0.118 ppm)>root(0.031 ppm), that of Cd, leaf(0.004 ppm)>root(0.001 ppm), that of Hg, leaf(0.004 ppm)>root(0.001 ppm). As the results, it could be thought that root is the organ which doesn't accumulate the heavy metal ions, but absorb them.

Heavy Metal Contents of Vegetables Available on the Markets in Seoul (서울에서 유통 중인 채소류의 중금속 함량에 관한 연구)

  • This study was conducted to estimate the heavy metal contents of vegetables available on the markets in Seoul area. Concentrations of mercury (Hg), lead (Pb), cadmium (Cd), arsenic (As), chrome (Cr), nickel (Ni), copper (Cu), and zinc (Zn) were measured in 300 samples using a mercury analyzer and inductively coupled plasma optical emission spectrometer (ICP-OES) after wet digestion. The average values of heavy metals in vegetables were as follows [mean (minimum~maximum), mg/kg]; Hg: 0.0005 (N.D~0.007), Pb: 0.011 (N.D~0.259), Cd: 0.012 (N.D~0.188), As: 0.002 (N.D~0.142), Cr: 0.100 (0.019~0.954), Ni: 0.093 (0.003~1.231), Cu: 1.098 (0.072~36.29), and Zn: 3.48 (0.485~21.31). The heavy metal contents of vegetables available on the markets in Seoul were almost the same as or lower than those reported in other studies. The weekly average intakes of mercury, lead and cadmium from vegetables take 0.44~7.71% of PTWI (Provisional Tolerable Weekly Intakes) that the FAO/WHO Joint Food Additive and Contaminants Committee sets for evaluation of food safety.

Cooking Properties of Fresh Pasta Using Korean Wheat and Durum Rimachinata (우리밀과 Durum Rimachinata를 이용한 생면 파스타의 조리특성)

  • This study investigated the physicochemical characteristics of Korean wheat flour substituted for 0%, 15%, 30%, 45%, and 60% durum rimachinata wheat in order to develop a Korean wheat pasta suitable for consumer-preferred soft textures. The particles of Korean wheat that were less than 250 ${\mu}M$ were 87.03% of all particles, while 68.7% of durum rimachinata had particles more than 250 ${\mu}M$ in size. Durum rimachinata had more protein (13.84${\pm}$0.03) and ash (0.70${\pm}$0.02) than Korean wheat. In farinograph characteristics, water absorption, development time, stability, and weakness increased as the amounts of substituted Korean wheat flour increased. Also, the gelatinization characteristics of the amylograph exhibited an increase of gelatinization temperature and decrease in maximum viscosity. However, maximum viscosity was shown to be more than 550 B.U. until 30% of the substitution level of Korean wheat flour to durum rimachinata wheat. Also, it did not affect the texture of the noodle product. We could make pasta with softness and springiness with less than a 15% substitution level of Korean wheat flour due to similar characteristics in cooking properties such as weight, volume, water absorption, turbidity, and cooking loss when compared to the control. L and a values increased, and the b value decreased in color as substitution amounts of Korean wheat flour increased. The hardness and adhesiveness of cooking noodles was shown to be a low value at more than a 30% substitution level of Korean wheat flour, and springiness, gumminess, and chewiness all exhibited high values. In a sensory evaluation, overall acceptability was shown to have the highest score in control. More than 30% of substitution of Korean wheat flour showed high preferences. Therefore, 15% of the substitution level of Korean wheat flour could be adapted in dough and cooking properties for making pasta-substituted Korean wheat. However, a texture analyzer and sensory evaluation of cooked pasta was shown to have a good quality at more than 30% substitution level of Korean wheat flour.

  • Effect of microwave radiation on physico-chemical properties of cor'n starches was studied. Waxy com, com and high amylose com starches of varying moisture content(20~35%) were subjected to microwave processing(2450MHz) at $120^{\circ}$ and the experimental starch samples were examined by a X-ray diffractometry, rapid viscosity analyzer(RVA) and. with the samples in temperature was observed and the peaks of high amylose com starches at $2^{\circ}$=5.0, 15.0 and $23.0^{\circ}$, were disappeared indicating the melting of crystallines while those of com and waxy com had not changed. A change in gelatinization pattern was observed in the case of corn starches from type A with nearly no peak-viscosity and breakdown to type C. Except a decreased viscosity, no change was observed in those of waxy com starches.

Monitoring soybean growth using L, C, and X-bands automatic radar scatterometer measurement system (L, C, X-밴드 레이더 산란계 자동측정시스템을 이용한 콩 생육 모니터링)

  • Soybean has widely grown for its edible bean which has numerous uses. Microwave remote sensing has a great potential over the conventional remote sensing with the visible and infrared spectra due to its all-weather day-and-night imaging capabilities. In this investigation, a ground-based polarimetric scatterometer operating at multiple frequencies was used to continuously monitor the crop conditions of a soybean field. Polarimetric backscatter data at L, C, and X-bands were acquired every 10 minutes on the microwave observations at various soybean stages. The polarimetric scatterometer consists of a vector network analyzer, a microwave switch, radio frequency cables, power unit and a personal computer. The polarimetric scatterometer components were installed inside an air-conditioned shelter to maintain constant temperature and humidity during the data acquisition period. The backscattering coefficients were calculated from the measured data at incidence angle $40^{\circ}$ and full polarization (HH, VV, HV, VH) by applying the radar equation. The soybean growth data such as leaf area index (LAI), plant height, fresh and dry weight, vegetation water content and pod weight were measured periodically throughout the growth season. We measured the temporal variations of backscattering coefficients of the soybean crop at L, C, and X-bands during a soybean growth period. In the three bands, VV-polarized backscattering coefficients were higher than HH-polarized backscattering coefficients until mid-June, and thereafter HH-polarized backscattering coefficients were higher than VV-, HV-polarized back scattering coefficients. However, the cross-over stage (HH > VV) was different for each frequency: DOY 200 for L-band and DOY 210 for both C and X-bands. The temporal trend of the backscattering coefficients for all bands agreed with the soybean growth data such as LAI, dry weight and plant height; i.e., increased until about DOY 271 and decreased afterward. We plotted the relationship between the backscattering coefficients with three bands and soybean growth parameters. The growth parameters were highly correlated with HH-polarization at L-band (over r=0.92).

Studies on the Solubility Phenomenon and Activities of Silk Cocoon Sericin through the Filature Water Conditions. (제사용수의 수질이 견층세리신의 용해에 미치는 거동구명에 관한 연구)

  • This study was carried out ill an attempt to investigate the properties and activities of sericin obtained from silk cocoon shells in silk reeling water through various instrumental analyses. In addition, the effects of the characteristics in sericin solubility on the reeling process and silk qualities were also studied on the basis of tile above conditions. The results obtained are as follows: I. The sericin properties and activities through various analytical instruments. 1. The water solubility of each amino acid such as serine, glycine. glutamic and aspartic acids against the pH modified water by using automatic amino acid analyzer, showed the lowest solubility at PH 5, but it increased according to drifting toward the alkalinity. 2. When the obtained sericin particles by water pH variation were observed with the electronic microscope, it was found for the sericin Particles to expand in the alkaline regions. 3. The IR spectrum results showed the differences among the pH modified sericin solutions at the range of 2,100cm-1 and 1.890cm-1 of wave number. 4. The existence of sericin with in silk fabrics made differences in the X-ray interference intencity, that is, the non-degummed fabrics had the interference peak at 2$\theta$=14$^{\circ}$, 17$^{\circ}$, 24$^{\circ}$, and the degummed ones had it at 2$\theta$=17$^{\circ}$, 18$^{\circ}$, 20$^{\circ}$, 23$^{\circ}$, 26$^{\circ}$. II. The results of sericin solubility for silk reeling process. 1. The sericin solubility and swelling had a tendency to increase up with high M-alkalinity and pH value of water. But in case of acidity. water hardness and concentration of ethylene glycol were high, the sericin solubility and swelling were decreased. 2. With the filature experiments, the best conditions of filature orator are summarized as fellows ; a. pH; 6.9${\pm}$0.2 d. acidity; below10ppm b. total hardness; 55:5ppm e. Fe ion; none c. M-alkalinity; 40${\pm}$10ppm

Occurrence Characteristics and Existing Forms of U-Th Containing Minerals in KAERI Underground Research Tunnel(KURT) Granite (한국원자력연구원 지하처분연구시설(KURT) 화강암의 U-Th 함유광물 산출특성 및 존재형태)

  • Occurrence characteristics and existing forms of U-Th containing minerals in KURT (KAERI Underground Research Tunnel) granite are investigated to understand long-term behavior of radionuclides in granite considered as a candidate rock for the geological disposal of high-level radioactive waste. KURT granite primarily consists of quartz, feldspar and mica. zircon, REE(Rare Earth Element)-containing monazite and bastnaesite are also identified. Besides, secondary minerals such as sericite, microcline and chlorite including quartz vein and calcite vein are observed. These minerals are presumed to be accompanied by a post-hydrothermal process. U-Th containing minerals are mainly observed at the boundaries of quartz, feldspar and mica, mostly less than $30{\mu}m$ in size. Quantitative analysis results using EPMA (Electron Probe Micro-Analyzer) show that 74.2 ~ 96.5% of the U-Th containing minerals consist of $UO_2$ (3.39 ~ 33.19 wt.%), $ThO_2$ (41.61 ~ 50.24 wt.%) and $SiO_2$ (15.43 ~ 18.60 wt.%). Chemical structure of the minerals calculated using EPMA quantitative analysis shows that the U-Th minerals are silicate minerals determined as thorite and uranothorite. The U-Th containing silicate minerals are formed by a magmatic and hydrothermal process. Therefore, KURT granite formed by a magmatic differentiation is accompanied by an alteration and replacement owing to a hydrothermal process. U-Th containing silicate minerals in KURT granite are estimated to be recrystallized by geochemical factors and parameters such as temperature, pressure and pH owing to the hydrothermal process. By repeated dissolution/precipitation during the recrystallization process, U-Th containing silicate minerals such as thorite and uranothorite are formed according to the variation in the concentrated amount of U and Th.

Phytic Acid Protects the Formation of Colonic Aberrant Crypt Foci Induced by Azoxymethane in Male F344 Rats (랫드에서 azoxymethane으로 유도된 대장 전암병변에 대한 피티산의 방어 효과)

  • Phytic acid(PA) (Inositol hexaphosphate, $IP_6$) is a naturally occurring polyphosphorylated carbohydrate that is present in substantial amounts in almost all plants and mammalian cells. Recently PA has received much attention for its role in anticancer activity. In the present study, the preventive effects of PA on colon carcinogenesis were investigated. Six-week old Fisher 344 male rats were fed a AIN-93G purified diet and PA(0.5% or 2% PA in water) for 8 weeks. The animals received two ($1^{st}\;and\;2^{nd}$ week) injections of azoxymethane(AOM, 15 mg/kg b.w.) to induce colonic aberrant crypt foci(ACF). After sacrifice, the total numbers of aberrant crypts(AC) and ACF in colonic mucosa were examined after staining with methylene blue. Blood and serum were analyzed with a blood cell differential counter and an automatic serum analyzer. AOM induced the total numbers of $142.3{\pm}22.3$ ACF/colon and $336.6{\pm}55.1$ AC/colon. PA at the doses of 0.5 and 2% decreased the numbers of ACF and AC/colon in a dose-dependent manner. The numbers of ACF/colon and AC/colon by PA at the dose of 0.5% were $124.4{\pm}28.5\;and\;302.7{\pm}67.3$, respectively. PA at the dose of 2% significantly decreased the ACF and AC numbers to $109{\pm}18.1\;and\;254.8{\pm}50.6$, respectively(p<0.01). Especially, 2% PA significantly reduced the number of large ACF(${\geq}4$ AC/ACF) from $26.8{\pm}6.2$ ACF/colon to $15{\pm}6.7$ ACF/colon(p<0.01). Although some parameters in blood counts and serum chemistry were changed compared with the control, no specific toxicity was found. These findings suggest that phytic acid can be a chemopreventive agent for colon carcinogenesis resulting from inhibition of the development of ACF in the F344 rat.
