LAS(Linear Alkybenzene Sulfonate)의 Plasmid에 의한 분해 (Degradation of MS(Linear Alkylbenzene Sulfonate) by Plasmid)

  • Microorganisms capale of utilizing linear alkylbenzene sulfonates(LAS) as sole carbon source were isolated from industrial effluent by using LAS agar plates. The isolated strains were identified as Salmonella sp(BC-2) and Escherichia sp.(BC-3) from the results of morphological, cultural and biochemical tests. The optimal condition for the growth and biodegradation of LAS was the initial pH 7.0 and LAS concentration 0.1%. The isolated BC-2 and BC-3 strains harbored plasmid and LAS-degrading activity was lost when the plasmids were cured by mitomycin C. The plasmids were transformed into E. coli and transformants have the LAS-degrading activity. Isolated strains were examined for primary biodegradation rate of LAS in the medium by methylene blueactive substance(MBAS) method. Of these isolates, BC-2 and BC-3 strains degradated LAS upto 60% and high resistant to CdCl$_{2}$ and HgCl$_{2}$. Isolated strains were sensitive to chloramphenicol, kanamycin, rifampicin, streptomycin and tetracycline but resistant to ampicillin and lincomycin.] Its minimal inhibitory concentration(MIC) for ampicillin was more than 1500 $\mu $g/ml.

  • A genetic map of the IncP-1 group plasmid pKU10 has been prepared through the construction of recombinant plasmids containing various fragments of pKU10. Phenotypic analysis of these derivatives has identified the location of genes encoding resistance to ampicillin, tetracyclin, and chloramphenicol. The region involved in conferring resistance to ampicillin was located around two PstI sites that are 1.0Kb apart. The tetracyclin resistance gene was mapped on the region of HindIII E fragment and a part of HindIII D fragment, and the determinant for chloramphenicol resistance gene was localized on HindIII D fragment.

Prevalence of Antibiotics in Nectar and Honey in South Tamilnadu, India

  • Reverse-Phase High-performance Liquid Chromatography (RP-HPLC) based technique is one of the most sensitive techniques to detect the antibiotics present in honey. In the southern part of Tamilnadu, India, majority of the farmlands are occupied by plantations such as coconut, banana and rubber. A variety of antimicrobial compounds and antibiotics, which have been reported in pollen, nectar and other floral parts of the plant, gets accumulated in honey through honeybees (Apis mellifera). We have collected the nectar samples from banana (Musa paridasiaca) and rubber (Ficus elastica) flowers and the honey from honey hives of banana and rubber cultivated areas. The extracted nectar and honey samples are subjected to RP-HPLC analysis with authentic antibiotic standards. Nectar and honey samples showed 4-17, 11-29 ${\mu}g/kg$ of streptomycin, 2-29, 3-44 ${\mu}g/kg$ of ampicillin and 17-34, 26-48 ${\mu}g/kg$ of kanamycin respectively.

가축 및 하천에서 분리한 대장균의 항균제내성 (Isolation and Resistance Patterns of E. coli from Domestic Animals and Dranage in 1987)

  • One hundred and sixteen strains of E. coli isolated from drainage in Seoul city in 1987 and 104 strains of E. coli isolated from stools of domestic animals in suburb of Seoul in 1987 were examined for susceptibilities to 10 antimicrobial agents by the agar dilution method. The susceptibilities of the two groups to each antimicrobial agent were compared and correlations in the antimicrobial susceptibility of the 220 strains of E. coli among the 10 antimicrobial agents were also analyzed. The frequency of resistance strains was highest to tetracycline with 77%, and followed by chloramphenicol, streptomycin, ampicillin, kanamycin and cephalosporin in the descreasing order, tanging from 62% to 10%. Strains resistant to tobramycin, nalidixic acid, gentamicin and amikacin occupied 3 strains, 2 strains, 2 strains and 1 strain respectively.

어패류에서 분리한 장염비브리오균의 분포 및 항균제 감수성 (Antibiotic Susceptibility and Distribution of Vibrio parahaemolyticus Isolated from the Seafood)

  • Vibrio parahaemolyticus was investigated for analysis of distribution and antibiotic susceptibility at the markets, Seoul in 2004. The average isolation rate was 26.8%(306/1,143) and the highest rate with 64.9% was obtained in August. The isolation rates from December to April was 0.0%. It is more effective to inoculate into the enrichment medium, alkaline pepton water by swabbing a sample with a cotton ball rather than pouring mixed sample. The isolation rates were from 24.4% to 37.1% in domestic samples and from 9.8% to 26.8% in imported ones. All of the isolates were resistent to ampicillin and ticarcillin and most of them were sensitive to chloramphenicol, tetracycline, nalidixic acid, amoxicillin, ceftriaxone, streptomycin, kanamycin, trimethoprim/sulfamethoxazol, ampicillin/sulbactam, ciprofloxacin.

Antimicrobial susceptibility and pathogenic genes of Staphylococcus aureus isolated from the oral cavity of patients with periodontitis

  • Purpose: The goal of this study was to characterize the patterns of antimicrobial resistance and virulence genes in samples of Staphylococcus aureus (S. aureus) isolated from periodontitis patients. Methods: From July 2015 to August 2015, oral saliva was collected from a total of 112 patients diagnosed with periodontitis, including 80 outpatients in dental hospitals and 32 patients in dental clinics located in Seoul and Cheonan. The samples were subjected to a susceptibility test to evaluate the prevalence of antimicrobial resistance, and the pathogenic factors and antimicrobial resistance factors in the DNA of S. aureus were analyzed using polymerase chain reaction. Results: A susceptibility test against 15 antimicrobial agents showed that 88% of cultures were resistant to ampicillin, 88% to penicillin, and 2% to oxacillin. Resistance to at least two drugs was observed in 90% of cultures, and the most common pattern of multidrug resistance was to ampicillin and penicillin. Enterotoxins were detected in 65.9% of samples. The cell hemolysin gene hld was detected in 100% of cultures and hla was detected in 97.6% of samples. All strains resistant to penicillin and ampicillin had the blaZ gene. The aph(3')IIIa gene, which encodes an aminoglycoside modifying enzyme, was detected in 46.3% of samples. Conclusions: In the treatment of oral S. aureus infections, it is important to identify the pathogenic genes and the extent of antimicrobial resistance. Furthermore, it is necessary to study patterns of antimicrobial resistance and cross-infection in the context of periodontological specialties in which antimicrobials are frequently used, such as maxillofacial surgery, where the frequency of antimicrobial use for minor procedures such as implant placement is increasing.

중이염 치료용 암피실린-폴리락트산 필름의 약물방출조건 (Controlled Drug Delivery of Ampicillin-Poly(L-lactic acid) Films for the Treatment of Otitis Media)

  • A new local drug delivery device to treat otitis media (OM) has been developed. This device consists of a biodegradable poly(L-lactic acid) (PLLA) film containing antibiotic (ampicillin, AMP), which can be placed into the middle ear cavity and release the therapeutic concentration of AMP for prolonged period. Biodegradable films containing AMP (10 w/w%) were prepared by solution casting method using a suspension of the drug in a $PLLA/CH_{2}Cl_{2}$ solution (molecular weight of PLLA, 100,000 (100 K) and 300,000 (300 K), respectively). PLLA-AMP films were characterized by FTIR, DSC, and SEM. In vitro release of AMP from AMP-PLLA films were examined. The release pattern of AMP from AMP-PLLA films remained consistent from 1 day to 14 days, and the release rates of AMP from AMP-100K-PLLA film and AMP-300K-PLLA film were $0.7384\;{\mu}g/ml/day$, $0.4107\;{\mu}g/ml/day$, respectively.

식중독 환자에서 분리한 황색포도상구균의 생물학적 특성 (Biological Characteristics of Staphylococcus aureus Isolated from Food-Borne Patients in Seoul)

  • 집단식중독 환자에서 분리한 Staphylococcus aureus에 대한 항생제 감수성, enterotoxin 형별을 PCR로 조사한 결과는 다음과 같았다. 1. 의심환자분변 413건중 105건(25.4%)에서 Staphylococcus aureus coagulase 양성 균주가 분리되었다. 2. enterotoxin 생산성을 PCR로 조사한 결과 총 45주(42.9%)에서 enterotoxin gene이 확인되었으며, 이 중 H형 단독생산주가 29주(64.4%)로 가장 많았으며. A형 단독생산주 5주(11.1%), A형 및 H형 혼합생산주 4주(8.9%), G형 생산주 2주(4.4%)및 I형 생산주 1주(2.2%)이었다. 3. 시험한 105주 중 48주(45.7%)에서 항생제 내성이 확인되었으며, 이 중 penicillin에 대한 내성이 38.1%로 가장 높았으며, ampicillin과 penicillin은 중등도 내성이 각각 42.9%, 61.0%이었다. chloramphenicol, clindamycin, methicillin. trimethoprim, vancomycin은 모두 감수성이었다. 또한 단제 내성이 85.4%를 차지하였으며, 2제 내성이 12.5%, 3제내성이 2.1%이었다.

젖소 유방원(乳房源) 병원세균(病原細菌)의 항생물질(抗生物質)에 대(對)한 감수성(感受性) (Sensitivity of Pathogens of Bovine Udder Origin to Antibiotics)

  • The sensitivities of 270 pathogens (124 Streptococci, 118 Staphylococci, 10 Corynebacterium pyogenes and 18 Escherichia coli) isolated from clinical or subclinical cases of bovine mastitis during lactation to 11 antibiotics were determined by the agar plate dilution method. All cultures of Streptococci were inhibited at $1.56{\mu}g/ml$ of ampicillin and 1.56 units/ml of penicillin G. Most of the cultures were inhibited at $3.125{\mu}g/ml$ of leukomycin, but were resistant to kanamycin, streptomycin, chloramphenicol, erythromycin and colistin at concentration of $12.5{\mu}g/ml$. More than 93% of the Staphlococcal cultures were sensitive to kanamycin, leukomycin, ampicillin, tetracycline, ledermycin and minocycline at concentrations of $12.5{\mu}g/ml$ or less, and sensitive to penicillin at concentration of 3.125 units/ml, but for more than 71% of the cultures to chloramphenicol, erythromycin and colistin the concentrations required to inhibit growth were 100 ${\mu}g/ml$ or higher. All 10 cultures of Corynebacterium pyogenes were inhibited by leucomycin, ampicillin and minocycline at concentration of $1.56{\mu}g/ml$ and by penicillin G at concentration of 0.78 units/ml, but all the cultures required at least $400{\mu}g/ml$ or higher of streptomycin, erythromycin and colistin for inhibition. More than 83% of E. coli cultures were sensitive to erythromycin and minocycline at concentration of $12.5{\mu}g/ml$, but resistant to leucomycin and chloramphenicol at concentration of $100{\mu}g/ml$.
